We examined five clinical isolates formerly classified as Candida famata to clarify their taxonomic position by DNA-DNA reassociation studies and several phenotypic features. Four of the five isolates, M 5101, M 5102, M 5111 and M 5112, produced ascospores after being grown for 1 week on YM agar at 20 degrees C. By the API 20C system, three of the five isolates were identified as C. famata, and the other two strains as C. guilliermondii/C. famata. The antigenic patterns of all the isolates examined with the Candida Check kit were similar to those of C. guilliermondii. Specific antiserum against C. saitoana was obtained by adsorption. However, none of the clinical isolates tested reacted with this adsorbed serum. The mol% G + C of the four sporulated isolates ranged from 35.8 to 37.7 and that of the remaining, non-sporulated, isolate M 2054 was 43.4. The DNA reassociation value was determined by the spectrophotometric method. M 5111 and M 5112 were identified as Debaryomyces hansenii var. hansenii, M 5101 and M 5102 as D. hansenii var. fabryi and M 2054 as C. guilliermondii. The ascospore formation was exclusive to the four isolates related to D. hansenii. These findings suggest that with the exception of C. guilliermondii the other C. famata isolates examined all belong to a single species, D. hansenii.
We re-valued three antigenic types within the genus Trichosporon as the causative agents of summer-type hypersensitivity pneumonitis (SHP). Corresponding standard strains, TIMM 1573 (serotype I), TIMM 1318 (serotype II), and M9456 (serotype III) were assigned to species T. mucoides, T. asahii and T. montevideense, respectively, based on 95% or more DNA/DNA relatedness with each type of culture and other genetical, physiological and morphological characteristics. To confirm the significance of these serotypes, 98 other strains of Trichosporon isolated from patients' environments and 24 CBS strains, including type cultures of species described within the genus, were serotyped. Serum antibody analysis of 220 SHP patients against Trichosporon spp. were also examined to estimate the antigenic profile of SHP. The present results indicate that T. asahii and T. mucoides, which are the most common causes of trichosporonosis, appear to be the major causative agents of SHP. These observations suggest a common pathogenesis of Trichosporon inducing hypersensitivity and infection, depending on the immunological status of the host.
A monoclonal antibody that reacts with the amyloid in senile plaques in the brains of patients with Alzheimer's disease, was produced. The antigen was the crude amyloid protein (DAM, Pras et al. 1968) from the spleen of a patient with primary amyloidosis. The antibody crossreacts with DAM, but does not stain the amyloid of primary amyloidosis in tissue sections. The presence of a common antigenic site or sites in both the amyloid of senile plaques and amyloid of primary amyloidosis which react with this antibody is postulated.
Effects of in vivo exposure to the synthetic polyprenoic acid, 3,7,11,15-tetramethyl-2,4,6,10,14-hexadecapentaenoic acid (E-5166) using 6 groups of rats treated simultaneously or individually with phenobarbital (PB) or with p,p'-dichlorodiphenyltrichloroethane (DDT) on the gap junctions of hepatocytes were examined by freeze-fracture analysis. Experimental groups were as follows: Group 1, basal diet alone; Group 2, E-5166 treatment; Group 3, 0.05% PB diet; Group 4, PB diet and E-5166 treatment; Group 5, 0.05% DDT diet; and Group 6, DDT diet and E-5166 treatment. E-5166 was given by gavage (40 mg/kg body weight, 3 times/wk). Experimental diets and E-5166 were started at 6 wk of age. All animals were sacrificed at 2 wk after the start of the experiments. The density of gap junctions in PB- or DDT-treated rats was higher and that in E-5166-treated rats was lower than that in controls. The density of gap junctions in rats given PB and E-5166-/or DDT and E-5166-treated rats was slightly increased when compared to controls. The average area of gap junctions in DDT- or PB and E-5166-treated rats was significantly smaller than in controls (p < 0.005), although no statistical differences were found among PB-, E-5166-, and DDT and E-5166-treated rats and controls. Unit area of gap junctions to membrane area in rats given E-5166, DDT, or PB and E-5166 was lower than in controls. However, the unit area of gap junctions in DDT and E-5166-treated rats did not differ from in controls.(ABSTRACT TRUNCATED AT 250 WORDS)

BACKGROUND/AIMS: The clinical significance of the interleukin-6 (IL-6) response in patients with biliary obstruction was studied. METHODOLOGY: This study included 30 patients who had biliary obstruction and underwent percutaneous transhepatic biliary drainage. Cases complicated by cholangitis were excluded. The serum levels of IL-6 and standard liver function tests were examined before and after biliary drainage. RESULTS: Serum IL-6 levels were significantly higher in patients with biliary obstruction compared to control subjects (p<0.05). After drainage, serum IL-6 levels decreased significantly (p<0.01). The patients with serum IL-6 levels exceeding 10 pg/ml were characterized by lower hepaplastin values, lower serum levels of total protein and albumin, higher mean age, and more frequently positive cultures of bile compared to those with serum IL-6 levels of less than 10 pg/ml. Serum IL-6 levels were correlated negatively with hepaplastin values and serum levels of total protein and albumin (p<0.05). CONCLUSIONS: These results suggest that subclinical biliary infection, liver dysfunction and advanced age promote the IL-6 response in patients with biliary obstruction. Such a cytokine response may disturb the physiologic immune response after the ensuing major operation and may be related to some post-operative complications.
The cytologic appearance of a carcinosarcoma (malignant ameloblastoma and fibrosarcoma) of the maxilla in a 63-year-old man is described. On his first admission the diagnosis of malignant ameloblastoma was made on biopsy. After five surgical excisions, radiotherapy and chemotherapy, the diagnosis was changed to carcinosarcoma because the stromal cells of the tumor had become malignant. Aspiration biopsy cytology of the tumor, with a cystic lesion found at the left suborbital area, revealed malignant epithelial cells, indicating malignant ameloblastoma. Imprint smears of both surgically resected and autopsy material showed two types of malignant neoplastic cells, of epithelial and mesenchymal origin.
