Striated muscle in the biopsy specimen of the prostate.

We had a chance to see striated muscle fibers in the biopsy specimen of the prostate. We examined the prostate specimens of autopsied cases and biopsy specimens. The distribution of striated muscle fibers was studied with an immunohistochemical method for myoglobin to obtain their anatomical distribution in detail. We showed that the striated external sphincter muscle fibers were present in the peripheral zone of the prostatic gland (9.1%). Therefore, if striated muscle fibers are found in biopsy specimens of prostatic cancer, pathologists and urologists should not diagnose simply that the cancer invades beyond the prostatic capsule.

Mechanisms involved in the effect of M6434 on experimental hemorrhagic shock: I. Effects on myocardial contractility and venous return.

To elucidate the mechanisms of protective effect of M6434 on experimental shock, the authors examined the effects of this compound on the survival time and hemodynamic changes in severely hemorrhagic-shocked dogs. We also examined the effects of M6434 on contractile tension of isolated canine ventricular strips and on venous return in dogs with cardiopulmonary bypass in normal and shock state. Intravenous infusion of M6434 at 10 micrograms/kg/min prolonged survival and maintained mean arterial pressure, cardiac output, and first derivative of left ventricular pressure at higher levels than those in the control group, whereas dopamine (10 micrograms/kg/min) did not significantly affect survival time and hemodynamic parameters. M6434 did not change contractile tension in electrically stimulated canine ventricular strips. M6434 (20 micrograms/kg/min) increased the venous return of dogs with cardiopulmonary bypass in both shock and normal state. Phenylephrine (20 micrograms/kg/min) slightly increased venous return in normal state, but not during shock. Dopamine had no effect at 20 micrograms/kg/min, but it increased venous return in both states at 50 micrograms/kg/min. These results suggest that M6434 may improve the hemodynamic derangement in severe hemorrhagic shock through decreasing venous blood pooling.

Adverse effects during endocrine therapy for prostatic carcinoma with a high dose of estrogen.

A clinical study of the adverse effects induced by the endocrine therapy with a high dose of estrogen in 45 patients with stage C or D prostatic carcinoma is conducted. Transient decreases of hemoglobin and serum protein values were observed after administration of estrogen. The levels of glutamine-oxaloacetic and glutamic-pyruvic transaminase showed a transient increase. The value of serum total cholesterol and electrolytes showed no changes. Serial evaluations of electrocardiograms have played a minor role in the observation of cardiovascular disease. Up to date we have not experienced cardiovascular death in our cases during the endocrine therapy.

[Effects of Hachimijiogan on hypothalamo-pituitary-testicular system].

Effects of Kampo (Chinese) drugs on hypothalamo-pituitary-gonadal system were investigated in immature male rats of Holtzman strain. Hachimijiogan(H), its components and Ninjin(N) were administered orally to the animals for 28 days, and the weight of the various organs, pituitary content of FSH, LH and PRL, and serum concentration of FSH, LH, PRL and testosterone were measured. H caused a significant increase in the weight of seminal vesicles while H, Sanyaku(S) and N caused a significant increase in the weight of the prostate, respectively. The pituitary content of FSH was significantly increased by H and S, respectively, while the testosterone concentration was significantly decreased by Jio, S and N, respectively. No increase in the weight of the prostate or seminal vesicles followed the administration of H, S and N to orchiectomied immature rats. This result clearly shows that these three drugs have no androgenic activity. After the administration of H to immature male rats, the character of dihydrotestosterone(DHT) receptor was studied by the DCC method. The number of binding sites of DHT receptor was significantly increased by 32% (p less than 0.001) by H. These results demonstrate that H has a stimulatory effect on androgen target organs through the increase in androgen receptor in these organs.
