A clinical and immunologic study was performed comparing a group of French patients allergic to the pollens of cypress (Cupressus sempervirens, which belongs to the Cupressaceae family) and a group of Japanese patients allergic to the pollens of Sugi (Cryptomeria japonica, which belongs to the Taxodiaceae family). By skin testing, RAST, and RAST inhibition, clear cross-reactivity was detected between the two pollens. No cross-reactivity was detected between the pollens of Cupressus sempervirens and Sugi and the Pinaceae family. In addition, one can speculate that an antigen in Cupressus sempervirens is cross-reactive with SBP, the major allergen of Sugi, suggesting that there is a closer relationship between the Taxodiaceae family and the Cupressaceae family than between these two families and the other families of the gymnosperms. This finding throws new light on the taxonomy of the gymnosperms.

The effects of 11-oxo-11H-pyrido[2,1-b]quinazoline-2-carboxylic acid (Sm 857), a new antiallergic drug, on immunological reactions in mice and in humans were investigated. The mouse hemolysin formation against sheep erythrocytes was not suppressed by Sm 857 (10-100 mg/kg/d i.p., p.o.) for 3 days either before or after immunization. The formation of anti-dinitrophenyl (DNP) IgE antibodies in mice immunized with dinitrophenyl-conjugated ovalbumin (DNP-OA) and alum, was not suppressed by Sm 857 (10-100 mg/kg i.p., p.o.) for 3 days either before or after immunization, but the formation of anti-DNP IgG antibodies on 7 days after immunization was slightly suppressed in the group of mice treated i.p. with 100 mg/kg/d of Sm 857 for 3 days after immunization. On the other hand, both the proliferation of human peripheral blood lymphocytes (PBL) induced by concanavalin A, phytohemagglutinin-P, pokeweed mitogen (PWM), protein A (Pro A) and Cowan I, and the formation of IgM and IgG plaque forming cells by PBL stimulated with PWM or Pro A, were suppressed significantly only in the presence of Sm 857 (10(-4) g/ml). In addition, Sm 857 hardly affected the nitroblue tetrazolium reduction test, phagocytosis and chemotaxis in human neutrophils.
The formation of 5-lipoxygenase products of arachidonic acid, 5-HETE and 5,12-diHETE, was determined in 100,000 X g supernatant of polymorphonuclear leukocytes from 17 healthy subjects, 17 patients with extrinsic asthma and 15 patients with intrinsic asthma. After the supernatant was incubated with 14C-arachidonic acid in the presence of calcium and indomethacin, the lipoxygenase products of arachidonic acid were separated by thin layer chromatography. The results were expressed as the percentage conversion of 14C-arachidonic acid into the product per 10(7) cells. The formation of 5,12-diHETE, but not of 5-HETE, was significantly increased in the cells from the group of patients with extrinsic asthma (4.38 +/- 0.78%, mean +/- S.E.; p less than 0.01) and intrinsic asthma (6.09 +/- 1.11%; p less than 0.01), when compared to normal subjects (1.74 +/- 0.30%). Both extrinsic and intrinsic asthmatics had significantly enhanced 5-lipoxygenase activity, which was expressed as the sum of percentage conversion of 14C-arachidonic acid into 5-HETE and 5,12-diHETE. The percentage conversion in normal subjects was 4.19 +/- 0.39%, 6.24 +/- 0.84% for 17 patients with extrinsic asthma (p less than 0.05), and 8.59 +/- 1.29% for 15 patients with intrinsic asthma (p less than 0.01). There was no significant difference between these asthmatic groups. These results indicate that 5-lipoxygenase activity is increased in patients with bronchial asthma.
A homogeneous glycoprotein (mol. wt 40,000) containing 34% carbohydrate was isolated from Aloe arborescens var. natalensis. At a concentration of 5 micrograms/ml, this glycoprotein was shown to stimulate deoxyribonucleic acid (DNA) synthesis in baby hamster kidney (BHK) cells and to have the properties of a lectin which reacts with sheep blood cells. The chemical and physical properties of the glycoprotein (aloe lectin) are also discussed.

A Russian stapler and an EEA stapler were used for end-to-end anastomosis of the canine descending colon. The vascular structures of the anastomotic sites were investigated for postoperative changes at two, three, four, five, and six weeks after anastomosis by using the resin-casting method, with a scanning electron microscope. The two techniques of anastomotic stapling led to the following differences: at two weeks or more following anastomosis, it was found that, compared with the EEA stapler, the site stapled with the Russian device showed a slightly more markedly irregular microvascular pattern of the capillary network in the mucosal membranes along the anastomotic suture line and what were believed to be microulcerations as well. Samples of the Russian stapler at two and three weeks postoperatively showed that the fault zones had developed in the vascular casts of the mucosal capillary system between the area of irregular vasculature and the normal mucosa. It was clearly established, however, that the microvascular channels in the mucosal membrane of the anastomotic site across the orad and anad segments of the colon had already formed after two weeks with both of the stapling techniques. New microvessels grew within a range of about 10 mu in diameter from the cut-off surface of the mucosa and united. No necrotic destruction of tissue or deficiency findings had manifested themselves in the inverted cuff zone of the anastomotic site, and the staple had been enveloped by the blood vessels of the submucosal layer and embedded in the intestinal wall during the period of two to six weeks. New microvessels, 10 to 15 mu in diameter, were found to have already formed in the two-week samples, passing vertically across the lumen of the closed eye of the staple, and no differences in vascular structure were detected, depending on the different shapes of staples used. Five to six weeks after the anastomotic surgery, the mucosal membranes in the anastomotic site showed a normal vascular structure with only minute residual indentations.
Dissecting aneurysm of the left atrium is extremely rare. A patient with such an aneurysm, which developed after mitral valve replacement is described. The diagnosis was established by cineventriculography of the left ventricle and two-dimensional echocardiography. The aneurysm was successfully treated by closure of the communicating opening through the left thoracotomy incision.

Human eosinophils with a density of over 1.095 were isolated from peripheral blood by dextran sedimentation, centrifugation with Lymphoprep and density gradients with Percoll. After the cells (1 X 10(5)/ml) were incubated with 1 microgram/ml calcium ionophore A23187 for 20 min, leukotriene C4 (LTC4) content in the supernatant was measured by radioimmunoassay. The generation of LTC4 was significantly higher in the cells from extrinsic asthmatics (23.5 +/- 14.8 ng/10(6) cells, mean +/- SD, n = 26, P less than 0.01) and intrinsic asthmatics (24.6 +/- 20.6 ng/10(6) cells, n = 27, P less than 0.01 as compared with normal healthy subjects (8.3 +/- 7.7 ng/10(6) cells, n = 10). There was no significant difference in the generation of LTC4 between intrinsic and extrinsic asthmatics. These observations indicate that eosinophils from asthmatic patients have increased ability to release LTC4.
Human eosinophils and neutrophils were isolated from 20 ml of peripheral blood by dextran sedimentation, centrifugation with lymphoprep, and density gradient centrifugation with Percoll. The incubation of neutrophils of purity higher than 99% (density: 1.080-1.085 g/ml) with calcium ionophore A23187 for 20 min resulted in the release of a little amount of leukotriene C4 (2.3 +/- 0.6 ng/10(6) cells) as measured with a commercial radioimmunoassay kit. On the other hand, the release from eosinophils was over 10 times that from neutrophils. When the releasability of leukotriene C4 from eosinophils was examined in relation to the cell density, the eosinophils with lower density were observed to produce greater amounts of leukotriene C4.
