Ampicillin concentrations in human serum, gingiva, the mandibular bone, and dental follicle following a single oral administration.

Ampicillin (ABPC) concentrations in human serum, gingiva, the mandibular bone, and dental follicle after a single oral administration of ampicillin (500 mg) were assayed by the agar diffusion (paper disc) method. The peak times of all specimens were identical, being 120 min after administration. The peak concentrations of the respective specimens were 2.01 micrograms/ml, 1.03, 0.34, and 0.72 micrograms/g, respectively. The concentration ratios of gingiva, the mandibular bone, and dental follicle to their corresponding serum at the peak time were 0.51, 0.16, and 0.35, respectively.

Concentrations of ampicillin and cefadroxil in human serum and mixed saliva following a single oral administration of talampicillin and cefadroxil, and relationships between serum and mixed saliva concentrations.

The concentrations of ampicillin (ABPC) from talampicillin (TAPC) and cefadroxil (CDX) in serum and mixed saliva were assayed by the thin layer disc plate method. Talampicillin and cefadroxil (500 mg) were given by a single oral administration. The relationships between serum and mixed saliva ampicillin and cefadroxil concentrations were evaluated in the paired specimens collected from 10 different persons, respectively. The means of concentration ratios of mixed saliva to serum ampicillin and cefadroxil were 0.006 +/- 0.003 and 0.025 +/- 0.010 (mean +/- SD), respectively. Significant correlation coefficients between mixed saliva and serum concentrations were found for both ampicillin and cefadroxil, which were r = 0.78, P less than 0.001, and r = 0.67, P less than 0.001, respectively.

In vivo color analysis of human crystalline lenses.

Indirect measurement in vivo of nuclear color of 64 cataractous eyes was made using a method of image analysis of photographed lenses developed by the authors. The grade of yellow of lens color was characterized by its dominant wavelength and excitation purity in the CIE chromaticity diagram. The differences were small between nuclear color observed from in vivo and in vitro photographs.

Effects of proteinase inhibitors on preimplantation embryos in the rat.

Proteinase inhibitors of microbial origin were injected into the uterine horns of mated rats at 14:00 h on Day 5 of pregnancy (spermatozoa in vaginal smear = Day 1), and 5 or 6 h later the embryos were flushed from the horns and examined. Chymostatin and alpha-MAPI, inhibitors of chymotrypsin-like serine proteinase and thiol proteinases, as well as thiolstatin, an inhibitor of thiol proteinases, significantly inhibited embryo growth. The inhibitory activity of alpha-MAPI on embryonic growth was distinctly greater than that of thiolstatin, although the ID50 values of the two inhibitors to papain are similar. Antipain and leupeptin which are inhibitors of trypsin-like and thiol proteinases, and talopeptin, an inhibitor of metal proteinases, significantly interrupted the removal of the zona pellucida from expanding blastocysts. These results suggest that (1) a chymotrypsin-like proteinase seems to be important to the growth of the embryo, (2) a thiol proteinase may participate in embryonic growth, and (3) a trypsin-like proteinase and a metal proteinase are likely to participate in zonalysis.

Studies of an immunomodulator, swainsonine. I. Enhancement of immune response by swainsonine in vitro.

Swainsonine isolated from Metarhizium sp., was found to enhance the activities of the mouse immune system in vitro. Concanavalin A stimulated lymphocyte proliferation and proliferative response in mixed lymphocytes culture, which were suppressed by immunosuppressive factor obtained from serum of sarcoma 180 tumor bearing mouse, were restored to normal levels by the addition of swainsonine. Furthermore, the concanavalin A induced incorporation of [3H]thymidine into mouse spleen cells was remarkably increased by treatment with swainsonine over a wide dose range, From studies using fluorescence activated cell sorting, swainsonine was shown to enhance the expression of concanavalin A receptors of spleen cells.

Postoperative elevation of serum pancreatic secretory trypsin inhibitor.

Postoperative transient elevations of serum pancreatic secretory trypsin inhibitor are reported for the first time. The elevations were independent of the surgical procedures or sites, and the maximum serum pancreatic secretory trypsin inhibitor was seen on the 2nd to 4th postoperative day. The elevation of pancreatic secretory trypsin inhibitor was significantly correlated with the length of the operation time and the volume of blood loss during the operation. The change of serum pancreatic secretory trypsin inhibitor was also significantly correlated with those of white blood cell count, alpha 1-antitrypsin, and fibrinogen.

Elevation of serum pancreatic secretory trypsin inhibitor (PSTI) in patients with serious injury.

Pancreatic secretory trypsin inhibitor (PSTI) in serum was measured periodically in 15 patients with serious abdominal and non-abdominal injuries. From the second or third posttraumatic day, serum PSTI began to rise. Elevation of serum PSTI to more than twice the initial value was seen in 93% of the patients following serious injury. In uneventful cases, it declined and returned to the level at the time of admission within 14 days, but in the patients with serious inflammations and in the patient who died, it remained elevated. The change in the concentration of serum PSTI was significantly correlated with those of the acute phase reactants (fibrinogen and alpha 1-antitrypsin). There was no correlation between the level of serum PSTI and serum amylase activity or creatinine.
