[Long-term prognosis of Crohn's disease].

Two hundred and three patients with Crohn's disease seen at our clinics and affiliated centers from April 1973 to August 1988 were followed for 4.4 +/- 3.2 years (mean +/- SD), in order to evaluate the prognosis of Crohn's disease in Japan. These 203 patients (142 males and 61 females) fulfilled the following criteria; 1) they have been followed at the outpatients clinic for more than six months or 2) they have been admitted to us for more than a month. Of these, 83 (40.9%) had ileitis, 60 (29.6%) ileocolitis, 25 (12.3%) colitis, 15 (7.5%) miscellaneous types and the remaining 20 (9.9%) had undergone bowel resection. Cumulative survival rate and cumulative probability of surgery were calculated by life table method. Cumulative survival rate in these patients was compared with expected survival rate of sex and age matched general population. Cumulative survival rates five and ten years after diagnosis were 98.9% and 98.9%, respectively. There were no significant differences in the survival rates between two groups. Cumulative probability of surgery five and ten years after onset of symptoms were 16.2% and 39.1%, respectively. Cumulative probability of surgery five and ten years after diagnosis were 25.9% and 46.9%, respectively. From these results, the prognosis of Japanese patients with Crohn's disease appears to superior to that in European and American literatures.

[Broncholithiasis: clinical experience of three operated cases].

Three patients with broncholithiasis had an operation in our hospital. Patient 1 had no history of tuberculosis. She had the pulmonary stones removed bronchoscopically twice. The stones were composed of inflammatory granulomas. Patient 2 had a history of pulmonary tuberculosis. Endoscopical removal of the pulmonary stone was not successful. After 6 days, she coughed up a large amount of blood, and the operation was performed. Patient 3 coughed up blood for 10 years. The amount of blood gradually increased and the operation was performed. All three cases had calcification of the hilar lymph nodes. Broncholithiasis appears to be related not only to pulmonary tuberculosis but also to other chronic inflammatory diseases of the lung.

Involvement of a protein kinase C-dependent process in long-term potentiation formation in guinea pig superior colliculus slices.

Superior colliculus (SC) slices were prepared from guinea pig. Electrical stimulation was applied to the optic layer (OL) of the SC slices and the postsynaptic potential (PSP) was evoked in the superficial grey layer (SGL) of the SC. Tetanic stimulation to the OL evoked long-term potentiation (LTP) in the PSP. To investigate the involvement of the protein kinase C (PKC) system on the LTP formation in the SC, the effects of PKC activator (phorbol ester) and PKC inhibitors (polymyxin B, melittin and H-7) on the LTP formation have been studied. Application of phorbol ester to the medium at concentrations between 10(-8) and 10(-10) M increased the amplitude of the PSP. On the other hand, the presence of phorbol ester at a concentration of 10(-6) M increased the glutamate release from the SGL slices. Tetanic stimulation which could not induce LTP by itself could elicit LTP during application of phorbol esters at the low concentration (3 X 10(-12) M). PKC inhibitors such as polymyxin B (10(-7) M), melittin (10(-8) M) and H-7 (10(-4) M) prevented LTP formation. When H-7 was applied once after LTP was formed, the enhanced PSP reduced to the original level. These results strongly indicate the involvement of PKC system on the LTP formation in the SC slices.

Activin/EDF as an inhibitor of neural differentiation.

Activin/EDG, a stimulator of the secretion of follicle stimulating hormone (FSH) from pituitary gland and an inducer of erythroid differentiation for Friend leukemia cells, has since been implicated in a variety of biological roles. Here, we show some novel effects of activin on murine embryonal carcinoma cells (EC cells). First, activin acts as a growth factor on undifferentiated P19 cells, a well characterized EC cell line for the study of mammalian development. Second, activin inhibits the retinoic acid (RA) induced differentiation of P19 cells to neurons and glial cells. The inhibitory effect of activin on neural differentiation, which has yet to be proved in other physiological peptides, is confirmed also on the differentiation of various neuroblastoma cell lines. Our results suggest a possible role of activin as a negative regulator of neural differentiation in mammalian development.

Production of endothelin-1 by rat cultured mesangial cells.

We investigated whether ET-1 is synthesized by and released from mesangial cells. ET-1-like immunoreactivity (LI) released into medium increased time-dependently under a serum-free condition. The amounts of ET-1-LI released into the medium was augmented in the presence of fetal calf serum. Reverse-phase HPLC profile of the conditioned media revealed a major component coeluting with standard ET-1. Northern blot analysis of poly(A) +RNA extracted from mesangial cells showed a single major band corresponding to the size of preproET-1 mRNA (2.3 kb). These findings demonstrate that ET-1 is synthesized by and released from rat mesangial cells and suggest a possibility that it acts on their own cells as an autocrine factor.

Masking effect of NMDA receptor antagonists on the formation of long-term potentiation (LTP) in superior colliculus slices from the guinea pig.

After electrical stimulation of the optic layer (OL) of superior colliculus (SC) slices, the postsynaptic potential (PSP) was recorded in the superficial gray layer (SGL) of the SC. The degeneration studies of retinotectal or corticotectal inputs to the SGL of the SC indicated that this PSP evoked in the SGL of the SC slices was retinotectal in origin. Neurotransmission in this pathway may be mediated by glutamate, because the PSP amplitude was reduced and blocked by application of kynurenate or quinoxaline dione (DNQX) to the medium. Furthermore, the concentration of glutamate in the right SGL was significantly reduced by 32% after left optic denervation and by 30% after ablation of the right visual cortex, compared with that in the left SGL. Long-term potentiation (LTP) in the SGL was induced by tetanic stimulation (50 Hz, 20 s) to the OL. The LTP formation was facilitated by the removal of Mg2+ from the medium. The effects of glutamate antagonists D-amino-5-phosphonovalerate (D-APV), gamma-D-glutamylglycine (gamma-DGG), and (+)-5-methyl-10,11-dihydro-5H-dibenzo, a,d-cycloheptene-5,10-imine maleate (MK-801) on the induction of LTP were investigated. D-APV (100 microM) or gamma-DGG (1 mM) masked the expression of LTP by tetanic stimulation, however LTP was induced after removal of the agents. LTP formation was observed without further tetanic stimulation following the removal of D-APV from the medium even 80 min after the tetanic stimulation. LTP once formed was not influenced by application of D-APV.(ABSTRACT TRUNCATED AT 250 WORDS)

Simple and rapid purification of brevin.

Brevin or plasma gelsolin, a calcium dependent actin-binding and actin-severing protein, was purified from bovine plasma by a very rapid and simple procedure; ammonium sulfate fractionation and only one step of anion exchange column chromatography by a convenient use. It takes only 24 hrs to complete all the procedure. The purity of brevin prepared by this method was more than 95% on SDS-PAGE and total recovery was much better than previous preparation methods. This brevin preparation has about 8 isomers on 2-D PAGE and strong severing activity on F-actin under electron microscopic observation.

O2- generation and lipid peroxidation during the oxidation of a glycated polypeptide, glycated polylysine, in the presence of iron-ADP.

Oxidation of glycated polylysine, a model compound of glycated protein, caused O2- production even at physiological pH, which could be accelerated by Fe3(+)-ADP. An enediol structure in glycated polylysine and related compounds, which could be confirmed by I2 uptake, was related to their oxidizability. Glycated polylysine was easily coordinated with Fe3+ even in the presence of phosphate at pH 7.4 and the formation of the iron complex was prevented by desferrioxamine. The exposure of unsaturated phospholipid liposomes to glycated polylysine-Fe3(+)-ADP system caused the production of a thiobarbituric acid-reacting substance, which was completely inhibited by 5 microM alpha-tocopherol or 150 microM desferrioxamine and slightly by 0.5 microM SOD. Catalase (20 micrograms/ml) and 10 mM sodium-benzoate did not affect the iron-glycated polylysine-induced lipid peroxidation, indicating no participation of an OH. in this reaction. A ferrous ion-coordinated glycated polylysine may act as an initiator of phospholipid peroxidation in the presence of oxygen. A possible mechanism of the iron-glycated polylysine-induced lipid peroxidation was discussed.

Reduction of glutamate content in rat superior colliculus after retino-tectal denervation.

The effect of afferent lesions on glutamate content was measured in the lamina of the superior colliculus (SC) in the rat. The analysis was performed 12 days after unilateral enucleation (left eye), or ablation of visual cortex (right), or both enucleation and ablation. The glutamate contained in the superficial grey layer (SGL) and deep layer was measured in the sectioned freeze-dried sample using an enzymatic cycling method of NAD-NADH. The upper layer of SGL contralateral to enucleation exhibited a significant reduction (23%) in glutamate content. Combining enucleation and ablation further decreased (35%) glutamate content. Additionally, the synaptic potential evoked in the SGL of SC slices after stimulation of optic layer was blocked by the application of kynurenic acid (3 mM) or DNQX (30 microM). These results indicate that the retino-tectal pathway in the rat can be glutamatergic in nature.

A specific protein, p92, detected in flat revertants derived from NIH/3T3 transformed by human activated c-Ha-ras oncogene.

Total proteins from a mouse embryo fibroblast cell line NIH/3T3, NIH/3T3 cells transformed by human activated c-Ha-ras (EJ-ras) oncogene (EJ-NIH/3T3), and the two flat revertant cell lines, R1 and R2, were analyzed by two-dimensional gel electrophoresis (IEF and NEPHGE). Several hundred polypeptides were resolved as seen by silver staining. Common alterations in four polypeptide spots were observed in the revertants when compared with NIH/3T3 and EJ-NIH/3T3 cells. In these alterations, a new polypeptide spot p92-5.7 (designated by molecular weight x 10(-3) and pI) was detected only in the revertants and not in NIH/3T3 and EJ-NIH/3T3 cells. Furthermore, the expression level of p92-5.7 seemed to be associated with the flat morphology and the reduced tumorigenicity of the revertants. Polypeptide p92-5.7 was also not detected in the total proteins extracted from BALB/3T3 cells, NIH Swiss mouse primary embryo fibroblasts, NRK (normal rat kidney) cells, and L6 (rat myoblast). Subcellular fractionation of total protein from R1 cells revealed that the p92-5.7 was present in the cytosol. Western blot analysis using an anti-gelsolin antibody demonstrated that the p92-5.7 might be a variant form of gelsolin which is thought to be an actin regulatory protein or a gelsolin-like polypeptide. These results may suggest that the expression of p92-5.7 detected only in the revertants is associated, at least in part, with the reversion. This may be the first demonstration of specific protein expression in the flat revertants.

Distribution of a gelsolin-like 74,000 mol. wt protein in neural and endocrine tissues.

A Ca2(+)-dependent actin binding protein with a molecular weight of 74,000, was purified from bovine adrenal medulla by using deoxyribonuclease I affinity chromatography followed by ion-exchange chromatography and gel filtration. This protein broke actin filaments into fragments and promoted nucleation of actin polymerization in a Ca2(+)-dependent manner as effectively as gelsolin. Proteolytic and immunological comparison with gelsolin which is widely distributed actin-severing protein, indicated that the 74,000 mol. wt protein is a distinct protein, but its domain structure resembles that of gelsolin. Immunoblotting using antibody against this protein showed a tissue-specific distribution. The protein was detected in various endocrine, neuroendocrine and nervous tissues, but not in muscle tissues and plasma which contained relatively large amounts of cytoplasmic and plasma gelsolin. This fact might indicate that this actin-severing protein is involved in the regulation of the secretory process of endocrine and nervous tissues. In the exocytotic process regulated by Ca2+, this protein probably plays a role to free secretory organelles like vesicles from the cytoskeletal network, mainly F-actin, which prevents the movement of secretory vesicles in the resting state.
