Cytoplasmic microtubules were investigated with two different methods in Behçet's disease. First, with an electron microscope, the peripheral blood neutrophils of 6 patients with Behçet's disease at the exacerbation stage and those of 3 healthy controls were examined. The number of microtubules was counted on enlarged electron micrographs of the neutrophils. Each electron micrograph was taken at a direct magnification of 15,000, so that at least one centriole was contained in the photograph. It was enlarged twice the size of the negative and printed. The average number of microtubules in the neutrophils of the patients was 4.96 +/- 2.76 (SD) and 2.36 +/- 1.88 (SD) in the control subjects. This difference was statistically highly significant (p less than 0.0005). Secondly, the peripheral blood polymorphonuclear leukocytes of 24 patients and those of 13 healthy controls were examined with an indirect immunofluorescence technique. The distribution and the pattern of the microtubules differed between the patients at the exacerbation stage and normal subjects and also between the patients at the exacerbation stage and the remission stage. These results suggested that microtubules may play a significant role in the pathophysiology of Behçet's disease.
Cytoplasmic microtubules are one of the various components of the cytoskeleton. They are associated with transport of lysosomes and movement of the cell membrane. Augmentations of the lysosomal enzyme activity and chemotactic activity are found in the neutrophils of patients with Behçet's disease, and colchicine which induces microtubule depolymerization is effective in the treatment of this disease. We examined microtubules in the neutrophils of 24 patients with Behçet's disease, by an indirect immunofluorescence technique, to investigate the significance of the microtubules in this disease. The distribution of the microtubules was different between the patients in the exacerbation stage and normal subjects, and also between the exacerbation stage and the remission stage. These results suggested that microtubules play some significant role in the pathophysiology of Behçet's disease.

Fibroblasts in human conjunctiva were examined by electron microscopy. Their striated fibers were almost always associated with basal bodies or centrioles. The length and thickness of the striated fibers were variable. The average length was 0.48 +/- 0.38 (SD) micron; the maximum length was 2.1 microns. The average thickness was 62.3 +/- 26.4 (SD) nm; the maximum thickness was 157 nm. Each bundle of striated fibers consisted of several filaments of about 7 nm in diameter. The periodicity of the major crossband was 69.7 +/- 2.8 (SD) nm. At least six intraperiod subbands were seen between the electron-dense major bands. The functional significance of striated fibers was discussed.

Slow reacting substance of anaphylaxis (SRS-A) is an important chemical mediator of bronchial asthma. Leukotriene C4 is a component of SRS-A and is synthesized from arachidonic acid. Its synthesizing and releasing processes are found to be Ca2+-dependent. We developed an in vivo inhalation asthma model, mainly mediated by SRS-A, and elucidated the relationship between a Ca2+-antagonist, nicardipine, and SRS-A. In the asthmatic model, mediated by endogenous SRS-A induced by antigen inhalation, continuous intravenous infusion of nicardipine 7 micrograms/kg/min depressed the open airway pressure by about 60% compared with the saline-treated group. Inhibition of mean pulmonary resistance (RL) was about 50% and that of the inverted value of dynamic compliance (1/Cdyn) about 36%. However, the same concentration of nicardipine did not significantly effect the airway response in the asthmatic model induced by the inhalation of leukotriene C4. These results suggest that nicardipine, at the concentration used in the present study. did not block the direct effect of SRS-A on the smooth muscle, but blocked the Ca2+ influx required for the synthesis of SRS-A and its release.
Slow-reacting substance of anaphylaxis (SRS-A) is an important factor mediating bronchoconstriction in asthma. We developed a guinea pig model for SRS-A-mediated bronchoconstriction induced by antigen inhalation. Using this model, we investigated the effect of N-(3', 4'-dimethoxycinnamoyl) anthranilic acid (N-5'), a new anti-allergic drug, on the bronchoconstriction. FPL 55712 inhibited most of the bronchoconstriction induced by antigen inhalation. N-5' inhibited the antigen-induced bronchoconstriction in a dose-dependent fashion. Intraperitoneal administration of 200 mg/kg N-5' was effective for 40 min after antigen inhalation, while the effect of 60 mg/kg lasted only 7 min. On the other hand, 200 mg/kg N-5' showed no inhibitory effect on the bronchoconstriction caused by direct inhalation of leukotriene C4, a component of SRS-A. These findings indicate that one of the anti-allergic actions of N-5' is due to inhibition of synthesis and/or release of SRS-A.
