Comparison of spontaneous activity of mesencephalic reticular neurones in the waking state and during pentobarbital anaesthesia.

In rats immobilized by d-Tubocurarine the spontaneous activity of 100 mesencephalic reticular neurones was recorded extracellularly and statistically evaluated before and after repeated intravenous administration of 15 mg/kg doses of Pentobarbital. Number of spontaneously active neurones decreases quasi-linearly with repeated 15 mg/kg Pentobarbital doses. After a 75 mg/kg cumulative dose practically all neurones ceased firing spontaneously, whereas cortical EEG activity fully disappeared after the 90 mg/kg Pentobarbital dose. The firing rate was characterized by the mean interval with its standard deviation. Mean value for the total sample of spontaneously active neurones was 146.7 +/- 192.3 msec without Pentobarbital and increased to 302.7 +/- 367.5 msec after 15 mg/kg and to 400.6 +/- 452.5 msec after 30 mg/kg cumulative dose of Pentobarbital. The 15 mg/kg dose increased the frequency of firing in 5% of neurones only. The most often encountered type of interval histogram in the mesencephalic reticular formation was the exponential type (59% in unanaesthetized state), which was also most sensitive to Pentobarbital. Synchronized activity in bursts, characterized by periodical peaks and dips frequently occurred in neurones with the exponential-like interspike interval density after Pentobarbital administration. On the contrary, neurones with gamma-like and especially with symmetrical-like types of density were less influenced by Pentobarbital. In many neurones a periodical increase in the firing rate (with intervals of tens of seconds) related to the occurrence of spindles was present in the cortical EEG activity.

Two-dimensional probabilistic images discrimination. I. Simultaneously presented pairs of patterns.

Reaction time and judgment of similarity or dissimilarity were studied in an experiment on two-dimensional probabilistic images (TDPIs) composed of rectangular black and white cells with statistical distribution of these elements 0.5-0.5. The subjects were asked to report verbally whether pairs of TDPIs, presented for 700 ms, appeared to them similar or not. Three sets of TDPIs differed as to the size of their "grain". Within the pairs "physically identical patterns", "statistically same patterns" or "different patterns" have been used. The statistically same patterns pairs reached the lowest (39 percent) judgment correctness. Reaction times for these pairs were generally longer than for others. In the case of identical patterns and statistically same patterns pairs the results indicated a general increase of the processing time as the size of grain had increased. There was a general tendency of reaction time shortening in successive sessions. These results suggest that correct discrimination of TDPIs does not depend primarily upon their grain.

Hippocampal theta activity in the acute pretrigeminal cat.

In five cats with a pretrigeminal brainstem transection ocular behavior and cortical and hippocampal EEG activity were recorded. The ECoG desynchronization was usually accompanied by clear hippocampal theta activity. The episodes of ECoG desynchronization and hippocampal theta activity lasted in some cases for several hours. Visual stimuli evoked two types of hippocampal EEG response: (I) increase of regularity and frequency of theta activity (II) EEG desynchronization. The intensity of these responses was positively correlated with the intensity of ocular and ECoG components of the orienting reflex (the most conspicuous phenomenon being their habituation with stimulus repetition). Electrical stimulation of the midbrain reticular formation evoked theta activity of a frequency proportional to stimulus voltage; for maintaining it constant stimulus strength had to be elevated gradually. Depriving the brain of influences from the lower brainstem does not influence qualitatively the function of reticulo-septo-hippocampal system.

A device for evaluation of the dominant component of the EEG activity.

A setup composed of a bioanalyser and additional electronic circuits is described allowing to measure the instantaneous frequency and amplitude of the dominant component of EEG activity. Besides recording the above values on paper by means of the EEG apparatus, functions of probability density of both the amplitude and the frequency of EEG activity are estimated on-line during preadjusted time intervals, and displayed on the screen of the bioanalyser. The mutual relationship between instantaneous EEG amplitude and frequency can be expressed in two-dimensional histograms displayed on a memory scope. The system described is suitable for laboratories having no possibility to use general purpose computer on-line.

Influence of increasing doses of pentobarbital on the mesencephalic reticular formation in rats. Spontaneous firing of neuronal pairs and activity evoked by polarization.

In rats immobilized by D-tubocurarine the spontaneous activity in pairs of mesencephalic reticular neurons was recorded by means of one microelectrode and differentiated by amplitude discriminators. Spontaneous activity of neuronal pairs and activity evoked by electrical polarization of cells (5-235 nA) through the recording microelectrode was evaluated in the curarized state and after cumulative doses of 15 mg/kg pentobarbital. No correlation between mean interspike interval duration in pairs of adjacent neurons was found in the unanesthetized state. After pentobarbital a slight dependence was observed between the values. Furthermore, it is probable that adjacent neurons will both cease firing at the same dose of pentobarbital. Cross-correlation of the spike trains of two adjacent neurons was found in the range of minus 10 msec to plus 10 msec (which is probably an expression of direct interaction or of a common input) in 6 of 50 pairs analyzed in the unanesthetized state. This dependence fully disappeared after 15-30 mg/kg of pentobarbital. After 15 mg/kg of pentobarbital in 17 recorded pairs (out of 50 analyzed) peaks in the cross-correlograms appeared in a range of about 100 msec and at a distance of 150-300 msec. Such a relationship was already present in 4 pairs of neurons in the unanesthetized state. The similarity of autocorrelation and cross-correlation histograms in this case favors the hypothesis that spontaneous activity of many mesencephalic reticular neurons is synchronized after pentobarbital administration. In many reticular neurons the firing may be evoked by direct cell polarization through the recording microelectrode even at cumulative doses of pentobarbital much higher than those sufficient to block spontaneous activity.

A technique for on-line classification of evoked potentials into two groups according to subjective interpretation of the stimulus.

A technique is described for on-line classification of evoked potentials (EPs) into distinct groups for separate averaging according to a criterion available after the beginning of the EPs. It is suitable for analysing psychophysiological correlations (relationship between EPs and subjective perception). The system consists of a slightly modified NTA 512 B (Hungarian) average response computer (however, any CAT-computer-like device can be used in a similar way) and of an additional device. It can be used in all cases when the number of subjective judgements according to which EPs have to be classified can be limited to two (or three, when, by slightly changing the system, all quarters of memory are used).
