Comparison of phosphofructokinases in submandibular glands of immature and adult rats.

1. Phosphofructokinases (PFKs) in immature and adult rat submandibular glands were purified to near homogeneity, and their properties were compared. 2. PFK in immature gland was less sensitive to inhibition by ATP than adult PFK. 3. Saturation curve for fructose 6-phosphate of PFK in immature gland was less sigmoidal than that of adult PFK indicating the lower cooperativity of subunits in immature PFK. 4. Fructose 2,6-bisphosphate relieved PFK from inhibition by ATP in adult gland, but a similar effect was not clearly observed in immature gland PFK. 5. Adult PFK was a heterotetramer consisting of C-, M-, L-subunits, but in immature PFK another type of subunit, which was slightly smaller than L-subunit, existed in addition to C-, M- and L-subunits.

Composition of glycosaminoglycans in human pancreatic cancer.

Five glycosaminoglycans were isolated from tryptic digestion of both cancerous and normal tissues of the human pancreas and were assayed by determining the carbohydrate content of materials. Separation of these five polymers was achieved by Dowex 1-X2 column chromatography and fractionation with Benedict's solution. They were identified as hyaluronic acid, heparan sulfate, dermatan sulfate, chondroitin-4-sulfate, and chondroitin-6-sulfate, respectively. The total amount of glycosaminoglycans in cancer tissue increased in comparison to the controls. The increase in tissue content of glycosaminoglycans was accompanied by increases in chondroitin-4-sulfate and chondroitin-6-sulfate levels.

Vectorcardiographic criteria for diagnosis of high lateral infarction--supplement for Chou's criteria.

The concept of high lateral myocardial infarction (HLMI) has not been clearly defined, so criteria for its vectorcardiographic (VCG) diagnosis have had no firm basis. However, we have reported that HLMI, expressed as abnormal Q waves in lead aVL on the electrocardiogram, corresponds to necrosis of the area usually supplied by the diagonal branches of the left anterior descending coronary artery. Here, we evaluated conventional VCG criteria for the diagnosis of HLMI on the basis of angiographic findings, and selected the criteria of Chou as typical. The frontal plane VCG was analyzed in 46 patients with HLMI on angiograms; other subjects were 233 healthy controls and 194 patients with left ventricular hypertrophy. Chou's criteria had good specificity, but sensitivity was unsatisfactory (32/46, or 70%). Accordingly, we examined various parameters of the QRS and T loops and found that the addition of four new criteria to Chou's improved sensitivity (46/46, or 100%) without impairment of specificity. The additional criteria are; 1) initial counterclockwise rotation of the QRS loop, 2) the ratio of the maximal QRS magnitude to the maximal T magnitude less than 4.5, 3) direction of the maximal T between +60 degrees and +180 degrees, 4) the QRS-T angle between 40 degrees and 135 degrees, all in the frontal plane.

[A case of membranous septal aneurysm associated with infective endocarditis].

A 43-year-old man with membranous septal aneurysm associated with infective endocarditis was reported. In this case, the stream of the shunt through VSD was directly oriented toward the atrial surface of the tricuspid anterior cusp and made it unusual form. The focus of infective endocarditis was located on that cusp, and he underwent tricuspid valve replacement with VSD closure. The post-operative course was uneventful. We discussed the tricuspid anterior cusp deformed by the jet stream of the shunt and the relationship between the aneurysm and the infective endocarditis.

Alpha 1-antitrypsin and hepatic fibrosis.

The administration of alpha 1-antitrypsin, both sialo and asialo types, to rats with chronic liver injury accelerated hepatic fibrosis. More fibrosis was seen histologically and the amount of hydroxyproline in the liver increased. Moreover, the growth of HEL cells (human embryonal lung fibroblast) in culture was promoted by alpha 1-antitrypsin though the action of the asialo type was very weak by contrast with the sialo type.

[A case of a small hepatoma with multicentric development in precirrhotic fibrosis].

Multicentricity of hepatocellular carcinoma (HCC) is considered, especially in patients with liver cirrhosis. This paper describes an operative case of a male patient with probable multicentric development of HCC in precirrhotic fibrosis. The main tumors inside the capsule were completely necrotic due to transarterial embolization (TAE). Histologic examination disclosed discrete tiny nodules of HCC that were not detected grossly. They showed highly differentiated trabecular arrangements: Edmondson I. A scirrhous type was noted in the center of the tumors. At the borders of the tumors the carcinoma cells exhibited replacing growth patterns and it was thought that they developed multicentrically. Partial resection must be performed as extensively as possible in a case such as the present one.

Inhibition by gossypol of tumor promoter-induced arachidonic acid metabolism in rat peritoneal macrophages.

Rat peritoneal macrophages were prelabeled with [3H]arachidonic acid. The release of radioactivity into the medium was increased by treatment with TPA-type tumor promoters, such as TPA, teleocidin and aplysiatoxin, and the non-TPA-type tumor promoter, thapsigargin. Gossypol, at concentrations of 3 and 10 microM, inhibited the release of radioactivity stimulated by both types of tumor promoter, although the mechanism of stimulation of arachidonic acid metabolism is different in the two types of tumor promoter. Stimulation of prostaglandin E2 production by these tumor promoters was also inhibited by treatment with gossypol. Calcium ionophore A23187-stimulated release of radioactivity and prostaglandin E2 production were also inhibited by gossypol treatment. The mechanism of inhibition by gossypol of prostaglandin E2 production is discussed.

Assessment of biological age by principal component analysis.

A method of assessing biological age by the application of principal component analysis is reported. Healthy individuals (462) randomly selected from about 6000 men who had taken a 2-day health examination were studied. Out of the 30 physiological variables examined in routine check-ups, 11 variables were selected as suitable for the assessment of biological age based on the results of factor analysis and the physiological meaning of each test. This variable set was then submitted to principal component analysis, and the 1st principal component obtained from this analysis was used as an equation for assessing one's biological age. However, the biological age calculated from this equation is expressed as a score, so the estimated score was transformed to years (biological age) using the T-score idea. The biological age estimated by this method is practically useful and theoretically valid in contrast with the multiple regression model, because this approach eliminates and overcomes the following 2 big problems of the multiple regression model: (1) the distortion of the individual biological age at the regression edges; and (2) a theoretical contradiction in that a perfect model will merely be predicting the subject's chronological age, not his biological age.

A glycopeptide from adenocarcinoma tissue of human lung.

A glycopeptide from adenocarcinoma tissue of human lung was extracted by protein digestion with papain (EC 3.4.22.2) and trypsin (EC 3.4.21.4). This component was isolated by Pevikon block electrophoresis. It possessed hexose, glucosamine, fucose, galactosamine, and sialic acid. In its carbohydrate composition and also the abundance of threonin in the peptide moiety it was quite similar to the glycoprotein from gastrointestinal tract. The physical characteristic of the two, such as their optical rotation and viscosity, were also very similar each other.

Comparison of glycopeptide in lung cancer tissues of different histological types.

The glycopeptide was studied in the pulmonary cancer tissues of three different histological types including squamous cell carcinoma, undifferentiated cell carcinoma, and adenocarcinoma. The glycopeptide preparation was made by digesting the lung cancer tissues with proteolytic enzymes followed by removal of enzymes and undigested protein. The comparison of the relative proportion of the glycopeptide, which migrated toward the anode slower than hyaluronic acid, and was stained with both alcian blue and PAS, and contained higher sialic acid, was carried out among three different histologic types of lung cancer. This glycopeptide was highest in adenocarcinoma, followed by undifferentiated cell carcinoma, and was lowest in squamous cell carcinoma.

Tissue content of glycosaminoglycans in the diffuse idiopathic interstitial fibrosis patient.

Quantitative changes of glycosaminoglycans in lung tissues of patients with diffuse idiopathic interstitial fibrosis and patients with bronchial asthma were studied. The total amount of glycosaminoglycans in lung tissue of patients with diffuse idiopathic interstitial fibrosis increased in comparison to the lung tissue of those patients with bronchial asthma. The increase in tissue content of glycosaminoglycans was accompanied by an increase in dermatan sulfate level. The increases in total amount of glycosaminoglycans in relative proportion of dermatan sulfate were closely related to the clinical findings, including vital capacity of the lung, PaO2, serum lactate dehydrogenase, and to the pathologic pictures of the lung.

Acid-base changes in ischemic myocardium and intervention with hypothermia or bicarbonate.

Effects of ischemia and reperfusion on acid-base changes in relation to myocardial contractility, and the effects of correcting H+ were studied by lowering PCO2 or increasing HCO3- levels. The hearts were perfused by working heart mode and whole heart ischemia was induced by use of a one way valve followed by myocardial warming (37 degrees C, normothermia), cooling (18 degrees C, hypothermia) or warming plus 2.1 mM NaHCO3 for 15 min. The hearts were then reperfused for 20 min. Coronary effluent was collected through pulmonary artery cannulation and used for the measurement of acid-base changes. There were close correlations between the decrease in coronary flow and LV pressure, LV dP/dt. Close correlations were also observed between the decline in LV pressure and the rise in PCO2, H+, and the decline in HCO3-. A highly significant correlation was seen between H+ and lactate production. Myocardial contractility decreased to the same extent in 3 groups during ischemia, whereas its recovery rate in both the hypothermia and HCO3- -treated groups were significantly higher than in the normothermia group. The increment of H+ was significantly less in both the hypothermia and HCO3- -treated groups than in normothermia. These results indicate that lactate production is the major H+ producing source and the correction of H+ could minimize the ischemic insult and at the same time contribute to the reperfusion injury.

Alpha 1-acid glycoprotein and hepatic fibrosis.

The relationship between alpha 1-acid glycoprotein (alpha 1-AG) and liver fibrosis was studied. Immunoperoxidase staining of liver specimens from patients with chronic hepatitis showed large amounts of alpha 1-AG to be located primarily in hepatocytes adjacent to areas of piecemeal necrosis, bridging fibrosis and fibrous septa. In patients with severe chronic active hepatitis, hepatocytes throughout the liver stained similarly to those adjacent to areas of piecemeal necrosis. In cell cultures of human embryonal lung (HEL) fibroblasts, addition to the culture medium of alpha 1-AG promoted cell growth. It is known that alpha 1-AG is also produced in the liver. It thus appears likely that alpha 1-AG is a promoter of hepatic fibrosis in chronic hepatitis.

The binding rate of sialic acid to serum alpha 1-acid glycoprotein in patients with chronic hepatic injury.

The level of serum orosomucoid, the amount of sialic acid binding to it, and the binding rate of sialic acid to orosomucoid in normal individuals and in patients with chronic hepatitis and liver cirrhosis were determined by affinity column chromatography, SDS electrophoresis and the method of Warren. Although no apparent changes in the level of serum orosomucoid or the amount of sialic acid binding to it in each group were seen, the binding rate of sialic acid to orosomucoid in chronic persistent hepatitis and chronic active hepatitis showed a remarkable increase, compared with normal individuals and patients with liver cirrhosis. Furthermore, the binding rate of sialic acid in liver cirrhosis was the lowest in each group.
