Enhanced neutrophil migration in vivo HLA B27 positive subjects.

Chemotaxis of polymorphonuclear leucocytes in vivo was studied in patients with previous yersinia arthritis and in healthy subjects with or without HLA B27 by means of a skin chamber technique. Irrespective of previous arthritis the number of neutrophils in the chamber media was significantly higher in HLA B27 positive subjects than in those without HLA B27. The amounts of prostaglandins E2, F2 alpha, and 6-keto-F1 alpha in the chamber media correlated positively with the corresponding cell counts. The present results give credence to the view that the hyperreactive neutrophils and the vasodilatory prostaglandins produced by them can together trigger a vicious circle which results in increased inflammatory symptoms in patients with yersinia arthritis who have HLA B27 as compared with those who lack this antigen.

Hangover headache and prostaglandins: prophylactic treatment with tolfenamic acid.

Tolfenamic acid (TA), a potent inhibitor of prostaglandin (PG) biosynthesis and action, was tested prophylactically against hangover symptoms in 30 healthy volunteers in a double-blind cross-over study. One capsule of TA (200 mg) or placebo was taken before starting to drink alcohol and another before going to bed. The hangover symptoms were evaluated in the morning. TA was found significantly better than placebo in the subjective evaluation of drug efficacy (p less than 0.001) and in reducing the reported hangover symptoms in general (p less than 0.01). In the TA group, significantly lower symptom scores were obtained for headache (p less than 0.01), and for nausea, vomiting, irritation, tremor, thirst and dryness of mouth (all p less than 0.05). In a separate study with eight participants, plasma levels of PGs were followed during ingestion of alcohol with or without TA. The plasma concentrations of PGE2 and TXB2 (a metabolite of thromboxane A2) were lower in the TA group during alcohol ingestion, while PGF2 alpha and 6-keto-PGF1 alpha (a metabolite of prostacyclin) were unaffected. TXB2 correlated with blood alcohol levels in a U-shaped manner.

Neutrophil function and HLA-B27.

The pathogenesis of HLA-B27 linked diseases, including reactive enteroarthritides, is not known. Differences in immune response, cross-reaction between microbial antigens and host cell structures, attachment of microbial components on the host cell surfaces with subsequent autoimmune reaction, and inflammatory responsiveness of the host may be involved. In this contribution we present evidence to show that HLA-B27 positive subjects may be prone to an exaggerated inflammatory response, as compared with HLA-B27 negative subjects.

The effect of age on circulating 6-keto-prostaglandin F1 alpha in humans.

The effect of age on the production of the antiaggregatory prostacyclin (PGI2) was studied by measuring 6-keto-PGF1 alpha in the plasma of 140 subjects (50 males and 90 females) between 10-90 years of age. The 6-keto-PGF1 alpha concentrations (mean +/- SD) in subjects between 10-20 years of age were higher (126 +/- 63 pg/ml, n = 24) than the levels in subjects between 21-30 years of age (88 +/- 30 pg/ml, n = 16), or in subjects between 51-70 years of age (88 +/- 30 pg/ml, n = 17), whereas in subjects between 71-90 years of age, the 6-keto-PGF1 alpha levels 100 +/- 59 pg/ml, n = 36) did not differ from those in subjects under 20 years of age. The 6-keto-PGF1 alpha levels in women over 70 years (118 +/- 66 pg/ml, n = 20) were higher (p = less than 0.05) than those in men of the same age (78 +/- 42 pg/ml, n = 16), but no other sex-related connections in 6-keto-PGF1 alpha concentrations were seen. Our data suggest that the PGI2 generation in vivo, as measured by 6-keto-PGF1 alpha levels in peripheral plasma, is higher in adolescence and elderly females than in the healthy adults.

Effect of iron on the absorption and distribution of clodronate after oral administration in rats.

The effect of iron on the absorption and distribution of disodium clodronate in rats after oral administration was studied. Disodium clodronate (300 mg/25 microCi/kg) was given both alone and with an equivalent amount of ferrous sulphate. The radioactivity in plasma and various tissue was measured. Concentration of clodronate in plasma was also determined with the GC-mass-selective detection method and the values compared with those measured with the isotope method. After administration, clodronate was rapidly cleared from plasma. Most of the dose was taken up by bone and only small amounts were found in non-calcified tissues. Concurrent ingestion of iron caused a marked decrease in the absorption of clodronate.
