Three-way differentiation of Chinese hamster ovary chromosomes by immunoperoxidase technique using a monoclonal anti-bromodeoxyuridine antibody.

A new permanent staining procedure for sister chromatid differentiation (SCD) in cultured Chinese hamster ovary cells has been established by combining the three-way differentiation in third mitosis (M3) chromosomes and the immunoperoxidase reaction developed with 3,3'-diaminobenzidine using a monoclonal anti-bromodeoxyuridine (BrdU) antibody. This procedure allows SCD at very low BrdU concentrations, and the evaluation of the sister chromatid exchange frequencies on a per cell-cycle basis.

Poly-D-lysine in G2 potentiates chromosome damage induced by X-rays and mitomycin C in CHO cells.

A number of reports suggest that the role of radiation-induced G2 arrest is to allow repair of potentially lethal damage in the cells before it comes to mitosis. Though the exact nature of the damage undergoing repair during the delayed G2 is not known, the yield of chromosomal aberrations observed in metaphase seems to be a good parameter to predict reproductive death of cells. In a previous paper, we have shown that poly-D-lysine, acting in a fashion reminiscent of that of caffeine in mammalian cells, is able to induce a premature onset of mitosis concomitant with an increase in the frequency of chromosomal aberrations in mutagen-treated plant cells. Cultured CHO cells were pre-exposed to either X-rays or mitomycin C and given different doses of the polycationic compound during G2 in order to analyze any effect on the frequency of chromatid-type aberrations as well as any modification of cell-cycle kinetics. A potentiation of chromosome damage and a premature arrival at mitosis were observed for both mutagens, though the effect was more evident in X-irradiated cells.

Brain mapping in dysphonemic dyslexia: in resting and phonemic discrimination conditions.

Topographic mapping of EEG bands and alpha asymmetry were analyzed during resting conditions and an auditory phonemic discrimination task (APD) in carefully screened samples of 9.0- to 12-year-old control and dysphonemic-sequencing dyslexic children. The EEG was recorded over the entire scalp, with a linked ears reference. We found: (1) no differences between groups during resting conditions in any frequency band analyzed; (2) a left hemisphere alpha responsiveness in dyslexic children during the APD condition; and (3) a beta 2 decrease in the left posterior quadrant in dyslexics during the APD condition. We conclude that the lower alpha and beta 2 responsiveness may be due to a diminished cortical reactivity and may be related to intrinsic disorganization of the neural processors associated directly with the APD task demands, the alpha activity reflecting poor attention to an external stimuli source and the beta 2 decrease reflecting an unusual processing associated with the linguistic cognitive requirements.

Chromosome damage induced by restriction endonucleases recognizing thymine-rich DNA sequences in electroporated CHO cells.

The influence of BrdU substitution of DNA in Chinese hamster cells on the frequencies of chromosomal aberrations induced by three restriction endonucleases which recognize thymine-rich sequences in DNA has been studied. The restriction enzymes chosen were Eco RI (recognition site G/AATTC), Sca I (AGT/ACT), and Dra I (TTT/AAA). A restriction enzyme that does not have thymine in the recognition sequence, Hae III (GG/CC), was also tried. These enzymes were introduced into cells by electroporation after two cell cycles of BrdU substitution and the aberration yields compared with that observed in non-substituted cells. Our results seem to indicate that the BrdU-substituted chromatin becomes resistant to the chromosome-breaking activity of the restriction enzymes recognizing thymine-rich DNA sequences. These observations are compared with the patterns of cutting of isolated DNA as shown by agarose gel electrophoresis.

Induction of chromosomal aberrations in the CHO mutant EM9 and its parental line AA8 by EcoRI restriction endonuclease: electroporation experiments.

EcoRI restriction endonuclease (RE), which produces cohesive-ended double-strand breaks (dsb) in DNA, was tested in the ethyl methanesulfonate- and X-ray-sensitive CHO mutant EM9 and its parental cell strain AA8 for its chromosomal aberration-inducing effect. The RE was efficiently introduced by electroporation into AA8 cells, while the mutant cells showed a very high sensitivity to electroporation, which consistently resulted in cell death. Nevertheless, the incubation of EM9 cells in the presence of EcoRI, without electroporation, was sufficient to induce about three times the chromosome damage observed in the electroporated parental cell line AA8 for any given dose of the RE.

G2 effects of DNA-repair inhibitors on chromatid-type aberrations in root-tip cells treated with maleic hydrazide and mitomycin C.

In recent years the existence of a DNA-repair process in G2 has been proposed to explain the potentiating effects of DNA-repair inhibitors given in G2 on chromatid aberrations (CA) induced by S-dependent as well as S-independent DNA-damaging agents. In the present report, root-tip cells of Allium cepa were exposed to maleic hydrazide (MH) or mitomycin C (MMC) and post-treated in G2 with caffeine (Caff) and various inhibitors of DNA synthesis. No enhancement of chromosome damage was observed when Caff was present in G2, but hydroxyurea (HU) or 5-fluorodeoxyuridine (FdUrd) potentiated the frequencies of CA. A slight additional increase of CA frequencies was observed following treatment with Ara C and excess thymidine in G2. When MH-damaged cells were pulse-treated with Caff earlier during recovery, the yield of CA was enhanced. The earlier Caff was present following MH treatment, the stronger was the potentiation.

Different distribution of ELI-perikarya in rat mesencephalon using various injection sites for colchicine.

In the present work, using the indirect peroxidase-labelled antibody technique, we made a comparative study about the distribution of Met-enkephalin-like-immunoreactive (ELI)-cell bodies in rat mesencephalon; comparing animals that received colchicine into the lateral ventricle and animals that got the colchicine injected into different cerebral nuclei. We discuss the possibility that the local intracerebral injections are more effective than the intraventricular ones and we show that the results depend on the injection place. So, we think that there are possible met-enkephalinergic connections between the injected region and their corresponding immunoreactive mesencephalic nuclei.

Effects of caffeine and inhibitors of DNA synthesis on chromatid-type aberrations induced by acetaldehyde in root-tip cells.

Root-tip cells of Allium cepa were exposed to acetaldehyde (AA) and post-treated with caffeine and 3 inhibitors of DNA synthesis, namely hydroxyurea (HU), 5-fluorodeoxyuridine (FdUrd), and arabinofuranosylcytosine (araC). Caffeine strongly potentiated the frequency of chromatid-type aberrations when given immediately after the AA treatment or as a 5-h treatment starting 10 h before the addition of colchicine. In contrast, no enhancement was observed when caffeine was present for the last 2.5 h, simultaneously with colchicine. The inhibitors of DNA synthesis were given following this last schedule. Both HU and FdUrd clearly enhanced the yield of AA-induced chromatid aberrations, while no enhancement of chromosome damage was observed after exposure to araC.

Enhanced androgen production by rabbit adrenocortical cells stimulated chronically with corticotropin: evidence for increased 17 alpha-hydroxylase activity.

The effects of prolonged treatment with corticotropin (ACTH1-24, 200 micrograms s.c. daily during 12 days) on the production of androgens and glucocorticoids were studied on rabbit dispersed adrenocortical cells. The steroidogenic capacity of adrenocortical cells, expressed in terms of the maximal response to ACTH of glucocorticoid (i.e. corticosterone and cortisol) production, was significantly increased after treatment with ACTH. This was associated with a loss of sensitivity to this peptide: indeed, the concentration of ACTH required to induce a half maximal secretory response was one order of magnitude higher with cells from ACTH-treated animals. Among the C21 steroids measured the changes observed involved the 17 alpha-hydroxylated compounds (cortisol, cortisone, 11-deoxycortisol) while corticosterone production was significantly depressed. This effect of prolonged ACTH treatment on steroidogenic pathways involving 17 alpha-hydroxylation, was further evidenced by a clear-cut enhancement in androgen secretion (dehydroepiandrosterone, androstenedione and testosterone) by adrenocortical cells from ACTH-treated animals. The changes observed after treatment of the animal with ACTH were equally obvious, whether the adrenocortical cells were incubated with ACTH or with dibutyryl-c-AMP.

[Neuropsychology and cognitive deterioration in epilepsy].

OBJECTIVE: To carry out a critical evaluation of the international literature on the neuropsychology of epilepsy. The variables which lead to cognitive deterioration in epilepsy and the effect on cognition of different treatments available (pharmacological and surgical). DEVELOPMENT: We evaluate the influence of different neurological variables on the higher functions (aetiology, age of onset, type of crises, duration of illness, frequency and anti-epileptic drugs). We also describe the cognitive functions most affected in epilepsy (memory, attention, executive function, language). Studies of the surgery of epilepsy show that this may lead to both beneficial and undesirable effects on cognition. CONCLUSIONS: The variables which in general most affect higher functions are the duration of the illness and the frequency and types of crises. The commonest neuropsychological effects are those of memory deficit. These studies show that neuropsychological studies should be done from the time of onset of the disorder. Finally, from the surgical point of view, young patients with considerable crisis reduction or free of crises, with moderate preoperative deterioration of memory have the greatest possibility of post-operative improvement in cognition. Older persons with intact cognitive function and major surgical resection have more possibilities of post-surgical deterioration.

[Psychometric validation of the telephone memory test].

INTRODUCTION: Several pathologies (i.e. Alzheimer's disease) that courses with memory alterations, appears in a context of impaired cognitive status and mobility. In recent years, several investigations were carried out in order to design short batteries that detect those subjects under risk of dementia. Some of this batteries were also design to be administrated over the telephone, trying to overcome the accessibility limitations of this patients. METHODOLOGY: In this paper we present a battery (called Autotest de Memoria) essentially composed by episodic and semantic memory tests, administered both over the telephone and face to face. This battery was employed in the cognitive assessment of healthy controls and subjects diagnosed as probable Alzheimer's disease patients. RESULTS: Results show the capability of this battery in order to discriminate patients and healthy controls, a great sensibility and specificity, and a nearly absolute parallelism of telephone and face to face administrations. CONCLUSION: These data led us to claim the usefulness and practicality of our so called < >.
