A cleavage product of mouse C3 adsorbable to (1----3)-beta-D-glucan from Alcaligenes faecalis var. myxogenes IFO 13140.

By using affinity chromatography of TAK, a gel-forming antitumor (1----3)-beta-D-glucan produced by Alcaligenes faecalis var. myxogenes IFO 13140 also known as an activator of the alternative complement pathway (ACP), we obtained from mouse serum a cleavage product of C3 (tentatively designated as C3X). Binding between C3X and TAK seemed to be noncovalent. In contrast to C3, C3X had an intact beta chain (70,000 daltons) and instead of an alpha chain, two peptide chains (40,000 and 30,000 daltons) linked by disulfide bonds. C3X retained a portion of C3 antigenicity. By trypsinolysis C3 yielded a fragment similar to C3X on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. These results suggest that C3X is C3c. Since C3X was obtained by elution with a high salt solution from TAK incubated with ethylenediaminetetraacetate (EDTA)-serum (serum containing EDTA) but not from TAK incubated with EDTA-plasma, C3X could already be present in the serum before the activation of ACP by TAK.

Liquid-chromatographic measurement of creatinine in serum and urine.

We describe the adaptation of a "high-performance" liquid chromatographic method for determination of creatinine in serum and urine. The proposed method is simple, rapid, precise, and accurate. The retention time for creatinine can be varied simply by changing the KH2PO4 concentration in the mobile phase: acetonitrile/aqueous KH2PO4 (1/4 by vol). Within-day precision (CV) was 1.2-3.6% in serum chromatographed with an internal standard, and 2.3-2.8% in serum when an external standard was used. Between-day precision (CV) was 1.3-2.1% in serum and 1.3-2.7% in urine (with an external standard). Analytical recoveries of creatinine added to serum were 94-100% for the method with an internal standard, 95-103% with an external standard.

Pharmacokinetics of the cephalosporin SM-1652 in mice, rats, rabbits, dogs, and rhesus monkeys.

The pharmacokinetics of SM-1652 were studied in mice, rats, rabbits, dogs, and rhesus monkeys. The plasma half-lives of SM-1652, administered intravenously at a dose of 20 mg/kg, were 11.0 min in mice, 26.0 min in rats, 65.8 min in rabbits, 72.6 min in dogs, and 150.9 min in monkeys. The 24-h urinary excretion of SM-1652 was 30 to 35% of the dose in mice and rats, 70 to 75% in rabbits and dogs, and 45% in monkeys. Biliary excretion of the antibiotic over a 24-h period was 60 and 19% in rats and rabbits, respectively; it was 19% in dogs over a 9-h period after SM-1652 administration. Approximately 95% of the intravenous dose of SM-1652 was recovered as the unchanged form in the urine and bile of rats and rabbits. The binding of SM-1652 to serum protein was 44.0% in mice, 46.0% in rats, 90.4% in rabbits, 93.2% in monkeys, 30.0% in dogs, and 96.3% in humans.

Development of a hemolytic assay for mouse complement components in sera and the variation of their levels with age.

The hemolytic activity of C1, C4, C2 and C3-9 of mouse complement in serum could be measured by using each intermediate cells with guinea pig or human complement components. C3 was assayed by immune adherence. Sheep erythrocytes (SRBC) sensitized with rabbit IgM antibody produced no or feeble lysis in the assay for C4 and C2, whereas SRBC sensitized with IgG antibody or unfractionated antiserum did significant lysis. Using these assays, the activity of each component in sera of several H-2 congenic strains was examined. The age-dependent changes of each component levels in BALB/c mice of both sexes were also examined. The differences in levels of C4 and C2 in sera from several strains were not significant in both sexes, while the levels of C3-9 were higher in males than in females. The levels of each component increased with age. The levels of C4 activity and the Ss protein showed good correlation in males but not in females. The correlation between the levels of C3-9 activity and the MuB1 protein was detected in both sexes.

Studies on protein nutrition of Papua New Guinea highlanders: nitrogen balance and hematological studies.

Studies were made in August 1978 on the N-balance and hematological characteristics of 18 men living in the village of Kalugaluvi (altitude: 1,500m) near Lufa, which is 60km from Goroka, in the Eastern Highland Province of Papua New Guinea. The average daily protein intake on 3 consecutive days was 95.2 +/- 29.3 mgN/kg (35.2 +/- 10.7 g protein/day), and 32.6 +/- 18.7% of the total protein intake was in the form of animal proteins. The calculated nutritional value of the dietary protein, scored according to the 1973 FAO/WHO pattern, was 83.7 +/- 10.6. Urinary and fecal N excretions during the same period were 81.9 +/- 18.7 and 26.3 +/- 11.8 mgN/kg, respectively, giving a nitrogen balance of -13.5 +/- 24.9 mgN/kg. From N-balance data on individuals, the N-intake for maintenance of a zero N-balance was estimated as 116.3 +/- 48.6 mgN/kg (mean 95% confidence interval). Hematological data showed normal or rather high values for hemoglobin (16.90 +/- 1.05 g/100 ml) and serum proteins (8.05 +/- 0.40 g/100 ml). The increased level of serum proteins was associated with an increased globulin level.

Nutritional status of Papua New Guinea highlanders.

A nutritional survey was held in August, 1978, at Kalugaluvi (altitude: 1,500m) near Lufa, which is 60 km from Goroka, in the Eastern Highland Province of Papua New Guinea. Anthropometric measurements were carried out on 55 males and 37 females aged from 7 to 64 years. whereas the physiques of the children looked as good as those of Japanese of a comparable age, the adult men were shorter than Japanese males, but body weight and chest girth were similar. The skinfold thickness was less than that of the Japanese. From the data collected, it was shown that the physique of the Highlanders was more muscular than that of the Japanese. The food intakes and energy balances of 18 healthy men (20-40 years old) were measured over 2 or 3 consecutive days. The average consumption of sweet potatoes, the staple food, was 956 +/- 305 g per day. The average consumption of taro and yam was 93 +/- 124 g/day and 36 +/- 99 g/day, respectively. Various green leaves, sugar canes, corn, bananas and other foods. (i.e., rice and tinned fish) purchased from trade stores were sometimes eaten. The mean daily energy intake was 2.390 +/- 540 kcal, which was about the same as the daily energy expenditure. The daily protein intake was 35.2 +/- 10.7 g. These results are probably exceptionally high, because the survey was unfortunately held during the yearly festival season of the village when the people often ate fatty port. Nevertheless, it is noteworthy that the growth of children and the physique of adults are normal in spite of the extremely low intake of protein.
