Surface membrane nucleoside triphosphatase activity and tumorigenicity of cultured liver epithelial cells.

A cell surface-located nucleoside triphosphatase activity can be assayed in liver epithelial cultures in situ with the incubation of intact cells in a medium containing [gamma-32P]adenosine triphosphate and correlated with the tumorigenicity of these cells in neonatal Wistar rats. The ectoenzyme activity of normal diploid cell lines is minimal, whereas a considerably high activity has been found in all tumorigenic cell lines tested. The optimum condition for the adenosinetriphosphatase activity is physiological with regard to osmolarity, ionic composition, pH, and substrate concentration in the medium. The enzyme is significantly stimulated by Ca2+, and its activation is controlled by Mg2+. Histochemical examinations indicate that glutaraldehyde-fixed cells of tumorigenic lines have Ca2+-stimulated adenosinetriphosphatase activity on the external surface. The isotopic assay of adenosine triphosphate hydrolysis by intact cells may provide a rapid method for screening oncogenesis in vitro of liver epithelial cells.

Detection of oncornavirus-like particles in normal and tumorigenic rat liver cell cultures.

Six epithelial cell cultures established from adult Wistar rat liver were examined by electron microscopy for oncornavirus-like particles. Two heteroploid and tumorigenic cultures demonstrated many cells with virus-like particles of type-A and type-C morphology. The remaining four, composed of diploid nontumorigenic cells, demonstrated similar virus-like particles but in few cells and only after bromodeoxyuridine treatment. The virus-like particles in the tumorigenic cultures were identified as a rat type-C virus by serological assays.

Tyrosine transaminase induction by dexamethasone in a new rat liver cell line.

A cell line derived from normal, adult rat liver has been established; the cells are similar to hepatocytes, as shown by electron microscopy. The addition of dexamethasone to the culture medium induced a three- to sixfold increase in the specific activity of tyrosine alpha-ketoglutarate transaminase; this increase was inhibited by the simultaneous addition of cycloheximide or actinomycin D. The latter, when added to cells given prior treatment with dexamethasone, further enhanced the transaminase activity. Contact-inhibited cells showed a lower response to dexamethasone than exponentially growing cells.
