Familial tricho-rhino-phalangeal syndrome Type II.

Two cases, a father and daughter, with all the principal signs of tricho-rhino-phalangeal syndrome Type II are described, although nine previously reported cases were all sporadic. It is suggested that these patients have a genetic disorder with an autosomal dominant mode of inheritance. It may be reasonable to assume that a patient with relatively mild mental retardation, such as the father in the present report, could marry and have off-spring. Generalized aminoaciduria was found in the affected daughter.

High performance liquid chromatographic determination of aclacinomycin A and its related compounds. I. Normal phase HPLC for monitoring fermentation and purification processes.

A normal phase high performance liquid chromatographic method is presented for the separation and quantitative determination of aclacinomycin A (ACM) and its related compounds classified as anthracycline antibiotics. The sensitive, reliable and reproducible chromatography was achieved on a mu-Porasil column with a mobile phase consisting of chloroform-methanol-acetic acid-water-triethylamine (68:20:10:2:0.01, v/v). By use of MA144 M1, one of the ACM analogues, as an internal standard, selectivity of the HPLC method could be retained by virtue of the adjustment of the detecting wavelength to 436 nm. The HPLC method was applicable to monitoring the fermentation and purification processes.

High performance liquid chromatographic determination of aclacinomycin a and its related compounds. II. Reverse phase HPLC determination of aclacinomycin A and its metabolites in biological fluids using fluorescence detection.

A sensitive, simple and selective reverse phase high performance liquid chromatographic method was applied to the determination of aclacinomycin A (ACM) and its metabolites in plasma. The chromatography was performed on a microparticulate phenethyl derivative of silica using a mobile phase of acetonitrile-0.03 M ammonium formate buffer (pH 5.0), (50:50, v/v) and monitoring with fluorescence detector (excitation 435 nm-emission 505 nm). High reproducibility and linear calibration curves were obtained from 20 ng/ml to 100 ng/ml of ACM and its metabolites in plasma by use of internal standard methods.

New anthracyclines, feudomycins, produced by the mutant from Streptomyces coeruleorubidus ME130-A4.

Various blocked mutants were isolated from Streptomyces coeruleorubidus ME130-A4 by NTG and UV treatments. Among them, mutant strain 4N-140 produced new anthracycline feudomycins A and B having new aglycones in which the side chain at C-9 position of daunomycinone was ethyl and acetonyl, respectively. New aglycones feudomycinones C and D having methyl at C-9 and additional groups at C-10 of daunomycinone were also isolated from this strain.

New anthracycline metabolites from mutant strains of Streptomyces galilaeus MA144-M1. I. Isolation and characterization of various blocked mutants.

During genetic study on obtaining high-yield variants of aclacinomycin A-producer, a variety of blocked mutants were isolated from Streptomyces galilaeus MA144-M1 and were characterized. The isolated mutants included those which accumulated only specific components of parental glycosides, those which produced new aklavinone glycosides devoid of parental rhodosamine or both rhodinose and cinerulose, those which produced non-glycosidic aglycones, and antibiotic-negative mutants, some of which were able to glycosidate exogenous aklavinone. By biotransformation with the aglycone feeding culture, the precursor activity of new aglycones was also tested. From the results and in relation to the characterization of isolated mutants, the biosynthetic pathway of aclacinomycin A and related antibiotics is discussed.
