Cardiac lesions in collagen disease.

Clinical and pathological abnormalities of the heart that may have major functional significance in those conditions were studied in collagen disease (total 242 cases). 1. atypical verrucous endocarditis (Libmann-Sacks) were found in 2 cases with loupus erythematosus (25 cases) and in 1 case with scleroderma (3 cases). 2. Rheumatoid nodules were not seen in the heart of rheumatoid arthritis patients (9 cases). 3. The most frequent findings in the heart with collagen diseases were pericarditis, fibrous scarring of the heart muscle, focal inflammation and vasculitis is similar to periarteritis nodosa or endoarteritis obliterans. 4. Morphological changes of the A-V node were found in patients who had A-V dissociation or A-V block before death. Heart muscle infarctions were found in 2 cases.

Effects of verapamil on SA and AV nodal action potentials in the isolated rabbit heart.

In the isolated rabbit heart, it was shown that verapamil suppressed the electrical activities of the SA and AV nodes without preventing regenerative responses in atrial muscle fibers. It was concluded that the different effects of verapamil on SA and AV nodal action potentials resulted from the operation of a different ionic channel (slow channel) from that of ordinary atrial muscle.

Studies on pharmacological and biochemical properties of deamino-dicarba-[GLY-7]-oxytocin (Y-5350).

Pharmacological and biochemical properties were investigated on deamino-dicarba-[Gly-7]-oxytocin (Y-5350). This is a newly developed compound, in which the disulfide bond and [Pro-7] of deamino-oxytocin are substituted by an ethylene linkage and glycine respectively. Bioassayed Y-5350 exhibited 202.6 u/mg of oxytocic activity, 4.6 u/mg of avian depressor activity, 441.2 u/mg of rat milk-ejecting activity and 0.02 u/mg of rat antidiuretic activity, however, adepressor activity was also evident in rats. In particular, the diuration of antidiuretic activity was short. Moreover, the oxytocic activity in pregnant rats and rabbits was weak in comparison with oxytocin. Cumulative dose-response studies were carried out on the isolated rat uterus using van Dyke-Hasting solution. The intrinsic activity was the same level as that of oxytocin, and the pD2 value was 8.62. Oxytocic activity was much enhanced by the existence of 0.5 or 2mM magnesium ion in vitro. However, the agreement between in vivo and in vitro oxytocic activity was not complete when assay was carried out in the prescence of 2 mM magnesium ion. Oxytocin was inactivated by the serum of pregnant women. In the experiment using rats, oxytocin was also destroyed by the extracts of uterus, kidney and liver. In contrast, Y-5350 was not destroyed by any of the enzyme solutions mentioned above.

Responses of Clostridium botulinum type B and E progenitor toxins to some clostridial sulfhydryl-dependent proteases.

Sulfhydryl-dependent proteases produced by Clostridium botulinum types A, B, and F, Clostridium histolyticum, Clostridium sporogenes and Clostridium perfringens activate preferentially type E over type B progenitor toxin but less efficiently than trypsin. The results explain why activable toxin is demonstrable in culture of a strongly proteolytic type B strain.

Common atrium associated with anomalous high insertion of the inferior vena cava.

The case is reported of a 38-year-old housewife with a common atrium associated with anomalous high insertion of the inferior vena cava, absence of the coronary sinus, and drainage of a left hepatic vein into the left side of common atrium. Surgical repair was performed successfully. The procedure was closure of a nearly total atrial septal defect with a Dacron patch. Embroyologically, the development failure of the sinu-atrial fold appears to be the basic causal factor.
