A leukocytosis-promoting substance was purified from a crude bovine parotid gland extract. The purified substance was proved to be a single component by polyacrylamide gel disc electrophoresis. It stimulates an increase of peripheral leukocyte numbers in rabbits. The molecular weight of the physiologically active component was estimated to be 4.5 . 10(4), and the component was found by sodium dodecyl sulfate polyacrylamide gel electrophoresis to be dissociated into two subcomponents.
Human antibodies to porcine erythrocytes were found to bind to porcine but not to human zonae pellucidae, whereas human isohemagglutinins bound to human but not to porcine zonae. On the basis of the binding behavior of agglutinins, it was found that the immunofluorescence of porcine zonae produced by selected sera from infertile women was due to an antibody of different specifity from that which agglutinated pig red blood cells. However, no serum component other than heteroagglutinin was contained in selected sera from control subjects which fluoresced porcine zonae. Since the component was present in the immunoglobulin G fraction, bound to human zonae, and was reactive with zona-specific antigen(s), it was judged an autoantibody to zonae. Thus a method for specific detection of autoantibodies to zonae has been developed by indirect membrane immunofluorescence using porcine ova as targets.
Paraformaldehyde-fixed mKSA tumor cells of BALB/c mice were shown to retain tumor-associated transplantation antigen (TATA) activity to a degree comparable to that of X-ray-inactivated tumor cells under the optimal conditions of 1% paraformaldehyde for 30 min at 37 degrees. Unexpectedly, the TATA activity of cells fixed below 10 degrees was greatly reduced. This temperature effect was reversible. TATA activity was restored if the cells were returned to 37 degrees before fixation. Fixation at all temperatures for longer than 2 hr or at paraformaldehyde concentrations greater than 1% also caused a decrease in immunogenicity. Spleen cells from mice immunized with tumor cells fixed at 37 degrees were able to more effectively neutralize tumor growth in the Winn assay compared with those from mice immunized with cells fixed at 0 degrees. Immunization with paraformaldehyde-fixed tumor cells was completely specific. Mice immunized with an antigenically unrelated tumor were not rendered immune to tumor challenge. Fixed tumor cells could be stored for at least 1 month without loss of TATA activity.
Having resistance to beta-lactamase-producing strains and showing resistance to not only cephalosporin resistant strains of E. coli and Klebsiella but also to Citrobacter, Proteus and Enterobacter, Cefuroxime (CXM) was used in pediatric field for both fundamental and clinical studies. CXM was found to be a useful antibiotic in views of high clinical efficacy rate obtained and no side effect noted. As for the dose, the single dose of 25 mg/kg achieved sufficient blood levels. Also in view of good clinical effect, the dose of 25 mg/kg three or four times daily seems appropriate for treatment of children.
A new colorimetric determination for serum phospholipid is described. Firstly, serum phospholipid is incubated with phospholipase C from Bacillus cereus, and then the released diglyceride and triglyceride are hydrolyzed completely to fatty acid and glycerol by lipoprotein lipase from Pseudomonas fluorescens. Secondly, the glycerol produced is enzymatically determined by glycerol dehydrogenase in the presence of NAD+, using phenazine methosulfate-nitro blue tetrazolium as color reagents. The absorbance at 570 nm is recorded. The amount of the glycerol from phospholipid is calculated by subtracting the amount of glycerol from triglyceride from the amount of total glycerol. The present method requires only 20 microliter of serum and a 40 min incubation and is highly reproducible. The results obtained show good correlation with those obtained by a chemical method (correlation coefficient, 0.925) or the phospholipase D-choline oxidase method (correlation coefficient, 0.936). These results strongly suggest that the proposed method can be utilized as a routine clinical test.
Of 52 serum samples from infertile women tested against isolated porcine oocytes by immunofluorescence, 8 produced intense reactions in zonae pellucidae, while only one of 52 sera from control subjects showed the reaction. Autoantibodies to the zona may be present in these women.
The effect of 25 mg of chlorpromazine (CPZ) on the contingent negative variation (CNV) at Cz and Oz was studied in nine male subjects. Two kinds of warning stimulus (S1)--imperative stimulus--and motor response (MR) were used. A click as S1 was common to both, while either flashes or electrical stimulation was used as S2. Simultaneously with CNV recording, the reaction time of the finger movement as MR to S2 was measured in five of nine subjects. The results obtained were as follows. 1) CNV amplitudes after CPZ administration were decreased significantly both at Cz and Oz when S2 was flashes and only at Cz when S2 was electrical stimulation. 2) Significant positive correlations of CNV amplitudes between Cz and Oz were found both before and after the drug administration. 3) Reaction times to flashes and electrical stimulation as S2 were not affected by CPZ administration. 4) The reaction time to flashes was significantly shorter than that to electrical stimulation irrespective of pre- and post-administration of CPZ.

The tumor-associated transplantation antigens (TATA) of crude membrane extracts from SV40-transformed BALB/3T3 tumor cells lytically infected with influenza virus were markedly more immunogenic than were extracts from uninfected cells measured either by the ability to induce heightened resistance to tumorgraft challenge or by heightened lymphocyte-mediated cytotoxicity against tumor cells in vitro. When intact tumor cells (as opposed to membrane extracts) were productively infected with Moloney sarcoma virus, they were made so immunogenic that they would only grow in X-irradiated syngeneic animals. Yet crude membrane extracts from the Moloney sarcoma virus-infected tumor cells showed no increase in TATA activity analogous to that seen after infection with influenza virus. Thus, influenza virus augmentation of tumor membrane TATA may operate by a different mechanism than does the oncornavirus augmentation of intact tumor cell TATA reported by others. It appears that Moloney sarcoma virus and possibly other oncornaviruses cannot be used to augment the TATA activity of tumor cell membranes in the same way that other surface-budding viruses can.

Regional cerebral blood flow (rCBF) was measured in patients with no neurological deficit and no abnormal findings of cerebral angiograms except aneurysm itself with Xenon-133 injection technique by using the computerized 32-detector rCBF system (rCBF-322, Meditronic, Denmark). This system consists of a 32-detector head with a concave surface and a 32 pulse height analyser and analog-digital ratemeter as the accumulator connected to display units with an oscilloscope for 32 clearance curves and also a microcomputer programmed for calculating the rCBF initial with the initial slope analysis, for calculating the rCBF 10 with the height-over-area method and the rCBF gray, rCBF white, weight gray and weight white with the SHAM method. The results obtained were as follows: 1) The mean value and SD of rCBF initial was 58.4 +/- 6.2 ml/100 g brain/min; rCBF 10, 50.6 +/- 5.0 ml/100 g brain/min; rCBF gray, 78.0 +/- 14.0 ml/100 g brain/min; rCBF white, 22.1 +/- 6.6 ml/100 g brain/min; weight gray, 49.0 +/- 3.7%; weight white, 51.0 +/- 3.7%. These data should be considered normal values as reported also by others. 2) Reproducibilities were estimated by measuring twice the rCBF initial and the rCBF 10 at rest with closed eyes. The reproducibilities of the rCBF initial eyes 13.55% (P less than 0.05) and one of the rCBF 10 was 8.85% (P less than 0.05). It was concluded that this system should be widely used for measuring the rCBF in patients with various cerebral diseases.
