Effects of tripelennamine, N,N-dimethyl-N',N'-dibenzylethylenediamine (DBED) and N,N-dibenzyl-N',N'-dimethyl-1,2-propanediamine (DBPD) on the isotonic contractions of isolated vas deferens of guinea pig were examined. These ethylenediamines, except DBPD, induced slight but significant increase in sensitivity of vas deferens to K+. Tripelennamine induced dose-dependent potentiation to epinephrine and norepinephrine, but tripelennamine-induced augmentation of acetylcholine-contractions was weak and dose-independent. DBED potentiated both catecholamines and acetylcholine in a dose-dependent way and in the same degree. The degree of DBPD-induced augmentation of epinephrine- and norepinephrine-induced contractions was dose-independent and much weaker than that of acetylcholine-contractions. Tripelennamine and DBED did not affect the contractile response to tyramine, while DBPD potentiated the contractile response to tyramine. These results suggest the following possibilities: 1) tripelennamine affects both amine uptake mechanism and beyond receptor mechanism of contractile processes and induced supersensitivity of various stimulants. 2) DBED affects mainly beyond receptor mechanism of contractile processes and augments the contractile responses to various stimulants. 3) The mechanism of DBPD-induced supersensitivity remained obscure, but the inhibition of the metabolic degradation of stimulants was proposed as one of the possible mechanism of supersensitization.

Ten different alloantisera absorbed with red blood cells (RBC) from appropriate strains of rats detected a series of B cell alloantigenic specificities by complement dependent cytotoxicity test, and that could be divided into three groups, presumably coded for by at least three different loci. The three loci were designated provisionally as Ba-1, Ba-3 and Ba-2 having two, three and six alleles, respectively, by analyzing strain distribution of Ba antigens. Each specificities were also designated Ba-1.1, 1.2, Ba-3.1, 3.2, 3.3, Ba-2.1, 2.2, 2.4, 2.9, 2.6, 2.7. The specificities from Ba-1 and Ba-3 groups were distributed in many strains of rats, and the specificities from the Ba-2 group were distributed in a limited strains of rats. By immunogenetical study, associations of the Ba specificity with RT1-Aa specificities, and of the Ba specificity with MLCR phenotype were observed. Immunochemically, Ba-1, Ba-3, Ba-2 antigenic specificities were studied using the B cell leukemia, KNL-14 cells, which was induced in the WKA rat with 1-buthy-1-nitrosourea, and it was clarified that as far as Ba-1.2, Ba-2.7, Ba-3.3 specificities were concerned, the three antigen specificities were found to be on the different rat Ia-like molecules, thus supporting a postulation of the existence of three loci in the RT1-B region, which was suggested by serological analyses.
Five rat leukemia cell lines induced with chemical agents and/or leukemia viruses were investigated in terms of their surface antigens. One of them, KNL-14, which was induced with 1-butyl-1-nitrosourea in WKA rats had RT1-B region-associated antigens on the cell surface. In addition, Thy-1.1 antigens and surface and cytoplasmic mu chains were detected by serological and immunochemical means. From the results it was concluded that this leukemia originated from immature B cells. In the present work, RT1 antigens on the KNL-14 cells were further investigated. The class I and class II antigens were examined and KNL-14 cells were found to have phenotypical antigens and immunogenicity similar to those of B cells from the WKA lymph nodes. As for class II antigens, which are responsible for various immunologic functions, two specificities were detected, Ba-1.2 and Ba-2.7. These KNL-14 cells should be useful for studying the process of rat B cell differentiation and for immunochemical studies of the rat MHC gene products.
Three different electric stimulation methods--direct current, electric Teflon film and PMLG piezoelectric film--could induce electrical osteogenesis around Donryu rat femur. The configuration of each type of electrical osteogenesis was different but the histological findings were essentially the same. 3H-thymidine autoradiographic studies were carried out on rat femurs stimulated by direct current to elucidate some mechanisms of electrical osteogensis. The results disclosed that the electric stimulation affected not only proliferation of osteoprogenitor cells from the periosteal cells but also differentiation of osteoprogenitor cells to osteoblasts and finally to osteocytes.

A radioimmunoassay procedure for plasma 11-deoxycortisol (S) was developed using an antiserum prepared by immunizing rabbits with S-21-hemisuccinate bovine serum albumin and S-3-oxime-bovine serum albumin. Thereafter plasma S, cortisol (F) and adrenocorticotropic hormone (ACTH) responses to metyrapone were investigated in 13 normal adult males and 39 patients with prostatic cancer. The results were as follows: 1) The antiserum against S-3-oxime-bovine serum albumin had less cross reactivity (less than 10%) with other steroids than that against S-21-hemisuccinate bovine serum albumin and obtained a good standard curve. The intra-assay variance and interassay variance of this method using the former antiserum (N = 10) were 12.4% asd 14.9% respectively, and the blank value was 3.7 +/- 1.6 pg. 2) Basal levels of S. F and ACTH in plasma from 13 normal adult males, ranged 21 approximately 80 years, old, were 98.4 +/- 15.7 ng/dl (mean value +/- S.E.), 12.7 +/- 0.78 micrograms/dl and 30.6 +/- 3.02 pg/ml respectively. Those level increased to 7060 +/- 598 ng/dl, 24.3 +/- 1.69 micrograms/dl and 24.3 +/- 1.6 pg/ml at 9 a.m. following oral administration of metyrapone (30 mg/kg b.w.) at midnight. 3) Both basal levels and responses of plasma S and F to metyrapone increased remarkably, while those of ACTH were within the normal range in prostatic cancer patients during the estrogen therapy. It was considered that protein-bound S and F increased following elevation of corticosteroid binding globulin but returned to the normal range about 2 weeks after discontinuation of the therapy. 4) In case treated wih estrogens, plasma, S, F and AC normal range in prostatic cancer patients during the estrogen therapy. It was considered that protein-bound S and F increased following elevation of corticosteroid binding globulin but returned to the normal range about 2 weeks after discontinuation of the therapy. 4) In case treated wih estrogens, plasma, S, F and ACTH responses to metyrapone were unchanged compared to normal adult males 2 approximately 4 weeks after discontinuation of the therapy, and this data suggested that estrogens had no inhibitory effect on the pituitary-adrenal axis. However, in cases treated with progestational agents over a long-term period, plasma S and ACTH responses to metyrapone decreased slightly but returned to the normal range 2 approximately 4 weeks after discontinuation of the therapy. This suggested that the inhibitory effect of these agents on the pituitary-adrenal axis was mild and reversible.

Computer processed adrenal imaging using dual-radioisotopes, 6 beta-iodomethyl-19-nor-cholest-5(10)-en-3 beta-ol-131I and 99mTc-phytate was performed in 12 patients with primary aldosteronism and 4 with Cushing's syndrome due to adrenocortical tumor. Adreno-photoscanning and hepato-photoscanning were performed in the same position 2-4 days following intravenous administration of radiocholesterol. The scintigraphic information was stored on cassettes and scan subtraction and a digital-computer method for data smoothing were performed on an oscilloscope. The tumor site could be determined in all cases until day 4 by this computer processed image.

A sequential radioimmunoassay procedure for unconjugated and conjugated estrone, estradiol-17 beta and estriol in male human plasma was developed. The blank values in this assay for unconjugated estrone, estradiol-17 beta, estriol conjugated estrone, estradiol-17 beta and conjugated estriol were 0.36 +/- 1.14 pg, 3.90 +/- 2.75 pg, 2.25 +/- 2.08 pg, 0.92 +/- 1.51 pg, 5.02 +/- 2.86 pg and 3.12 +/- 2.97 pg, respectively. Mean values of unconjugated estrone, estradiol-17 beta, estriol, conjugated estrone, estradiol-17 beta and conjugated estriol in plasma from 28 normal adult males were 38.4 +/- 13.4 pg/ml, 32.6 +/- 9.90 pg/ml, 4.06 +/- 3.68 pg/ml, 34.2 +/- 13.8 pg/ml, 40.4 +/- 12.3 pg/ml and 31.8 +/- 7.41 pg/ml, respectively. Both unconjugated and conjugated estrogen levels in patients with liver cirrhosis were elevated and conjugated estrogen, especially estriol levels, in patients with renal insufficiency were markedly elevated.
