Detection of human T-cell leukemia virus type I (HTLV-I) provirus in an infected cell line and in peripheral mononuclear cells of blood donors by the nested double polymerase chain reaction method: comparison with HTLV-I antibody tests.

Human T-cell leukemia virus type I (HTLV-I) provirus DNA from the cultured cell line HUT 102 and from peripheral mononuclear cells (PBMC) of anti-HTLV-I antibody-positive Japanese blood donors was detected by the nested double polymerase chain reaction (PCR) method. This procedure consists of a first amplification and a second amplification with the products of the first amplification and primers interior to the first primers. Using this method, we demonstrated that it is possible to detect single-template DNA. Polyacrylamide gel electrophoresis of the nested double PCR products, with our primers, revealed three bands with excess amounts of template DNA, two bands with moderate amounts, and a single band with limited amounts. The amount of provirus in PBMC was roughly estimated from the results of the nested double PCR. Particle agglutination (PA) assays and indirect immunofluorescence testing (IF) with mixed MT-2 cells and Molt-4 cells as targets to detect anti-HTLV-I antibody were performed, and the results were compared with those of the nested double PCR of the pX region. None of the 101 PA-negative samples were positive in either the IF or PCR test. Of the 155 samples that were antibody positive by the PA assay, 57 were positive by both PCR and IF. Furthermore, the results of the IF and PCR tests coincided completely. It was therefore concluded that the IF method is most appropriate for confirmation of the PA assay currently used in most diagnostic laboratories and blood centers.

Effect of grinding on dehydration of crystal water of theophylline.

The effect of grinding on the dehydration of crystal water of theophylline has been studied. It was observed that the water content of theophylline hydrate decreased with increased grinding time. As the grinding time proceeded, the results of differential scanning calorimetry (DSC) indicated that crystal water of ground theophylline hydrate dehydrated in three steps at ca. 58, 44, and 17 degrees C, respectively. Powder X-ray diffraction study revealed that the crystal lattice of theophylline monohydrate collapsed by grinding, and part of the theophylline molecules subsequently rearranged the collapsed lattice to form theophylline anhydrate. The result of Fourier transformed infrared spectroscopy demonstrated that the hydrogen bonds between crystal water molecules and theophylline molecules were weakened or destroyed to some extent by grinding. It was supposed that crystal water in the ground theophylline hydrate might exist at least in three molecular states of different hydrogen-bonding. From DSC study, it was suggested that the ruptured hydrogen bonds of water molecules in the ground theophylline hydrate were strengthened after storage under 96.5% relative humidity at 30 degrees C.

[The effects of a recipient's openness and conveyance to a third party of the self-disclosure on changes in the discloser's liking and self-disclosure].

One hundred and ninety-nine university students (101 males, 98 females) served in a study designed to examine various factors affecting a discloser's degree of liking of and self-disclosure to a recipient. Specifically, the effects of (1) the recipient's degree of openness (high, low), (2) whether or not the recipient conveyed the discloser's self-disclosure to a third party (yes, no), and (3) the intimacy of self-disclosure by the discloser (high, low) were assessed in terms of (a) change in self-disclosure by the discloser and (b) change in liking by the discloser. Results confirmed the hypotheses that: (1) the degree of openness of the recipient and the conveyance of self-disclosure by the recipient would interact with change in the discloser's liking of and self-disclosure to the recipient, and, (2) the degree of openness of the recipient and the intimacy of self-disclosure would interact with change in the discloser's liking of and self-disclosure to the recipient. Moreover, females were found to be more sensitive than males to conveyance by the recipient.

[Epidural anesthesia with high dose fentanyl for abdominal surgery].

An epidural catheter was inserted at T9-L2 interspace and 10 micrograms.kg-1 fentanyl with (E+) or without (E-) epinephrine 1:100,000 was given for 82 elective abdominal surgeries. N2O 66%, enflurane and muscle relaxant were used as needed. The onset and the duration of the action were estimated to be approximately 15 minutes and 4 hours, respectively. Anesthesia was maintained with enflurane below 0.4% (0.22 +/- 0.09%) in 70 patients (85.4%). E+ group needed significantly lower concentration of enflurane than E- group. There was no severe hemodynamic change during the operation. Systolic pressure, diastolic pressure and heart rate during the operation were 115.2 +/- 16.0 mmHg, 69.4 +/- 10.8 mmHg and 74.2 +/- 11.4 min-1, respectively, each of which was about 18% less than the values on arrival in the operating room. Sixty-one patients (82.5%) woke rapidly. Almost all patients felt well and had no pain during the recovery period. Naloxone 0.05-4 mg was administered intravenously in 21 patients (31.7%) whose respiratory rate was below 10 min-1. The patients with shorter operation time (shorter than 2.5 hours) needed more naloxone. Troubles of respiratory depression did not occur in the recovery room and in the ward in both naloxone and non-naloxone groups. This anesthesia method which induces mild depression of blood pressure and heart rate may be indicated for patients with ischemic heart disease or with poor cardiac function, but has no advantages in patients with poor respiratory function who need early extubation after a short operation.

Effects of sevoflurane on myocardial metabolism during postischemic reperfusion in the rat.

In experiments on isolated rat heart lung preparation, the effects of sevoflurane on myocardial metabolism during postischemic reperfusion were evaluated with intramyocardial high energy phosphates, lactate and glycogen. Hearts were perfused for 10 min initially and made globally ischemic for 8 min. Afterwards, they were reperfused for 12 min. Sevoflurane was administered from 5 min after the start of perfusion to the end of reperfusion. There was no significant difference in myocardial lactate levels between the sevoflurane (S) and control groups. However, the myocardial ATP level in Group S was significantly higher than that in control (17.45 +/- 1.51 vs 15.50 +/- 0.87 : P < 0.01). The administration of sevoflurane to the isolated rat heart during pre- and post-ischemia enhanced metabolic recovery in the postischemic state.

[The relation of person's motives for self-disclosure and the congruency of personality judgment by discloser and recipient].

This study examined the relationship of a person's motives for self-disclosure and degree of self-disclosure to the congruency of personality judgment by the discloser and recipient. The subjects used were 76 pairs of close friends, members in each pair being of the same sex. Personality judgments of disclosers were executed by the disclosers themselves as well as the recipients; and disclosers were also asked to fill out (SMI) and (JSDQ). Results demonstrated that (JSDQ) had no relation to the congruency of personality judgments, but the motives had. In particular, persons who were apt to disclose based on mostly the emotional motive had the minimum congruencies on their personality judgment by disclosers and recipients.

[A case report of hypoxia in a child after renal transplantation].

A 4 year-old boy underwent renal transplantation and the kidney was given by his mother. Anesthesia was induced slowly with halothane and nitrous oxide in oxygen. The trachea was intubated with pancuronium and the anesthesia was switched to neurolept anesthesia. Blood, plasmanate and crystalloid were infused in order to increase the circulatory blood volume measuring the CVP before the kidney was transplanted because it was so large for the child that it could not be perfused adequately. The spontaneous breathing and soon the recovery followed immediately after the end of operation. However, the patient became hypoxic on the next day probably due to the aspiration of gastric juice. He recovered from hypoxia after 3 days since the aspiration pneumonia was not severe. When the adult kidney is transplanted into a very small child, gastric juice can easily be regurgitated because of the high intraabdominal pressure. Therefore, we must be careful when a child undergoes renal transplantation.

Positive inotropic and chronotropic effects of pancuronium and vecuronium in the canine blood-pefused papillary muscle and sinoatrial node preparations.

The positive inotropic and chronotropic potencies of pancuronium (Pc) and vecuronium (Vc) were compared with developed tension of the isolated papillary muscle (DT) and sinoatrial rate of the sinoatrial node preparation (SAR). Both Pc and Vc caused dose-dependent positive inotropic and chronotropic effects. Pc showed much more potent effect than Vc on DT (0.1, 0.3 mg; P << 0.05) or SAR (0.03, 0.1, 0.3 mg; P << 0.05). DT and SAR were increased by Pc and Vc, and the increase in DT was inhibited by propranolol or tetrodotoxin. These results suggest that the cardiac effects of Pc or Vc may be mediated by release of norepinephrine from the synpathetic nerve endings.

[Bilateral generalized convulsion of split brain cats caused by the amygdaloid kindling].

Previous studies have shown that the corpus callosum is involved in the propagation of epileptic discharge between two hemispheric areas of the brain. It is generally admitted that integrity of the corpus callosum is necessary for the bilateral clinical seizure manifestation. After investigating the role of the forebrain commissures in the development of the kindled amygdaloid seizure in cats, some authors insist that following the bisection of the corpus callosum, ordinary bilateral amygdaloid kindling convulsion becomes into hemiconvulsion and its EEG becomes also unequal; the kindling side showed clear seizure pattern, the other side remained normal. In our former experiment, we intended to compare the half-brain loss hemispherectomized cats with the intact ones on the time latency for the completion of the amygdaloid kindling effect as well as on the developmental change of the clinical seizure manifestation. The time latency of the operated cats was significantly shorter than that observed in the control cats and all the examined cats always showed typically bilateral symmetrical convulsion from the first kindling. Our results are contradictory to those of the split brain experiments whose cat's brain is only locally bisected. We reviewed the related papers and knew that not a few authors omitted to mention the operation technique and did not give the histological evidence of cutting. So we tried to contrive the best split brain cutting after studying previous authors' operation techniques. Ten cats weighing from 2.7 to 3.3 kg aged above 12 months were used. Flagellation of Salmonella typhimurium treated with nalidixic acid.

Filamentous cells of Salmonella typhimurium, obtained after treatment with nalidixic acid in the exponential phase of growth, elongated up to 10 micron, corresponding to 4 unit cell lengths, per nucleoid. During elongation, division of nucleoids and septum formation did not occur, but de novo formation of flagella continued. Most of the filamentous cells were motile, and flagella were evenly distributed on their surface.
