Matlystatins, new inhibitors of typeIV collagenases from Actinomadura atramentaria. I. Taxonomy, fermentation, isolation, and physico-chemical properties of matlystatin-group compounds.

During the course of a screening for inhibitors of typeIV collagenases, new metabolites, designated matlystatins, have been isolated from an actinomycete strain, which was identified as a strain of Actinomadura atramentaria. Matlystatins were composed of five congeners, which were separated and purified by n-butanol extraction and chromatography.

Matlystatins, new inhibitors of typeIV collagenases from Actinomadura atramentaria. II. Biological activities.

Matlystatin A, the main component of matlystatins, inhibits 92 kDa and 72 kDa typeIV collagenases with IC50 values of 0.3 microM and 0.56 microM, respectively, while 7- to 11-fold greater concentrations are required to inhibit thermolysin and aminopeptidase M. The inhibition is reversible and competitive with respect to gelatin. It inhibits the invasion of basement membrane Matrigel by human fibrosarcoma HT1080 dose-dependently with an IC50 value of 21.6 microM.

[Rehabilitation in rheumatoid arthritis].

In the patient with rheumatoid arthritis, muscle weakness and joint contracture and deformity develop, despite the most carefully directed programs of medical care. At the acute phase of this disease, the immobilization or stabilization of arthritic joints is effective for the improvement of inflammation without risk of contracture. At the chronic phase, the patients should be carefully instructed as to the importance of the joint mobility exercise and muscle strengthening exercise. Finally, we must consider that patients with rheumatoid arthritis suffer not only from painful joints but also from psychological and socio-economical damage.

[Causes of death in patients with rheumatoid arthritis--analysis of 117 cases for 13 years].

One hundred seventeen deaths of RA patients (30 males and 87 females) at National Sagamihara Hospital for 13 years (1975-1988) were analysed. The average duration of disease were 10.5 years in male patients and 17.7 years in female. The average life span of the patients with RA, revealing 65.8 years in male and 63.7 years in female, were much shorter than of general population. The causes of all deaths were investigated by ourselves and/or autopsy. The autopsy was performed in 56.6%. The most common causes of death in RA patients were infectious diseases (20.5%), respiratory diseases (16%, mainly interstitial pneumonia and chronic obstructive lung diseases), and gastrointestinal diseases (14.7% chiefly perforation or bleeding of peptic ulcer). The distribution of causes of death in RA patients was quite different from in general population. The gastrointestinal disease decreased from 20% in the early half (1975-1981) to 12.6% in the latter half (1982-1987). It seems likely that H2-receptor antagonist played a major role for preventing the death by perforation or bleeding of ulcer, because the drug has been used since 1982 in Japan. Renal insufficiency including amyloidosis increased markedly in the latter half (14.9%) compared with in the early half (6.7%). Frequency of infectious diseases, respiratory diseases, and basilar impression remain unchanged in all course. Although our study are case-analysis in only one institute and further study will be necessary, the accumulation of the data investigated by rheumatologist will be helpful to grasp correct cause of death in patients with RA.

Adjuvant for IgE antibody and islet-activating protein in Bordetella pertussis.

A new purified protein, with a molecular weight of 77,000, isolated from the supernatant of Bordetella pertussis culture fluid and called islet-activating protein (IAP), had adjuvant activity for IgE antibody in addition to leukocytosis-promoting, histamine-sensitizing and islet-activating activities. None of the three subunits (F1, F2 and F3) was biologically active by itself. All of the four activities were recovered in the complexes of subunits F1.3 and F2.3. These results suggest that the activities are brought about by a common factor inherent in B. pertussis.

IgE levels in nude mice.

IgE levels in nude mice were estimated by the one-step single radial radiodiffusion method antisera prepared by immunization of guinea pigs with an IgE-rich fraction obtained from sera of normal mice infected with Nippostrongylus brasiliensis and immunized with DNP-ovalbumin in alum gel. 3 out of 8 nude mice had IgE levels significantly higher than those of normal mice.

Effect of levamisole on the auto-antibody formation in nude mice.

The effects of levamisole on the formation of antibody to double-stranded DNA (ds-DNA), an auto-antibody, and to some thymus-dependent antigens were studied in nude (nu/nu) mice. The antibody titre to ds-DNA in nu/nu mice treated with levamisole was lower than that in control animals. The number of Thy-1-antigen-positive (Thy-1+) cells in the spleen from nu/nu mice treated with levamisole was higher than that from non-treated animals. It is suggested that the Thy-1+ cells were induced by levamisole in nu/nu mice and these cells suppressed the formation of antibody to ds-DNA, whereas the formation of antibody to a DNP-conjugate (OVA) and SRBC, thymus-dependent antigens, was not evoked in nu/nu mice after the treatment with levamisole.

Antibody formation to double-stranded DNA in nu/nu and nu/+ mice.

Antibody formation to double-stranded (ds-) DNA in nude mice (nu/nu) and normal littermates (nu/+) was studied using a Millipore filter method. The mean antibody titre to ds-DNA in the sera of nu/nu mice was generally higher than that in nu/+ mice raised in the same environment. However, in both genotypes, anti-ds-DNA titres varied according to the environmental condition in which the animals were raised. Mice maintained germ-free showed no significant antibody formation to ds-DNA. Nu/+ thymus and spleen cells passively transferred to nu/nu mice reduced the antibody titre to ds-DNA in the recipients. Immunochemical studies indicated that the antibodies to ds-DNA were IgM. It is suggested that T cell-mediated regulatory mechanisms control antibody formation to ds-DNA and that some infections trigger antibody formation to ds-DNA.

Preferential enhancement of IgE antibody formation by Bordetella pertussis.

Preferential enhancement of IgE antibody response was observed in BALF/c mice by the administration of Bordetella pertussis with antigen (DNP-Salmonella). Correlation between B cell mitogenic activity and adjuvant action among B. pertussis, Salmonella, lipopolysaccharide of Escherichia coli and Ficoll was examined but was not found. Thymus-derived cells seemed necessary to develop adjuvant action of B. pertussis since antibody response in athymic nude mice was not influenced by B. pertussis. Helper function of adoptively transfered spleen cells was enhanced by immunization of the donor mice with carrier antigen in the presence of B. pertussis. The magnitude of enhancement was greatest in IgE class. The results indicated that preferential enhancement of IgE antibody formation by B. pertussis is mediated by the augmentation of carrier-specific helper function.
