Partial purification of Simian virus 40 large T antigen by immunoaffinity chromatography and its detection by enzyme-linked immunosorbent assay.

Simian virus 40 (SV40) large T antigen was partially purified from small amounts of SV40-infected and SV40-transformed cells by immunoaffinity chromatography with high recovery. T antigen, in both crude and partially purified states, was detected rapidly by a sensitive and quantitative enzyme-linked immunosorbent assay (ELISA). Stability of the partially purified T antigen was found to increase by addition of 0.01% bovine serum albumin (BSA).

Differential sensitivity to aphidicolin of replicative DNA synthesis and ultraviolet-induced unscheduled DNA synthesis in vivo in mammalian cells.

In vivo in mammalian cells, ultraviolet-induced unscheduled DNA synthesis was less sensitive to aphidicolin than was replicative DNA synthesis. Replicative DNA synthesis in HeLa, HEp-2, WI-38 VA-13 and CV-1 cells was inhibited more than 97% by aphidicolin at 10 micrograms/ml, whereas aphidicolin inhibition of DNA synthesis in ultraviolet-irradiated cells varied between 30% and 90% depending on cell types and assay conditions. Aphidicolin inhibition of unscheduled DNA synthesis (UDS) in HeLa cells increased gradually with increasing aphidicolin concentration and reached approximately 90% at 100 micrograms/ml aphidicolin. A significant fraction of UDS in ultraviolet-irradiated HEp-2 cells was resistant to aphidicolin even at 300 micrograms/ml. Considered along with related information reported previously, the present results suggest that both aphidicolin-sensitive and insensitive DNA polymerases, DNA polymerase alpha and a non-alpha DNA polymerase (possibly DNA polymerase beta), are involved in in situ UDS in these ultraviolet-irradiated cells. Comparison of staphylococcal nuclease sensitivity between DNAs repaired in the presence and in the absence of aphidicolin in HEp-2 cells suggested that the involvement of DNA polymerase alpha in UDS favored DNA synthesis in the intranucleosomal region.

Effect of coenzyme Q10 on the survival time and lipid peroxidation of adriamycin (doxorubicin) treated mice.

The effect of coenzyme Q10 (Co Q10) was examined on the survival time and lipid peroxidation of adriamycin (ADM)-treated ICR mice. Co Q10 showed a protective effect against a subacute toxicity in mice induced by two intraperitoneal administrations of ADM (15 mg/kg). The group treated orally with 10 mg/kg of Co Q10 showed the longest survival time of all the groups studied (16.81 +/- 10.29 days, mean +/- S.D.) and a significantly longer survival time (p less than 0.001) than the ADM-alone group (7.48 +/- 1.99 days). The inhibitory effect of Co Q10 on the plasma and tissue lipid peroxidation levels did not correlate with the effect of prolonging the survival time of mice. Co Q10 tended to inhibit rises in plasma and liver lipid peroxidation levels induced by ADM administration, but there was no statistically significant difference between treatments. There was a statistically significant different inhibitory effect in the kidney lipid peroxidation levels, but was not in those of the heart.

Capillary blood flow in the amelanotic melanoma of the hamster after isovolemic hemodilution.

In malignant tumors, spontaneous arteriolar vasomotion disappeared already during earliest tumor growth (tumor weight less than 200 mg) suggesting that arterioles supplying a malignant tumor with blood are maximally dilated from the very beginning of tumor growth. As a result, the viscosity of blood becomes a limiting factor of tissue perfusion. To evaluate the effects of isovolemic hemodilution in a malignant tumor, a transparent chamber model was used in conjunction with a subcutaneously growing amelanotic melanoma of the hamster. Quantitative intravital microscopy (video image analysis) and a platinum multiwire electrode were utilized to study the tumor microcirculation. Isovolemic hemodilution was performed by exchange of blood vs. Dextran 60 to lower the systemic hematocrit to 30% in 21 animals. Capillary flow increased significantly and improved tissue pO2 on the tumor's surface without any change in capillary density. Since tumor growth was accelerated after hemodilution, these data suggest to combine with hemodilution other treatment modalities to improve blood-mediated tumor therapy.

DNA replication of simian virus 40 chromatin in digitonin-treated and saponin-treated permeable cells.

Permeable cell systems have been developed by treatment of simian virus 40 (SV40)-infected cells with digitonin or saponin for studying in vitro DNA replication of chromatin. SV40 DNA replicated efficiently in these systems in the presence of the four deoxyribonucleoside triphosphates, ATP, Mg2+, and the proper ionic environment. Analysis of the time course of the replicated SV40 DNA I, II and replicating intermediates by gel electrophoresis and autoradiography indicated that SV40 DNA replicated completely in the form of nucleoprotein complexes (SV40 chromatin).

A newer hyperthermic therapy with a bacterial immunopotentiator (OK432) for chemotherapy-resistant ovarian cancer.

Systemic hyperthermic therapy was applied to two patients with chemotherapy-resistant malignancy. To obtain hyperthermia, a pyretic effect of a bacterial immunostimulant (OK432) was utilized. In a case of pseudomyxoma peritonei, her distended abdominal girth showed a remarkable decrease during this therapy. In another case, an endodermal sinus tumor, the serum alpha fetoprotein (AFP) level of the patient was lowered temporally and leukopenia induced by prior cytotoxic chemotherapies was significantly recovered. Because of the anticancer effect of OK432 itself, an accurate evaluation of this is difficult. At a moderate estimate, however, this method seems to be feasible as a useful adjuvant therapy in the management of cancer.

Prognostic significance of glioblastoma element in ovarian immature teratoma.

A case of ovarian immature teratoma containing glioblastoma element with increased serum alpha fetoprotein (AFP) is demonstrated. In spite of a series of postoperative chemotherapeutic programs reported to be effective against gonadal germ cell tumors, the patient's condition continued to worsen even though the level of AFP had decreased to the normal range after the first line therapy with vincristine, actinomycin D and cyclophosphamide. The serum AFP titer in this case was unreliable as a tumor marker. Histologic examination of recurrent and metastatic tumors obtained at autopsy showed that other teratomatous tissues were widely infiltrated by a proliferated glioblastoma element. When considering the prognosis of ovarian immature teratoma, it is important to realize the aggressive potential of glioblastoma multiforme as a component.

[Amenorrhea due to weight loss].

The cessation of the normal menstrual cycle caused by rapid loss of weight was defined as amenorrhea due to weight loss. All patients developed secondary amenorrhea and 80% of 243 patients were young, unmarried women. More than half of the patients had consciously restricted food intake for cosmetic reasons. LH-RH test revealed that, at onset, the basal levels of LH were low and the LH responses were impaired. The basal and the stimulated levels of FSH were comparable to normal. As the amenorrhea improved, the basal and the stimulated levels of LH rose to the normal range while the FSH responses became greater than normal. There was no significant difference between the rate of weight loss and the responsiveness to LH-RH. The rate of induction of ovulation with clomiphene was 95% of 40 patients with 1st grade amenorrhea, while only 3.9% of 54 patients with 2nd grade amenorrhea ovulated following treatment clomiphene. 80 percent of 19 patients with 2nd grade amenorrhea ovulated following gonadotropin therapy. Within a year after LH-RH stimulation therapy, a resumption of the normal ovulatory cycle or a return to 1st grade amenorrhea was observed in 60%.

[Evaluation of cancer chemotherapy in primary ovarian cancer in rats--observation of histological effect by light and electron microscopy].

In order to evaluate the effects of ifosfamide as an anticancer agent, ifosfamide was administered to rats with ovarian cancer induced by DMBA (7,12-dimethylbenz (a) anthracene). Four to five weeks after the chemotherapeutic treatment 16 rats were sacrificed and then tumor sizes were compared before and after the treatment, and in addition morphologic changes were investigated by light and transmission electron microscopy. Of 16 rats, the reductions in tumor size were recognized in six cases. A histologic effect was observed in both the tumor decreased group and the increased group. One of the latter group showed excellent histological chemotherapeutic effect. On light microscopic observation, the histologic effects seen were reduction in the amount of neoplastic tissues and proliferation of connective tissues. On transmission electron microscopic observation, destruction of tumor cells and a reduction in the amount of ribosome and chromatin were recognized as the effects of the anticancer drug. Multinuclear giant cells, presumably derived from the mesenchym, were found among the connective tissues in one case showing a remarkable histologic effect.

Fine localization of serine:pyruvate aminotransferase in rat hepatocytes revealed by a post-embedding immunocytochemical technique.

Immunocytochemical localization of serine:pyruvate aminotransferase (SPT) in rat hepatocytes was studied using a protein A-gold technique. Rat liver was fixed by perfusion. Vibratome sections (100 micron thick) of the liver were embedded in Epon or Lowicryl K4M. Ultrathin sections were incubated with antiSPT, followed by protein A-gold complex. Gold particles representing the antigenic sites for SPT were seen in three subcellular compartments, peroxisomes, mitochondria, and cytoplasm. In the control experiments the specificity of the immunolabelling was confirmed. Quantitative analysis of the labelling density showed that main subcellular compartments containing SPT are mitochondria and peroxisomes. In addition, the gold particles distributing in the cytoplasm were 16%-29% of the total labelling. The result indicated that the cytoplasm also contains SPT with a low density.

[Studies on tobramycin by the intravenous drip infusion method].

Fundamental and clinical efficacy evaluation has been made in the study on the therapy with intravenous drip infusion of tobramycin (TOB) which is one of the aminoglycoside antibiotics and which has been recently approved of intravenous administration. The fundamental evaluations were made with intravenous drip infusion of 60 mg of TOB for 1 hour and that of 120 mg of TOB for 2 hours to 4 healthy adult volunteers on crossover method. Follow-up determinations were made for comparison on its concentrations in blood and urine samples which were collected both at intervals thereafter, using 2 assay methods of the bioassay and the EMIT methods. The maximum blood level of TOB based on the intravenous drip infusion of 60 mg for 1 hour was 5.9 micrograms/ml and 4.6 micrograms/ml by the bioassay and the EMIT methods at the end of intravenous drip infusion, respectively, and 0.3 micrograms/ml and 0.4 micrograms/ml, respectively, at 6 hours after the drip infusion. The T 1/2 value was 1.81 hours with bioassay and 2.41 hours with the EMIT method. The maximum blood level of TOB through drip infusion method of 120 mg of TOB for 2 hours was 7.5 micrograms/ml by bioassay and 5.8 micrograms/ml by EMIT method. The T 1/2 value was 1.87 hours and 1.99 hours, respectively. The recovery rate of TOB from urine until 24 hours after the administration was 83.0% and 94.4% with the doses of 60 mg and 120 mg, respectively, which were determined only with the bioassay. The correlation coefficient between the agar well method and the EMIT method was 0.963 and 0.972 in the groups of intravenous drip infusion of 60 mg for 1 hour and 120 mg for 2 hours, respectively, showing proximity to each other. TOB was kept administered with a daily dose of 120 mg at a time to 11 cases and at 2 times to 1 case with chronic complicated infections of urinary tracts (comprising 9 cases with chronic complicated cystitis, 2 with chronic complicated pyelonephritis, and 1 with acute epididymitis). Of the 12 patients, 8 could meet the criteria for evaluation of drug efficacy and 6 of them proved to be effective, while 2 were ineffective. The efficacy rate was so high as 75%. The rate of disappearance of bacteria was 80.0%, which was very high. No side effect was specifically found out, except 1 case showing slight rise in S-GOT.
