Echographic findings and histological feature of the thyroid: a reverse relationship between the level of echo-amplitude and lymphocytic infiltration.

In 43 patients with Graves' disease, 5 patients with painless thyroiditis and 30 patients with Hashimoto-thyroiditis ultrasonographical observations and histological examinations by needle biopsy of the thyroid were carried out simultaneously. In all cases the level of echo-amplitude was well correlated with the rate of lymphocytic infiltrations and fibrosis. In cases which exhibited marked lymphocytic infiltrations in the thyroidal biopsy specimen, no apparent echoes or very low amplitude echoes were observed in the whole thyroid and in cases in which replacement with lymphocytic infiltration was observed in almost a half part of the thyroid, several sonolucent regions were observed in the thyroid and in cases in which lymphocytic infiltration or fibrosis was observed sporadically, low-amplitude and uniform echoes were observed in the whole or several regions of the thyroid. In cases with no lymphocytic infiltration in the histological specimen, diffuse high-amplitude and uniform echoes were observed throughout the whole lobe of the thyroid. In patients with painless thyroiditis, the amplitude of echo was low when the level of lymphocytic infiltration was high and the echo-amplitude showed a tendency to increase along with the decrease in the rate of lymphocytic infiltration. From these observations it is concluded that echo-amplitude is well correlated with lymphocytic infiltration and fibrosis in patients with Hashimoto-thyroiditis, Hashitoxicosis and painless thyroiditis.

Effects of insulin, amino acids and fasting on myofibrillar protein degradation in perfused hindquarters of rats.

The rate of 3-methylhistidine (3-MH) release from rat perfused hindquarters was measured to investigate the effects of insulin, amino acids and fasting on myofibrillar protein degradation. Since release of 3-MH into the perfusate increased linearly and the pool of free 3-MH in the perfused muscle did not change significantly during 2 h of perfusion, it was concluded that 3-MH release reflected the rate of myofibrillar protein degradation. Tyrosine release in the presence of cycloheximide represented the degradation rate of total muscle protein. Insulin suppressed the net release of tyrosine in normal rats, but did not affect the rate of release of 3-MH and tyrosine in the presence of cycloheximide. 3-MH release was not influenced by perfusate amino acid concentrations at zero to 5 times the normal plasma levels. When rats were fasted for one and two days, 3-MH release increased 1.7 and 2.6 times, respectively, compared with the fed rats, which showed that the rate of degradation of myofibrillar protein in skeletal muscle rose just after the beginning of fasting.

The effect of gizzerosine, a recently discovered compound in overheated fish meal, on the gastric acid secretion in chicken.

The effect of gizzerosine [2-amino-9-(4-imidazolyl)-7-azanonanoic acid], which causes gizzard erosion in chicks, on gastric acid secretion was studied in young chicks. Three-day-old broiler chicks given a diet containing synthetic DL-gizzerosine showed severe symptoms of gizzard erosion after 7 days. The pH of the gastric and duodenal contents of these chicks was lower than that of the control chicks, demonstrating that acid secretion was enhanced in gizzerosine-fed chicks. The pH of gastric contents was lower and the amount of total gastric acid was higher in 1-day fasted chicks injected with gizzerosine intravenously than in those injected with physiological saline solution. The intramuscular injection of cimetidine, H2-receptor antagonist of histamine, blocked the stimulation of acid secretion by gizzerosine and by histamine. Cimetidine added in feed also prevented gizzard erosion, which was induced by feeding overheated mackerel meal. The results showed that gizzard erosion developed after oral or intravenous administration of gizzerosine is caused at least in part by the enhanced gastric acid secretion. Furthermore, it was suggested that gizzerosine acts on H2-receptor of histamine and stimulates the gastric acid secretion in broiler chicks.

Bestatin, a microbial aminopeptidase inhibitor, inhibits DNA synthesis induced by insulin or epidermal growth factor in primary cultured rat hepatocytes.

Bestatin, a microbial aminopeptidase inhibitor, inhibited insulin- or epidermal growth factor-induced DNA synthesis and cell proliferation in primary cultured hepatocytes of rats. The aminopeptidase inhibitor also affected the growth of FM3A or LOBN cells of mice, when it is included in the culture media at the concentration of 0.5 mg/ml. These results suggest the important role of the bestatin-sensitive aminopeptidase(s) in the process of DNA synthesis and proliferation of animal cells.

Circulating blood group-related antigen(s) in cancer patients detected by the monoclonal antibodies produced against hepatocellular carcinoma cell line.

Two murine monoclonal antibodies (MAbs) S1 and S3 produced against a human hepatoma cell line were found to recognize both H (type II) antigen and difucosyl type II structure (Y-antigen). Both MAbs reacted with hepatoma tissues obtained from surgical resection. A double determinant enzyme immunoassay (DDEI) employing these MAbs successfully detected the corresponding antigen(s) in the supernatant which is either released or shed from the immunizing hepatoma cell line. DDEI also revealed that these antibodies reacted with the corresponding antigen(s) in the sera from cancer patients. Among the sera from O blood group donors, individuals with primary hepatoma showed markedly higher levels of antigen concentration than the normal control group. Several cases with gall bladder, lung and pancreas carcinomas, which are of O blood type, also had significantly higher levels of antigen. One case with ovarian cancer of B blood type showed higher antigen concentration than normal O blood group donors suggesting a change in the carbohydrate structure of the corresponding antigen(s) in the serum. These data suggest that the release or the shedding of the antigen(s) from the cells may increase due to the malignant transformation, resulting in higher amounts of the antigen(s) in the serum of certain cancer patients.

A recessive mutation (ter) causing germ cell deficiency and a high incidence of congenital testicular teratomas in 129/Sv-ter mice.

The genetic basis of a germ cell deficiency, accompanied by a high incidence of testicular teratomas, was studied in strain 129/Sv-ter mice (formerly designated "129/terSv"). Germ cell deficiency became more severe with advancing age in males, and they were sterile whether or not they had bilateral teratomas. Germ cell-deficient testes were smaller than normal except when the testes had teratomas. In females the ovaries were smaller than normal, but the germ cell deficiency did not progress and most were fertile. The germ cell deficiency resulted from the homozygous state of a recessive mutant gene designated "teratoma (ter)." Matings between females with small ovaries and homozygous normal males produced no germ cell-deficient offspring. When F1 offspring with normal gonads were mated together, germ cell-deficient F2 animals appeared at a frequency close to 1 in 4. When females with small ovaries (ter/ter) were mated with heterozygous males (ter/+), half of the offspring were germ cell deficient. It was concluded that the genetic factor is a single recessive gene. The incidence of teratomas in +/+ strain 129/Sv-ter males was 1.4% (3/216), and all teratomas were unilateral. Seventeen percent (20/117) of heterozygous males had teratomas, of which 18 were unilateral and 2 were bilateral. Ninety-four percent (167/178) of homozygous ter/ter males had teratomas, of which 75% were bilateral. Introduction of the mutant gene ter onto the C57BL/6 genetic background resulted in germ cell deficiency in homozygotes, but it reduced considerably the teratoma incidence in ter/ter males.

[Adrenal myelolipoma: a case report].

A 51-year old obese man had been followed up for diabetes mellitus and hypertension. Hepatomegaly was noted on routine examination without any complaints in 1981. Through further investigation of the liver, a large hyperechoic mass lesion was shown between right kidney and the liver on ultrasonogram. On admission, diabetes was controlled by diet alone and blood pressure was 160/90 mmHg. without medication. There were no abnormal findings in laboratory data including hormone assays. The mass showed fat density on CT scan and hypovascularity on angiogram. The mass was diagnosed as benign non-functioning right adrenal tumor, most likely myelolipoma. By operation, the tumor was removed through long subcostal incision. The tumor was well defined and encapsulated, 14 X 8 X 8cm. in size and 480g. in weight. Microscopically the tumor was typical myelolipoma with a mixture of hematopoietic and adipose tissue. Postoperative course was uneventful and the patient discharged on 17-th postoperative day. Myelolipoma of the adrenal gland is a rare non-functioning benign tumor consisting of fat cell and bone marrow element. Only 30 resected cases of adrenal myelolipoma have been reported in the world including 7 cases in our country.

Suppression of human melanoma growth in nude mice injected with anti high-molecular-weight melanoma-associated antigen monoclonal antibody 225.28S conjugated to purothionin.

The cytotoxic agent purothionin purified from barley was covalently conjugated to the anti-human high-molecular-weight melanoma-associated antigen (HMW-MAA) monoclonal antibody (MoAb) 225.28S by utilizing water-soluble carbodiimide. Injection of the conjugate (0.1 mg/injection) significantly increased the life span of nude mice injected with ascitic-form human melanoma cells (Colo 38), and caused 40% inhibition on day 25 of the growth of solid-form human melanoma cells in nude mice. The effect is specific and is markedly influenced by the site of growth of tumors and by the schedule of administration of the conjugate.

Liquid-chromatographic assay of diazepam and its major metabolites in serum, and application to pharmacokinetic study of high doses of diazepam in schizophrenics.

In this rapid, sensitive method for simultaneously determining diazepam and its biologically active metabolites--nordiazepam, oxazepam, and temazepam--in serum, 250 to 1000 microL of serum is extracted with ether and the extracted compounds are quantified by "high-performance" liquid chromatography on a Shimpack FLC-C8 microparticulate column. Absorbance of the effluent is monitored at 254 nm. The limit of detection in serum is about 10 micrograms/L for each drug. Analytical recovery of each drug added to the serum varied from 91 to 102% for oxazepam and temazepam, from 82 to 99% for nordiazepam and diazepam. Within-day and between-day CVs ranged from 2 to 5% for oxazepam and temazepam, and from 2 to 7% for nordiazepam and diazepam. We also report results on using this assay in a pharmacokinetic study of high doses of diazepam used to treat chronic schizophrenic patients.
