[Clinical trial of fosfomycin for Campylobacter enteritis].

Forty-three children and 4 adults with Campylobacter enteritis were studied in the treatment of fosfomycin (FOM). FOM was administered per orally in doses ranging from 50 to 100 mg/kg/day for children and 3 g/day for adults for 5 days. Main symptoms such as diarrhea and fever were disappeared within 2 days on the average. Campylobacter jejuni in stool specimen disappeared within a week in 95% of these patients. The duration of main symptoms and the period of positive stool culture were evidently shortened in FOM-treated group compared with non-treated group. All of the isolated strains were sensitive to FOM by mono-concentration disk method. MIC50 of these strains remained between 1.56 and 3.13 micrograms/ml. None of these MIC was beyond 12.5 micrograms/ml.

[Kinetics of protein synthesis in the human granulosa lutein cell during pregnancy as revealed by electron microscopic autoradiography of 3H-leucine].

Kinetics of protein synthesis in the human granulosa lutein cell during pregnancy were investigated using the electron microscopic autoradiography of 3H-leucine. The corpus luteum of early pregnancy was chased at 10 and 30 min, 1, 3, 6, 12, 24 and 48 h after pulse labelling (10 min) with 100 microCi of 3H-leucine. Although the whole silver grains were very few at 10 min postpulse, silver grains were localized mainly over the rough endoplasmic reticulum (r-ER, 34.3%) and there were no silver grains over the smooth endoplasmic reticulum (s-ER). At 30 min postpulse, the ratio of the grain number to r-ER is decreased (18.7%) and silver grains begin to appear over the s-ER (5.4%). The total number of silver grains over the cell increases with the lapse of time, the ratio of the grain number over the r-ER slightly increases and then decreases afterwards, while that over the s-ER continues to increase gradually. From these facts, it is felt that some protein synthesized on the r-ER might be transported to the s-ER in the human granulosa lutein cell during pregnancy showing steroidogenic activity.

[Histogenesis on the ovarian Leydig cell tumor--light microscopic dry-mounting radioautography for 3H-cholesterol and electron microscopic cytochemistry for 3 beta-hydroxysteroid dehydrogenase activity].

A case of a virilizing ovarian Leydig cell tumor in a 38 year-old woman with a markedly elevated plasma testosterone level was investigated using light microscopic dry-mounting radioautography and electron microscopic cytochemistry. Following total abdominal hysterectomy and bilateral salpingo-oophorectomy, the plasma testosterone level fell abruptly. Light microscopic dry-mounting radioautography for 3H-cholesterol showed silver grains mainly localized over the cytoplasm of the tumor cells. In the Leydig cell tumor there were three different cell types: Fibroblast-like cells, Leydig cells (steroid-secreting cells) and transitional cells (partially or incompletely differentiated Leydig cells). Reaction products for 3 beta-hydroxysteroid dehydrogenase activity were localized on tubular or lamellar cristae and inner membranes of the mitochondria, and on the membranes of smooth endoplasmic reticulum in the transitional cell as well as in the Leydig cell. From these facts, it is suggested that the Leydig cell tumor is derived from the fibroblast-like cell and the Leydig cell, already has a steroidogenic activity in secreting testosterone.

Electron microscopy of cytodifferentiation and its subcellular steroidogenic sites in the theca cell of the human ovary.

The cytodifferentiation and subcellular steroidogenic sites in the theca cell of the human ovary during the follicular phase were investigated using the electron microscopic cytochemistry. Only fibroblast-like cells were seen around or near the primordial follicle. In the theca interna of the secondary and Graafian follicle however there were three different cell types: fibroblast-like cells, theca gland cells (steroid-secreting cells) and transitional cells (partially or incompletely differentiated theca cells). On the other hand the theca externa of these follicles consisted mainly of fibroblast-like cells. The hallmarks of the cytodifferentiation of the theca cell were: 1) the appearance of lipid droplets, 2) a structural change of the mitochondrial cristae from lamellar to tubular form and 3) the appearance and development of smooth endoplasmic reticulum. Reaction products of 3 beta-hydroxysteroid ferricyanide reductase, indicating 3 beta-hydroxysteroid dehydrogenase activity, were localized on tubular or lamellar cristae and inner membrane of the mitochondria, and on the membranes of smooth endoplasmic reticulum in the transitional cell as well as in the theca gland cell of the secondary and Graafian follicle. From these data, it is suggested that the transitional cell has a steroid-secreting activity and also plays an important role in follicular development in human reproduction.

In vitro induction of segregational errors of chromosomes by natural cannabinoids in normal human lymphocytes.

It has been shown that segregational errors (SE) of chromosomes can be induced by olivetol and several halogenated inhalation anesthetics. The purpose of this study was to examine the effects of natural cannabinoids - including delta-9-tetrahydrocannabinol (THC), cannabinol (CBN), and cannabidiol (CBD) - on chromosome segregation. Lymphocytes obtained from healthy adult males were incubated with various concentrations of natural cannabinoids for 72 hours. Anaphase preparations were made from these cultures. A statistically significant increase in the incidence of SE of chromosomes was observed in the lymphocytes exposed to THC at a concentration of 3.2 x 10(-6) M, but not to CBN or CBD. A greater incidence of bridge formations, anaphase lags, micronuclei, and unequal segregations in bipolar divisions and multipolar divisions were observed in THC-treated lymphocytes, compared with the controls. However, only anaphase lags and unequal segregations in bipolar divisions reached statistically significant levels. It appears that THC affects the formation of microtubules and spindles and may be considered as a mitotic poison. The value of examining SE as a part of cytogenetic studies on chemical mutagens is emphasized. A description of a classification system of SE developed in our laboratory is also presented. This system can be applied to studies using both normal human lymphocyte cultures and lymphoid cell lines.

Changes in amplitude of the EEG induced by a photic stimulus.

Occipital EEG responses to a single photic flash were studied by using a complex demodulation technique. Individual EEGs were first digitally filtered by making use of the Ormsby band-pass filter. Envelopes of the alpha (8-12 c/sec) and of the low frequency (3-7 c/sec) activities were obtained in terms of the demodulation calculation. Both individual and averaged data of raw, filtered, and demodulated EEGs were utilized for the present study. Two typical kinds of EEG responses were observed. One showed clearly the blocking as well as the enhancement of the alpha activity. The alpha activity first exhibited a reduction in amplitude in the range 50-200 msec after the stimulus (the minimum amplitude at about 150 msec). Then an evocation, rather than merely a recovery, of the alpha was observed in the range 500-800 msec (maximum amplitude at about 650 msec). The other typical example showed the low frequency evoked potential prominently. This response appeared almost locked to the stimulus and showed roughly a bi-phasic pattern in the time range where the alpha blocking took place (30-200 msec). Phases of the evoked low frequency component were almost locked to the stimulus but no distinct phase coherence was observed for the enhanced alpha wavetrains. Detection by demodulation of the phase change of the alpha was critical for a single EEG. It is pointed out that changes of the alpha must be studied on individual EEGs.
