[The study of Chinese herbal medicinal prescription with enzyme inhibitory activity. VII. The study of inchinko-to with adenosine 3',5'-cyclic monophosphate phosphodiesterase].

A Chinese herbal medicinal prescription, Inchinko-to, was studied for the inhibitory activity on adenosine 3',5'-cyclic monophosphate (cAMP) phosphodiesterase (PDE). The inhibitory activity on cAMP PDE of the extract of each constituent from the crude drug and of several compounds isolated from the drug was studied. By changing the combination ratio of two kinds of the crude drugs from 1:1 to 1:5, the inhibitory activity on cAMP PDE was estimated and it was found to be highest at the ratio of 1:3 in Rhubarb and Gardenia fruit, and at 1:4 in Rhubarb and Capillaris buds. These were the same ratio which is now commonly used for Chinese herbal medicinal prescription in Japan. Similar results were obtained in the combination test of the components of the crude drug such as emodin, geniposide, capillarisin and dimethylesculetin. The effects of Inchinko-to and its prescription which lacks one of the constituents of the crude drugs on carbon tetrachloride inducing liver injury in mice were also studied. Among the four prescriptions the antihepatotoxic activities were observed in the prescriptions containing Capillaris buds. Correlation between the administrated content of dimethylesculetin and antihepatotoxic activity was not recognized.

Primitive neuroectodermal tumors of bone and soft tissue. With reference to histologic differentiation in primary or metastatic foci.

Primitive neuroectodermal tumors (PNET) of the bone and soft tissue were reviewed by immunohistochemistry and partly by morphometry, focusing particularly on histologic changes in recurrent or metastatic foci, in order to elucidate their probable histogenetic relationship with Ewing's sarcoma (ES) and its extraskeletal counterpart (EES). Eleven cases of bone tumor (average patient age; 15.1 yr) and 12 cases of soft tissue tumor (average patient age; 22.1 yr) which disclosed unequivocal Homer-Wright rosettes and/or at least foci of ganglion cell differentiation either in a given primary tumor or metastatic (or recurrent) foci were selected from small round cell tumors primarily categorized as ES or EES. Most of the cases for which follow-up biopsy samples were available disclosed prominent Homer-Wright rosettes in the metastases, whereas the primary tumors showed features of ES and lacked rosettes. In only one case, Homer-Wright rosettes were absent in the metastatic tumor. Most cases had been treated by combined intensive chemotherapy and radiotherapy, which might have influenced cell differentiation. Neural markers (neuron-specific enolase, neurofilament protein and others) were positive in most cases. Three cases with otherwise typical histologic features of ES or EES showed minute foci of ganglion cell differentiation, as confirmed by morphometry and neural markers. These results suggest that ES (or EES) and PNET are histogenetically related, but represent different stages of cell differentiation.

Activation of human CD4 T lymphocytes. Interaction of fibronectin with VLA-5 receptor on CD4 cells induces the AP-1 transcription factor.

Fibronectin synergized with anti-CD3 antibody to promote CD4 cell proliferation in a serum-free culture system whereas no proliferation was observed when CD4 cells were cultured with anti-CD3 alone or fibronectin alone. In addition, anti-CD29 (integrin beta 1) as well as anti-VLA-5 (human fibronectin receptor) antibodies blocked this CD4 cell activation in this system. Although anti-CD3 alone or fibronectin alone cannot induce IL-2 message by CD4 cells, the combination of anti-CD3 plus fibronectin induced IL-2 message by CD4 cells. In an analysis of the molecular mechanism by which IL-2 message was generated, we showed that a fibronectin-VLA-5 fibronectin receptor interaction may contribute an independent signal distinct from the CD3 pathway of activation by the induction of an AP-1 transcriptional factor. Thus the VLA-5 fibronectin receptor on CD4 cells can play a complementary role in CD3-TCR-mediated signal transduction through its interaction with fibronectin.

Case report 679. Central low-grade osteosarcoma with foci of dedifferentiation.

An infrequently encountered case of well-differentiated CLOS containing dedifferentiated tumor foci in a 28-year-old Japanese man is reported. The patient died from widespread metastases with a microscopic appearance identical to the anaplastic dedifferentiated lesions of the original tumor. This is, to our knowledge, the first report of a case of CLOS with dedifferentiation at the time of initial surgery. The literature concerning CLOS was reviewed and the diagnostic features of CLOS discussed.

Molecular cloning of transcripts that accumulate during the late G1 phase in cultured mouse cells.

To identify previously undetected genes that might be involved in later stages of the transition from a quiescent state (G0) to the DNA synthetic phase (S) of murine cells, we set out to isolate cDNA clones derived from mRNAs that appear late in G1 phase in serum-stimulated cells. A lambda-cDNA library was prepared using poly(A)+ RNA from chemically transformed Balb/c 3T3 cells (BP/A31) that had been brought to quiescence and subsequently stimulated for 12 h with serum. From the first screening of approximately 21,000 recombinant phage plaques, about 100 clones were isolated that hybridized to a single-stranded cDNA pool derived from stimulated-cell RNA but not to DNAs made from resting-cell RNA. Eventually, six different clones were identified. The mRNAs from five of these genes increased gradually during the G0 to S transition, in contrast to the "immediate-early" rise of c-myc mRNA or the later rise of thymidine kinase mRNA. These six clones were sequenced and compared to the GenBank database. Clones LG-80, LG-2, and LG-69 are highly homologous to beta-actin, lactate dehydrogenase, and alpha-tubulin. Clones LG-5, LG-61, and LG-74 had no significant homologies to known sequences. A subtractive cDNA library was used to isolate two additional clones, Sub-S1 and Sub-S2; these have homologies to enolase and ribosomal protein L32. Additional studies that examine the function and regulation of these newly identified "late response" genes in the pre-DNA synthesis pathway are in progress.

Photocure bonding agent containing phosphoric methacrylate.

Effective photocure phosphoric methacrylate bonding agents which bonded a dental composite to tooth substrates were proposed. Methacryloyloxydecyl phosphoric acid (MDP) or 2-methacryloyloxyethyl phenyl phosphoric acid (Phenyl-P) was dissolved in triethyleneglycol dimethacrylate (TEGDMA) with camphorquinone (CQ) and N-phenylglycine (NPG). The combination of CQ and NPG was a good visible-light initiator for the polymerization. The highest bond strength was 5 MPa to dentin and 10 MPa to enamel when the adhesive contained Phenyl-P in TEGDMA. Dentin and enamel were recommended for demineralization with an aqueous mixture of 0.3 mol/L EDTA diammonium salt and 0.2 mol/L EDTA ammonium iron salt for generation of better bond strength.

Ovarian sex cord tumor with annular tubules. Report of a case.

A 23-year-old woman underwent resection of a tumorous right ovary after long-standing irregular genital bleeding. The tumor measured 50 x 30 mm and had a yellowish-white solid cut surface with scattered small cysts. The light microscopic diagnosis was a sex cord tumor with annular tubules. Ultrastructurally, the tumor cells had Charcot-Boettcher filaments and showed vimentin positivity by immunohistochemistry. A three-dimensional reconstruction study proved that the tumor cell nests had a ovoid outer margin and contained blind-ended ellipsoid hyaline bodies. This tumor is presumed to be a variant of sex cord/stromal tumor showing differentiation predominantly to Sertoli cells.

Inhibition of adenosine 3',5'-cyclic monophosphate phosphodiesterase by flavonoids from licorice roots and 4-arylcoumarins.

Isoliquiritigenin, glabridin, licoarylcoumarin and licoricidin were identified as strong inhibitors of adenosine 3',5'-cyclic monophosphate (cAMP) phosphodiesterase in waste materials which were obtained during the industrial extraction of glycyrrhizin from licorice roots. The structure-activity relationships of 12 flavonoids from licorice roots and 34 4-arylcoumarins were studied. In 4-arylcoumarins, 5,7-dihydroxy derivatives were generally highly inhibitory towards cAMP phosphodiesterase.

[The study of Chinese herbal medicinal prescription with enzyme inhibitory activity. V. The study of hange-shashin-to, kanzo-shashin-to, shokyo-shashin-to with adenosine 3',5'-cyclic monophosphate phosphodiesterase].

Fifty-nine species of extracts of Chinese herbal medicinal prescription were tested for inhibitory activity of adenosine 3',5'-cyclic monophosphate (cAMP) phosphodiesterase (PDE). Kanzo-shashin-to showed the highest activity in these prescriptions. Kanzo-shashin-to, Hange-shashin-to and Shokyo-shashin-to, whose contracting crude drugs were very similar, were especially studied among these prescriptions. Pinellia tuber acted as an ascent component for Scutellaria root and a mitigatory component for Giycyrrhiza. Jujube acted as a mitigatory component for Glycyrrhiza. Ginger acted as an additional component for Scutellaria root in cAMP PDE test. This additional effect of 6-gingerol, 6-shogaol from Ginger and baicalin from Scutellaria root were investigated.

Effect of sulfonamides and 4-MET on adhesion to tooth substrates.

Adhesion of a composite to tooth substrates was studied. The bonding liner was a mixture: camphorquinone, 0.5%; one of four sulfonamides (A, B, C, and D), 0.5%; 4-methacryloxyethyl trimellitate (4-MET), 0, 2, and 5%; in triethyleneglycol dimethacrylate (TEGDMA). The best tensile bond strength to bovine dentin treated with 0.3 M EDTA 2 Na-0.2 M EDTA FeNa (EDTA 3-2) was 7.5 MPa. The bond strength to bovine dentin decreased with the addition of 4-MET. The bond strength to bovine enamel treated with 65% phosphoric acid (H3PO4) was 12.0 MPa. The tensile bond strength to bovine enamel treated with EDTA 3-2 was lower than that to enamel treated with 10% citric acid-3% ferric chloride (10-3) and with H3PO4. The polymerization of these liners was characterized by differential scanning calorimetry (DSC). The photocurable bonding liner studied was adequate for clinical studies.

Cloning and nucleotide sequence of cDNA for Ki antigen, a highly conserved nuclear protein detected with sera from patients with systemic lupus erythematosus.

Patients with systemic lupus erythematosus (SLE) produce autoantibodies against a variety of nuclear antigens including Ki antigen. Although anti-Ki autoantibodies were found in a significant number of SLE patients, the nature of Ki antigen is poorly characterized. By using anti-Ki serum as a probe we have cloned a bovine cDNA directing the synthesis in Escherichia coli of a polypeptide immunologically indistinguishable from the authentic Ki antigen. A homologous human cDNA was also cloned and its nucleotide sequence predicted the entire primary structure of a novel nuclear protein with a molecular weight of 29 508 and with highly hydrophilic and weakly acidic character. The gene is highly conserved not only in the coding region but also in the 3'-untranslated region. The bacterially produced Ki antigen would be valuable for diagnosis of SLE.

[Clinical courses and pathological findings in two gay male patients with acquired immunodeficiency syndrome infected in Japan].

This is a report on the clinical courses and pathological findings in two gay male patients with acquired immunodeficiency syndrome (AIDS) infected in Japan. Case 1. A 39 year-old Japanese homosexual male was diagnosed as amebic dysentery complicated with liver abscess on admission. He was placed on Metronidazole with complete relief. Serological tests was positive for AIDS. On second admission, he was found to have pneumocystis carinii pneumonia (PCP) and cytomegalo-viral uveitis. Administration of Pentamidine was partially effective, however the therapy with Azidothimidine was discontinued by bone marrow suppression. On his third admission, he suffered from cryptococcal meningitis and therapy-resistant fungusemia. Finally he died of recurrent pneumonia regardless of appropriate therapies. Autopsy proved extended cryptococcal infection in the brain, meninx, lungs, liver and kidney, and cytomegalo-infection in the lungs, liver and kidney. Furthermore, atypical mycobacteriosis was found in the lymph nodes. There was no active findings compatible with PCP. Case 2. A 44 year-old Japanese homosexual male was admitted with oral candidiasis and diagnosed as AIDS related complex. He suffered from pneumonia with marked improvement on sulfamethoxazole-Trimethoprim. On his second admission, he developed diarrhea and was found to be infected with Giardia lambia. In addition, cytomegalo-viral infection damaged his eye sight. He died of pneumonia and meningitis shortly there after. Autopsy proved a cytomegalo-viral infection in the lung and colon, old lesions possibly caused by PCP in the lungs, and suppurative meningitis in the meninx. These experiences confirm that AIDS patients can be exposed to several opportunistic infections at the same time in the multiple organs. Furthermore, it is suggested that homosexual patients with AIDS may have unique opportunistic infections such as amebic dysentery or Giardia lamblia unlike other AIDS patients related to hemophilia.

Ultrastructural localization of fibronectin and laminin in human granulation tissue in relation to capillary development.

The distribution of fibronectin (FN) and laminin (LM) at developing capillaries during various developmental stages, from capillary sprouts to relatively developed capillaries, was studied by light- and electron-microscopy immunocytochemistry. By light-microscope, FN immunoreactivity was diffusely distributed throughout the stroma of the granulation tissues, while for LM it was preferentially distributed at the perivascular region with the various developmental stages of the immature capillaries. Ultrastructural study revealed that capillary sprouts were closely surrounded by plentiful deposits of immunoreaction with the FN, but only faintly for LM. Relatively developed capillaries with large and tall endothelium were surrounded by plentiful immunoreactive products with both FN and LM, and immunoreactivities in the cisternae of rER of the endothelium and/or the pericytes were also shown. Cytoplasmic interdigitations between the endothelium and the pericyte of developing capillaries were recognized without an immunoreaction for FN and LM. These results mean that the capillary sprouts are associated with a prepatterned FN-rich and LM-poor perivascular matrix, whereas relatively developed capillaries are associated with a FN- and LM-rich perivascular matrix which would have been produced by the capillary endothelium and/or pericytes.

[The study of Chinese herbal medicinal prescription with enzyme inhibitory activity. IV. The study of the prescription containing mineral drug with adenosine 3',5'-cyclic monophosphate phosphodiesterase].

Twenty five Chinese herbal medicinal prescriptions containing gypsum, kaolin, longgu, oyster shell and sodium sulfate were studied for the inhibitory activity of adenosine 3',5'-cyclic monophosphate phosphodiesterase. The inhibitory activity of 15 prescriptions without mineral drug was higher than that of each original prescription. On the contrary, four were lower and six were not recognized to be different. All 11 prescriptions containing gypsum with an exception increased the inhibitory activity by removing gypsum. The half prescriptions containing kaolin or sodium sulfate also increased the inhibitory activity by removing the drug.

[The study on Chinese herbal medicinal prescription with enzyme inhibitory activity. III. The study of mao-to with adenosine 3',5'-cyclic monophosphate phosphodiesterase].

Mao-to, a Chinese herbal medicinal prescription was studied for the inhibitory activity of adenosine 3',5'-cyclic monophosphate (cAMP) phosphodiesterase. The inhibitory activity for this enzyme depended mainly on Ephedra herb and Glycyrrhiza in this prescription. Apricot kennel acted as a mitigatory component for Ephedra herb in cAMP phosphodiesterase inhibitory test. In ephedrine and the related compounds the inhibitory activity of cAMP phosphodiesterase was not shown.

[Effect of adhesion promoting monomers on adhesion to ground dentin].

Effect of adhesion promoting monomers dissolved in photocurable bonding agents on adhesion to ground dentin was investigated. They were MDP, Phosmer-M and 4-MET. The effect of those monomers was compared with that of Phenyl-P. The bonding agents contained campherquinone (CQ) as a photosensitizer, N-phenylglycine (NPG) as a reducing agent and TEGDMA as a base monomer. Phenyl-P was the best adhesion promoting monomer among those studied monomers. MDP could not polymerize well enough to give good bond strength. Phosmer-M could permeate through the smeared layer but could not make a stable resin reinforced dentin. 4-MET did not permeate through the smeared layer and did not adhere to ground dentin.

Immunocytochemical and ultrastructural studies of the histogenesis of Ewing's sarcoma and putatively related tumors.

The histogenesis of Ewing's sarcoma (EW) and extraskeletal Ewing's sarcoma (EEW) is still disputable. Their relationship to the so-called Askin's tumor, neuroectodermal tumor of bone, and peripheral neuroblastoma remains to be established. In an attempt to clarify these points, immunocytochemical and ultrastructural studies were done on tissues from 14 cases of EW, 4 cases of EEW, and 9 cases of primitive neuroectodermal tumor (PNET) and compared with neuroblastoma and olfactory neuroblastoma. Six tumors categorized initially as EW and EEW on biopsy, turned out to be PNET by extensive histologic and/or ultrastructural observations. Abundant glycogen was recognized not only in 16 of 18 cases of EW and EEW, but also in seven of nine cases of PNET. Fine fibrillar cell processes were seen between tumor cells, at least in limited areas even in cases of EW and EEW. Immunocytochemically, neuron-specific enolase (NSE), neuroblastoma cell surface antigen (NBCA), neuron cell surface antigen (NCSA), and neurofilament (NF) were demonstrated not only in neuroblastoma, but also frequently in cases of EW, EEW, and PNET. The results seem to suggest that EW and EEW represent the most immature forms of neuroectodermal tumor. Electron microscopic study showed predominantly primitive cells with occasional areas of cell processes, neurosecretory granules, and microtubules, suggesting a neuroectodermal origin.
