Role of specific determinants in mannan of Candida albicans serotype A in adherence to human buccal epithelial cells.

Candida albicans serotype A (C. albicans A) possesses a specific antigen, designated antigen 6, which resides in mannans on the cell surface. To determine the role of the mannan moiety of the C. albicans cell wall in adherence to buccal epithelial cells, we used antigen 6-deficient mutants which had been isolated by screening with an agglutinating monoclonal antibody against antigen 6 (MAb-6). 1H nuclear magnetic resonance spectral analysis of the purified mannans from the mutants showed a loss of the signals related to that beta-linkage of the side chains. Moreover, acetolyzed fragments of the mutant mannans showed a decreased amount of mannohexaose and mannopentaose. The mutant yeast cells exhibited significantly reduced ability to adhere both to exfoliated buccal epithelial cells and to a human buccal cell line. A number of strains of C. albicans A, C. tropicalis, and C. glabrata, all of which bear antigen 6, showed significantly higher adherence to the cell line than did those of C. albicans serotype B, which lack antigen 6. The whole mannan from the C. albicans A parent inhibited the adherence of C. albicans A to epithelial cells dose dependently, whereas mannan from a mutant strains did not. Moreover, C. albicans A treated with MAb-6 or polyclonal factor 6 serum showed reduced adherence. A close correlation was found between adhesive ability and agglutinability with MAb-6 in the C. albicans A parent, the antigenic mutants, and their spontaneous revertants. These results suggest that so far as mannan adhesion is concerned, serotype A-specific determinants are largely involved in the mechanisms of adherence of C. albicans A to human buccal epithelial cells.

[CML with autoimmune thrombocytopenia observed in the course of IgG (kappa) type monoclonal gammopathy].

An 80-year-old male with IgG (kappa) type benign monoclonal gammopathy was admitted to our hospital because of marked leukocytosis. At the time of admission, thrombocytopenia was also noted. A bone marrow aspirate showed marked granulocytosis with a normal megakaryocyte count. PAIgG was elevated and the NAP score was low. Ph1 chromosome and rearrangement of the breakpoint cluster region were detected. On the basis of these findings, he was diagnosed as having CML with autoimmune thrombocytopenia. This case was of interest with respect to blood cell differentiation and immunological findings.

Bacterial Accumulation of Metallic Compounds.

Xanthomonas maltophilia accumulated metals or metallic compounds within the cell in the form of one or a few particles, in a peculiar condition similar to that of an electrolyte solution in an electric cell. The particles consisted of many little granules, most of which were assumed to be composed mainly of a compound of zinc and phosphorus in an amorphous state.

[IgA-lambda type myeloma complicating with autoimmune hemolytic anemia].

A 66-year-old female was admitted to our hospital with lumbago. On admission, laboratory examination revealed hemolytic anemia. Direct Coombs' test was positive, and also direct monospecific-Coombs' test by anti-C3d serum was positive. Immunoelectrophoresis showed IgA-lambda type M proteins in serum. Bone marrow aspiration disclosed increased atypical plasma cells. X-ray of skull showed punched-out lesion. From these findings, she was diagnosed as IgA (lambda) myeloma complicating with autoimmune hemolytic anemia. Hemolysis was improved by chemotherapy. It was thought that IgA and C3 were related well to hemolysis in this case.

[Hemophagocytic syndrome in two elderly men].

Case 1: A 68-year-old male, who was admitted to our hospital because of massive intestinal bleeding, died 1 day after admission. Case 2: An 80-year-old male, who was admitted to our hospital because of a loss of consciousness, died 4 days after admission. In both cases prodromic infection was not detected. At autopsy, phagocytosis of RBC and WBC by histiocytes was shown markedly in the spleen in case 1 and in the bone marrow in case 2. So diagnosis of virus-associated hemophagocytic syndrome was made. It might be related to some underlying immunologic derangement, because both cases were in advanced age.

A new platelet-specific antigen, Naka, involved in the refractoriness of HLA-matched platelet transfusion.

Serum from a thrombocytopenic patient who was refractory to the transfusions of HLA-matched platelets contained a platelet-specific alloantibody, anti-Naka. Immunofluorescence analyses revealed that the Naka antigen defined by the serum was expressed exclusively on platelets and its distribution was different from P1A1, Baka, Yuka or Yukb. Analysis by Dr. von dem Borne's group revealed the Naka was also different from Koa, Kob or Zwb. Family studies showed that the Naka antigen was inherited as an autosomal codominant trait. Its antigen frequency in the Japanese population was over 97%. The results of the enzyme immunoassay using monoclonal antibodies for antigen immobilization showed that the Naka epitope did not appear to reside on GPIIb/IIIa or Ib. The transfusions of Naka-compatible platelets improved the patient's thrombocytopenia.

[Chronic disseminated intravascular coagulation due to abdominal true aneurysm in two cases--effect of anti-platelet and anti-fibrinolytic therapy].

Case 1: 75 years old male was admitted to our hospital with anterior chest subcutaneous bleeding. Coagulation study revealed that fibrinogen and alpha 2-PI decreased, and FDP, FPA, B beta 15 approximately 42 and D-dimer increased. Case 2: 78 years old male was admitted to Shingu City Hospital with a left hip subcutaneous hematoma. Coagulation study revealed that fibrinogen, ATIII and alpha 2-PI decreased, and FDP increased. US and CT showed abdominal true aneurysm in both cases. Either severe infection or malignancy was not found. Ticropidine and T-AMCHA were medicated for 8 days in case 1, and for 18 months in case 2. Symptom and coagulation study improved in these cases. Due to some side effects such as appetite loss and liver dysfunction in case 1, and diarrhea in both cases, we changed the therapy to mini-dose heparin therapy. This therapy also proved effective. It is concluded that anti-platelet and anti-fibrinolytic therapy are effective for chronic DIC with abdominal true aneurysm.

Validity of Candida natalensis van der Walt et Tscheuschner.

In order to verify whether Candida natalensis is a synonym of Candida sake or not, we investigated DNA base composition, DNA-DNA hybridization, the electrophoretic patterns of enzymes, and proton magnetic resonance (PMR) spectra of cell wall mannans of representative strains of C. natalensis and C. sake. No significant differences in DNA base compositions were found among strains of C. natalensis and C. sake. Very low DNA relatedness was detected between the type strain of C. natalensis and the type strain of C. sake, and little DNA relatedness was demonstrated between a strain of C. natalensis and the type strain of C. sake. Furthermore, little DNA relatedness was found between the type strain and a strain of C. natalensis. These results well correlated with those of the electrophoretic patterns of seven enzymes and PMR spectra of cell wall mannans. C. natalensis is a distinct species from C. sake. Also, one strain, which was previously identified as C. natalensis, is a different species from C. natalensis and C. sake.

Sporobolomyces yuccicola, a new species of ballistosporous yeast equipped with ubiquinone-9.

A hitherto undescribed ballistosporous yeast was isolated from a dead leaf of Yucca sp. in Canada. This yeast produces apiculate or short-ellipsoidal ballistospores, produces pale colored colonies, has Q-9 as the major ubiquinone, and does not contain xylose in the cells. This new yeast is described as Sporobolomyces yuccicola Nakase et Suzuki. Sporobolomyces yuccicola is the sixth species of the intermedius group, a group of atypical species of the genus Sporobolomyces equipped with Q-9.

[Experience of HLA-matched platelet transfusion in patients who are refractory to random donor platelets].

Platelets collected from random and multiple donors were as effective on transfusion as those obtained from single donor by platelet pheresis if patients were not alloimmunized. In alloimmunized patients, effects of platelet transfusion were correlated with the HLA-matching grade; 100% effective in A-match, 92% in B-match and 65% in C-match. This correlation was not observed if the patient had infection. The transfusion of A- or B-matched platelets resulted in a lowering or disappearance of HLA antibody activities in most patients. However, the transfusion of C-matched platelets caused increasing or appearance of HLA antibody activities in some patients. The ABO-incompatibility did not show a significant effect on platelet transfusion when anti-A or anti-B antibody activities in donors and antiglobulin tests on patients' red cells were negligible. Non HLA, platelet specific antibodies were produced in some patients and presumed to be participated in the destruction of transfused HLA-matched platelets.

Sporobolomyces inositophilus, a new species of ballistosporous yeast isolated from a dead leaf of Sasa sp. in Japan.

A new species of ballistosporous yeast, Sporobolomyces inositophilus was isolated from a dead leaf of Sasa sp. collected at a mountain in Japan. This yeast is assumed to have a close relationship to Sporobolomyces singularis since it produces bilaterally symmetrical ballistospores and pale-coloured colonies and resembles Sporobolomyces singularis in these respects. These two species differ from typical species of Sporobolomyces and seem to constitute a natural taxonomic group.

Metabolic fate of ethyl O-[N-(p-carboxyphenyl)-carbamoyl] mycophenolate (CAM), a new antitumor agent, in experimental animals.

Absorption, distribution, metabolism and excretion of ethyl O-[N-(p-carboxyphenyl)-carbamoyl] mycophenolate (CAM), a new antitumor agent, was studied in mice using 14C-labeled CAM and species difference of metabolism was studied in mice, rats, guinea pigs, rabbits and dogs. CAM was rapidly absorbed from the gastrointestinal tract of mice. Concentration of radioactivity in the plasma of mice showed two peaks reflecting enterohepatic circulation of metabolites, the first was 30 min and the second was 6 h after dosing. Extremely high concentration of radioactivity was observed in the bile as about 100 times of the radioactivity detected in the plasma. Wide distribution of radioactivity was observed in the whole body tissues of the tumor-bearing mice. Strong radioactivity was presented in the liver, kidney, bile and gastrointestinal content. Radioactivity in the tumor was higher than those in the liver and kidney 24 h after dosing. About 45% of the dose and 30% of the dose were excreted in the urine and feces, respectively, during 24 h after administration of 14C-CAM to mice. The metabolites of CAM were only mycophenolic acid (MPA) and its glucuronic acid conjugate (MPA-G) in all tested animals. Converting ratio of MPA to MPA-G differed greatly among animal species. In the plasma of rats and mice, MPA was presented as higher level than MPA-G at all times. While, in the plasma of rabbits, MPA-G was presented exclusively and MPA was observed only slightly.

DNA base composition of species of the genus Saccharomyces.

DNA base compositions (GC content) of Saccharomyces species are reported and discussed. Several amendments of the four groups given by van der Walt are suggested, viz. the transfer of S. kluyveri to group 1, and of S. eupagycus, S. cidri, S. montanus, S. microellipsodes and S. florentinus to group 2. The synonomy of S. amurcae and S. cidri is suggested. The DNA base compositions revealed two possible pairs of sibling species: S. elegans and S. bailii, with a difference in GC content of 4.1%; S. dairensis and S. servazzii with a difference in GC content of ca. 3%. S. mrakii had a GC content of 47.3-48.5% the highest encountered in this genus and similar to that of Kluyveromyces thermotolerans.
