[Potentiation of cytotoxic effects of 5-fluorouracil by inosiplex on cancer cells].

Inosiplex, a 3:1 molarcomplex of N, N-dimethylamino-2-propanol-p-acetamidobenzoate and inosine, which has been reported to exhibit antiviral activity in vitro and in vivo, enhanced cytotoxic effects of 5-fluorouracil (5-FU). Effects of inosiplex on the potentiation of cytotoxicity of 5-FU were investigated in vitro and in vivo. In vitro studies demonstrated that cytotoxic effects of 5-FU on cloning efficiency of HeLa cells were prominently enhanced by inosiplex, while inosiplex alone showed no cytotoxicity at the concentrations examined. Further, the survival time of mice intraperitoneally inoculated with Ehrlich ascites tumor cells was investigated. The mean survival time of mice treated with the combination of 5-FU and inosiplex significantly prolonged as compared with that of control animals or mice treated with inosiplex alone or 5-FU alone. Since inosiplex has been clinically studied as an antiviral agent and proved to be harmless to humans, the present results indicate that a combined administration of 5-FU and inosiplex may be effective in the treatment of human cancers.

Role of the central and peripheral retina upon optokinetic nystagmus. Foveal and peripheral nystagmus.

The optokinetic (OK) nystagmus induced by OK stimulus on the foveal area (foveal nystagmus) or on the peripheral retina (peripheral nystagmus) are discussed. Separation of the visual field can be achieved by a combination of a projection type OK stimulator and a masking cylinder driven by a d.c. ENG. Foveal and peripheral nystagmus became increasingly difficult to elicit in tact with narrowing of the visual field or increase in stimulus velocity. The influence of velocity was much more noticeable in the foveal nystagmus. The characteristics of foveal and peripheral OK nystagmus are elucidated by the overlapping method. The foveal nystagmus has a rather distorted form, but its slow-phase eye velocity is the same as the target velocity. It has a tendency to start and reverse with slow phase. Its beating field deviates in the direction of the slow phase. The peripheral nystagmus, on the contrary, has a typical saw-tooth form and its slow-phase eye velocity is slower than the target velocity, even at the low speed stimulus. It has a tendency to start and reverse with the quick phase. Its beating field deviates in the direction of the quick phase.

Hypertrophic neuropathy with diabetes mellitus.

A 52-year-old male with diabetes mellitus showed sensorimotor disturbance of symmetrical glove and stocking distribution. Electromyography demonstrated signs of denervation, and motor nerve conduction velocities could not be obtained because the muscle action potential was not evoked by the electrical stimulation of the nerves. Quantitative histologic and ultrastructual studies were performed in the sural nerve biopsy. Determination of fiber densities revealed a striking decrease of both myelinated and unmyelinated fibers and remarkable increase of the onion bulb formations. Hypertrophic changes with onion bulb formations have been observed in various clinical conditions, particularly in hereditary disorders, but uncommon in diabetic neuropathy. Our case was non-familial and may form one atypical type of diabetic neuropathy.

Interferon potentiates cytotoxic effects of 5-fluorouracil on cell proliferation of established human cell lines originating from neoplastic tissues.

A potentiation of the cytotoxic effects of 5-fluorouracil (5-FU) on human tumor cells by interferon was examined. The human neoplastic cell lines used were HeLa (uterine cervical cancer), MCF-7 (mammary cancer), WI-38 CT-1 (embryonic lung fibroblasts transformed in culture by Co-60 gamma-ray irradiation), KMM-1 (myeloma) and Raji (Burkitt's lymphoma). The normal human cell strain used was WI-38 (normal human lung fibroblasts). The cytotoxic effects were determined by colony formation for HeLa, MCF-7, WI-38 CT-1 and WI-38 cells, and by cell growth for KMM-1 and Raji cells. Each cell line was different in sensitivity to interferon or 5-FU. Interferon potentiated synergistically the cytotoxic effects of 5-FU on HeLa, WI-38 CT-1 and KMM-1 cells. In the case of Raji cells, the cytotoxic effects of the combination of interferon and 5-FU were additive. Neither synergistic nor additive lethal effects of the combination of the 2 agents were observed in MCF-7 and WI-38 cells. The present results indicate a possibility that interferon and 5-FU can mutally reduce the amount of the other needed to treat cancer patients.

Seeding of gallbladder carcinoma along the tract after percutaneous transhepatic choledochoscopy.

An implant metastasis developed along the tract after percutaneous transhepatic cholecystoscopy 5 months after initial placement of the catheter into the gallbladder. Percutaneous transhepatic cholecystoscopy is conducted by insertion of choledochofiberscope into the gallbladder through the dilated sinus tract established around a catheter inserted into the gallbladder. The catheter had been placed for total of 15 days. Two days after the removal of the catheter, cholecystectomy supplemented by partial resection of the liver was carried out because of the extent of a gallbladder carcinoma. The choledochofiberscope used was Olympus CHF type 4B, with a thinner flexible part 5.0 mm in diameter. It is not surprising that carcinoma of the biliary tract or pancreas may seed along the tract of a biliary drainage catheter, but only few reports describing such incidence related to percutaneous transhepatic biliary drainage have previously been published. Physicians should be aware of this complication whenever a firm nodule develops at the site of previous entry of a biliary drainage catheter in a patient with malignant obstruction.

[Clinical evaluation of latamoxef in the perinatal period].

Latamoxef (LMOX), a new oxacephem antibiotic with high activity against Gram-negative bacteria has been investigated for use in No. of 58 mothers in perinatal period, and obtained following results. Concentration of LMOX in maternal serum was 43.4 micrograms/ml at the 1 hour after intravenous administration of 1 g. In umbilical cord serum and amniotic fluid, LMOX showed good translation after intravenous administration of 1 g into the mother, but no adverse effect appeared in the neonate. LMOX is highly useful antibiotic in perinatal infections, and the safe dose of LMOX to the mother in perinatal period is 1--2 g per day considerably.
