SAL-TOL in vivo recombinant plasmid pKF439 was characterized in a strain from a mixed culture of bacteria harboring various degradative plasmids. Analysis of the gene organization of pKF439 revealed that the 57-kilobase TOL fragment, including the 40-kilobase TOL metabolic region, was inserted into the complete SAL replicon at the position of SmaI-C within XhoI-B of SAL. The molecular size of pKF439 was calculated to be 138 kilobases. pKF439 could be transferred to Pseudomonas putida and Pseudomonas aeruginosa at high frequency, and the transconjugants gained the ability to grow with m-xylene, m-toluate, and salicylate as the sole carbon source.
The biological and immunomodulating activities of polysaccharide fraction (GF-1), an antitumor poysaccharide fraction from cultured fruiting bodies of Grifola frondosa, was examined in mice. GF-1 showed no cytocidal effect on culturing tumor cells. However, GF-1 induced resistance against Sarcoma 180 in ICR mice which had completely regressed from the tumor by the effect of GF-1. The administration of GF-1 into mice increased the weights or cell numbers of spleen, and peritoneal cavity. GF-1 enhanced the antigen specific antibody response and carbon clearance activity, whereas GF-1 did not show polyclonal B cell activation and mitogenic activities, and the effect on delayed type hypersensitivity.
The effect of Vesiculogen (a hot-water extracted B cell mitogen from Peziza vesiculosa) on antibody production in mouse spleens was studied. The number of plaque-forming cells (PFC) elicited by injection with Vesiculogen and/or 2,4,6-trinitrophenyl substituted sheep red blood cells (TNP-SRBC) were comparatively assayed. In the mice injected with 100 micrograms of Vesiculogen (i.p.), an obvious increase in the numbers of anti-SRBC and anti-TNP SRBC PEC in spleens was observed after 2 d of injection, and the numbers of PFC reached a maximum on day 3. Injections to 1-1000 micrograms of Vesiculogen were effective. This effect was shown when Vesiculogen was administered by i.p. or i.v. injections. In some cases, the numbers of PFC in mice injected with Vesiculogen and antigen exceeded the sum of each number of PFC elicited by polyclonal B cell activation activity of Vesiculogen and by antigenic stimulation of TNP-SRBC. Pretreatment with Vesiculogen within 4 d before immunization markedly reduced the PFC response. These results suggest that the effect of Vesiculogen on antibody response in mice attributes to its activities as a polyclonal B cell activator and adjuvant.

The difference between the right (RV) and the left ventricle during progressive pressure loading is that the RV changes to cylindrical form without change in its enddiastolic pressure (EDP). We investigated this particular phase experimentally and clinically. In experimental animal studies (dogs), the gradual construction of the main pulmonary artery (PA) resulted in an elevation of RV systolic pressure (RVSP) without changes in RVEDP and PA flow (FPA). This was followed by an increase in RVEDP and a decrease in FPA. In this initial phase, the RV increased in its dimension from the free wall to the septum. In 155 clinical cases with RV pressure loading, the relationship between RVEDP and RVSP suggested that the RV compliance of the cases with chronic RV pressure loading was less than that with subacute loading. However, these studies did not supply the expected data on the phase we mentioned. In 2 patients with primary pulmonary hypertension who showed spontaneous remission during our observation, a decrease in RVSP without change in RVEDP occurred with a change in RV form detected by 2-dimensional echocardiography. These data suggested the existence of a phase in which the RV showed a character of plasticity during pressure loading, but direct documentation was not successful.

High ferritin levels in the aspirate of thyroid cyst (Six yellow clear, 4 yellow turbid and 10 chocolate colored turbid) without apparent sings of malignancy were found. The mean concentration in the 3,000 X g supernatant of the fluid was 40,116 ng/ml, and the 3,000 X g precipitate was 11,147 ng/ml. All cases showed normal levels of serum ferritin. Con A binding with ferritin was distributed from low to high. These ferritins showed a molecular weight of approximately 450,000 which was the same as found in human spleen and liver. The continuous increase in ferritin levels in thyroid cyst fluid was found by a chronological study in some cases. When tumor markers such as CEA, NCA, CA 19-9 and alpha 1-acid glycoprotein (alpha 1-AG, acute phase reactant) were examined simultaneously, an increase in some of the cyst fluid was observed. However, the incidence and the rate of increase of these tumor markers and acute phase reactant were low compared to ferritin. Neither a correlation between ferritin and CEA, nor between ferritin and CA 19-9 was found. The increase in ferritin in thyroid cyst fluids may be due to the increased synthesis, release and storage by the inflammatory cells.
After a large amount of aldosterone was injected into a male rabbit, urine was collected for 48 h. Separation of urinary aldosterone metabolites into monoglucosiduronate fraction and monosulphate fraction was carried out by a combination of countercurrent distribution and DEAE-Sephadex A-25 column chromatography. Each fraction was hydrolyzed with enzyme and free steroids released were separated by Sephadex LH-20 column chromatography. The free steroid was then identified by gas chromatography-mass spectrometry. In monoglucosiduronate fraction, 3 alpha, 5 beta-tetrahydroaldosterone and 3 beta, 5 alpha-tetrahydroaldosterone were found. On the other hand, 3 alpha, 5 beta-tetrahydroaldosterone was the only aglycone detected in monosulphate fraction. These findings comfirmed results in the preceding paper, where the free steroid was characterized on the basis of the mobility of the steroid and its derivatives on paper chromatography.
Analysis of urinary metabolites of [1, 2-3H]-aldosterone and [1, 2-3H]-3 alpha, 5 beta-tetrahydroaldosterone was performed in male rabbits. The preliminary separation of urinary metabolites was carried out by submitting these metabolites to countercurrent distribution. Further separation of each fraction thus obtained was achieved by means of DEAE-Sephadex A-25 column chromatography. The separated peak was then hydrolyzed with the enzyme and the free steroid released was identified on the basis of the mobilities of the steroid and its derivatives on paper chromatography. After the injection of [1, 2-3H]-aldosterone, a major urinary metabolite was characterized as monosulphate of 3 alpha, 5 beta-tetrahydroaldosterone. In addition, a small amount of the monoglucosiduronate fraction was found in the urine. 3 alpha, 5 beta-tetrahydroaldosterone and 3 beta, 5 alpha-tetrahydroaldosterone were detected as aglycones in this fraction. After the injection of [1, 2-3H]-3 alpha, 5 beta-tetrahydroaldosterone, a similar pattern of urinary radiometabolites was observed. The close similarity between the profile of urinary metabolites of [1, 2-3H]-aldosterone and that of [1, 2-3H]-3 alpha, 5 beta-tetrahydroaldosterone suggests that the conversion of aldosterone to 3 alpha, 5 beta-tetrahydroaldosterone is needed before the conjugation processes take place.
Lysosomal arylsulfatase B of human leukocytes consisted of two forms; a basic form (B) and a variant form (B1) which is phosphorylated at the carbohydrate chains of the B form (Uehara, Y., Gasa, S., Makita, A., Sakurada, K., and Miyazaki, T. (1983) Cancer Res. 43, 5618-5622). The amounts of the variant form relative to the basic form were considerably increased in leukocytes of chronic myelogenous leukemia (CML). The present communication demonstrates that, upon chemotherapy of the patients with CML, degree of phosphorylation as well as the relative amounts of the phosphorylated variant form of CML leukocytes are markedly decreased concomitantly with an increase of the basic, less phosphorylated form. This effect of chemotherapy on the variant form preceded to clinical improvement of the CML patients, suggesting that the relative amount of the phosphorylated enzyme will be a potential prognostic indicator for the therapeutic effect of CML.

Sera and peripheral blood lymphocytes in 40 clinical cases of adult T-cell leukemia (ATL) and 66 cases of mature T-cell malignancies in Hokkaido district, the most northern part of Japan, were examined for the infection with ATL virus (ATLV). All of the 40 ATL patients (100%) had antibodies to ATLV-associated antigen (ATLA) and 13 (19.7%) of the 66 patients with mature T-cell malignancies other than ATL were also positive for anti-ATLA. When the peripheral ATL lymphocytes were assessed after short time cultivation, both ATLA antigen and C type virus particles were detectable. In contrast to the high prevalence of ATLV antibodies in these patients, the positivity in 30,000 control individuals in this district was 0.73%. It was calculated that 8 patients of a 40,000 seropositive population are diagnosed annually as ATL in this particular northern part of Japan.
Family members of non-endemic Hokkaido patients with adult T-cell leukemia (ATL) were assessed for the prevalence of ATL virus (ATLV) infection. Immunofluorescence test showed that 53 (39.8%) of 133 healthy family members of 23 ATL patients were positive for antibodies of ATLV-associated antigen (ATLA). When seropositive T-cell malignancies other than ATL and healthy controls in Hokkaido were examined, 10 of 33 family members (30.3%) of 9 patients and 3 of 18 family members (16.7%) of 5 donors had ATLA antibodies, respectively. In sharp contrast, the overall seropositivity in this northern part of Japan was 0.73%. None were seropositive (0%), when 26 family members of 6 patients with acute myeloid leukemia sero-converted by blood transfusion during the course of the disease.
