[A study on the relation between stomatognathic system and the systemic condition, concerning the influence of experimental occlusal interference on upright posture, particularly on gravity fluctuation and the antigravity muscles].

The purpose of this study is to reveal the relation between stomatognathic system and the systemic condition. In the present study, experimental occlusal interference was given to the first molar on main mastication side of 6 healthy subjects and the influence on the upright posture was evaluated through simultaneous measurements of changes in activity of antigravity muscles via electromyography, other than the measurement of loci of the gravity fluctuation for stabilograph before and after the interference was provided. The following results were obtained, 1. Loci of gravity fluctuation 1) All parameters tended increase 24 hours after the interference was provided. 2) The decreasing trend was noted 24 hours after the interference was removed. 3) At one week after the interference was removed all analysis items tended to restore to the normal range. 2. Activity of antigravity muscles In some of the subjects, the muscular activity showed the same trend as the changes of analysis items of gravity fluctuation. 3. The above results suggest that the evaluation of the loci of the gravity fluctuation may be helpful to assess the therapeutic effect of malocclusion.

Determination of the molecular nature and cellular localization of Thy-1 in human renal tissue.

The molecular nature and cellular localization of Thy-1 antigen in human renal tissue were studied. Strong immunohistochemical staining was observed in frozen sections of human kidney using monoclonal anti-human Thy-1 antibody; this reaction was almost completely abolished by pretreating the kidney section with phosphatidyl inositol (PI)-specific phospholipase C (PI-PLC). Immunohistochemical analysis revealed that the Thy-1 antigen is localized on the proximal tubular epithelial cells and the Bowman's capsule of the glomerulus. Northern blot analysis of renal mRNA using a cloned human Thy-1 gene revealed the presence of human Thy-1 mRNA of a similar size to the one in human brain. When a human kidney cDNA library was screened with the same probe, a cDNA of human Thy-1 was isolated. Moreover, human Thy-1 protein with a molecular weight (MW) of 21,000 was detected in renal tissue by gel electrophoresis and Western blot analysis using monoclonal anti-human Thy-1 antibody. These data demonstrate for the first time the production of human Thy-1 as a PI-anchored protein with a unique cellular location in human renal tissue.

[Statistical studies of student clinical practice. 1. Clinical practice of operative dentistry from 1983 to 1989].

Clinical practice has been assigned as a final and very important aspect of study and practice of dental education. In our university, the students are exposed the clinical practice from the latter term of fifth year. This report investigated the protocols of clinical cases from the first to the seventh class of graduates. The results obtained were as follows: 1) The graduates of first class averaged 9 clinical cases. This decreased with the years and the fourth class averaged 5.5 cases. From the sixth class simulator training and mutual practice among the students was introduced and the total average of seventh class was 5.1 cases. 2) The dental practice centers on restoration and 20 percent of all cases were amalgam fillings from the first to four the class. However this decreased from the fifth class and does not take place at present. Composite resin restorations increased. Cast restorations occupied 20 percent of all cases and the proportion hasn't changed. 3) Clinical cases of graduates of fifth class who were first to include treatment between students averaged 7 cases. Mutual treatment practice is useful because of the increase exposure to clinical practice.

[A study for non-asbestos casting liners. An experimental study of the adaptability of MOD casting to die].

Asbestos liners for investment metal casting has frequently been used in dental laboratory work. In recent years, the dangerous properties of asbestos to the human body were reported in Europe and the United States, and casting liners without asbestos were developed and marketed by many manufacturers. These are so-called non-asbestos liners. This study evaluated the adaptability of MOD inlay castings to dies using 8 kinds of commercial non-asbestos liners of ADA standard No. 2 specifications. The results were as follows; Single and double layers of Casting ribbon, New Asbestos Ribon, Shofu experimental, and KAOLIN showed good adaptation. Shofu experimental with a single layer showed the best marginal adaptation (10 microns). OVAL LINERS and CASTING LINERS with single layer showed good adaptation, but with double layers the adaptation was poor. In FLASK LINER, the double layer showed better adaptation than that with a single layer. Most non-asbestos liners used in this study are adequate in useful to MOD inlay castings.

[The influences of habitual side chewing and the character of food on the masticatory muscle activity in children].

The purpose of this study was to investigate the influences of habitual side chewing and the characteristics of food on the masticatory muscle function in children. A total of thirty persons were subjected to the study; 10 with deciduous dentition (designated as G1), 10 with mixed dentition (as G2), 10 with permanent dentition (as G3), none of whom had any gnatho-facial dysfunction. EMGs were recorded from the temporal anterior muscles and masseter muscles on both sides, and the mandibular movement on the frontal plane was recorded during chewing of boiled fish cake paste, chewing-gum and chewing of dried cuttlefish on both the habitual side and the non-habitual side. EMG activities obtained and periods of closed phase, occlusal phase and open phase in the pathway of the mandibular movement were measured with a personal computer system. The relationship between the total muscle activities at clenching in the intercuspal position and the total muscle activities at chewing were also evaluated. The results obtained in the present study were as follows: 1) Influence of habitual side chewing on the rhythm of the mandibular movement during chewing of chewing-gum was not found in all groups. Influence on EMG activities was not found in G1 and G2, but were presented in G3. 2) Influence of the character of food on rhythm of mandibular movement was not clear in G1, but as found in the other groups, while EMG activities were influenced by the character of food in all groups. 3) The MA Index (the ratio of total muscle activities while chewing to total muscle activities at clenching) showed a decrease with age. As far as the results of the present study is concerned, it could be suggested that the proficiency of the chewing ability is dependent on the dentitional growth. Furthermore, the MA Index was considered to be appropriate for evaluating the masticatory muscle function in children.

[Effect of PMUE therapy (CDDP, MMC, UFT, etoposide) in terminal gastric cancer].

CDDP, MMC, UFT and Etoposide (PMUE)-combined therapy was applied to 60 cases of terminal gastric cancer to examine its effectiveness. PMUE therapy consists of i.v. injection of CDDP 75 mg/body and MMC 10 mg/body on day 1, i.v. injection of Etoposide 50 mg/body on days 3, 4 and 5 and consecutive daily administration of UFT 400 mg/body, with 3 weeks as one course. Of 42 cases having estimable lesions, 23 (53.8%) showed high rate of effectiveness (PR). Especially, of 23 cases receiving no previous treatment, 15 (65.2%) benefitted by the therapy (PR) and 9 (69.2%) of 13 non-resected cases, to a wonderful extent. Five non-resected cases showed such a reduction in tumor size as made gastrectomy possible. As for the prognosis, one year-survival rate was 34.3, 49.0 and 16.0% for all 42 cases, 23 effective cases and 19 ineffective cases, respectively, with significant (p less than 0.001) prognostic prolongation for effective cases compared with ineffective ones. Side effects were digestive symptoms (85.7%), epilation (81.0%) and myelopathy (73.8%), which were all transitory and recovered. The present PMUE therapy was regarded as one of the best combined chemotherapies for terminal gastric cancer.

Potassium channels from NG108-15 neuroblastoma-glioma hybrid cells. Primary structure and functional expression from cDNAs.

The complete amino acid sequences of two potassium channel proteins from NG108-15 neuroblastoma-glioma hybrid cells have been deduced by cloning and sequencing the cDNAs. One of these proteins (NGK2) is structurally more closely related to the Drosophila Shaw gene product than to the Shaker and Shab gene products, whereas the other (NGK1) is identical with a rat brain potassium channel protein (BK2) which is more closely related to the Drosophila Shaker gene product. mRNAs derived from both the cloned cDNAs, when injected into Xenopus oocytes, direct the formation of functional potassium channels with properties of delayed rectifiers.

Blood clotting factor IX BM Nagoya. Substitution of arginine 180 by tryptophan and its activation by alpha-chymotrypsin and rat mast cell chymase.

Factor IX BM Nagoya (IX Nagoya) is a natural mutant of factor IX responsible for severe hemophilia B. A patient with this mutant is characterized by a markedly prolonged ox brain prothrombin time. IX Nagoya was purified from the patient's plasma by immunoaffinity chromatography with an anti-factor IX monoclonal antibody column. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis showed that treatment of IX Nagoya with factor XIa/Ca2+ resulted in cleavage only at the Arg145-Ala146 bond. Reversed-phase high performance liquid chromatography of a trypsin digest of IX Nagoya showed an aberrant peptide, which was further digested with proteinase Asp-N. Primary structure analysis of one of the Asp-N peptides revealed that Arg180 is replaced by Trp. An essentially complete (99%) amino acid sequence of IX Nagoya was obtained by sequencing fragments derived from a lysyl endopeptidase digest in which no other substitutions in the catalytic triad or substrate binding site were found. We also found that IX Nagoya is activated by alpha-chymotrypsin or rat mast cell chymase by monitoring the rate of factor X activation using a fluorogenic peptide substrate in the presence of factor VIII, phospholipids, and Ca2+. These results indicate that the substitution of Arg180 by Trp impairs the cleavage by factor XIa required for activation of this zymogen and that the substitution causes hemophilia BM.

Structurally similar but functionally distinct factors, IRF-1 and IRF-2, bind to the same regulatory elements of IFN and IFN-inducible genes.

Viral infections commonly induce expression of type I interferon (IFN) genes. The induction is transient and involves transcriptional activation wherein a positive factor, IRF-1, binds to upstream regulatory cis elements. In the present study we report the isolation of a cDNA encoding a novel factor, termed IRF-2, that interacts with the same nucleotide sequence elements as IRF-1. Both genes are inducible not only by virus but also by IFN. Unlike IRF-1, IRF-2 does not function as an activator; rather, it suppresses the function of IRF-1 under certain circumstances. Our results suggest that transcription of the IFN and IFN-inducible genes is regulated by two similar trans-acting factors that apparently compete for the same cis-acting recognition sequences, but which have opposite effects.

Endotoxaemia, pulmonary complications, and thrombocytopenia in liver transplantation.

Plasma endotoxin was measured in 64 patients undergoing primary liver replacement. Endotoxin concentrations increased during the anhepatic phase of the operations, and remained high for several days. Although the severity of endotoxaemia did not correlate with duration of the anhepatic phase, there was a correlation between endotoxaemia and the need for perioperative platelet transfusions, ventilator dependency postoperatively, and one-month case-fatality.

Possible finger structure in gene A protein of Microviridae.

Microvirid phages alpha 3 and phi K encode for A protein which functions in initiation of the viral DNA synthesis. By nucleotide sequencing analysis, we have found that each gene A protein has 'finger motif structure' which conserves two cysteine and histidine residues similar to the consensus sequence deduced from more than thirty finger motifs reported from many eukaryotic regulatory proteins. In closely related phages, phi X174 and G4, we have detected the same structures in their gene A proteins also.

Interleukin-2 receptor beta chain gene: generation of three receptor forms by cloned human alpha and beta chain cDNA's.

Interleukin-2 (IL-2) binds to two distinct receptor molecules, the IL-2 receptor alpha (IL-2R alpha, p55) chain and the newly identified IL-2 receptor beta (IL-2R beta, p70-75) chain. The cDNA encoding the human IL-2R beta chain has now been isolated. The overall primary structure of the IL-2R beta chain shows no apparent homology to other known receptors. Unlike the IL-2R alpha chain, the IL-2R beta chain has a large cytoplasmic region in which a functional domain (or domains) mediating an intracellular signal transduction pathway (or pathways) may be embodied. The cDNA-encoded beta chain binds and internalizes IL-2 when expressed on T lymphoid cells but not fibroblast cells. Furthermore, the cDNA gives rise to the generation of high-affinity IL-2 receptor when co-expressed with the IL-2R alpha chain cDNA.

Nucleotide sequences of immunoglobulin-epsilon pseudogenes in man and apes and their phylogenetic relationships.

To understand the phylogenetic relationships between hominoids, the nucleotide sequences of immunoglobulin-epsilon processed pseudogenes from chimpanzee, gorilla and orangutan were determined. The basic structures of these processed pseudogenes agreed with their human counterpart. Although the degrees of nucleotide differences between man and the African apes had no statistical significance, all the analytical data examined supported the theory that chimpanzee is the closest relative of man. This result was consistent with that deduced by our recent qualitative study. Studies on the nucleotide sequences of globin genes have suggested that the molecular clock runs more slowly in hominoids than in non-hominoid primates. According to the present data, however, further retardation of the evolutionary rate was not observed in the human lineage. Assuming that orangutan diverged 14 million years ago and that the evolutionary rate between the orangutan lineage and the lineage leading to the other three species is constant, the divergence dates of chimpanzee and gorilla were estimated to be 4.9(+/- 0.9) and 5.9(+/- 0.9) million years ago, respectively.
