Cellular elements in the resistance to candida infection in mice. I. Contribution of T lymphocytes and phagocytes at various stages of infection.

Live organisms (cfu) of Candida albicans per organ were counted 1 hr and 1 to 20 days after an intravenous inoculation into various groups of mice which had distinct levels of immunologic or non-immunologic defense mechanisms. a) The number of cfu in the liver decreased progressively in normal mice, but those in the kidney maintained a constant level during the observation period. b) The number of cfu in the liver decreased progressively also in nude mice. In their kidneys, however, cfu increased progressively at a late stage of infection. c) In lethally irradiated AKR of nude mice in which phagocyte functions were severely depressed, the number of cfu increased progressively in both liver and kidney from the initial stage of infection. d) In immunized AKR mice, growth of C. albicans was suppressed at late stages of infection. Such protective immunity could be transferred partly with immune lymphoid cells but not with hyperimmune serum in the experimental system employed. In protection against candida infection, non-immune phagocytosis and T cell-mediated immunity appear to be required at the early and late stages of infection, respectively.

A synthesis of 3',4'-dideoxykanamycin B.

3',4',-Dideoxykanamycin B, the kanamycin B derivative that is active against resistant bacteria, was prepared from kanamycin B via N-tosylation, 3',4'-O-sulphonylation, 3',4'-unsaturation, and hydrogenation. The unsaturated intermediate was obtained from the 3',4'-di-O-sulphonyl derivatives by the action of sodium iodide in N,N-dimethylformamide; if zinc dust was added in this reaction, aziridine derivatives were formed. Removal of the tosyl group was successfully performed by using sodium in ammonia-ethylamine.

New phosphoramidates as protecting groups in ribooligonucleotides synthesis.

N, N-Dimethyl-p-phenylenediamine, glycine amide and p-methylthioaniline were condensed with uridine 5'-phosphate and the phosphoramidates obtained were tested for their stability in anhydrous pyridine, 50% aqueous pyridine or 80% acetic acid. The p-methylthioanilidate (IIc) was oxidized to give p-methylsulfoxylanilidate of uridine 5'-phosphate (IId) which was found to be 5 times more stable than the p-methylthio compound. The p-methylsulfoxylanilidate of 2'-O-benzoyluridine 3'-phosphate was condensed with the mononucleotide to yield the dinucleotide, MMTrU(OBz)-p-U(OBz)-p in 28% yield.

Tolerance to the respiratory effects of marijuana in man.

The respiratory depressant effect of marijuana was studied by means of carbon dioxide respiratory response curves in eight healthy male subjects participating in a study of the effects of a 90-day ad libitum intoxication to marijuana. Subjects were studied in the morning after refraining from marijuana use since the previous evening. Respiratory response curves were done before smoking and at 15, 30 and 60 minutes after smoking two 900-mg cigarettes containing 2.2% delta9-tetrahydrocannabinol. Studies were conducted 2 to 3, 5 to 6 and 8 to 9 weeks after onset of intoxication. There was a significant change from base-line controls, indicating respiratory depression following marijuana smoking. A significant change in the time-effect curve over these three study sessions, indicative of tolerance, was noted. The degree of pulse elevation also decreased over the three study sessions, but this decrease in effect was not statistically significant.

Chronic mucocutaneous candidiasis accompanied by enhanced antibody production.

In four cases of chronic mucocutaneous candidiasis (CMCC) enhanced antibody production against candida antigen was detected in association iwth depressed cellular immunity. These patients showed extremely high agglutinin titre to candida antigen, while they were unable to mount delayed hypersensitivity reactions to candida antigen, tuberculin-purified protein derivative (PPD) or to dinitrochlorobenzene (DNCB). They also showed prolonged survival of skin homografts. There was no correlation between migration inhibitory factor (MIF) production and blast transformation in response to candida antigen.
