A gel mobility shift assay was used to examine nuclear extracts of muscle and nonmuscle cell lines for a factor or factors that interact with the functional promoter segment of both the human cardiac and skeletal alpha-actin genes. A single major band of altered mobility was seen with nuclear extracts from C2 myoblasts and myotubes and L8 myoblasts and myotubes as well as several non-muscle cell lines. Competition experiments with nuclear extracts from both muscle and non-muscle cells localized the region of binding to a 10-base pair sequence of the cardiac alpha-actin gene containing the functional CArG box promoter domain. Similar results and binding site specificities were obtained with a DNA fragment from the human skeletal alpha-actin promoter. The CArG box binding factor was shown to be distinct from a CAAT box binding factor. A molecular weight of 67 kDa for the CArG box binding factor was determined by photoaffinity labeling of the complex and sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Protease treatment resulted in a smaller fragment that still bound the cardiac alpha-actin promoter fragment and may represent a structurally distinct DNA-binding domain. We discuss how muscle-specific expression of the sarcomeric alpha-actin genes might be achieved with a ubiquitous transcription factor.

Three cases of inter-sterno-costo-clavicular ossification (ISCCO) associated with chronic tonsillitis are reported. Two cases had additional symptoms of pustulosis palmaris et plantaris (PPP). All the patients underwent tonsillectomy. The histological changes of the palatine tonsils of case 1 and case 2 were characterized by so-called focal small lacunar ulcers and periarteriolar fibrosis (onion-skin lesion). Some multinucleated giant cells were observed in case 2. Following tonsillectomy, symptoms in the clavicular region and the PPP improved. It is possible to consider that ISCCO is caused by focal tonsillar infection.
Proliferative cell indices (PCI) in 20 cases of endometrial adenocarcinoma were obtained by staining DNA polymerase alpha. The PCI ranged from 11.1 to 42.1, averaging 24.7 +/- 8.7. Four cases of stage III all exhibited fairly large PCI (30.4-32.9). In contrast, in 16 cases of stage I the values were spread over a wide range. In 13 cases with histological grade (HG) 1 of stage I, larger PCI were obtained in the nuclear grade (NG) 2 group; the mean PCI values of HG 1-NG 1 and HG 1-NG 2 were 16.5 +/- 4.6 and 25.6 +/- 5.2, respectively. Because of the good correlation between PCI and HG or NG, PCI may be useful as an additional prognostic factor in endometrial adenocarcinoma, especially in stage I cases.
A long-term cell culture epitheloid cell line (NPC-F/L) was established from the fibroblast-dominant cell line derived from nasopharyngeal carcinoma tissue by cocultivation with an Epstein-Barr virus (EBV)-transformed lymphoblastoid cell line of adenoid origin. The cells were investigated by karyotyping, by immunofluorescent testing for EBV-associated nuclear antigen (EBNA), early antigen (EA), and viral capsid antigen (VCA) and by a light microscopy. The cells were shown to be epithelial not only by morphology but also by immunocytochemical staining with monoclonal antibody to cytokeratins and they expressed EBNA and EA but not VCA. The cells developed histologically undifferentiated squamous cell carcinoma when inoculated subcutaneously into the back of athymic nude BALB/c(nu/nu) mice, which were similar to the human tumors from which the fibroblastoid cell line was established. The presence of EBNA in touch smears prepared both from the biopsy tissue of human and nude mice was demonstrated. Karyotyping analysis indicated an aneuploid chromosome with a model number of 74.
Morphometric evaluation of 20 rhesus articular cartilage samples were correlated with hexosamine and an 18-hour incorporation of 35SO4- as a measure of proteoglycan production on samples from the same joint. Reduced chondrocyte cellularity was the basis of the reduced maintenance of the matrix by the chondrocyte. In the more cellular cartilage, the matrix/lacunae area ratio was less than 25, and the less cellular group had a ratio of greater than 40. An inverse correlation existed between morphometric cartilage matrix/lacunar area ratio and hexosamine content. A significant difference of 35SO4 incorporation was not seen between the three morphometric grades. Morphometric assessment reduces the subjectivity of articular cartilage evaluation.

The inhibitory action of pirenzepine on acid secretion of isolated guinea pig parietal cells was investigated by the aminopyrine method. Pirenzepine markedly inhibited acid secretion of isolated parietal cells induced by carbachol in a dose-dependent manner but showed no inhibition on acid secretion stimulated by histamine. These results may suggest a direct action of pirenzepine on muscarinic receptors in parietal cells.
We analyzed various biochemical variables of the bones in fetal rats whose dams were protein-energy malnourished. Dams were randomly divided into two groups and fed either a 6% protein diet as a malnourished group or a 20% protein diet as a control, from day 13 of gestation to day 22, when fetuses were removed. Hexosamine and hydroxyproline contents of the calvaria and hexosamine contents of long bones were greater in the malnourished group than in the controls. Sequential extractability of collagen differed among various bones in the malnourished group and controls. The ratio of alpha:beta obtained from SDS-polyacrylamide gel of neutral salt-soluble collagen tended to increase in the long bones and mandible, and decrease in the calvaria and ribs in the malnourished group. Also, the ratio of alpha 1:alpha 2 tended to be lower in the malnourished group than in the control group in all bones. Protein-energy malnutrition during pregnancy has shown to affect biochemical composition of various fetal bones.
Although the topical treatment with steroids is the widespread method for olfactory disorders in human, the reports are limitted to clinical observations. The aim of this report is to evaluate the effect of topically applied of steroids in mice on the repairing course by the experimental olfactory disorders caused by zinc sulfate. The olfaction of mice was estimated by the record of behavioral task, using Cycloheximide, commonly used as repellent in rodents. When mice taste Cycloheximide for the first time, immediately the mice experience a very disagreeable taste leading to the simultaneous memory of the odor of Cycloheximide. Those who once experienced the taste and odor of Cycloheximide, will avoid simply by the odor next time. Two bottles were offered to mice, one containing distilled water and the other 0.01% Cycloheximide. When the mouse drank the distilled water, the response was interpreted as a correct response. When the Cycloheximide solution was chosen, an error. Twenty four mice were tested at ten times during a 10-minute trial, and the rates of correct response were counted. From the next day after the treatment of zinc sulfate, nasal application of 0.1% beta-methasone (group A), or nasal application of distilled water (group B) were applied to each group of twelve mice for five days. The results were as follows. 1. In the first experiment, the correct response was 85% (group A), and 80.8% (group B), however at the second trial, all responses were correct.(ABSTRACT TRUNCATED AT 250 WORDS)
Histidine decarboxylase (HDC) activity in the oxyntic gland and gastric volume were measured in rats treated with tetragastrin, cimetidine or omeprazole. HDC activity was dose dependently activated by not only tetragastrin but also cimetidine and omeprazole treatment. Histamine concentration in the oxyntic gland was reduced, but the amount of histamine in the gastric contents was increased by tetragastrin treatment. In rats premedicated with cimetidine or omeprazole, histamine concentration in the oxyntic gland and the amount of histamine in the gastric contents were not changed by administration of tetragastrin. It was concluded that tetragastrin activated HDC which increased histamine release into the gastric contents. Cimetidine and omeprazole induced the secretion of endogenous gastrin, leading to the activation of HDC.
Polyamines are compounds required for initiation of rapid cellular growth and differentiation in many cell types. Ornithine decarboxylase is the rate limiting enzyme in polyamine synthesis. Fasting and refeeding regulates the activity of ornithine decarboxylase and polyamine content in the intestinal tract. We tested the hypothesis that polyamines regulate cell growth via the Na+/H+ exchanger which is believed to be intimately involved in cell growth. Ileal Na+/H+ activity was therefore examined in control, fasted, refed fasted, and in rats given the specific inhibitor of ornithine decarboxylase alpha-difluoromethylornithine. A well-validated ileal brush border membrane vesicles for the study of Na+/H+ exchange activity was utilized. Fasting markedly decreased while refeeding stimulated Na+/H+ exchange activity at all times studied (P less than 0.05-0.001). Maximal uptake of Na+ at 5 min was 3.12 +/- 0.05, 2.5 +/- 0.05 and 2.22 +/- 0.05 nmol/mg protein in refed, control and fasted rats respectively. Kinetics of amiloride sensitive Na+/H+ exchanger showed a Vmax of 17.1 +/- 3.5, 8.0 +/- 0.64 and 4.7 +/- 1.1 nmol/mg protein per 5 s in refed fasted, control and fasted rats respectively Km values were not significantly different between the groups studied. 2% alpha-difluoromethylornithine given in the drinking water abolished the stimulation in Na+/H+ exchange activity in refed fasted rats. These results suggest a close relationship between polyamines and Na+/H+ activity in the intestinal mucosa of rats.
It was confirmed that histamine, cholinergic drugs and gastrin separately produce acid secretory effects of isolated parietal cells. However, the acid secretory response of isolated parietal cells to gastrin stimulation is known to be weaker than that to histamine or cholinergic drugs, which is different from the acid secretory response found in vivo studies. Tetragastrin and pentagastrin were less potent than histamine and carbachol in stimulating acid secretion in our previous study performed using parietal cells isolated from guinea-pigs. In this study, the effect of gastrin extracted from the pyloric antrum of the guinea-pig on parietal cells isolated from the guinea-pig was investigated based on the assumption that the acid secretory response of isolated parietal cells to gastrin stimulation will be especially good if gastrin isolated from the same species is used.
The antibody activities against Aspergillus fumigatus and Candida albicans by indirect hemagglutination (IHA) and counterimmunoelectrophoresis (CIE) were examined twice at a month's interval in 251 sera from 169 male and 82 female patients admitted to the Higashi Nagoya National Hospital for pulmonary diseases. The patient population was composed of 226 patients with active or cured pulmonary tuberculosis including 25 patients complicated with pulmonary aspergillosis and 25 other lung diseases. In our tests, antigens used were crude supernatants of culture-broth which are the generous gifts of Torii Pharmaceutical Company. The antibody activity against Aspergillus by IHA was positive in 2.9% of the sera in the first and in 0.9% in the second test and was positive against Candida by IHA in 44.9% in the first and in 44.0% in the second test (Table 1). As regards patients with pulmonary tuberculosis, the population whose serological reaction was positive against Candida did not increase with age (Table 2). Two results of the antibody activities against Aspergillus and Candida by CIE were the same. The CIE results were positive in 19.5% against Aspergillus and in 16.3% against Candida (Table 3). In CIE test, 4.3% were positive against both Aspergillus and Candida. The causes of high positive rate in IHA against Candida may be due to (1) difference in the antibody, namely, antibody measured by IHA mainly composed of IgM and that by CIE composed of IgG and (2) difference in the virulence between Candida and Aspergillus.(ABSTRACT TRUNCATED AT 250 WORDS)
A multicentre, randomized, double-dummy control trial compared the efficacy of two famotidine dosage regimens for the treatment of acute gastric mucosal lesions. A total of 209 patients, aged 16-85 years, were randomized to treatment with either 10 mg famotidine twice daily or a 20 mg bedtime dose. Treatment was continued for 2 weeks unless healing occurred earlier. Endoscopy was performed before treatment and at the end of weeks 1 and 2 of treatment. Pain and haemorrhages disappeared in greater than 90% of patients after 2 weeks of treatment with either famotidine dosage. Mucosal erosions healed in more than two out of three patients by the end of the treatment period. No statistical difference was seen between the two dosage groups. Since 20 mg famotidine given at bedtime was as effective as twice daily dosing, it is recommended for the treatment of acute gastric mucosal erosions.
The association of an arteriovenous malformation (AVM) and a cerebral aneurysm is not so rare. Many reports have analyzed its localized relationship and studied hemodynamically the development of the cerebral aneurysm. However its therapeutic measures have not been established so far. At this time, authors have experienced 9 cases of AVM associated with cerebral aneurysm, and have studied suitable therapeutic measures. All 9 cases developed with intracranial hemorrhage, and in 6 of 9 cases, cerebral aneurysm was recognized in the feeding arteries or feeding system of the AVM (Group I). In the other 3 cases, localization of cerebral aneurysm was hemodynamically considered to have less of a relationship with AVM (Group II). It was characteristic that cerebral aneurysm in Group I was multiple, and developed frequently in the peripheral regions of the cerebral artery, especially in feeding system of the AVM itself. An operation was carried out in all 9 cases, and in Group I, 2 of 6 cases were operated on in one stage. The other 4 cases because of difficulty of management in the same operative field, were divided into two stages. In cases with clearly hemorrhagic sources, operation was firstly performed on the hemorrhagic source, and its residual focus was operated on after several months. However, in cases with unknown hemorrhagic source, cerebral aneurysm existing on the proximal side was operated on firstly. On the other hand, in Group II, when the operation was performed at one or two stages, the hemorrhagic source was dealt with first.(ABSTRACT TRUNCATED AT 250 WORDS)
The refractive state in darkness (dark focus of accommodation) was measured in 224 eyes of 112 young subjects with a Canon Autoref R1 and differences of the refractive state in dark and bright rooms were studied. The dark focus of accommodation was also compared with refractive error after instillation of 1% cyclopentlate hydrochloride. Accommodation was almost entirely responsible for myopic shift in the dark focus of accommodation. Myopic shift was larger in hyperopic eyes than in myopic eyes, but hyperopic shift occurred in markedly hyperopic eyes. In emmetropic and myopic eyes, the degree of hyperopic shift by cycloplegia had no parallel relationship with that of myopic shift in the dark focus of accommodation.
