Effects of concentration and duration of toluene exposure on Sidman avoidance in rats.

Fifteen rats in which responding under the Sidman avoidance schedule had been established were first exposed to air for 4 hr, then randomly to 350, 750 or 1,500 ppm of toluene vapor for 1, 2 or 4 hr, following which the effects on lever pressing behavior were investigated for 80 min after the termination of toluene exposure. The effect of concentration, exposure duration and test period on lever pressing of toluene was revealed clearly in the test. Using the technique adopted in this experiment, one could be determining the minimal effective exposure condition in terms of toluene concentration and exposure duration is 1,500 ppm for 2 hr. These findings suggest the usefulness of this technique in assessing the behavioral toxicity of toluene.

[An experimental study of nursing work load on the low back (author's transl)].

The relation between work postures and muscle load, the differences of muscle load in holding two different shaped objects, a human model and a box each weighting 20 kg, and the effects of wearing a belt and a corset to reduce the low back muscle load were studied in the experimental nursing work on physically and/or mentally handicapped patients. Four female healthy college students exercised a static and a dynamic nursing work designed for the experiment. E.M.G., heart rate, subjective symptoms and maximum sustaining time were measured during the work. Results are as follows: 1) The low back muscle load increased only in sustaining the forward bending posture. 2) The forward bending posture with a load weighing 20 kg caused shortening of the time to sustain the same posture. 3) The muscle load by holding the human model was larger compared to that by holding the box, and the muscle load was different according to the shape of the holding objects. 4) Wearing a belt and a corset was effective to reduce the low back muscle load by raising the abdominal pressure.

[Evaluation of toluene toxicity using Sidman avoidance behavior by lever pressing in rats (author's transl)].

Effects of single exposure to toluene on the operant behavior in rats were studied. The animals, in which the behavioral base line had been established, were exposed to 0, 1,000 and 3,000 ppm of toluene vapor for four hours, and the effects on the behavioral pattern of lever pressing were investigated for one hour after the end of exposure using Sidman avoidance test. 1) Exposure to 0 or 1,000 ppm of toluene did not affect the established operant behavior of the rats. 2) Exposure to 3,000 ppm produced increased responses, shortening of the inter-response-time, and no change in shock counts at the early stage of Sidman avoidance test when compared with those before exposure. 3) The behavioral change indicated here appears to be caused by a temporary breakdown of the established timing of lever pressing. 4) The effect of toluene on the operant behavior was strongest immediately after the aviodance test begun and the behavior recovered in about an hour. From the results of our experiment it can be said that the experimental methods used in this study is useful to evaluate the toxicity of toluene to the central nervous system of the animals.

Effects of single exposure to toluene on Sidman avoidance response in rats were studied. The animals, in which the behavioral base line had been established, were exposed 0, 1,000 and 3,000 ppm of toluene vapor for 4 hours and the effects on the behavioral pattern were investigated using Sidman avoidance test for one hour after the termination of toluene exposure. 1) Exposure to 0 or 1,000 ppm of toluene did not produce marked change on the operant behavior. After the exposure to 3,000 ppm, the animals produced increased responses with shortening of the inter-response-time (IRT) and no changes in shock counts from the beginning to the 50 min. zone of the Sidman avoidance test when compared with those before exposure. The behavioral change indicated here appears to be caused by a temporary breakdown of the established timing of lever pressing. 2) The effect of toluene on the operant behavior was strongest immediately after the termination of the exposure and the behavior recovered in about an hour. This result suggested that toluene is rapidly transfered to and removed from the brain.

A family of Holt-Oram syndrome.

A family of Holt-Oram syndrome is reported. Twenty of 41 living members in the last 3 generations were examined. The propositus expressed typically the cardinal features of this syndrome: atrial septal defect, patent ductus arteriosus, cardiac arrhythmias and conduction block of various types, and hypoplasia of the left thumb. Two individuals were clinically diagnosed to have atrial septal defect without any upper limb anomaly, and 3 showed upper limb anomaly without any sign of congenital heart disease. Other 5 members showed only minor abnormalities, such as funnel chest or mild right ventricular hypertrophy on ECG and VCG. Some of the skeletal abnormalities observed were hitherto undescribed ones. These include generalized hypoplasia of left hand and downward displacement of right sterno-clavicular joint.

Development of systemic immunologic responses against hepatic metastases during gene therapy for peritoneal carcinomatosis with retroviral HS-tk and ganciclovir.

Gene therapy with retroviral mediated gene transfer of the herpes simplex thymidine kinase (HS-tk) gene into a tumor mass confers sensitivity of the tumor cells to ganciclovir (GCV). Tumor-specific immunologic responses may develop following treatment of the primary tumor with retroviral HS-tk and GCV. In the present study we assessed whether GCV treatment of HS-tk transduced colon cancer (TK+) implanted in the peritoneal cavity induced a systemic antitumor response that would inhibit growth of a second wild-type (TK-) tumor implanted in the liver. DHDK12 rat colon cancer cells were transduced in vitro with the retroviral HS-tk vector and established as a permanent cell line (TK+ cells). TK+ or TK- DHDK12 cells (6x10(6) cells) were injected intraperitoneally on day 0 into BD-IX rats. On day 10, TK- cells (3x10(6) cells) were injected into the liver in all the groups. The animals were then treated with GCV (150 mg/kg) for 13 days. TK+ peritoneal tumors underwent significant regression during therapy with GCV (0.05+/-0.004 g; n=7) compared to wild-type (TK-) tumors (2.2+/-0.7g; n=6) (P<0.05). The volume of TK- tumors in the liver was significantly lower in GCV-treated rats with TK+ peritoneal tumors (12.5+/-8.3 mm3) compared to rats with TK- peritoneal tumors (96.7+/-18.1 mm3) (P<0.05). Histology of the liver tumors in the TK+ groups showed a dense monocytic infiltrate with fibrosis and only occasional viable tumor cells. Gene therapy with retroviral HS-tk vectors may provide a novel approach to treatment of gastrointestinal cancer by both direct cytotoxicity and an indirect mechanism that may include enhanced immuno logic responses against disseminated disease.

The effect of neutrophil depletion on reperfusion arrhythmias during intracoronary thrombolysis using the leukocyte removal filter.

To evaluate the effects of neutrophil depletion on reperfusion arrhythmias using a leukocyte removal filter (LRF), the authors examined several cardiovascular variables in three patients with myocardial infarction who underwent successful thrombolytic therapy with LRF compared with control patients without LRF. Coronary angiography performed within 4 hr of onset revealed total occlusion in the proximal segment of the left anterior descending (LAD) coronary artery, without collaterals, in all patients. Thrombolysis was performed by intracoronary administration of urokinase (10,000 IU/ml, 1,000,000 IU total) mixed with filtered autologous blood. LRF was composed of a nonwoven polyester fabric (1.8 microns, 4.6 g). Removal rates of leukocytes and platelets by LRF were 98-100% and 98%, respectively. Average duration of the ischemic episodes, maximum ST segment elevation, and integral of T segment depression did not differ in the two groups. However, the frequency of ventricular premature beats (VPB) during the first 30 min after reperfusion were significantly lower in the LRF treatment group (3.8%) compared with the control group (10.7%). Segmental shortening, measured by echocardiography one hour after recanalization, exhibited higher values in the LRF treated patients than control patients, without significant difference. Data suggested that LRF is effective in reducing reperfusion arrhythmias during intracoronary thrombolysis and activated neutrophils may play an important role in stunned myocardium.

Successful treatment of angioimmunoblastic lymphadenopathy with dysproteinemia by local irradiation.

We report an 85-year-old man presenting with angioimmunoblastic lymphadenopathy with dysproteinemia (AILD) of the palatine tonsil with cervical and axillary lymph node enlargement, pleural effusion and hypergammaglobulinemia. The patient was treated in Internal Medicine with combined chemotherapy with steroids which resulted in a complete remission. However, he returned almost 1 year later with an ulcerative lesion in the left palatine tonsil, with no lymph node involvement. Considering that the patient was an elderly person, chemotherapy had been administered during the initial admission, tumor cells appeared to originate from T cells and relapse occurred only in the local lesion, the lesion was treated by local irradiation using 60Co at a dose of 40 Gy, which produced a complete resolution. Radiotherapy appeared to be effective for the treatment of localized lesions of AILD.
