Characterization of two monoclonal antibodies which recognize different subpopulations of chicken T lymphocytes.

Distribution among peripheral T lymphocyte subpopulations and biochemical properties of the chicken lymphocyte surface antigens defined by monoclonal antibodies (mAbs) Lc-4 and Lc-6 were examined. Two-color immunofluorescence analysis revealed that Lc-4 and Lc-6 antigens were expressed on mutually exclusive subpopulations of peripheral T lymphocytes but not on B lymphocytes. Lc-4 mAb precipitated a polypeptide with apparent molecular mass of 35 and 65 kilodalton under reducing and non-reducing conditions, respectively. These results indicated that the antigen recognized with Lc-4 was closely similar in tissue distribution and biochemical property to mammalian CD8 antigen.

[Etoposide delivery to malignant brain tumors by induced hypertension with angiotensin II].

Etoposide delivery to 9L gliosarcoma model in Fischer 344 rats was evaluated. Etoposide 2 mg/body was given intravenously (IV) or intracarotidly with (IHIC) or without (IC) intravenously administrated angiotensin II. Mean etoposide concentration of tumors was 5.08 micrograms/g for IHIC group, 2.27 micrograms/g for IC group and 0.70 micrograms/g for IV group. The difference in etoposide concentration between IHIC group and IV group was statistically significant (p less than 0.05). In addition, etoposide concentrations of the normal brain tissues were lower than concurrent plasma concentrations in all groups. The etoposide concentration was increased in the tumor and very low in the normal brain tissues in Fischer 344 rats with 9L gliosarcoma by induced hypertension with angiotensin II. These studies might suggest that intraarterial chemotherapy by induced hypertension with angiotensin II was useful on treatment of malignant brain tumors.

Preliminary comparisons of the biological properties of two strains of feline immunodeficiency virus (FIV) isolated in Japan with FIV Petaluma strain isolated in the United States.

Two strains of feline immunodeficiency virus (FIV) were isolated directly from peripheral blood mononuclear cells (PBMCs) of Japanese domestic cats or indirectly from PBMCs of specific pathogen free (SPF) cats inoculated with whole blood from naturally infected cats with FIV by cocultivation with primary PBMCs from SPF cats. Two isolates, designated as FIV TM 1 and FIV TM 2, had a lentivirus-like morphology by electron microscopy, a tropism for interleukin-2 dependent T-lymphocytes and Mg2+-dependent reverse transcriptase activity. By immunoblotting the isolates gave bands at 130, 48, 44, 40, 28, 17, and 13 kDa, and these bands except 130 kDa were detected in FIV Petaluma strain when reacted with the plasma of cats infected naturally with FIV TM 1 strain.

Establishment of a feline T-lymphoblastoid cell line highly sensitive for replication of feline immunodeficiency virus.

Interleukin-2 dependent feline T-lymphoblastoid cells designated as MYA-1 cells were established. The cells were free from exogenous retroviruses and sensitive for replication of feline immunodeficiency virus (FIV). FIV can grow more efficiently in MYA-1 cells than in feline primary peripheral blood mononuclear cells. This line of cells will be useful not only for isolation and propagation of FIV, but also for further investigation of properties of FIV.

Serological and genetical relationships of three herpesvirus strains from salmonid fish.

Serological and genetical relationships of three salmonid herpesviruses [strain H-83, OO-7812 strain of Oncorhynchus masou virus (OMV) and Herpesvirus salmonis] were investigated. Although the serological relationships of these viruses were not distinctly demonstrated by cross-neutralization test, cross enzyme-linked immunosorbent assay revealed that strain H-83 and OMV appeared to be serologically more closely related to each other than to H. salmonis. Strain H-83 was genetically similar to OMV but distinct from H. salmonis by restriction endonuclease cleavage analysis and DNA-DNA hybridization test. These results indicated that strain H-83 should be classified under the same group as OMV, and H. salmonis belong to another group of salmonid herpesviruses.

Activation of trout macrophages and production of CRP after immunization with Vibrio anguillarum.

To examine the mechanism of the protection of rainbow trout (Salmo gairdneri) against Vibrio anguillarum in the early stage of immunization, the activation of macrophages and production of C-reactive protein (CRP) were investigated. Fish immunized with formalin-killed bacteria emulsified in Freund's complete adjuvant (FCA) resisted intraperitoneal challenge with living bacteria seven and ten days after immunization. The activation of macrophages was demonstrated by a significant increase of the chemiluminescent (CL) response and phagocytic activity. These fish also showed a significant increase of the CRP level in sera. Fish immunized with V. anguillarum alone or injected with FCA, however, did not resist the challenge. Though FCA itself increased CRP level and the sera enhanced phagocytic activity, increase of CL activity was weak. These results indicated that the increase of CL activity and opsonising effect of CRP on the phagocytosis of specifically activated macrophages concern to host defense in the early stage of infection.

Protective effect of N-acetyl chitohexaose on Listeria monocytogenes infection in mice.

A water-soluble oligosaccharide, N-acetyl chitohexaose (NACOS-6) was able to enhance the protecting effect of BALB/c male mice against Listeria monocytogenes infection, when administered intraperitoneally 24 hr before the challenge with this microbe. Significant decrease in number of microbes within the peritoneal cavity, spleen, and liver from the mice of NACOS-6-administered group was not observed 1 day after the infection but 4 days after the infection. Administration of NACOS-6 enhanced the delayed-type hypersensitivity response against sheep red blood cells (SRBC) or heat-killed L. monocytogenes. Splenic T lymphocytes from mice administered NACOS-6 released macrophage activating factor (MAF). These results suggested that NACOS-6 was also able to elevate the function of cellular immunity. Macrophages treated with a combination of NACOS-6 and the culture supernatant of splenic T lymphocytes from mice administered NACOS-6, "NACOS-6 sup," were found to exert a fairly strong growth-inhibitory effect on L. monocytogenes. Interferon-gamma (IFN-gamma) and interleukin 2 (IL-2) were able to enhance the growth-inhibitory effect on L. monocytogenes by the NACOS-6-treated macrophages.

Development of a ceramic heart valve.

A durable and thromboresistant ceramic heart valve comprised of a single crystal alumina disk and titanium nitride (TiN) valve ring has been developed. Blood compatibility was examined by scanning electron microscopy (SEM) examinations of the valves implanted in sheep for 35 (#1), 26 (#2), 20 (#3), 23 (#4), and 26 (#5) days. The single crystal alumina and TiN surfaces were free of platelet aggregation or fibrin networks, except for some depositions of fibrin and platelets on the outflow TiN ring in #3, and isolated red cells on the outflow TiN ring in #5. Durability testing under high pressure (1750 mmHg = 233 KPa) pulsatile conditions showed that the safety factor of the ceramic valve was more than seven times greater than anticipated. The ceramic valve is promising as an artificial heart valve.

An eight year experience with gradually longer interval postremission therapy for adults with acute leukemia.

Between January 1980 and March 1983, a study was conducted into the effects of postremission therapy on 20 patients with acute leukemia who had achieved complete remission through induction therapy. Postremission therapy consisted of cyclic administration of six combination therapies given at gradually longer intervals. Postremission therapy used DCMP (D, daunorubicin; C, cytosine arabinoside; M, 6-mercaptopurine; P, prednisolone), DCyMP (Cy, cyclocytidine), DCVP (V, vincristine), BHAC-DMP (BHAC, behenoyl-ara-c), BHAC-AMP (A, aclarubicin) and ACM-MP (ACM, aclacinomycin). Six combinations were given sequentially starting at one month interval, and then at 2, 3, 4, 5 and eventually 6 month intervals until 5 year survival was reached. The median remission duration was 38 months for acute myelogenous leukemia (AML), and 17 months for acute lymphoblastic leukemia (ALL). The median survival was 66 months for AML, and 33 months for ALL. The survival rate at 5 years was 60% for AML., 40% for ALL, and 50% in all 20 patients. Methotrexate and prednisolone were administered intrathecally for prophylaxis of CNS leukemia on Day 4 of each stage of postremission therapy. There was no CNS leukemia. This postremission therapy was shown to be effective in improving the prognosis of adults with acute leukemia.

[Attempt of analyzing the mandibular movement in three dimensions by computer graphics].

In clinic, simplified mandibular movement measuring devices are used for diagnosis of patients with stomatognathic dysfunction and recording maxillomandibular relation. It is difficult to understand mandibular movement in three dimensions from the path on the three planes, because the path of mandibular movement is in three dimensional space with time, not separately on each plane. The purpose of this study was to develop the three dimensional analyzing system by computer graphics. Mandibular movement was recorded by Mandibular Kinesiograph. The signals were sent to personal computer and the data were processed by the software. The basic specifications of the program were as follows: 1. Data could be observed from various viewpoints. 2. Necessary section of the simultaneous data could be displayed. 3. "C" was used for the software describing language. Following results were obtained: It was possible to analyze the path of mandibular movement in other coordinate axis and understand the path of mandibular movement in three dimensions more accurately.

[Left ventricular diastolic flow dynamics in dilated cardiomyopathy: a color flow imaging and pulsed Doppler echocardiographic study].

Left ventricular filling flow was evaluated at the mitral orifice and in the left ventricle (LV) using color flow imaging and pulsed Doppler echocardiography in seven patients with dilated cardiomyopathy (DCM) and 10 normal subjects. Using the apical approach, filling flow in early and late diastole was observed by two-dimensional color and M-mode color flow imaging, and flow velocity patterns at the mitral orifice and in the LV, 3 cm below the orifice, were analyzed by pulsed Doppler echocardiography. Color flow imaging disclosed slower movement of the inflow signals toward the apex in DCM than in the normal. With pulsed Doppler echocardiography, the delay in timing of peak velocity in the LV was distinctly greater in DCM than in the normal, although the mitral inflow velocity did not differ between DCM and the normal. The width of the filling flow, the rate of velocity reduction and the flow velocity in the LV did not differ between DCM and the normal. Thus, in DCM, the timing of filling flow in the LV is clearly delayed compared to the timing at the mitral orifice, and this cannot be explained solely by the mitral flow velocity pattern.

Optical measurement of dental cast profile and application to analysis of three-dimensional tooth movement in orthodontics.

Dental casts are frequently used for measuring the size, shape and position of a tooth. We have developed an optical apparatus for measuring a three-dimensional profile of a dental cast. The measurement is based on the triangular method which detects a laser spot on a cast by an image sensor. The apparatus is computer-controlled and designed to achieve a total measuring depth of 40 mm, a data acquisition time of 25 ms per point and a minimum scanning pitch of 0.1 mm. The depth and lateral resolutions are 0.05 and 0.1 mm, respectively, within a depth range of 25 mm. As a particular application of the apparatus, three-dimensional movement of teeth, including rotational movement, during orthodontic treatment, has been quantified by means of the superimposition of tooth profiles using a computer.

Antitumor effect of recombinant human lymphotoxin on a tumor line of human malignant glioma.

We examined the antitumor effect of recombinant human lymphotoxin (rHuLT) on a xenotransplantable human malignant glioma line. Tumor-bearing nude mice were treated with rHuLT for three weeks following four schedules: intratumoral injection of rHuLT 20,000 units once a week, twice a week, intravenous injection once a week and twice a week. The inhibition rate of tumor growth was 98.8%, 99.1%, 92.1% and 98.8%, respectively. Histologically, necrotic lesions were observed in the tumors of all treated mice. Thrombo-obstructive changes of tumor vessels were also seen in the tumors of mice after intravenous injection of rHuLT. None of the mice died as a result of this treatment in spite of significant body weight loss. These results indicate that rHuLT has a strong antitumor effect on a xenotransplantable human malignant glioma line.

[Clinical study of hypertensive cerebellar hemorrhage: surgical indication and measurement of volume of hematoma].

There have been a few reports available for determining surgical indications in hypertensive cerebellar hemorrhage based on volume of hematoma on computerized tomography (CT). The authors then studied the clinical results of hypertensive cerebellar hemorrhage, and the surgical indication based on clinical findings and volume of hematoma on CT scan was considered. Forty-five patients with hypertensive cerebellar hemorrhage diagnosed by CT scan who were hospitalized to Matsue Red Cross Hospital from January 1980 up to December 1986 were studied. The 25 male and 20 female patients ranged in age from 52 to 85 years, and 16 of them were operated on. The results were as follows: 1) The Kanaya's neurological grading tended to be high in the patients with cerebellar vermis hemorrhage or a large volume of hematoma (greater than or equal to 30 ml). 2) In patients with grade I or II and a moderate volume of hematoma (15-30 ml), the patient complicated with hydrocephalus should be treated with ventricular drainage. The patients with grade III and IVa should be treated with surgical therapy (suboccipital craniectomy and evacuation of the hematoma). The patients with cerebellar vermis hemorrhage should be treated with surgical therapy. The patients with a large volume of hematoma (greater than or equal to 30 ml) should be treated with surgical therapy. The patients with grade IVb and V should not be treated actively because the prognosis is bad.(ABSTRACT TRUNCATED AT 250 WORDS)

[Acute myelomonocytic leukemia with inv (16) (p13 q22) disappeared abnormal karyotype during complete remission].

A 21-year-old man was admitted to our hospital because of anorexia and general malaise in July, 1988. On admission, the white blood cell count of 18,600/microliters with 72% leukemic cells. The bone marrow aspirate showed 76.8% immature monocytes, 10% mature and immature eosinophils. Leukemic cells were 66.6% myeloperoxidase positive cells, and 20.6% naphthylbutyrate esterase positive cells. The lysozyme activity in urine was high. Cytogenetic analysis revealed the presence of 46 XY inv (16) (p13 q22). Under the diagnosis of acute myelomonocytic leukemia with eosinophilia (M4Eo) associated with inv (16) (p13 q22), one course of DCMP induction therapy was performed. After complete remission, the bone marrow aspirate showed disappearance of inv (16) (p13 q22), and associated with decreased residual leukemic cells.

[Intra-arterial infusion chemotherapy of advanced breast cancer--effects and side effects of adriamycin, 4'-epi-adriamycin and THP-adriamycin].

In 34 patients with primary advanced breast cancer, intra-arterial administration of ADR (50 mg X 3, total dose 150 mg, 10 cases), 4' epi ADR (50 mg X 3, 150 mg, 8 cases; 70 mg X 3, 210 mg, 10 cases) and THP-ADR (50 mg X 3, 150 mg, 6 cases) was performed, and its effects and side effect were analyzed. The clinical and histological response rate were superior in the ADR (150 mg) regimen and 4'-epi-ADR (150 mg) regimen. Signs of systemic toxicity such as gastrointestinal disorders, leukocytopenia and thrombocytopenia were the side effects in patients treated with THP-ADR, but the frequency of alopecia was lower. No cardiotoxicity was recorded in any of the patients. These results indicated that 4'-epi-ADR given the total dose of 150 mg in a single dosage of 50 mg was the most effective agent in intra-arterial infusion chemotherapy for advanced breast cancer.
