Phenotypic differentiation-linked growth inhibition in human leukemia cells by active vitamin D3 analogues.

Active vitamin D3 induced phenotypic differentiation of the human promyelocytic leukemia cell line, HL-60, cultured in serum-free medium. Upon exposure to 10(-10)-10(-7) M 1,25-(OH)2D3, 1,24S-(OH)2D3 or 1,24R(OH)2D3, monocyte-granulocyte-associated plasma membrane antigens of HL-60 cells detected by monoclonal antibodies, OKMI, 63D3 and Mo2, quantitated by fluorescence-activated cell sorter analysis, were increased time- and dose-dependently. After expose to 1,25-(OH)2D3, promotion of this antigenic expression was detected within 16 h, and the induction of differentiation continued until 96 h. The number of cells bearing transferrin receptors recognized by the monoclonal antibody, OKT9, and its density on the surface of HL-60 cells were decreased symmetrically. These effects appeared in parallel with the inhibition of cell growth, poly(ADP-ribose) content and de novo DNA-RNA synthesis. These findings indicate that 1,24S-(OH)2D3 stimulates differentiating induction of HL-60 like 1,25-(OH)2D3 in vitro and that the decrease of transferrin receptor is apparently correlated with the inhibition of proliferation.

Effect of tumor promoters on human T cell leukemia/lymphoma virus (HTLV)-structural protein induction in adult T-cell leukemia cells.

We assayed the capacity of tumor promoters to induce human T-cell leukemia/lymphoma virus (HTLV) structural proteins p19 and p24 from the HTLV genome-carrying adult T-cell leukemia (ATL) cell lines, MT-1 and KH-2Lo, and fresh ATL cells. Among the tested substances, 12-O-tetradecanoyl phorbol-13-acetate (TPA), 12-hexadecanoyl-phorbol-13-acetate (HPA) and teleocidin induced HTLV structural protein p19 and p24. This suggests that certain environmental substances, especially those known to be tumor promoters, may activate the HTLV-gene in ATL cells.

Isolation and characterization of proteinase from Candida albicans: substrate specificity.

Candida albicans was able to produce a keratinolytic proteinase (KPase) when cultivated in a medium containing human stratum corneum as a nitrogen source. The KPase was purified to 108.5-fold by ion-exchange chromatography and gel filtration. The molecular weight of the enzyme was estimated to be 42,000 by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and gel filtration through Sephacryl S-200, while the isoelectric point was determined to be at pH 4.5. The enzyme had an optimum pH of 4.0 and was "inactive" below pH 2.5 and above pH 6.0. The activity of KPase after preincubation at various temperatures was stable up to 50 degrees C. The keratinolytic activity was not affected by the addition of nonionic detergents and divalent cations. The enzyme was a glycoprotein and contained a high content of aspartic acid residues (172/1000). Pepstatin and chymostatin inhibited the activity in a dose-dependent manner; however, neither the other group specific inhibitors tested nor the pepsin specific inhibitors, DAN or EPNP, showed any effect on the enzyme. From these inhibitory profiles, this enzyme was determined to be a carboxyl proteinase such as cathepsin D. Among the various substrates for proteolytic enzymes, KPase digested human stratum corneum as much as albumin and hemoglobin. In the three fractions (water soluble, keratin filamentous, and membranous) prepared from human stratum corneum, the keratin filamentous fraction was more susceptible to degradation by KPase than the other two fractions were. KPase also digested much less human fingernail (13%) than human stratum corneum, but did not show any signs of there being any digestion of human scalp hair. These studies suggest that KPase from C. albicans may play an important role in superficial infection by affecting the human stratum corneum of the skin and nail.

Effect of calcitonin on salivary phosphorus and calcium excretion in sheep.

Three thyroidectomized sheep were infused intravenously with porcine calcitonin at a rate of 30 mi.u./h per kg and three sham-operated sheep were infused with vehicle for 5 h. Saliva was collected from the left parotid duct by cannulation for 10 min in every hour. Salivary secretion rates were not changed in either thyroidectomized or sham-operated sheep throughout the experiment. Before the infusion, salivary phosphorus excretion was less in thyroidectomized sheep than in sham-operated animals. Calcitonin infusion increased salivary phosphorus excretion and decreased serum phosphorus concentrations in thyroidectomized sheep. Vehicle infusion did not affect salivary phosphorus excretion in sham-operated sheep. Serum concentrations and salivary excretion of calcium were decreased by calcitonin infusion into thyroidectomized sheep but were not changed in sham-operated sheep infused with vehicle. Calcitonin infusion increased serum parathyroid hormone concentrations in thyroidectomized sheep after the decrease of serum calcium concentrations. However, vehicle infusion did not affect serum parathyroid hormone concentrations in sham-operated sheep. There was little change of cyclic AMP excretion during the experiment in either thyroidectomized or sham-operated sheep. It is concluded that calcitonin increases salivary phosphorus excretion in sheep.

A new agglutination test for serum antibodies to adult T-cell leukemia virus.

A new gelatin particle agglutination test was developed for assay of natural antibodies to adult T-cell leukemia virus (ATLV/HTLV-I). Partially purified viral antigen from culture fluid of a virus-producer cell line was coated on artificial gelatin particle carriers. A high correlation was observed between the titers of antibodies determined by the agglutination test and by indirect immunofluorescence assay. The agglutination test is simple, sensitive and specific, and should be useful for mass screening of human sera for viral antibodies.

Clinical trial of the Oka strain of live attenuated varicella vaccine on healthy children.

Clinical and serological follow-up studies were made on 257 healthy children who had received live varicella vaccine (strain Oka) in Showa Hospital. Good antibody responses were shown with a seroconversion rate of 98.4% (253/257) by the immune adherence hemagglutination test. Mild adverse reactions were observed in 11 of the vaccinated children. During observation periods of 6 months to 4 years, 6 of the 253 children who were successfully vaccinated contracted mild varicella, while all 4 vaccinees who showed no primary immune response contracted mild to moderate clinical varicella. It is concluded that this vaccine is highly immunogenic and causes few clinical reactions in normal children.

[Immunochemotherapy of human gliomas transplanted into nude mice].

In the first part of this paper, various chemotherapies were performed against oligodendrogliomas subcutaneously transplanted in to nude mice. Vincristine (VCR), adriamycin, and 1000 rads irradiation were effective against this tumors. Concerning these two drugs, dose response effect was observed. And the effect of 1 mg/kg injection of VCR roughly corresponded to that of five weekly injections of 0.2 mg/kg. In the second part of this experiment, single or combined effects of VCR and immunotherapeutic agents including OK-432 (OK), PSK, and recombinant leucocytic interferon (IFN) were examined. Two glioma lines including oligodendroglioma and glioblastoma were used. Following results were obtained from this experiment: 1) Effect of OK and VCR against oligodendrogliomas were as follows: control less than OK local injection (Local) less than OK intraperitoneal injection (IP); VCR; OK (IP X 2) less than VCR + OK(IP) less than VCR + OK (IP X 2). Effects of OK and VCR were expressed in order of their effects against glioblastomas: control less than VCR less than OK (IP); OK(IP X 2); OK(Local) less than VCR + OK (IP); VCR + OK (IP X 2). Effects of PSK and VCR against glioblastomas were as follows: control; PSK (Local) less than VCR less than VCR + PSK (Local) less than VCR + PSK (IP). Effects of IFN and VCR against oligodendrogliomas were as follows: control; IFN (IP) less than VCR less than IFN (Local) less than VCR + IFN (IP); VCR + IFN (Local). Effects of IFN and VCR against glioblastomas were as follows: control less than IFN (IP) less than VCR; IFN (Local); VCR + IFN (IP).(ABSTRACT TRUNCATED AT 250 WORDS)

T lymphocyte subsets in osteoporosis. Effect of 1-alpha hydroxyvitamin D3.

In order to explore the possible immunological abnormalities as the background for the development of postmenopausal and senile osteoporosis, T lymphocyte subsets were identified by monoclonal antibodies of the Orthoclone OKT series. The OKT4/OKT8 (helper-inducer/suppressor T cell) ratio was significantly higher in osteoporotics with spinal compression fracture than in non-osteoporotics without such fracture in similar age. Total lymphocyte and T cell counts were similar between the two groups. Young control subjects had an OKT4/OKT8 ratio indistinguishable from elderly non-osteoporotics. Administration of 0.5 microgram/day 1 alpha-hydroxyvitamin D3 for 2 months resulted in a significant fall of the OKT4/OKT8 ratio in osteoporotics. While the significance of elevated OKT4/OKT8 in osteoporotics remains unknown, the fall of this ratio in response to administration of 1 alpha-hydroxyvitamin D3 suggests vitamin D dependence of this immunological abnormality associated with osteoporosis.
