Synergistic induction by calcium ionophore and phorbol ester of interleukin-2 (IL-2) receptor expression, IL-2 production, and proliferation in autoimmune MRL/MP-lpr mice.

MRL/MP-lpr/lpr (MRL/l) mice spontaneously develop an age-related autoimmune disease concomitant with interleukin-2 (IL-2) defects. Induction of IL-2 receptor (IL-2R), IL-2 production and subsequent de novo DNA synthesis in MRL/l mice by the tumour-promoting phorbol ester 12-o-tetradecanoyl phorbol 13-acetate (TPA) and calcium ionophore (A23187) were examined. These two compounds given together induced significant IL-2R expression, IL-2 production, and de novo DNA synthesis of spleen cells from this murine strain, as did concanavalin A (Con A) plus TPA. TPA and A23187 may bypass the early steps of activation by mitogens in murine lymphocytes. However, even though these IL-2 defects could be overcome to some extent, the response of MRL/l mice to these stimuli was considerably lower than the enhanced IL-2R expression and IL-2 production of MRL/MP-+/+(MRL/n) control mice. These results suggested that the failure to respond to mitogens in these mice may be due, at least in part, to failure of receptor signal transduction, and to defects of molecular and biochemical reactions following signal transduction.

A novel agglutination test for the human immunodeficiency virus antibody: a comparative study with enzyme-linked immunosorbent assay and immunofluorescence.

A new serological test, the gelatin particle agglutination (PA) test, was developed for the detection of antibody to human immunodeficiency virus (HIV). The sensitivity of the PA test was the same as that of enzyme-linked immunosorbent assay (ELISA) among 761 US and Japanese sera tested. However, false-positive cases were found in 1.2% of all sera in ELISA but in 0% in the PA test (P less than 0.05). There was 100% agreement of seropositivity between PA and a confirmatory indirect immunofluorescence (IF) test. In addition, the agglutination test was reproducible. Thus, the test should be useful for the mass screening of anti-HIV antibody in blood donors.

Inhibition of phorbol ester-induced phenotypic differentiation of HL-60 cells by 1-(5-isoquinolinylsulfonyl)-2-methylpiperazine, a protein kinase inhibitor.

To identify the possible role of calcium ions in cell differentiation, we studied the extracellular Ca2+ requirement and the effect of Ca2+/phospholipid-dependent protein kinase (protein kinase C) inhibitor on proliferation and differentiation of human promyelocytic leukemic HL-60 cells. HL-60 cells grew equally well in 0.1 and 1.0 mM Ca2+ media. The addition of 12-O-tetradecanoyl-phorbol-13-acetate (TPA), 1,25-dihydroxyvitamin D3, and all-trans-beta-retinoic acid inhibited the cell growth and induced mature macrophage and granulocyte phenotypes in 1.0 mM Ca2+ medium. 1,25-Dihydroxyvitamin D3 and all-trans-beta-retinoic acid induced HL-60 differentiation to the same degree in 0.1 mM Ca2+ and 1.0 mM Ca2+ media. However, TPA failed to induce HL-60 differentiation or to inhibit proliferation in a 0.1 mM Ca2+ medium. The decrease of extracellular Ca2+ from 1.0 to 0.1 mM caused a significant drop in the intracellular Ca2+ level in undifferentiated and TPA-treated HL-60 cells, although no rapid change in cytosolic Ca2+ was detected in response to TPA addition. 1-(5-Isoquinolinylsulfonyl)-2-methylpiperazine (H-7), a protein kinase C inhibitor, inhibited proliferation of HL-60 cells in a dose-dependent manner. In contrast, H-7 selectively restored the proliferation of TPA-treated HL-60 cells and inhibited TPA-induced phenotypic differentiation. However, the same concentrations of 1-(5-isoquinolinylsulfonyl)-2,3-dimethylpiperazin and N-[2-(methylamino)ethyl]-5-isoquinolinesulfonamide, analogues of H-7 that inhibit protein kinase C more weakly, had no effect on the proliferation or differentiation induction. H-7 also suppressed 1,25-dihydroxyvitamin D3- and all-trans-beta-retinoic acid-induced phenotypic changes of HL-60 cells but did not eliminate the growth inhibition by these inducers. These results demonstrate the Ca2+ requirement and the protein kinase C involvement in phorbol ester-induced phenotypic differentiation of HL-60 cells.

Effects of alpha-difluoromethyl ornithine on alpha-fetoprotein and albumin secretion in human hepatoma cell line, PLC/PRF/5.

The in vitro effects of alpha-difluoromethyl ornithine (DFMO) on the growth and the secretion of alpha-fetoprotein (AFP) and albumin in human hepatoma cell line PLC/PRF/5 were studied. DFMO caused a marked reduction in the growth rate and de novo synthesis of DNA, whereas secretion of AFP and albumin was not altered. These data indicate that AFP and albumin secretion are neither linked to polyamine synthesis nor to cellular proliferation in human hepatoma cells.

1,25-Dihydroxyvitamin D3 regulates proliferation of activated T-lymphocyte subsets.

The biologically active vitamin D metabolite, 1,25-(OH) 2D3 suppressed phytohaemagglutinin (PHA)-induced lymphocyte proliferation dose-dependently (0.1 nM-100 nM), and decreased the OKT4+/OKT8+ ratio and transferrin-receptor-positive (OKT9+) cells. A possible parallelism between expression of 1,25-(OH) 2D3 receptors and interleukin 2 (IL2)-receptors recognized by anti-Tac antibody was not confirmed in this study. However, the addition of exogenous IL2 abolished the inhibitory effects of 1,25-(OH) 2D3 on PHA-stimulated T-cell proliferation, and the decrease of OKT4+ and OKT9+ T-cell in this population. Among various vitamin D3 analogues examined, 1,25-(OH) 2D3 was the most potent anti-proliferative effect, followed in order by 1,24S-(OH) 2D3, 1 alpha OH D3, 25 OH D3 and 24,25-(OH) 2D3.

Acute predominantly sensory neuropathy.

An account is given of an unusual patient with acute sensory neuropathy, leading within a few weeks to almost generalized sensory loss. Generalized sensory deficits were involved in the face, tongue, and palate. He had a drug allergy from chemotherapy during an early stage of illness. An albuminocytological dissociation of the cerebrospinal fluid was noted in a few months. A sural nerve biopsy after 2 months showed a marked reduction of fibers, and extensive invasion of phagocytes throughout the endoneurium. This patient recovered poorly with profound sensory ataxia of the limbs and tongue. A possible explanation for the clinico-pathological findings may be that acute idiopathic polyradiculoneuritis (Guillain-Barré syndrome) and toxic neuropathy exert an etiological role in the extramedullary sensory system.

Effect of 1,25-dihydroxyvitamin D3 on cytokine-induced thymocyte proliferation.

To clarify the mechanism of the inhibitory effect of 1,25(OH)2D3 on lymphocyte proliferation, the effect of 1,25(OH)2D3 on murine thymocyte proliferation induced by interleukin 1 (IL-1), or 2 (IL-2) was examined. Physiological concentrations of 1,25(OH)2D3 inhibited thymocyte proliferation induced by IL-1 and IL-2 in similar fashion suggesting an inhibition of the response to IL-2 by this hormone. In addition, cortisone-resistant thymocytes (including a majority of medullary thymocytes), which proliferate more vigorously in response to IL-1 than do untreated thymocytes, were more sensitive to 1,25(OH)2D3 inhibition. Therefore, the inhibition of IL-2 production of the mature medullary thymocyte by this hormone was also suggested.

Systemic malacoplakia in a breeding pig.

An adult breeding pig had multiple disseminated lesions in several organs of the thoracic and abdominal cavities. Histopathologically, the lesions consisted of accumulated histiocytic cells with a few giant cells and a mild infiltration of lymphocytes and plasma cells associated with concentric laminated spherical Michaelis-Gutmann bodies which stained strongly with haematoxylin and varied in diameter. The histological features of the lesions were similar to those of malacoplakia reported in man. The present case seems to be unique because the characteristic lesions were found in many organs including kidneys, liver, spleen, lungs, gall bladder, small intestine and several lymph nodes. The possibility of the haematogenous spread of malacoplakia was suggested by the frequent formation of the lesions in perivascular areas. The present case should be described as "systemic malacoplakia in a breeding pig".

Camptodactyly caused by an abnormal lumbrical muscle.

A thirteen-year-old girl, who complained of pain in the palm and difficulty in extension of the right little finger, had an abnormal insertion of the lumbrical muscle into the flexor digitorum superficialis tendon. The symptoms were relieved by detaching the lumbrical muscle and transferring its tendon into the lateral band.

Effect of serum albumin on free fractions of phenobarbital and valproic acid in patients with convulsive seizures.

The free fractions of phenobarbital and valproic acid were assayed with Free-Level system I (Syva) and by an enzymatic immunoassay technique (EMIT) in 186 patients under chronic anti-convulsant therapy at precisely 2 hrs after they had taken the medicine at breakfast. The free fractions of PB ranged from 49 to 53% in monopharmacy and from 50 to 55% in polypharmacy. Those of VPA ranged from 10 to 12% in monopharmacy and from 8 to 11% in polypharmacy. The regression lines of the free fractions in PB monopharmacy and PB polypharmacy against serum albumin concentration indicated a negative correlation. There was no difference between the free fractions in PB monopharmacy and polypharmacy at the same albumin concentration. The regression lines of the free fractions in VPA monopharmacy and polypharmacy against serum albumin concentration indicated a negative correlation. The free fractions of VPA did not vary under a total VPA concentration of 80 micrograms/ml, while those of VPA above a total VPA concentration of 90 micrograms/ml increased with the total concentration, in the monopharmacy groups. On the other hand, free fractions of VPA in the polypharmacy groups did not vary with the total VPA concentration.
