Infiltration of Leu-3a+3b-positive lymphocytes and expression of human leukocyte antigen DR on tumor tissue induced by radiation therapy.

Tumor infiltrating lymphocytes induced by radiotherapy were analyzed by the biotin-avidin-horseradish peroxidase method. At the early dose of 4 Gy (400 rad) of irradiation (2 Gy X 2), remarkable infiltration of lymphocytes was observed surrounding cancer cells. This infiltration was mainly composed of Leu-3a+3b-positive lymphocytes, and human leukocyte antigen DR-positive tumor and interstitial cells were remarkably observed.

Inositol 1,4,5-trisphosphate-induced contraction in skinned single smooth muscle cells from guinea-pig taenia coli.

Inositol 1,4,5-trisphosphate (IP3) caused a contraction in skinned single smooth muscle cells from the guinea-pig taenia coli. The contractile response to IP3 was rapid (half time less than 5 s), concentration-dependent (0.01-10 microM, EC50; 0.1 microM) and also depended on the concentration of Ca2+ during preloading. IP3 induced a rapid 45Ca2+ release from the skinned single cells which were preloaded with 10(-6) M Ca2+. The contractile response caused by IP3 was not enhanced by simultaneous application of caffeine (25 mM). The inhibitory effect of procaine on IP3-induced contraction was less than that on caffeine-induced contraction. These results suggest that IP3 also plays an important role in the contractile response in smooth muscle cells from guinea-pig taenia coli and that the mechanism of Ca2+ release from intracellular storage sites by IP3 is different from that by caffeine.

[Anti-cancer efficacy of peplomycin, adsorbed on activated carbon particles, against lymph node metastases in mice].

PEP-CH is comprised of activated carbon particles that adsorb peplomycin. Mice which were inoculated with 5 x 10(5) MH 134 tumor cells subcutaneously opposite the foot-pad of the left hind paw on day 0, received a subcutaneous injection of peplomycin, 0.25 mg/mouse, in the form of PEP-CH or a peplomycin aqueous solution administered at the foot-pad of the left hind paw on day 10. The left popliteal lymph nodes then were transferred intraperitoneally to normal mice on day 14. In the PEP-CH group, the survival time of the recipients was statistically and significantly long, and the transferred tumor cell number, estimated with a calibration curve, was markedly small when compare to mice in the other treatment groups.

[Detection of mural thrombi and analysis of blood flow velocities in the left atrial appendage using transesophageal two-dimensional echocardiography and pulsed Doppler flowmetry].

To correlate blood flow velocities with thrombogenesis in the left atrium (LA) and left atrial appendage (LAA), we performed transesophageal two-dimensional echocardiography (Eso 2-D Echo) combined with pulsed Doppler flowmetry, which was developed in our laboratory. Thirty-eight patients were studied; 1) 15 who had sinus rhythm including four with mitral stenosis (MS) and 11 normal subjects, and 2) 23 with atrial fibrillation (Af). The patients with Af were classified as lone Af (nine cases); Af-CI: Af with cerebral infarction having no MS (four cases); and Af-MS: Af with MS (10 cases). In four of 10 patients with Af-MS, mural thrombi were detected in the LA or LAA by Eso 2-D Echo, but they were not visualized by conventional echocardiography. In lone Af and Af-CI, however, abnormal echoes in the LAA were not observed by Eso 2-D Echo. Thus, Eso 2-D Echo proved superior to conventional 2-D echocardiography in detecting mural thrombi in the LA or LAA. In sinus rhythm, a biphasic flow pattern followed the P wave on the ECG. It was observed in the vicinity of the junction between the LA and LAA. In Af a flow with a saw-tooth appearance was observed throughout the cardiac cycle. The peak flow velocity in the LAA was significantly less in Af than in sinus rhythm (p less than 0.05). Especially in Af-MS, the peak flow velocity was markedly decreased (1 +/- 4 cm/sec: mean +/- SD) compared with those of the other Af groups (p less than 0.01), and it was significantly less in Af-CI (11 +/- 5 cm/sec) than in patients with lone Af (21 +/- 9 cm/sec) (p less than 0.05). These results indicate that analysis of blood flow velocities in the LAA by Eso 2-D Echo is of great value in assessing thrombogenesis in the LAA, and the patients with Af, even without MS, who have markedly reduced peak flow velocities in the LAA, should be prophylactically anticoagulated, because stagnation of blood is strongly suspected.

[Antineoplastic effect of prostaglandins on human glioma in athymic nude mice].

The prostaglandins (PG) are known to have various physiological effects. Some series of prostaglandins such as PG D2 have been reported to inhibit growth of tumor cells. In this study, the growth-inhibitory effects of PG A2, PG D2, PG J2 and 6-keto PGE1 were investigated in nude mice receiving subcutaneous transplant of human brain tumor. One to two milligram of prostaglandins was given intraperitoneally every day for three weeks. Tumor volumes were measured twice weekly and the tumor reduction rates (treated/control) were evaluated. T/C rate treated with PG D2 or PG A2 was 50-60% respectively. The effectiveness of PG J2 or 6-keto PGE1 was inferior to that of PG A2 or PG D2. But in the evaluation of antitumor effects of PG J2, we must consider the fact that the activity of PG J2, is liable to be lost. The effect of PG D2 on proliferation of cultured glioma cells was also studied. At concentrations of 10 micrograms/ml, PG D2 strongly inhibited the proliferation of glioma cells. However, precise mechanism of prostaglandin action is presently unknown. Further studies are required to clarify the mechanism of antitumor effects of prostaglandins.

[Lateral coordinate of the thalamic Vim neurons in the cases with dilated third ventricle].

Precise radiographical measurement of the third ventricle in the first and essential procedure in stereotaxic surgery in order to determine the target point. This is done while referring to an available standard brain atlas such as Schaltenbrand & Bailey. However there have been no criteria established for determining the deviation of the coordinates of the thalamic nuclei, especially in their lateral coordinates when the third ventricle is highly dilated. Therefore, in 109 cases encountered recently (81 parkinsonism, 11 essential tremor, 10 cerebral palsy, 3 thalamic pain, 1 Benedikt's syndrome, 1 torticollis, 1 oral dyskinesia, 1 striato-nigral degeneration), we studied the correlation between the width of the third ventricle and the lateral coordinate of the thalamic point where kinesthetic neurons or tremor-synchronously discharging neurons were detected. These neurons were especially related to the arm. According to the width of the third ventricle, we classified the cases into three types: 18 cases with large ventricles (more than 10 mm), 37 cases with medium-sized ventricles (4-10 mm) and 16 cases with small ventricles (less than 4 mm). By plotting the lateral coordinate of the thalamic point where kinesthetic neuron of the upper extremity was recorded in reference to the radiogram of anteroposterior view, we found that the lateral coordinates of large ventricular cases generally tended to show more lateral displacement than cases where the ventricles were narrower. Their mean lateral coordinate was 16.9 mm from midline in contrast ot 14.0 mm in cases with small ventricular width. There was a linear correlation between the width of the third ventricle and the lateral coordinate of the kinesthetic neuron of the Vim nucleus.(ABSTRACT TRUNCATED AT 250 WORDS)
