Antihyperalgesic activity of epibatidine in the formalin model of facial pain.

A growing body of evidence supports a nicotinic cholinergic approach to pain management, as neuronal nicotinic receptor agonists have shown efficacy across animal models of both inflammatory and neuropathic pain. However, most of these investigations have focused on the spinal system, and there is to date no report of nicotinic receptor-mediated antinociception in any pain model involving the trigeminal field of innervation. Thus, the purpose of the present studies was to evaluate whether the neuronal nicotinic receptor agonist epibatidine possesses antihyperalgesic activity in the formalin model of facial pain. Adult, male, Sprague--Dawley rats received a 50-microl, subcutaneous injection of 5% formalin into one vibrissal pad and the consequent, facial grooming behavior was monitored. Consistent with previous investigations using the formalin model, animals exhibited two periods of nocifensive grooming: (1) an acute phase that began immediately, peaked at 3 min and almost completely abated by 6 min, and (2) a tonic phase that began between 6 and 9 min, peaked at 21 min and slowly diminished over the ensuing 24 min. The subcutaneous administration of epibatidine (1--5 microg/kg) 5 min prior to the formalin injection led to a significant, dose-dependent reduction of both the acute and tonic phases of hyperalgesia. Separate groups of animals receiving epibatidine either 15, 30 or 60 min prior to the formalin injection exhibited a progressively diminishing antihyperalgesic response that was no longer significant in either phase by 30 min. Finally, pretreatment with the selective neuronal nicotinic receptor antagonist mecamylamine completely abolished the antihyperalgesic effect of epibatidine in both phases. Taken together, these studies demonstrate that in both the acute and tonic phases of the formalin model of facial pain, epibatidine produces a neuronal nicotinic receptor-mediated antihyperalgesia that is both dose- and time-dependent. These results support the rationale for exploring the clinical efficacy of nicotinic agonists as analgesics to treat certain types of trigeminal pain in humans.

Factors influencing the 17O quadrupole coupling constant in bridging oxygen environments.

Ab initio calculations were performed on the series of clusters [(OH)3M-O-M(OH)3](-2) where M = B, Al, and Ga, (OH)3M-O-M(OH)3 where M = C, Si, and Ge, (OH)2OM-O-MO(OH)2 where M = P and As, and (OH)O2M-O-MO2(OH) where M = S and Se. These clusters were constructed to model the effect of changing the coordinating cation and coordinating cation-oxygen distances on the bridging oxygen 17O quadrupole coupling constant. For all clusters studied, we observed a general trend that the magnitude of Cq increases linearly with increasing cation-oxygen bond distance and cation group number, suggesting that group number and cation-bridging oxygen distance can serve as a better predictor of the bridging oxygen quadrupole coupling constant than electronegativity differences.

The electrical properties of the anterior stomach of the larval mosquito (Aedes aegypti).

The electrical properties of the anterior stomach of the larval mosquito (Aedes aegypti) were determined. At late times after cannulation, the intraluminal space constant was 936 microm, which is almost as long as the isolated tissue itself. At this time, the resistance of the apical cell membranes dominates the transcellular resistance; it is approximately 14 times the resistance of the basal cell membrane. Two physiologically distinct epithelial cell types were identified. One type has a stable basal potential of approximately 65 mV and responds to 5-hydroxytryptamine with hyperpolarization. The second cell type initially shows a basal potential of 100 mV. However, this basal potential decays in the first few minutes in parallel with the decay of the transintestinal potential. This latter cell type does not respond to 5-hydroxytryptamine.

Serum concentrations of IGF-I, estradiol-17beta, testosterone, and relative amounts of IGF binding proteins (IGFBP) in growing boars, barrows, and gilts.

The purpose of this research was to determine whether serum concentrations of steroids, IGF-I, and relative amounts of serum IGF-binding proteins (IGFBP) differ in growing boars (n = 11), barrows (n = 11), and gilts (n = 12) from 70 to 140 d of age. Pigs of similar age and weight were housed in pens of three or four and given ad libitum access to a 17% CP corn-soy diet and water. Pigs were weighed and blood samples were collected every 14 d beginning at 70 d of age. Serum concentrations of IGF-I and steroids were determined by RIA and relative amounts of IGFBP were determined by ligand blot analysis. By 84 d of age and continuing through 140 d of age, mean serum concentrations of IGF-I were greater (P < .05) in boars than in barrows or gilts. Relative amounts of 46-kDa IGFBP3 and 28-kDa IGFBP-4 were similar (P > .05) among pigs at 70 d of age; however, boars and barrows had greater (P < .05) relative amounts of 24-kDa IGFBP-4 and 41-kDa IGFBP-3 than gilts. Relative amounts of IGFBP-2 were greater (P < .01) in barrows than in gilts or boars at 70 d of age. From 84 d of age through 140 d of age, relative amounts of both forms of IGFBP-3 and the 28-kDa IGFBP-4 were greater (P < .05) in boars than in gilts or barrows. Relative amounts of IGFBP2 were greater (P < .05) in barrows than in gilts or boars at 98 d of age, but by 140 d of age relative amounts were greater (P < .05) in boars and barrows than in gilts. Mean serum concentrations of estradiol-17beta were similar (P > .05) between gilts and boars at 70 d of age, but by 98 d of age, and continuing through 140 d of age, mean serum concentrations of estradiol-17beta were greater (P < .05) in boars than in gilts. Mean serum concentrations of testosterone in boars increased (P < .05) with increasing age and were greatest at 128 and 140 d of age. Serum concentrations of testosterone were negatively correlated (P < .01) with relative amounts of serum IGFBP-2 but positively correlated (P < .01) with serum concentrations of IGF-I and estradiol-17beta. Serum concentrations of estradiol-17beta were positively correlated (P < .01) with serum concentrations of IGF-I in boars. Changes in serum concentrations of IGF-I and relative amounts of IGFBP resulting from changes in serum concentrations of estradiol-17beta and testosterone may contribute to growth differences observed among sexes.

The anterior and posterior 'stomach' regions of larval Aedes aegypti midgut: regional specialization of ion transport and stimulation by 5-hydroxytryptamine.

The 'stomach' region of the larval mosquito midgut is divided into histologically distinct anterior and posterior regions. Anterior stomach perfused symmetrically with saline in vitro had an initial transepithelial potential (TEP) of -66 mV (lumen negative) that decayed within 10-15 min to a steady-state TEP near -10 mV that was maintained for at least 1 h. Lumen-positive TEPs were never observed in the anterior stomach. The initial TEP of the perfused posterior stomach was opposite in polarity, but similar in magnitude, to that of the anterior stomach, measuring +75 mV (lumen positive). This initial TEP of the posterior stomach decayed rapidly at first, then more slowly, eventually reversing the electrical polarity of the epithelium as lumen-negative TEPs were recorded in all preparations within 70 min. Nanomolar concentrations of the biogenic amine 5-hydroxytryptamine (5-HT, serotonin) stimulated both regions, causing a negative deflection of the TEP of the anterior stomach and a positive deflection of the TEP of the posterior stomach. Phorbol 12,13-diacetate also caused a negative deflection of the TEP of the anterior stomach, but had no effect on the TEP of the posterior stomach. These data demonstrate that 5-HT stimulates region-specific ion-transport mechanisms in the stomach of Aedes aegypti and suggest that 5-HT coordinates the actions of the Malpighian tubules and midgut in the maintenance of an appropriate hemolymph composition in vivo.

Septic shock from Mycobacterium tuberculosis after therapy for Pneumocystis carinii.

Septic shock resulting from Mycobacterium tuberculosis (TB) occurs only rarely, even among patients infected with human immunodeficiency virus. We report a case of fulminant TB sepsis in the setting of human immunodeficiency virus infection. This case illustrates the hazards of corticosteroid use as a part of empirical treatment of Pneumocystis carinii pneumonia, as well as the unique appearance of TB on chest x-ray films.

Dose-dependent effects of CRF-like diuretic peptide on transcellular and paracellular transport pathways.

The mechanism of action of synthetic Culex corticotropin-releasing factor (CRF)-like diuretic peptide (CCRF-DP) was investigated in isolated, perfused Malpighian tubules of the yellow fever mosquito, Aedes aegypti. Low concentrations of CCRF-DP (10(-10) and 10(-9) M) caused depolarizing oscillations of the lumen-positive transepithelial voltage (Vt) in Malpighian tubules, whereas high concentrations (10(-8) and 10(-7) M) first depolarized and then transiently hyperpolarized Vt; CCRF-DP always lowered transepithelial resistance (Rt), regardless of voltage depolarization or hyperpolarization. The short-circuit current (Isc), an electrical estimate of active transepithelial transport of Na and K, remained unchanged at low concentrations of CCRF-DP, but Isc more than doubled at high concentrations. These effects of CCRF-DP suggest dose-dependent sites of action: low concentrations of CCRF-DP affect the paracellular pathway, and high concentrations affect both paracellular and transcellular pathways.

The concentration-dependence of CRF-like diuretic peptide: mechanisms of action.

The mechanism of action of synthetic CCRF-DP, the corticotropin-releasing factor (CRF)-related diuretic peptide of the salt marsh mosquito Culex salinarius, was investigated in isolated Malpighian tubules of the yellow fever mosquito Aedes aegypti. A low concentration of CCRF-DP (10(-9)mol l-1) caused a small but insignificant increase in transepithelial secretion of NaCl and fluid, but significantly reduced transepithelial voltage and resistance without a change in short-circuit current, pointing to the stimulation of passive Cl- transport through the paracellular pathway as the principal mechanism of a mild diuresis. Significant changes in voltage and resistance but not in short-circuit current were duplicated by the ionophore A23187 (0.4 micromol l-1), suggesting Ca2+ as a second messenger at 10(-9)mol l-1 CCRF-DP. A high concentration of CCRF-DP (10(-7)mol l-1) significantly increased transepithelial secretion of NaCl and fluid and significantly increased short-circuit current, pointing to the stimulation of active Na+ transport through the transcellular pathway as the mechanism of a strong diuresis. This effect was mimicked by dibutyryl-cAMP, suggesting cAMP as a second messenger at 10(-7)mol l-1 CCRF-DP. Dibutyryl-cGMP had no effects. These results suggest dose-dependent, receptor-mediated effects of CCRF-DP that target discrete transport pathways via discrete second messengers: low concentrations of CCRF-DP cause a mild diuresis, apparently via Ca2+-mediated effects on paracellular Cl- transport, and high concentrations cause a strong diuresis via cAMP-mediated effects on active transcellular Na+ transport in addition to the effects on the paracellular pathway.

Evaluation of a dual-channel full dynamic range compression system for people with sensorineural hearing loss.

This article describes an evaluation of an in the ear hearing aid, which applies fast-acting full dynamic range compression independently in two frequency bands. This can compensate for the loudness recruitment typically associated with sensorineural hearing loss. The crossover frequency between the two bands and the gain and compression ratio in each band are programmable to suit the individual patient. Twenty subjects with moderate sensorineural hearing loss were tested in a counterbalanced order using the aid programmed as a linear amplifier (condition L) and as a two-band compressor (condition C). All subjects were fitted binaurally. Subjects were also tested without hearing aids (condition U) and using the hearing aids that they normally wore (condition Own). Speech intelligibility was measured in quiet at three sound levels (50, 65, and 80 dB SPL), and speech reception thresholds (SRTs) in 12-talker babble were measured under monaurally and binaurally aided conditions, with the speech and babble both coincident and spatially separated. In condition C, speech intelligibility in quiet was high at all sound levels. Speech intelligibility at the two lower levels decreased in condition L, and decreased still further in conditions Own and U. Condition C gave, on average, better speech intelligibility in babble (lower SRTs) than conditions L, Own, or U. The advantage of condition C over condition L varied across subjects and was correlated with the dynamic range for tones at high frequencies; small dynamic ranges were associated with greater benefit from compression. A significant advantage for binaural aiding was found both when the speech and noise were spatially separated and when they were coincident. The binaural advantage was similar for the C and L conditions, indicating that the independent compression at the two ears did not adversely affect the use of binaural cues. Questionnaires on the subjects' experiences with the aids in everyday life indicated that they generally preferred condition C over condition L.

Digital overlay of fluorescein angiograms and fundus images for treatment of subretinal neovascularization.

A system for comparing sequential fundus photographs and angiograms has been developed using a personal computer and commercially available image processing software. This process provides rapid results and a high degree of accuracy in image correlation, even when images come from different cameras with different magnifications. Though no concentrated effort was made to compare the superiority of this method to the manual technique, the value of the manual results is fully acknowledged, and this technique is offered as another method of achieving the same end. This technique can be used to determine the adequacy of treatment of subretinal neovascularization, recurrence of subretinal neovascularization in subsequent angiograms, pigment epithelial creep, and changes in fundus lesions.

Patient experiences with multiband full dynamic range compression.

This investigation compared performance for 18 experienced hearing aid patients with a multiband full dynamic range compression hearing device and their previously worn linear or automatic gain control hearing aids. Performance was evaluated using a variety of clinical tests. These included functional gain and dynamic range measurements, speech recognition, and real ear measurements. Patients rated their performance with the multiband compression device and their previously worn aids on a rating scale pertaining to a variety of real life listening situations. Statistical analysis of data derived from test results shows significantly improved performance on all measures using the multiband compression aid over their conventional aids. Patients rated their performance higher with the compression instrument than with the conventional aids. Differences in performance on clinical tests and subjective patient responses appear to be related to the different signal processing schemes used in the two groups of devices.

Degradation of organic sulfur compounds by a coal-solubilizing fungus.

Paecilomyces sp. TLi, a coal-solubilizing fungus, was shown to degrade organic sulfur-containing coal substructure compounds. Dibenzothiophene was degraded via a sulfur-oxidizing pathway to 2,2'-dihydroxybiphenyl. No further metabolism of that compound was observed. Ethyl phenyl sulfide and diphenyl sulfide were degraded to the corresponding sulfones. A variety of products were formed from dibenzyl sulfide, presumably via free radical intermediates. Diphenyl disulfide and dibenzyl disulfide were cleaved to the corresponding thiols and other single-ring products. It was concluded that degradation of organic sulfur compounds by Paecilomyces involves an oxidative attack localized at the sulfur atom.
