The vacuolar proton pump of Dictyostelium discoideum: molecular cloning and analysis of the 100 kDa subunit.

The vacuolar proton pump is a highly-conserved multimeric enzyme that catalyzes the translocation of protons across the membranes of eukaryotic cells. Its largest subunit (95-116 kDa) occurs in tissue and organelle-specific isoforms and thus may be involved in targeting the enzyme or modulating its function. In amoebae of Dictyostelium discoideum, proton pumps with a 100 kDa subunit are found in membranes of the contractile vacuole complex, an osmoregulatory organelle. We cloned the cDNA that encodes this 100 kDa protein and found that its sequence predicts a protein 45% identical (68% similar) to the corresponding mammalian proton pump subunit. Like the mammalian protein, the predicted Dictyostelium sequence contains six possible transmembrane domains and a single consensus sequence for N-linked glycosylation. Southern blot analysis detected only a single gene, which was designated vatM. Using genomic DNA and degenerate oligonucleotides based on conserved regions of the protein as primers, we generated products by polymerase chain reaction that included highly variable regions of this protein family. The cloned products were identical in nucleotide sequence to vatM, arguing that Dictyostelium cells contain only a single isoform of this proton pump subunit. Consistent with this interpretation, the amino acid sequences of peptides derived from a protein associated with endosomal membranes (Adessu et al. (1995) J. Cell Sci. 108, 3331-3337) match the predicted sequence of the protein encoded by vatM. Thus, a single isoform of the 100 kDa proton pump subunit appears to serve in both the contractile vacuole system and the endosomal/lysosomal system of Dictyostelium, arguing that this subunit is not responsible for regulating the differing abundance and function of proton pumps in these two compartments. Gene targeting experiments suggest that this subunit plays important (possibly essential) roles in Dictyostelium cells.

African American and white differences in nutritional status among low-income women attending public health clinics.

Information concerning nutritional status and factors influencing dietary intakes among underserved populations is scarce. To obtain this information, data on nutritional status in a group of 726 white and African American women of low education and low income who were inner-city dwellers were analyzed. Dietary habits in all subjects were characterized by high intakes of fat, saturated fat, cholesterol, and salt and low intakes of fiber and folate. A comparison of dietary intake patterns of low-income white and African American women showed a trend toward poorer dietary habits among the white women. It is suggested that differences between African Americans and whites in most nutrient intakes were due to factors such as low levels of education and income rather than racial background alone. Results imply that disease prevention and health promotion programs should include efforts to increase awareness and practice of healthy diet habits among all low-income women.

[Clinical analysis of 62 pregnant women complicated with organic heart diseases].

OBJECTIVE: To investigate the factors which effect maternal-fetal outcomes in pregnant women complicated with organic heart disease. METHOD: Clinical data of 62 pregnant cases with either rheumatic or congenital heart diseases, collected from Beijing Anzhen Hospital during 1985-1994, were analysed retrospectively. RESULTS: The prevalence of organic heart diseases in total pregnant women during the same period was 0.97%. No significant difference was found in maternal-fetal complications between the two types of heart diseases. However, the incidences of maternal heart failure, premature birth, intrauterine growth retardation were significantly higher in patients without previous surgical treatment as compared with those with previous surgical therapy, and were also higher in patients with heart function of grade III-IV in comparison with those with grade I-II. CONCLUSIONS: Patients with organic heart diseases should be treated and operated on in order to achieve grade I-II heart function before pregnancy. Cesarean section is the better choice of delivery.

Characteristics of the inward-rectifying potassium current in mouse ventricular myocytes and its relation to early after-depolarization.

The properties of the inward-rectifying potassium current (IK1) were studied in the single myocytes isolated from adult mouse ventricles by the whole-cell patch-clamp technique for the first time. Most of the properties of IK1 including channel conductances, activation, inactivation, rectification and external K+ sensitivity in mouse ventricular myocyte were similar to those in other species, but the current-voltage (I-V) curve of mouse ventricular myocyte showed no negative slope, i.e the slope in the range of membrane potential 50 mV positive to the reversal potential (VRev) was virtually flat and remained at a low current level ((59 +/- 39) pA). Under the superfusion of Tyrode's solution with 3 mmol/L K+ and 3 mmol/L Cs+, IK1 in the above region nearly decreased to zero, and then the early after-depolarization (EAD) occurred. The results suggest that this distinctive characteristic of IK1 in mouse ventricular myocyte may relate to the high susceptibility to EAD in mouse myocardium. The inhibition of IK1 seems to be a prerequisite for the occurrence of EAD in this experiment.

In situ amplification of DNA fragments specific for human Y chromosome in cellular nuclei by PCR.

Using single primer pairs Y3 and Y4 in situ polymerase chain reaction (in situ PCR) was successfully performed on the specimen slides of peripheral leukocytes. By both of the direct digoxiginin-11-dUTP incorporation into PCR products with in situ PCR (direct in situ PCR) and in situ PCR followed by detection of in situ hybridization (indirect in situ PCR), DNA fragments specific for human Y chromosome were obviously amplified in cellular nuclei of specimens on the slides. The results were verified by Southern analysis. The methodology of in situ PCR and its application were discussed.

Intraperitoneal radioimmunotherapy of ovarian cancer with lutetium-177-CC49.

UNLABELLED: Twelve ovarian cancer patients who failed chemotherapy entered a Phase I trial of intraperitoneal 177Lu-CC49 antibody. METHODS: Patients had disease confined to the abdominal cavity +/- retroperitoneal lymph nodes, adequate organ function and no previous radiation. RESULTS: Side effects included mild discomfort with administration (1/12), delayed transient arthralgia (2/12), and mild marrow suppression (calculated marrow doses of 11-54 cGy). The maximum tolerated dose has not been reached with levels of 10, 18, 25 and 30 mCi/m2. Radioimmunoscintigraphy revealed localization consistent with tumor in 11 of 12 patients. One of eight patients with gross disease had > 50% tumor reduction after therapy, while six progressed and one went off study with stable disease. Of patients with microscopic or occult disease, one relapsed at 10 mo and three remain without evidence of disease after 18 mo. CONCLUSION: Intraperitoneal radioimmunotherapy with 177Lu-CC49 is well tolerated and appears to have antitumor activity against chemotherapy-resistant ovarian cancer in the peritoneal cavity.

Enhancement of radiolabeled antibody binding and tumor localization through adenoviral transduction of the human carcinoembryonic antigen gene.

Conventional radiolabeled antibody targeting utilized in radioimmunotherapy has resulted in limited success clinically due in part to inadequate tumor localization resulting from low expression of human tumor-associated antigens on target cells. We hypothesized that one could improve upon these limitations by genetically inducing tumor cells to express high levels of a new membrane-associated receptor with high affinity for a radioligand. As a preliminary strategy, we induced a human glioma cell line (D54 MG) to express human carcinoembryonic antigen (CEA) in vitro. To accomplish this, we constructed a recombinant adenoviral vector encoding the CEA cDNA inserted downstream of a cytomegalovirus (CMV) promoter (AdCMVCEA). D54 MG cells were transfected with AdCMVCEA or an adenoviral vector encoding lacZ reporter gene as a control (AdCMVlacZ). LS174T human colon cancer cells, known to express CEA constitutively, served as positive controls. Immunofluorescence and immunohistochemistry assays employing unlabeled anti-CEA COL-1 monoclonal antibody demonstrated expression of CEA antigen on the cell surface of transduced D54 MG cells in culture. In addition, assays utilizing 125I-labeled COL-1 indicated high binding to transduced D54 MG cells expressing CEA (4.7 +/- 0.5 x 10(5) COL-1 molecules bound per cell) as compared with minimal binding to nontransduced D54 MG cells. LS174T cells demonstrated only 2.7 +/- 0.5 x 10(6) COL-1 molecules bound per cell. Thus, AdCMVCEA was able to induce levels of cell surface CEA in target cells at a higher level than CEA-overexpressing tumor cells (P < 0.01). The efficacy of transduction of recombinant AdCMVCEA by direct intratumoral injection into D54 MG xenografts was investigated by immunohistochemical analysis, immunofluorescence and by measuring 131I-labeled COL-1 uptake through external scintigraphic imaging and biodistribution studies. Expression of CEA in the tumor xenografts by, and radiolabeled antibody tumor targeting to, AdCMVCEA transduced D54 MG xenografts was comparable to that seen with LS174T xenografts. Results of these studies indicate the potential of adenovirus-mediated delivery of targets to improve radiopharmaceutical tumor localization.

Approaches to enhance cancer radiotherapy employing gene transfer methods.

This review presents an overview and discussion of the potential synergistic strategies of radiation therapy and gene transfer for treating neoplastic disease. Topics discussed include radiation-inducible promoters coupled to genes which produce proteins that are cytotoxic or enhance radiosensitivity, employment of molecular chemotherapy approaches in conjunction with radiation therapy, and genetic induction of radiosensitization through modification of DNA repair, signal transduction, and cell cycle control genes. Additional topics discussed relate to gene transfer augmentation of radioimmunotherapy of cancer. Specifically, gene transfer methods to genetically induce tumor cells to express enhanced levels of cell surface antigens and receptors to increase radiolabeled antibody and peptide targeting and thus increase their therapeutic effect, selection of radionuclides for therapeutic ligand labeling, and computer simulation of genetic tumor-specific delivery of radiolabeled ligands are proposed.

[Clinicopathological evaluation on ovarian serous borderline tumours and their implants].

OBJECTIVE: To investigate the clinicopathologic features and the expressions of proliferation cell nuclear antigen (PCNA) in ovarian serous borderline tumours (OSBT). METHODS: The primary ovarian tumours, implants and recurrent specimens of 48 cases of OSBT were studied. Expression of PCNA was determined by avidin-biotin-peroxidase complex (ABC) method. Clinical data for all patients were abstracted from hospital records. RESULTS: While the primary ovarian tumours have the characteristic histological pictures of borderline tumour, three different types of histologic appearance including benign, noninvasive and invasive were shown by their peritoneal implants. All benign and most noninvasive implants were PCNA negative or weakly positive, and the 25 patients were free from tumour 5-28 years after their operations. Some of the noninvasive implants (9/22) were recurrent, and a few (3/22) underwent malignant transformation 10-23 years after surgery. The only one invasive implant was PCNA strongly positive and the patient died 3.5 years after surgery. CONCLUSION: The different histologic types of the peritoneal implants are of prognostic significance and are factors to be considered in determining the treatment project. The diagnostic value of PCNA immunoreactivity in implantations is confirmed.

[Detection of c-Ki-ras oncogene codon 12 point mutations in lung carcinoma, gastric carcinoma and hepatic carcinoma].

DNA extracted from paraffin-embedded tissues of 30 lung carcinomas, 40 gastric carcinomas and 22 hepatocellular carcinomas was tested for the presence of c-Ki-ras codon 12 point mutation by PCR-RFLP technique. The results showed that 20% (6/30) lung carcinomas and 2.5% (1/40) gastric carcinomas had Ki-ras codon 12 point mutation, but non of the 22 hepatocellular carcinomas displayed this point mutation. Among 6 cases of lung carcinomas of mutation, 4 were squamous carcinomas and 2 were adenocarcinomas.

[Studies on MDM2 oncogene expression and its effect on pancreatic carcinoma cells].

In order to study the interrelation and interaction between MDM2 oncogene and wild type p53 in human pancreatic cancer, we studied the expression and amplification of MDM2 oncogene and its antagonistic effect on wild type p53 by use of gene recombination, gene transduction and molecular hybridization techniques. The results showed that MDM2 oncogene could be detected in all 5 pancreatic cell lines, but MDM2 mRNA expression varied in the different cell lines. The recombinant vector pCMV-MDM2 was transduced into PC-2/s-wtp53 cell line (a transformed PC-2 pancreatic carcinoma cell line containing wild type p53 gene). The resultant cell line, PC-2/s-wtp53/pCMV-MDM2 showed rapid cell growth, a rate similar to that of the parent cell line PC-2. Our results verify the fact that MDM2 gene can abrogate the cell growth arrest mediated by wild type p53 and the antagonistic function of wild type p53.

[Clinical importance of p53 gene mutation and PCNA expression in ureteral and renal pelvic cancers].

In order to know the clinical importance of p53 gene mutation and PCNA expression in ureteral and renal pelvic cancers, research was carried out with immunohistochemistry S-P method in 45 cases. Mutant p53 nuclear staining was positive in 24 cases (53.3%) and correlated with PCNA expression, being higher in clinical stages T3 and T4 than in T1 + T2. They also correlated with the grade of cell differentiation, being higher in G3 than in G1 + G2.

[Reappraisal of the pathological criteria for uterine leiomyosarcoma].

In order to study the pathologic criteria for uterine Leiomyosarcoma, 42 cases of morphologically malignant or uncertain malignant potential uterine smooth muscle tumors which exhibited recurrences or metastasis, or were followed for a minimum of 2 years clinically, were analysed and reappraised. The results showed that in comparison with the leiomyoma group, active mitosis (mitotic index, MI > or = 10/10HPF), significant cytologic atypia and coagulative tumor cell necrosis (CTCN) were more common in the leiomyosarcoma group (71.4%, 92.9% and 71.4% respectively). Whereas 4 of the 14 leiomyosarcomas had MI < or = 4/10HPF, developed recurrences or metastasis, and 7 of the 28 leiomyomas had MI > or = 5/10HPF, including 2 with MI > 10/10HPF were alive and well 5.5-23.0 years after operation; hence a mitotic count is an unreliable prognostic criterion for uterine smooth muscle tumors. The data suggest that a trivariate rule using MI, the degree of atypia and CTCN best identifies the aggressive potential of uterine smooth muscle tumors.

[Construction of a retroviral vector expressing antisense N-myc gene].

In their previous studies the authors demonstrated that nerve growth factor-induced differentiation of neuroblastoma cell lines was closely related with nerve growth factor receptors and amplification of N-myc oncogenes. The authors have also successfully restored the nerve growth factor receptor expression in the cell lines which lacked expression of nerve growth factor receptors. In order to clearify the role of N-myc oncogenes during nerve growth factor-induced differentiation of neuroblastoma cell lines, a new retroviral vector was constructed to express antisense N-myc genes. This new vector contained a puromycin resistant gene, which allowed the authors to express another gene in the cell lines which had already been neomycin resistant.

[Expression and clinical significance of PCNA in renal pelvic and ureteral cancer].

To determine the relationship between PCNA expression and pathological stage, cellular grade, recurrence and prognosis of renal pelvic and ureteral cancer, research on PCNA proliferating index in 47 cases of renal pelvic and ureteral cancer we studied by immunohistochemistry S-P method. PCNA proliferating index of renal pelvic and ureteral cancer increased with pathologic stage and cellular grade. PCNA proliferating index of T3 + T4 was significantly higher than that of T1 + T2 (P < 0.01). PCNA proliferating index of G3 was significantly higher than that of G1 + G2 (P < 0.001). Those with higher PCNA proliferating index (III + IV) had higher bladder cancer recurrent rate after operation (60%, 12/20), those with lower PCNA proliferating index (I + II) had lower bladder cancer recurrent rate (22.2%, 6/27). P < 0.01. 35% (7/20) of those with III + IV PCNA proliferating index had simultaneous polyorganic urothalial cancer, 7.4% (2/27) of those with I + II PCNA proliferating index had simultaneous polyorganic urothelial cancer, P < 0.05. 7.4% (2/27) of those with I + II PCNA proliferating index had local recurrence and metastasis after operation, 40% (8/20) of those with III + IV had local recurrence and metastasis, P < 0.05. The 5-year survival rate of I + II was 86.7%, the 5-year survival rated of III + IV was 35.7% (5/14), P < 0.05. The findings suggest that PCNA proliferating index connects with pathological stage and cellular grade of renal pelvic and ureteral cancer. It is an important characteristic of low differentiation, invasiveness and simultaneous or sequentially polyorganic urothelial cancer. It may be an important prognostic indicator of renal pelvic and ureteral cancer.

Cataract extraction by phacoemulsification in situ and elliptical intraocular lens implantation.

OBJECTIVE: To investigate the technique of phacoemulsification in situ for cataract extraction and elliptical intraocular lens (IOL) implantation. METHODS: 60 eyes of 57 cases with cataract were included and the hardness of nucleus was grade-1-2 in 25 eyes, grade-3 in 27 eyes and grade-4 in 8 eyes. Tunnel incision was made and in situ phacoemulsification with elliptical (5 mm x 6 mm) IOL implantation was performed on all the cases. RESULTS: Phaco-spent-time was 45 seconds to 3 minutes 45 seconds (average 1 minute 52 seconds). Phacoemulsification in 53 eyes (88.3%) was successful in situ Visual acuities of 0.5 or better and 1.0 or better at postoperative one week were obtained in 75.0% and 56.7%, and at 1-12 month follow-up in 85.0% and 71.7% respectively. CONCLUSION: The technique of phacoemulsification in situ and elliptical IOL implantation is worthy to be spread due to minimum postoperative astigmatism and fast restoration of visual acuity.

Phase II study of dual 131I-labeled monoclonal antibody therapy with interferon in patients with metastatic colorectal cancer.

The combination of COL-1 (anti-CEA) and CC49 (anti-TAG-72) has shown an increase in binding and distribution in colon cancer by immunoperoxidase staining compared to either antibody alone. To overcome tumor heterogeneity and allow delivery of higher radiation dose, 131I-labeled COL-1 and CC49 at a total dose of 75 mCi/m2 (2775 MBq/m2) were simultaneously administered to 14 patients with metastatic colon cancer. alpha-IFN (3 x 10(6) IU) was given s.c. on days -5 to +3 to increase carcinoembryonic antigen and TAG-72 antigen expression. Most patients had mild symptoms during IFN therapy, including mild neutropenia, fever, and malaise, which rapidly subsided after IFN cessation. No acute allergic reactions occurred with radioimmunotherapy; two patients experienced transient, delayed grade 2 arthralgias. Transient neutropenia and/or thrombocytopenia, which was maximal at 4-6 weeks, were consistent side effects without adverse events. All patients had tumor localization, and 13 of 14 patients achieved 4+ (highest grade) localization readings to at least one known site of disease. No objective responses occurred; 4 patients were stable and 10 progressed. Tumor dose estimates varied from 393 to 1327 cGy, including liver and extrahepatic sites. Combining two complementary antibodies and IFN administration appeared to increase localization intensity and radiation doses at tumor sites as compared to historical controls. The amount of radiation delivered to tumor sites was still below that required to cause tumor regressions in metastatic colorectal cancer.
