Linear alkylbenzenes in muscle tissues of white croaker near a large ocean outfall in southern California, USA.

Muscle tissues of a bottom-dwelling marine fish, white croaker (Genyonemus lineatus), collected near a large wastewater outfall in southern California, USA, were analyzed for long-chain linear alkylbenzenes (LABs). Total LABs (summed concentrations of C11 through C14 isomers) were highest (166-748 ng g-1 wet wt) in individuals collected in the immediate vicinity of the Orange County Sanitation District (OCSD; Fountain Valley, CA, USA) outfall diffuser, whereas relatively lower concentrations occurred in fish from mid-shelf and inshore locations at distances of 2.5 and 5 km, respectively, from the outfall. Fish tissue LAB concentrations were roughly proportional to sediment LAB concentrations at the respective collection sites. The extent of LAB degradation, as determined by ratios of internal to external C12 isomers, did not appear to relate to LAB concentrations or sampling location. Tissue DDT and PCB concentrations were not significantly correlated with LABs and, thus, did not appear to relate to recent exposures to sewage residues from the OCSD discharge. Measurements of LAB concentrations in fish tissues may be widely applicable as a monitoring tool for interpreting exposures to sewage discharges.

Alterations in the adhesion behavior of osteoblasts by titanium particle loading: inhibition of cell function and gene expression.

Total joint replacement prostheses are required to withstand corrosive environments and sustain millions of loading and articulation cycles during their term of implantation. Wear debris generation has been implicated as one of the primary causes of periprosthetic osteolysis and subsequent implant loosening in total joint replacements. Particulate debris consisting of metals, polyethylene, ceramics, and bone cement have each been shown to provoke a biological response in joint tissues. The major cell types within the interfacial granulomatous fibrous tissues consist of fibroblasts, macrophages, lymphocytes, and foreign-body giant cells. Osteoblasts are one of the principal cell types in the bone tissue adjacent to prostheses, maintaining physiologic bone remodeling through the balanced coordination of bone formation and resorption in concert with osteoclasts. To date the phenomenon of osteoblast phagocytosis of titanium particles has been suggested, but has not been sufficiently studied or confirmed. This study seeks to clarify the influence of titanium particles on osteoblast adhesion, deformability, proliferation, and gene expression profile. These studies were accomplished by performing biorheological testing, Northern blot analysis and RNase protection assay. The uptake of metallic particles by the osteoblast resulted in a particle-filament complex formation, which induced a series of variations in cell function. Understanding these variations is critical to expanding our knowledge of implant loosening and elucidating the nature of prosthetic joint failure. This study suggests that the impact of titanium particles on osteoblast function and subsequent implant loosening may have been previously underestimated.

Automatic analysis of image of surface structure of cell wall-deficient EVC.

Some computer applications for cell characterization in medicine and biology, such as analysis of surface structure of cell wall-deficient EVC (El Tor Vibrio of Cholera), operate with cell samples taken from very small areas of interest. In order to perform texture characterization in such an application, only a few texture operators can be employed: the operators should be insensitive to noise and image distortion and be reliable in order to estimate texture quality from images. Therefore, we introduce wavelet theory and mathematical morphology to analyse the cellular surface micro-area image obtained by SEM (Scanning Electron Microscope). In order to describe the quality of surface structure of cell wall-deficient EVC, we propose a fully automatic computerized method. The image analysis process is carried out in two steps. In the first, we decompose the given image by dyadic wavelet transform and form an image approximation with higher resolution, by doing so, we perform edge detection of given images efficiently. In the second, we introduce many operations of mathematical morphology to obtain morphological quantitative parameters of surface structure of cell wall-deficient EVC. The obtained results prove that the method can eliminate noise, detect the edge and extract the feature parameters validly. In this work, we have built automatic analytic software named "EVC.CELL".

[A research on the quantification of retinal blood vessels in cases of arteriovenous crossing pressure syndrome caused by hypertension].

The method for quantification of retinal blood vessels in cases of arteriovenous crossing pressure syndrome caused by hypertension was studied. The matched filtering pre-processing of image enhancement for local retinal blood vessels and closing of mathematical morphology for image smoothing, and the method of scale-up lineation for contour determination of retinal blood vessels and optimal seeking algorithm with reversible quantification were used to measure the pathological deformation of vessel, thus providing a computer method for auxiliary diagnosis of relative diseases.

[Long-term morphological changes in mucosa of intestinal urinary reservoir].

OBJECTIVE: To investigate the relationship between long-term morphological changes in the mucosa of the intestinal urinary reservoir (IUR) and postoperative complications. METHODS: A total of 25 patients undergoing the replacement of bladder with IUR were followed up. Of these 25 patients, 15 had colonic pouch and 10 ileal pouch. Tissues of IUR were biopsied examined with microscope and transmission electron microscope. RESULTS: In patients with colonic pouch, the glandular depth was (0.17 +/- 0.04) mm and microvillous height (0.35 +/- 0.05) micron, which were significantly lower than those (0.42 +/- 0.04) micron and (0.69 +/- 0.08) mm in the control group. In those with ileal pouch, the microvillous height was (0.70 +/- 0.14) mm, (0.54 +/- 0.10) mm and (1.99 +/- 0.14) mm in the short-term group, long-term group and control group, respectively. As compared with the control group, mucous atrophy in these patients was more severe. Tight junctions of the epithelium in both kinds of pouches were well preserved even in the long-term cases. No remarkable malignant changes were found in the IUR mucosa in all the patients. Meanwhile, no special morphological changes were seen in the patients with metabolic acidosis or pouch calculus. CONCLUSIONS: IUR mucosa has some morphological "adaptive" changes after the operation. Intestinal segments are ideal bladder substitutions.

[Classification study on the marshland in endemic areas of Schistosoma japonicum using satellite TM image data].

OBJECTIVE: To create a land cover map of the marshland region of Schistosoma Japonicum using satellite TM data from Chayegang, Jiangxi Province. METHODS: Satellite data of three TM bands, namely TM3, TM4 and TM5, were selected to be used in classification study with Principal Components Analysis. Then TM satellite images were applied to false color composite and unsupervised classification with computer software to manage the TM data. Finally the resulting clusters were identified by undertaking site visits and a systematic sampling snail survey was also carried out in the study area in April, 2000. RESULTS: Three land cover classes were generated, including class 1, the carex zone that is both the dominant vegetation and main snail habitats of the study area. Based on the snail survey in the spring of 2000, the live snail density and infectious snail density of the carex zone were 2.51/0.11 m2 and 0.006 9/0.11 m2, respectively. The infection rate of the snail was 0.28%. No snail was found in land cover class 3, the mixed vegetation, because the altitude of this area is either above 17 m or under 14 m and therefore the vegetation in this area is unsuitable for snails to survive. CONCLUSION: Remote sensing can contribute the study on the distribution of snail habitats and become a promising epidemiological study method for schistosomiasis and other ecological diseases.

[Cytotoxic natural killer/T-cell lymphomas of the lymph nodes].

OBJECTIVE: To investigate the clinicopathologic features of cytotoxic natural killer (NK)/T-cell lymphomas of the lymph nodes. METHODS: Clinicopathologic observations and follow up on 5 cases of cytotoxic NK/T-cell lymphomas of the lymph nodes, immunohistochemical staining for CD45RO, CD8, CD56, CD30, CD20, TIA-1 and in situ hybridization for EBER1/2 were performed. RESULTS: (1) The clinicopathologic features of cytotoxic NK-T-cell lymphomas of the lymph nodes were summarized; (2) The tumor cells were positive for CD45RO in 4 of the 5 cases, in which 3 cases were also positive for CD56. One case was of null cell type. The tumor cells were positive for TIA-1 and EBER1/2 in all 5 cases. CONCLUSIONS: The clinicopathological features and special immunophenotypes of cytotoxic NK/T-cell lymphomas of the lymph nodes were present in all 5 cases. Clinically, this entity tends to be an aggressive process with poor prognosis.

[Optimized linear temperature programming for capillary gas chromatography by the method of gridding search].

Linear temperature programming of capillary gas chromatography was optimized by using a new method of gridding search. A point of gridding demonstrated a condition of temperature programmed and the resolution and the retention time were optimized at all gridding points. With a new design of calculation, the method not only considered directly all components that were separated, but the velocity of calculation was improved also. Twenty-one compounds of alkanes, alcohols, halo-hydrocarbons and aromatic hydrocarbons, of which Kovats Indexes are varying from 600 to 1,000, were separated by this new method on an OV-101 column under linear temperature programming. Good results were obtained. The time for optimizing the 21 compounds was less than 1 min.

[The photostability of hydrocortisone injection].

OBJECTIVE: This experiment was designed to make known wether the photostability of drugs in daylight can be obtained from lamplight exposure. METHODS: A self made cumulative illuminometer via pulse counting method was used for measuring cumulative illuminance of daylight to investige the photodegradation of Hydrocortisone injection in various lamp light and daylight; the equivalent influences of different light sources on the photostability of the injection were obtained, and the shelf-life in indoor daylight of the drug was predicted. RESULTS: The photodegradation of Hydrocortisone injection obeys zero order kinetics: C = C0-kEt; the shelf-life in indoor daylight of the injection in nude ampoules was predicted as 36 days and is comparable to 35 days in a long-term storage test. CONCLUSION: The photostability of drugs in daylight can be obtained from lamplight exposure experiment.

Unusual renal features of Lowe syndrome in a mildly affected boy.

The oculocerebrorenal syndrome of Lowe (OCRL) is an X-linked disorder characterized by congenital cataracts, mental retardation, and renal tubular dysfunction. The gene responsible for OCRL was identified by positional cloning and encodes a lipid phosphatase, phosphatidylinositol 4,5, bisphosphate [PtdIns(4,5)P2]5-phosphatase, which localizes to the Golgi apparatus and is suspected to play a role in Golgi vesicular transport [Suchy et al., 1995]. In addition to the ocular and renal manifestations, most boys with OCRL have cognitive problems and maladaptive behaviors including tantrums and stereotypies. We report a boy with a history of congenital cataracts and mild developmental delay who was also found to have hematuria with proteinuria but minimal signs of renal tubular dysfunction. Subsequent renal biopsy was compatible with a diagnosis of a noncomplement fixating chronic glomerulonephritis. Despite the atypical renal findings, skin fibroblast analysis for PtdIns (4,5)P2 5-phosphatase was performed, and enzyme activity was low, consistent with the diagnosis of OCRL. Western blot analysis from cell lysates showed the ocrl protein was decreased in size and amount. Our report shows atypical renal features of OCRL in a mildly affected boy. The possibility of OCRL should be considered in boys with cataracts and glomerular disease, even in the absence of renal tubular defects and frank mental retardation usually associated with the syndrome. 3D domain swapping modulates the stability of members of an icosahedral virus group.

BACKGROUND: Rice yellow mottle virus (RYMV) is a major pathogen that dramatically reduces rice production in many African countries. RYMV belongs to the genus sobemovirus, one group of plant viruses with icosahedral capsids and single-stranded, positive-sense RNA genomes. RESULTS: The structure of RYMV was determined and refined to 2.8 A resolution by X-ray crystallography. The capsid contains 180 copies of the coat protein subunit arranged with T = 3 icosahedral symmetry. Each subunit adopts a jelly-roll beta sandwich fold. The RYMV capsid structure is similar to those of other sobemoviruses. When compared with these viruses, however, the betaA arm of the RYMV C subunit, which is a molecular switch that regulates quasi-equivalent subunit interactions, is swapped with the 2-fold-related betaA arm to a similar, noncovalent bonding environment. This exchange of identical structural elements across a symmetry axis is categorized as 3D domain swapping and produces long-range interactions throughout the icosahedral surface lattice. Biochemical analysis supports the notion that 3D domain swapping increases the stability of RYMV. CONCLUSIONS: The quasi-equivalent interactions between the RYMV proteins are regulated by the N-terminal ordered residues of the betaA arm, which functions as a molecular switch. Comparative analysis suggests that this molecular switch can also modulate the stability of the viral capsids.

Role of acidic sphingomyelinase in Fas/CD95-mediated cell death.

Engagement of the Fas receptor has been reported to induce ceramide generation via activation of acidic sphingomyelinase (aSMase). However, the role of aSMase in Fas-mediated cell death is controversial. Using genetically engineered mice deficient in the aSMase gene (aSMase(-/-)), we found that thymocytes, concanavalin A-activated T cells, and lipopolysaccharide-activated B cells derived from both aSMase(-/-) and aSMase(+/+) mice were equally sensitive to Fas-mediated cell death, triggered by either anti-Fas antibody or Fas ligand in vitro. Similarly, activation-induced apoptosis of T lymphocytes was unaffected by the status of aSMase, and aSMase(-/-) mice failed to show immunological symptoms seen in animals with defects in Fas function. In vivo, intravenous injection of 3 microg/25 g mouse body weight of anti-Fas Jo2 antibody into aSMase(-/-) mice failed to affect hepatocyte apoptosis or mortality, whereas massive hepatocyte apoptosis and animal death occurred in wild type littermates. Animals heterozygous for aSMase deficiency were also significantly protected. Susceptibility of aSMase(-/-) mice to anti-Fas antibody was demonstrated with higher antibody doses (>/=4 microg/25 g mouse). These data indicate a role for aSMase in Fas-mediated cell death in some but not all tissues.

The binding of Na(+) to apo-enolase permits the binding of substrate.

Enolase from rabbit muscle (betabeta-enolase) is inactivated by NaClO(4). Enolase free of divalent cations is more susceptible to inactivation by NaClO(4) than is enolase in the presence of Mg(2+). We find that substrate protects apo-enolase against inactivation, indicating that substrate can bind to enolase in the absence of a divalent cation. This binding is not due to contamination by trace levels of divalent cations since (1) it occurs even in the presence of EDTA or EGTA and (2) metal analysis by ICP (inductively coupled plasma) mass spectrometry did not reveal sufficient contamination to account for the protection. The binding of PGA to apo-enolase did require Na(+). When TMAClO(4) was used instead of NaClO(4), there was no protection by PGA. Protection was restored when TMAClO(4) plus NaCl were used. The inactivation of apo-enolase by NaClO(4) is due to dissociation into inactive monomers. We conclude that Na(+) binds to apo-enolase, permitting substrate to then bind. Of the three known Me(2+) binding sites on enolase, we believe the most likely binding site for Na(+) is the carboxylate cluster of site 1, the highest affinity site of enolase.

Titanium particles inhibit osteoblast adhesion to fibronectin-coated substrates.

To illuminate the effect of titanium particles on osteoblast function, we compared the adhesion force of neonatal rat calvarial osteoblasts on fibronectin-coated glass after incubation with titanium particles (80% had diameters of less than 5 microm). The cells were incubated with the particles for 1.5-72 hours. Using a micropipette single-cell manipulation system, we showed that the adhesion force of the osteoblasts to fibronectin-coated glass (1.0 microg/ml) was significantly affected by the presence of particulate debris. The adhesion force of the cells incubated with titanium particles for less than 4 hours was not significantly affected by exposure to the particles; after 4 hours, however, it was significantly reduced relative to that of controls. Aspiration of particle-challenged osteoblasts into the micropipette demonstrated that the particles were not stripped from the cell surface and therefore confirmed that the osteoblasts had ingested them. During aspiration, the particles traveled through the cytoplasm rather than on the cell surface. When the osteoblasts were exposed to the particles and cytochalasin D, they exhibited much lower adhesion forces than did the controls or the cells exposed to titanium particles only; this indicates an important role of actin filaments in the osteoblastic response to particles. Staining for F-actin also indicated an influence of internalized titanium particulate on cytoskeletal arrangement and cell spreading. Furthermore, with standard Northern blotting techniques, levels of mRNA for collagen type I and fibronectin were significantly reduced as early as 4 hours after exposure to particles compared with levels in controls, and this effect continued to 72 hours. These data indicate that direct exposure of osteoblasts to titanium particles, which we propose to be ingested by the osteoblasts, can significantly decrease osteoblast adhesion force; this may lead to decreased cellular activity and gene expression of fibronectin and collagen type I in the presence of titanium wear debris.

Time-dependent increases in type-III collagen gene expression in medical collateral ligament fibroblasts under cyclic strains.

Numerous studies have demonstrated the capacity of mechanical strains to modulate cell behavior through several different signaling pathways. Understanding the response of ligament fibroblasts to mechanically induced strains may provide useful knowledge for treating ligament injury and improving rehabilitation regimens. Biomechanical studies that quantify strains in the anterior cruciate and medial collateral ligaments have shown that these ligaments are subjected to 4-5% strains during normal activities and can be strained to 7.7% during external application of loads to the knee joint. The objective of this study was to characterize the expression of types I and III collagen in fibroblast monolayers of anterior cruciate and medial collateral ligaments subjected to equibiaxial strains on flexible growth surfaces (0.05 and 0.075 strains) by quantifying levels of mRNA encoding these two proteins. Both cyclic strain magnitudes were studied under a frequency of 1 Hz. The results indicated marked differences in responses to strain regimens not only between types I and III collagen mRNA expression within each cell type but also in patterns of expression between anterior cruciate and medial collateral ligament cells. Whereas anterior cruciate ligament fibroblasts responded to cyclic strains by expression of higher levels of type-I collagen message with almost no significant increases in type-III collagen, medial collateral ligament fibroblasts exhibited statistically significant increases in type-III collagen mRNA at all time points after initiation of strain with almost no significant increases in type-I collagen. Furthermore, differences in responses by fibroblasts from the two ligaments were detected between the two strain magnitudes. In particular, 0.075 strains induced a time-dependent increase in type-III collagen mRNA levels in medial collateral ligament fibroblasts whereas 0.05 strains did not. The strain-induced changes in gene expression of these two collagens may have implications for the healing processes in ligament tissue. The differences may explain, in part, the healing differential between the anterior cruciate and medial collateral ligaments in vivo.

Choroidal vascular permeability in visually regulated eye growth.

The choroidal thickness fluctuates both diurnally and in response to changes in visual input. The fluctuations may represent a physiologic means of aligning the retinal photoreceptors with the focal position of distant images during the emmetropization process. To evaluate the basis for choroidal thickness changes, we studied the sources of the extravascular fluid in the chick choroid in two visually-regulated ocular growth conditions: accelerated ocular growth in goggle-induced form-deprivation myopia and ocular growth retardation in the recovery from myopia after goggle removal. Two week old chicks, controls, myopic and those recovering from myopia, received fluorescein dextran (MW = 140,000) as a tracer. It was given by intravenous injection to identify a potential vascular pathway and by intracameral injection to identify a potential pathway from the anterior chamber to the suprachoroidal space. Using a microscopically positioned needle, clear fluid was aspirated from the suprachoroidal space of the enucleated chick eye; this fluid presumably corresponds to the contents of the lacunae, prominent lymphatic-like structures of the chick choroid. Plasma, aqueous humor and suprachoroidal fluid were sampled 1 hr after injection and assayed for both protein content and the tracer dye. Sulfated glycosaminoglycans were assayed in the suprachoroidal fluid, choroid and sclera under each experimental condition. In control chicks, aqueous humor and suprachoroidal fluid protein concentrations were about 0.8 and 9% of plasma levels respectively. Aqueous humor protein concentration was unaltered in myopic or recovering eyes. Suprachoroidal fluid protein concentration in myopic eyes fell dramatically to 1.5% of plasma levels (P < 0.001). In contrast, recovery from myopia led to a marked increase in suprachoroidal fluid protein level to 30% of that in plasma (P < 0.001). None of the procedures affected suprachoroidal fluid protein in the contralateral control eyes. In all three groups of chicks, fluorescein dextran distribution in the suprachoroidal fluid at 1 hr after intravenous injection tracked protein levels, with reduced levels in myopic eyes and elevated levels in recovering eyes. After intracameral injection, suprachoroidal fluid dextran levels were higher in injected eyes of control chicks (P < 0.01) and in recovering eyes (P < 0.001) but lower in myopic eyes (P < 0.01), compared to the levels in the respective contralateral non-injected eyes in each group. Sulfated glycosaminoglycan levels were at the limits of detection in the suprachoroidal fluid under all conditions and, on a whole choroid basis, were unaltered in the choroid in either myopia or recovery. Suprachoroidal fluid is lymph-like in nature and largely derives from plasma. Sulfated glycosaminoglycan levels do not seem to regulate the fluid content of the choroid in either myopia or recovery. Instead, the changes in protein and marker dye levels in myopic and recovering eyes suggest markedly altered choroidal circulatory dynamics and capillary permeability in both conditions.

The effects of calcium phosphate cement particles on osteoblast functions.

Calcium phosphate cements (CPC) are increasingly used in the orthopedic field. This kind of cement has potential applications in bone defect replacements, osteosynthetic screw reinforcements or drug delivery. In vivo studies have demonstrated a good osteointegration of CPC. However, it was also observed that the resorption of CPC could create particles. It is known from orthopedic implant studies that particles can be responsible for the peri-implant osteolysis. Biocompatibility assessment of CPC should then be performed with particles. In this study, we quantified the functions of osteoblasts in the presence of beta-TCP, brushite and cement particles. Two particle sizes were prepared. The first one corresponded to the critical diameter range 1-10 microm and the second one had a diameter larger than 10 microm. We found that CPC particles could adversely affect the osteoblast functions. A decrease in viability, proliferation and production of extracellular matrix was measured. A dose effect was also observed. A ratio of 50 CPC particles per osteoblast could be considered as the maximum number of particles supported by an osteoblast. The smaller particles had stronger negative effects on osteoblast functions than the larger ones. Future CPC development should minimize the generation of particles smaller than 10 microm.
