Phe-46(CD4) orients the distal histidine for hydrogen bonding to bound ligands in sperm whale myoglobin.

The role of Phe-46(CD4) in modulating the functional properties of sperm whale myoglobin was investigated by replacing this residue with Leu, Ile, Val, Ala, Trp, Tyr, and Glu. This highly conserved amino acid almost makes direct contact with the distal histidine and has been postulated to affect ligand binding. The overall association rate constants for CO, O2, and NO binding were little affected by decreasing the size of residue 46 step-wise from Phe to Leu to Val to Ala. In contrast, the rates of CO, O2, and NO dissociation increased 4-, 10-, and 25-fold, respectively, for the same series of mutants, causing large decreases in the affinity of myoglobin for all three diatomic gases. The rates of autooxidation at 37 degrees C, pH 7.0 increased dramatically from approximately 0.1-0.3 h-1 for wild-type, Tyr-46, and Trp-46 myoglobins to 1.5, 5.2, 4.9, and 5.0 h-1 for the Leu-46, Ile-46, Val-46 and Ala-46 mutants, respectively. Rates of NO and O2 geminate recombination were measured using 35 ps and 9 ns laser excitation pulses. Decreasing the size of residue 46 causes significant decreases in the extent of both picosecond and nanosecond rebinding processes. High resolution structures of Leu-46 and Val-46 metmyoglobins, Val-46 CO-myoglobin, and Val-46 deoxymyoglobin were determined by X-ray crystallography. When Phe-46 is replaced by Val, the loss of internal packing volume is compensated by (1) contraction of the CD corner toward the core of the protein, (2) movement of the E-helix toward the mutation site, (3) greater exposure of the distal pocket to intruding solvent molecules, and (4) large disorder in the position of the side chain of the distal histidine (His-64). In wild-type myoglobin, the van der Waals contact between C zeta of Phe-46 and C beta of His-64 appears to restrict rotation of the imidazole side chain. Insertion of Val at position 46 relieves this steric restriction, allowing the imidazole side chain to rotate about the C alpha - C beta bond toward the surface of the globin and about the C beta - C gamma bond toward the space previously occupied by the native Phe-46 side chain. This movement disrupts hydrogen bonding with bound ligands, causing significant decreases in affinity, and opens the distal pocket to solvent water molecules, causing marked increases in the rate of autooxidation.(ABSTRACT TRUNCATED AT 400 WORDS)

Combination therapy with probucol prevents adriamycin-induced cardiomyopathy.

Adriamycin (doxorubicin) is a broad spectrum anti-tumor antibiotic used to treat cancer patients. However, the potential usefulness of this drug is currently limited by the development of a dose-dependent cardiomyopathic process terminating in severe heart failure. Although several mechanisms have been suggested to explain the pathogenesis of adriamycin-induced cardiomyopathy, free-radical induced oxidative stress appears to play an important role. A concise description of adriamycin-induced cardiomyopathy is provided. Various combination therapies which have been attempted in the past to modulate the adriamycin-induced cardiomyopathy are also discussed. Recently, it has been discovered that probucol, a lipid lowering agent and potent antioxidant, provides complete protection against adriamycin-induced cardiomyopathy in rats without interfering with the anti-tumor properties of this antibiotic. Clinical trials employing adriamycin therapy in combination with probucol are needed to determine the applied value of these laboratory findings.

Differences in eye growth and the response to visual deprivation in different strains of chicken.

Several laboratories studying visual deprivation myopia in the domestic chick report varying degrees of axial elongation and myopia induced by similar visual deprivation techniques. In this study we tested the hypothesis that in different strains of chick the eyes respond differently to visual deprivation. We compared under identical conditions two strains of White Leghorn chick commonly used in ocular development research--the Cornell-K strain (K) and Washington H & N Strain (H/N). The normal development of the eye was found to vary significantly between these strains of White Leghorn chicks. The K strain normally develops flatter corneas, thicker lenses, and larger eyes than the H/N strain. The response to visual deprivation also varies significantly between strains. For example, we find that 2 weeks of visual deprivation in the K strain results in less elongation of the vitreous chamber and flattening of the cornea yielding lower levels of induced myopia compared to the H/N strain. Our results show that while visual experience clearly affects normal ocular development in both strains of chick, the nature of the effect depends upon not only the type and duration of the experience but the genetics of the subject population as well.

Constant light produces severe corneal flattening and hyperopia in chickens.

In this study we report on the effects of constant light (CL) on the refractive development and ocular morphology of White Leghorn chicks (Cornell K-strain). Refractive state and corneal curvature were measured by IR photoretinoscopy and IR keratometry respectively. The axial lengths of the ocular components were measured by A-scan ultrasonography. We find that constant light produces significant hyperopia compared to controls in as few as 10 days (7.4 vs 4.0 D). This is apparently the result of flatter than normal corneal curvature (radius of curvature: 3.22 vs 3.08 mm) as vitreous chamber depth is significantly deeper in CL eyes than controls at that age (5.6 vs 5.1 mm). In contrast to other reports, if CL rearing is continued for longer periods the hyperopia progresses, even though vitreous chamber depth continues to increase. After 11 weeks of CL severe hyperopia was observed (18.2 vs 2.8 D). Long term CL is also found to produce shallow anterior chambers, corneal thickening, lenticular thinning and cataracts, and damage to the retina, pigment epithelium, and choroid.

Activation of Bruton's tyrosine kinase (BTK) by a point mutation in its pleckstrin homology (PH) domain.

Bruton's tyrosine kinase (BTK) is a nonreceptor tyrosine kinase critical for B cell development and function. Mutations in BTK result in X-linked agammaglobulinemia (XLA) in humans and X-linked immunodeficiency (xid) in mice. Using a random mutagenesis scheme, we isolated a gain-of-function mutant called BTK* whose expression drives growth of NIH 3T3 cells in soft agar. BTK* results from a single point mutation in the pleckstrin homology (PH) domain, where a Glu is replaced by Lys at residue 41. BTK* shows an increase in phosphorylation on tyrosine residues and an increase in membrane targeting. Transforming activity requires kinase activity, a putative autophosphorylation site, and a functional PH domain. Mutation of the SH2 or SH3 domains did not affect the activity of BTK*. Expression of BTK* could also relieve IL-5 dependence of a B lineage cell line. These results show that transformation activation and regulation of BTK are critically dependent on the PH domain.

Simultaneous determination of guanine, uric acid, hypoxanthine and xanthine in human plasma by reversed-phase high-performance liquid chromatography with amperometric detection.

A reversed-phase high-performance liquid chromatographic method with amperometric detection is described for the separation and quantification of uric acid, guanine, hypoxanthine and xanthine. The isocratic separation of a standard mixture of the compounds was achieved in 5 min on a Spherisorb 5 C18 reversed-phase column, with a mobile phase of NaH2PO4 (300 mmol dm-3, pH 3.0)-methanol-acetonitrile-tetrahydrofuran (97.8 + 0.5 + 1.5 + 0.2). Uric acid, guanine, hypoxanthine and xanthine were completely separated, with detection limits in the range 2-20 pmol per injection. The effect of pH and the composition of the mobile phase on the separation are described. The hydrodynamic voltammograms of these compounds were recorded at a glassy carbon electrode. The linear range of the calibration graph for each compound was: uric acid, 1-5000 mumol dm-3; guanine, 0.5-2000 mumol dm-3; hypoxanthine, 0.1-500 mumol dm-3 and xanthine, 0.5-5000 mumol dm-3. The within- and between-day precision was good. The uric acid and hypoxanthine content in human plasma was measured using the proposed method. Good recoveries of uric acid (97.9-103%), hypoxanthine (98.0-99.2%), guanine (96.0-98.3%) and xanthine (96.0-102%) were obtained from human plasma. The results of electrochemical detection were in good agreement with those of UV detection.

Molecular genetics of retinitis pigmentosa.

Hereditary degenerations and dysfunctions of the retina are an extremely heterogeneous group of diseases. This summary deals with recent advances in the molecular genetics of a subset of those disorders, namely, those encompassed under the diagnosis 'retinitis pigmentosa'. Over 20 loci where mutations cause retinitis pigmentosa have been mapped; the review focuses on the seven retinitis pigmentosa loci that have been identified.

Comparative plasma and tissue pharmacokinetics and drug residue profiles of different chemotherapeutants in fowls and rabbits.

Blood and tissue pharmacokinetics and drug residue profiles of six chemotherapeutants were studied. Ceftriaxone (CEF), intravenously at 50 mg/kg, sulfamonomethoxine (SMM) and sulfaquinoxaline (SQ), orally at 200 mg/kg, and olaquindox (OLA), orally at 50 mg/kg, were administered to young broilers. Penicillin (PEN), intramuscularly at 200,000 U/kg, and albendazole (ALB), orally at 20 mg/kg, were given to rabbits. For each drug, 13-18 groups (n = 5-10 individuals/group) of the dosed animals were killed at different post-dosing times. Drug and/or metabolite concentrations in plasma, liver, kidney, heart, lung, and muscle tissues were analysed by HPLC procedures. Multi-exponential kinetic models were fitted to the observed tissue concentration-time data by applying a non-linear least-squares regression computer program. Tissue half-life, peak tissue concentration, and time of peak tissue concentration were determined. Half-life of CEF, SMM, SQ, OLA, PEN, ALB, and two metabolites of ALB (sulfoxide and sulfone) in various tissues ranged 0.6-1.4, 4.7-9.0, 4.5-18.9, 1.8-3.1, 0.9-3.0, 3.4-9.6, 5.0-16.1 and 7.4-12.2 h. The times required for CEF, SMM, SQ, OLA, PEN, and ALB residue concentrations to decline to 0.1 microgram/g in various tissues ranged from 5.0-11.6, 70.0-110.5, 114.0-179.8, 21.3-30.3, 4.1-24.8 and 47.8-84.4 h. Drug kinetic characteristics in tissues differed significantly from those in plasma, and also varied from tissue to tissue. It is necessary, therefore, to evaluate tissue kinetics when designing dosage regimens in tissue infection chemotherapy with these drugs. Knowledge of tissue kinetics is also important in predicting and controlling drug residues in edible tissues of food-producing animals.

Concomitant immunity against tumor development is enhanced by the oral administration of a kampo medicine, Hochu-ekki-to (TJ-41: Bu-Zhong-Yi-Qi-Tang).

The oral administration of a kampo herbal medicine, Hochu-ekki-to (TJ-41: Bu-Zhong-Yi-Qi-Tang) using a water-supplying bottle resulted in a slight but significant inhibition of Meth A growth. The oral administration of TJ-41 with gastric gavage significantly enhanced the specific antitumor activity against Meth A at rechallenge on day 9. In a tumor-neutralizing assay, the tumor draining LN cells of the TJ-41 administered mice showed an antitumor activity against Meth A. In a cytolytic assay, the anti-Meth A specific cytolytic T lymphocyte activity was not detected in the spleen cells of the Meth A bearing and TJ-41 administered mice. The oral administration of TJ-41 enhanced the natural killer (NK) activity of the spleen cells in naive mice but could not improve the decreased NK activity of spleen cells from the tumor bearing mice. In a cytostatic assay, the peritoneal exudate cells from the Meth A bearing and TJ-41 administered mice showed a significantly higher amount of cytostatic activity against Meth A than that from either Meth A bearing or TJ-41 administered mice. These results indicate that the oral administration of TJ-41 into the tumor bearing mice may thus be able to enhance concomitant antitumor immunity through the augmentation of the cytostatic activity.

[The protective effect of IL-1 beta on stress-induced gastric mucosal damage in rat].

The effect of recombinant IL-1 beta on stress-induced gastric mucosal lessions was studied in rats. Pretreatment with IL-1 beta prevented formation of gastric mucosal damage in a dose-dependent manner. IL-1 receptor antagonist peptide (IRAP) could totally reverse the protective effect of IL-1 beta. IL-1 fragment peptide (163-171) had no effect on gastric ulcer formation in the experimental model, whereas sulfhydryl blocker N-ehtylmaleimide partially blocked the protective effect of IL-1 beta. The concentration of protein and noneprotein sulfhydryls in the gastric mucosa was significantly decreased 3 h after stress, and this decrease was partially prevented by IL-1 beta. IL-1 beta also decreased the concentration of molondialdehyde (MDA) in the gastric mucosa after 3 h stress. The results suggest that IL-1 beta could effectively lessen the degree of stress-induced gastric mucosal damage, due possibly to the production of endogenous sulfhydryl compounds in the gastric mucosa.

[Long-term effect of leprosy control in both highly endemic prefectures, Weifang of Shandong and Wenshan of Yunnan].

The long-term effect of leprosy control was studied in two prefectures of different geographical conditions and socio-economic status. During 1955-1985, the incidence rate in Weifang of Shandong decreased by 94.5% (9.1-0.5/10,000) and became evident after the introduction of MDT in 1986 (0.04/10,000, 1993). In Wenshan of Yunnan, however, it remained at about 20/10,000 before 1980. After the introduction of rifampin plus dapsone in 1979 and fixed duration MDT in 1986, it decreased by 94% (20-1.2/10,000), whereas the detection rate remained constant (+5/100,000). In comparison with the features of new patients detected since 1980, we noted a steady increase of MB rate and voluntary reporting, and a decrease of deformity rate, in the last 8 years in Weifang. It is estimated that by the year 2000 Weifang may have the detection rate of 0.001260/10 thousand and the incidence rate of 0/00757/10 thousand in contrast to the detection rate of 0.2980/10 thousand and the incidence rate of 0.316/10 thousand in Wenshan.

[Preliminary studies on microbe-mediated N-nitrosamide synthesis].

Chemical synthesis of N-nitrosamide and influences on it by bacteria and fungi isolated from human gastric juice were studied with trace analysis method to explore the pathway of its endogenous formation. Results showed in synthesis of N-methyl-N-nitrosourea (MNU) with precursors of methylurea (0.42 mmol/L) and sodium nitrite (25 mmol/L) in a condition simulating human stomach, pH of the reaction system played an important role in the synthesis, the amount of synthetic chemical maximized at a pH of 1-3 and almost no synthesis was found at pH of 5-7. A study on the ability to synthesize MNU catalyzed by bacteria isolated from the patients with stomach cancer living in a high-risk area showed its synthesis could be accelerated by Pseudomonas aeruginosa at pH of 6-7 with its catalyzing activity existed in heat liable component of living bacteria. This is the first report showing microorganisms to catalyze the formation of N-nitrosamide from its precursors.

[The effect of intralipid on the membrane fluidity of lymphocytes from human blood].

We observed the effect of 10% intralipid on the membrane fluidity of lymphocytes by means of fluorescent polarization measurement. After in vitro incubation with intralipid in concentration 0, 10, 20mg/ml, the lymphocytes from healthy adult blood showed no changes in the membrane fluidity (P > 0.05). In vivo, the membrane fluidity of lymphocytes did not alter significantly during and after infusion of 500ml 10% intralipid at a speed of 100ml per hour into healthy adult (P > 0.05). The level of blood triglyceride was elevated significantly during the infusion (P < 0.01), but decreased rapidly after the infusion. With 500ml per day of infusion (contral group), the surgical patients showed significant decrease in membrane fluidity of lymphocytes after operation (P < 0.01). But there was no significant difference between the study group and control group. The membrane fluidity might be affected by many factors. One of them is membrane lipid. Intralipid mainly contains triglycerides composed of polysaturated fatty acids and lecithin as emulsifier. The result showed that intralipid may neither alter the lipid composition of lymphocytes membrane nor the membrane fluidity. Furthermore, in case of modulation of membrane lipid composition by intralipid occurs, cellular self-regulation may be accomplished by fatty acid synthesis to keep the membrane fluidity stable.
