Nine healthy and seven mite-sensitive allergic subjects were examined by skin sensitivity and IgE antibody to fractions from Dermatophagoides farinae, including high and low allergenic fraction to human. In the intracutaneous test, all healthy subjects reacted to 200 ng and in particular, five of them to 0.2 ng of Fr void on Sephadex G-100 filtration which was a low allergenic fraction. Furthermore, end points of intracutaneous test and specific IgE antibody titers did not correlate on Fr void. Conclusively, Dermatophagoides farinae extracts contain a nonimmunologically highly skin-reactive fraction.

In this paper, the effect of acute human growth hormone (GH) administration on erythrocyte insulin binding in GH deficient children (N = 6) was studied. Following GH (0.25 U/kg) administration, the blood levels of GH peaked within 4 to 8 h and returned to basal levels 24 h later. However, the changes in somatomedin activity, free fatty acid (FFA), urea, blood glucose and 125I-insulin binding to erythrocyte were observed around 24 h following the injection, and there was a converse relationship between maximum percent 125I-insulin binding (IBmax) and FFA (P less than 0.02). By Scatchard analysis it was found that the decrease in IBmax is mainly due to the change in the number of insulin receptors. These results suggest that GH may possibly affect the insulin binding to erythrocyte indirectly through metabolic changes as a result of hormonal changes in GH deficient children.

We have studied the precipitation test as a diagnostic procedure for hypersensitivity pneumonitis or associated allergic states using Sitophilus granarius (SG) antigen. The incidence of positive reaction in the precipitation test against SG is extremely high in children and adults, not only asthmatics, but also in normal, healthy subjects. In experiments with radioimmunoassay using SG paper discs and 125I-labelled IgG fragments, SG was more strongly bound to the Fab than to the Fc fragment. However, the binding was not mediated by a true antigen-antibody reaction because the reaction was inhibited by both kappa- and lambda-type Bence-Jones proteins. Nonspecific immunoglobulin binding substance (IBS) was separated from crude SG extract by affinity chromatography using IgG immunosorbent. IBS was monocomponent on polyacrylamide gel electrophoresis in sodium dodecyl sulfate, and the molecular weight was about 12,000-14,000. We believe that IBS might be a valuable tool for immunological experiments, comparable with staphylococcal protein A.
Thymidine factor was studied in human cord sera from 44 neonates with gestational ages ranging between 36 and 43 weeks. Thymidine factor was determined by the uptake of 6-3H-thymidine using isolated chick embryo cartilage cells. The thymidine factors increased with gestational age and those of neonates at 40 weeks of gestation were 0.89 +/- 0.17 U/ml (mean +/- SD), but were significantly lower after 42 weeks of gestation. There was no correlation between thymidine factor in cord sera and birth weight or size. There was a positive correlation (p less than 0.02) between insulin level and thymidine factor in cord sera. Our data suggest that somatomedin measured as thymidine factor might not have a major role in fetal growth and insulin might be an important factor in the synthesis of somatomedin by the fetus.
Concanavalin A (Con-A)-treated human lymphocytes or their culture fluid suppressed protein A-induced lymphocyte colony formation by more than 40%. The suppressor factor was produced primarily by Con A-stimulated T lymphocytes and maximal production occurred on the 2nd day of culture with 10 microgram Con A, a condition optimal for the induction of suppressor T cells. This suggests that the factors are released by suppressor T cells. We hypothesize that suppressor T cells exert a negative control on clonal proliferation of lymphocytes.
We have studied cyclic adenosine 3':5'-monophosphate (cAMP) and cyclic 3':5'-guanosine monophosphate (cGMP) concentration in cord blood lymphocytes (CBL) and in adult peripheral blood lymphocytes (PBL) after stimulation with thymosin V-1 and V-2. At concentrations of 100 micrograms/ml, thymosin V-1 produced an 85% increase of cAMP in adult PBL, alterations being demonstrated within 1 min, whereas it did not affect the cAMP level in CBL which have a nine times higher basal level than PBL. On the other hand, thymosin V-1 did not have any effect on cGMP in CBL nor in adult PBL. Thymosin V-2 which inhibits E-rosette formation also increased cAMP in adult PBL like thymosin V-1. The increasing effect of thymosin V-1 on cAMP was observed even when cAMP phosphodiesterase was completely inhibited with isobutyl methylxanthine. From these data it was assumed that thymosin probably acts through the stimulation of adenylate cyclase.
Anti-human delta chain antibody was prepared and specific IgD antibodies to Mycoplasma pneumoniae (MAC strain) antigens were studied by means of the RAST. M. pneumoniae antigens were chromatographically separated by Sephadex G-25 and Sephadex G-200 gel filtration. Specific IgD antibody titers were significantly higher in patients with primary atypical pneumonia due to M. pneumoniae infection than those of healthy and disease controls. The specific IgD antibodies increased at an earlier stage of the disease and decreased rapidly. Further studies are needed to clarify the role of the IgD antibodies in regulating antibody production during infection.

Allergenic activities of major component proteins of soybean 11S-, 7S- and 2S-globulins were examined by the radioallergosorbent test (RAST) and by RAST inhibition experiments with the sera from soybean-sensitive patients. The reactivity with specific IgE antibody was found in all fractions. There was considerable cross-reactivity among three fractions, and 2S-globulin had the highest allergen potency to inhibit the RAST against 11S, 7S- and 2S-globulin. When the fractions were heat-treated at 80, 100 and 120 degrees C for 30 min, the RAST reactivity of 2S-globulin heated at 80 degrees C for 30 min was slightly enhanced, while that of others was reduced to 39--75% of that of the native globulin. IgE antibody could be produced in BALB/c mice only with one of the three fractions, i. e. 2S-globulin, and the produced IgE antibody lacked reactivity with 11S- and 7S-globulin.
