A gene coding for a xylanase activity of alkalophilic Aeromonas sp. no. 212 (ATCC 31085) was cloned in Escherichia coli HB101 with pBR322. Plasmid pAX1 was isolated from transformants producing xylanase, and the xylanase gene was located in a 6.0 kb Hind III fragment. The pAX1-encoded xylanase activity in E. coli HB101 was about 80 times higher than that of xylanase L in alkalophilic Aeromonas sp. no. 212. About 40% of the enzyme activity was observed in the periplasmic space of E. coli HB101. The pAX1-encoded xylanase had the same enzymic properties as those of xylanase L produced by alkalophilic Aeromonas sp. no. 212, but its molecular weight was lower (135 000 vs 145 000, as estimated by SDS polyacrylamide gel electrophoresis).
A gene for the beta-lactamase from alkalophilic Bacillus sp. strain 170 was cloned in a functional state on a 1.0 kb DNA fragment and its nucleotide sequence was determined. The coding sequence showed an open reading frame of 257 amino acids, which represents the beta-lactamase precursor protein. It is considered that the signal peptide consisted of 30 amino acids including 12 hydrophobic amino acids.
An alkalophilic Bacillus DNA bank cloned in an expression probe plasmid, pGR71, was screened for the presence of developmentally regulated genetic elements. A 508-base pair HindIII fragment isolated from this bank in plasmid pGR71-5 expressed plasmid-encoded chloramphenicol resistance only during the sporulation phase of a Bacillus subtilis host grown on Schaeffer medium. This developmentally regulated expression was altered in spo0E and spo0H mutants which had very low levels of chloramphenicol acetyltransferase activity relative to the wild type or other spo0 mutants. We determined the nucleotide sequence of the entire 508-base pair fragment and located the site of regulated transcription initiation by high-resolution S1 nuclease mapping of the in vivo transcript. The deduced promoter sequences upstream from this start site were 5'C-G-A-A-T-C-A-T-G-A3' at -10 and 5' A-G-G-A-A-T-C3' at -35. This transcript was not detected in spo0E or spo0H mutants, indicating that the products of these genes control developmentally regulated chloramphenicol acetyltransferase expression at the level of transcription.
The gene for xylanase A of alkalophilic Bacillus sp. strain C-125 was cloned in Escherichia coli with pBR322. The plasmid pCX311 contained 2.6- and 2.0-kilobase-pair HindIII fragments. The characteristics of the purified pCX311-encoded xylanase were the same as those of purified xylanase A from alkalophilic Bacillus sp. strain C-125.
H and A antigens expressed only in the RBC were quantitated using specific antisera, and the amount of the antigens was compared with that of total H and A antigens shared by the RBC and saliva. Total H antigen decreased in the order of O, A, B and AB blood groups. However, H antigen specific for the RBC distributed similarly among four blood groups. In addition, the amount of total A antigen in A group was more than that of AB group, but the difference in the amount of A antigen specific for the RBC was not significant between the two blood groups. B antigen specific for the RBC was not examined because of failure in preparing specific anti-B serum. In conclusion, the quantitative difference of the total H or A between A and AB groups might be due to the difference in the amount of antigen common to the RBC and saliva but not one specific for the RBC.
Catecholamines were measured in the amniotic fluid and in the first voided newborn urine obtained from appropriate-for-date infants of term deliveries. Catecholamine values in the amniotic fluid and urine were nearly equal when expressed in terms of creatinine. Significant positive correlations were observed between the amniotic fluid and urine of norepinephrine and epinephrine. In normal cases (n = 32) that underwent uneventful vaginal delivery, the 95% confidence limits for norepinephrine and epinephrine in the amniotic fluid were 1.53 to 2.33 ng/ml and 0.16 to 0.30 ng/ml, respectively. In cases of moderate stress (n = 12), only norepinephrine showed significantly higher values than the normal cases, while in cases of severe stress (n = 12), norepinephrine became more significantly high, and epinephrine was found to be elevated significantly. A significant difference was noted in the incidence of fetal stress between the infants with more than and those with less than 2.30 ng/ml of norepinephrine, the upper limits of the normal 95% confidence limits. However, for epinephrine such a significant difference was not noted. It was concluded that amniotic fluid catecholamines are of fetal origin and reflect fetal sympathoadrenal activity directly, even during labor, and that their level may be a good indicator of fetal condition and stress.

The characteristics of the infiltration of bile duct carcinoma along the wall were studied from 25 cases with resection. The histologic mode of the spread of carcinoma was the mucosal infiltration in 1 case, mucosal and transmural infiltration in 2 cases, transmural infiltration in 22 cases. The average distance from the gross margin of the tumor to the margin of spread of growth in the bile duct as determined microscopically was 19.1 mm in the direction of the liver (the longest was 85 mm), and 5.8 mm toward the duodenum (the longest was 20 mm). The distance of the polypoid or nodular type of the gross appearance of the tumor is shorter than that of the infiltrating type. The polypoid type showed inverted U-type in obstructive or stenotic feature of bile duct radiologically and the infiltrating type showed U-type, V-type or constricted type. The distance of the tubular adenocarcinoma in histological cellular type tended to be longer than that of papillary or undifferentiated type. The severer the invasion of the carcinoma to the lymphatic vessels was, the longer the distance was.
