Screening the p53 status of human cell lines using a yeast functional assay.

We have screened the p53 status of 156 human cell lines, including 142 tumor cell lines from 27 different tumor types and 14 cell lines from normal tissues by using functional analysis of separated alleles in yeast. This assay enables us to score wild-type p53 expression on the basis of the ability of expressed p53 to transactivate the reporter gene HIS3 via the p53-responsive GAL1 promotor in Saccharomyces cerevisiae. Of 142 tumor cell lines, at least 104 lines (73.2%) were found to express the mutated p53 gene: 94 lines (66.2%) were mutated in both alleles, three lines (2.1%) were heterozygous, and no p53 cDNA was amplified from seven lines (4.9%). Of the 14 cell lines originating from normal tissues, all the transformed or immortalized cell lines expressed mutant p53 only. Yeast cells expressing mutant p53 derived from 94 cell lines were analyzed for temperature-sensitive growth. p53 cDNA from eight cell lines showed p53-dependent temperature-sensitive growth, growing at 30 degrees C but not at 37 degrees C. Four temperature-sensitive p53 mutations were isolated: CAT-->CGT at codon 214 (H214R), TAC-->TGC at codon 234 (Y234C), GTG-->ATG at codon 272 (V272M), and GAG-->AAG (E285K). Functionally wild-type p53 was detected in 38 tumor cell lines (26.8%) and all of the diploid fibroblasts at early and late population doubling levels. These results strongly support the previous findings that p53 inactivation is one of the most frequent genetic events that occurs during carcinogenesis and immortalization.

Advanced endometrial cancer detected at 7 months after childbirth.

It is very rare to find endometrial cancers arising within a short period following childbirth, presumably because pregnancy has a protective effect against endometrial cancer mediated by elevated secretion of progesterone. We present the case of a 30-year-old Japanese woman with stage IIIC endometrial cancer that was found 7 months after childbirth. The patient was treated with surgery followed by 6 cycles of intravenous combination chemotherapy and oral administration of medroxyprogesterone acetate. Histopathological examination of surgical specimen revealed poorly differentiated adenosquamous carcinoma of the endometrium with deep myometrial invasion, cervical stromal involvement, and pelvic and paraaortic lymph node metastases. The disease progressed rapidly and the patient died of the disease.

Clinical assessment of Hokkaido University PACS.

The paper describes the hardware and software used in the Hokkaido University picture archiving and communication system (HUPACS), which has been in use for the past two years. An evaluation of the system is also included. Fuji computed radiography completely replaces conventional X-ray radiographs, and 10:1 data compression is routinely employed. The PACS LAN is connected to the hospital information system through the main-frame computer and it is linked, not only to the radiology department, but also to the outpatient clinics. Image data have been steadily stored in the optical disc library, which contains more than 200,000 images. The original image can be accessed within 1 min. Image quality on a cathode ray tube (CRT) is clinically acceptable. Nevertheless, the transfer time of a newly made image is prolonged in some cases, during periods of heavy usage of the system. During scheduled radiology conferences, image reading tends to take more time on a CRT than on film. An improvement in the ease of operation of the workstations is necessary for full acceptance of CRT diagnoses. Generally speaking, however, HUPACS is working well and is appreciated by most of the clinicians.

Three cases of familial ulcerative colitis--in a mother and two of her sons.

A pedigree of familial ulcerative colitis with their HLA haplotypes is reported. The mother and two children, the eldest and second of three sons were affected. The mother developed proctitis at age 35, and the lesion extended to the entire colon at the time of a relapse. The two sons developed ulcerative colitis in a similar fashion; total colitis, of moderate severity, developing at age 16 in both. The analysis of HLA haplotypes of all five family members suggested that the HLA responsible for ulcerative colitis in this family was not HLA B, C, or DR, but Aw24 and DQw1. The third son had the same HLA haplotype as the second son, and it was presumed that he would develop ulcerative colitis in the future. These cases, together with other cases of familial ulcerative colitis indicate that Aw24 and DQw1 are critical phenotypes for ulcerative colitis in Japan.

Midazolam reverses histamine-induced bronchoconstriction in dogs.

PURPOSE: Midazolam has been used clinically as a sedative and as an anaesthetic induction agent. However, the bronchodilating effects of midazolam have not been comprehensively evaluated. We sought to determine relaxant effects of midazolam on the airway. METHODS: After our Animal Care Committee approved the study, eight mongrel dogs were anaesthetized with 30 mg.kg-1 pentobarbitone iv, and were paralysed with 200 micrograms.kg-1.hr-1 pancuronium. The trachea was intubated with an endotracheal tube (ID 7 mm) that had a second lumen for insertion of a superfine fibreoptic bronchoscope (OD 2.2 mm) to measure the bronchial cross-sectional area (BCA) continuously. The tip of the bronchoscope was placed at the level of the second or third bronchial bifurcation of the right bronchus. A videoprinter printed the BCA which was then measured with a NIH image program. Bronchoconstriction was produced with histamine (H) 10 micrograms.kg-1 followed by 500 micrograms.kg-1.hr-1. Thirty minutes later, 0 [saline], 0.01, 0.1 and 1.0 mg.kg-1 midazolam and 25 micrograms.kg-1 flumazenil were given. The BCA was assessed before (basal area) and 30 min after the start of H infusion, and was also measured five minutes after each midazolam and flumazenil iv. At the same time, arterial blood was sampled for plasma catecholamine measurement. RESULTS: Histamine infusion decreased BCA to 49.7 +/- 17.3% of basal BCA. More than 0.1 mg.kg-1 midazolam increased BCA up to 71.7 +/- 15.3% of the basal (1.0 mg.kg-1) (P < 0.01). Plasma adrenaline concentration was decreased from 6.9 +/- 3.8 to 3.7 +/- 1.9 ng.ml-1 by 1.0 mg.kg-1 midazolam (P < 0.05). Flumazenil did not antagonize the relaxant effect of midazolam but reversed the inhibitory effect of midazolam on histamine-induced adrenaline release. CONCLUSION: Midazolam has a spasmolytic effect on constricted airways but this bronchodilatation was not reversed by flumazenil.

K+/H+ antiporter in alkaliphilic Bacillus sp. no. 66 (JCM 9763).

A K+/H+ antiport system was detected for the first time in right-side-out membrane vesicles prepared from alkaliphilic Bacillus sp. no. 66 (JCM 9763). An outwardly directed K+ gradient (intravesicular K+ concentration, K(in), 100 mM; extravesicular K+ concentration, K(out), 0.25 mM) stimulated uphill H+ influx into right-side-out vesicles and created the inside-acidic pH gradient (delta pH). This H+ influx was pH-dependent and increased as the pH increased from 6.8 to 8.4. Addition of 100 microM quinine inhibited the H+ influx by 75%. This exchange process was electroneutral, and the H+ influx was not stimulated by the imposition of the membrane potential (interior negative). Addition of K+ at the point of maximum delta pH caused a rapid K+-dependent H+ efflux consistent with the inward exchange of external K+ for internal H+ by a K+/H+ antiporter. Rb+ and Cs+ could replace K+ but Na+ and Li+ could not. The H+ efflux rate was a hyperbolic function of K+ and increased with increasing extravesicular pH (pH(out)) from 7.5 to 8.5. These findings were consistent with the presence of K+/H+ antiport activity in these membrane vesicles.

Involvement of the claustrum in the convulsive evolution of temporal limbic seizure in feline amygdaloid kindling.

The effects of unilateral lesioning of the claustrum (CL) either prior to or following completion of amygdaloid (AM) kindling was examined in cats. CL lesioning ipsilateral to the kindling or the kindled AM caused the emergence of a mirror image pattern of expected or established AM onset convulsive seizure originating in the contralateral hemisphere. This finding suggests that: (1) the CL is involved in the convulsive evolution of AM onset non-convulsive limbic seizure; (2) an alternative transhemispheric route is available for the convulsive evolution of AM onset limbic seizure when the ipsilateral CL is disrupted; and (3) the CL is not critical for secondary convulsive generalization.

Immunohistochemical analysis of glutathione S-transferase mu expression in ovarian tumors.

OBJECTIVE: To explore the association between Glutathione S-transferase (GST) -mu expression and clinicopathologic features in ovarian tumors. STUDY DESIGN: Immunohistochemical study was made to investigate GST-mu expression in diverse ovarian tumors. RESULTS: All 75 ovarian tumors expressed GST-mu. There was no significant association between GST-mu immunopositivity and clinicopathological features. In serous cystadenocarcinoma, however, patients with weak GST-mu expression survived longer than those with moderate or strong GST-mu expression. Three of the 8 tumors that had expressed weak GST-mu initially increased GST-mu expression after chemotherapy. CONCLUSION: GST-mu expression is common in ovarian tumors and malignant tumors expressed more GST-mu than benign tumors. GST-mu might play a major role in the development of drug resistance in certain ovarian tumors and could be a useful marker of natural resistance and their future outcome.

Effect of callosal bisection on seizure development and interhemispheric transfer effects in feline motor cortical kindling.

Motor cortical (MC) kindling was carried out in 12 adult cats, seven with the corpus callosum (CC) intact and five with the CC bisected, to study interhemispheric transfer effects and the effect of callosal bisection on both seizure development and interhemispheric transfer effects. MC kindling developed from partial motor seizures to partial onset generalized convulsions. Interhemispheric negative transfer effect, but not positive, existed in secondary site kindling and primary site retest of the CC-intact group, as shown by: (i) electroencephalographically lateralized seizure development in the stimulated hemisphere; (ii) delayed generalization of partial onset generalized convulsions; and (iii) a markedly unstable generalized convulsive seizure state. The CC-bisected group showed: (i) significantly delayed seizure development from partial motor seizure stage to generalized convulsive seizure stage in primary and secondary kindling; (ii) facilitated intrahemispheric seizure development; (iii) the diminution of interhemispheric negative transfer effect; (iv) modified generalized convulsions showing extremely asymmetrical generalized convulsions shifting from contralaterally dominant convulsions to ipsilaterally dominant ones (n = 2) or alternate generalized convulsions changing from contralateral hemiconvulsions to ipsilateral ones (n = 3). The results obtained suggest that the CC plays a major role in interhemispheric seizure propagation as well as interhemispheric negative transfer effects in MC kindling and may have suppressive effect on intrahemispheric motor seizure development in MC kindling. However, interhemispheric seizure propagation and interhemispheric negative transfer effects were mediated via other structures, possibly subcortical structures, when the CC was bisected.

Effect of motor cortical kindling on subsequent ventral hippocampal kindling and the role of the corpus callosum in the cat.

The effect of bilateral motor cortical (MC) kindling on subsequent unilateral ventral hippocampal (VHIPP) kindling was studied in four cats with the corpus callosum (CC) intact and five cats with the CC bisected, compared with nine cats with unilateral VHIPP kindling. Subsequent VHIPP kindling in CC-intact cats resulted in the modified development of limbic seizures to ipsilateral, not contralateral, focal motor seizures in one of four cats, significantly greater seizure regressions from generalized convulsive seizure stage to earlier seizure stages and delayed onset of focal motor seizures and generalized convulsions in partial onset generalized convulsions. CC bisection reduced the degree of seizure regression from generalized convulsive seizure stage to earlier stages, facilitated the development of the last limbic seizure to the first generalized convulsive seizure, accentuated hemiconvulsions and asymmetrical generalized convulsions and delayed the onset of generalized convulsions in partial onset generalized convulsions. The modified seizure development was also induced in three of five CC-bisected cats. Results indicate that bilateral MC kindling induces inhibitory effects on subsequent unilateral VHIPP kindling and the modified ictal progress from the VHIPP to the contralateral hemispheric motor structures and also that CC bisection interferes with the bilateralization and synchronization of convulsions, but reduces the inhibition of previously established MC kindling against VHIPP kindling and facilitates the development of focal motor seizures to secondarily generalized convulsions.

Studies on the binding of [3H]amethocaine to rat cerebrocortical membranes.

We have examined the binding of the local anaesthetic agent [3H]amethocaine to rat cerebrocortical membranes. All studies were performed in Tris buffer 50 mmol litre-1 at pH 7.4. Bound and free radioligand were separated by rapid vacuum filtration. [3H]Amethocaine binding at room temperature was dose-dependent and saturable, with mean Kd and Bmax values of 153 (SEM 18) nmol litre-1 and 9.4 (1.6) pmol/mg protein, respectively. [3H]Amethocaine binding was displaced in a dose-dependent manner (pIC50) by unlabelled amethocaine (6.89), procaine (5.20), lignocaine (3.46) and prilocaine (2.81). Ropivacaine and bupivacaine did not produce 50% displacement at the highest concentrations used (10(-4) and 10(-3) mol litre-1, respectively). We examined the nature of the binding site further with a range of ion channel antagonists (nifedipine, verapamil, diltiazem, omega-conotoxin, tetrodotoxin, tetraethylammonium and 4-aminopyridine) and ion channel coupled receptor ligands (L-glutamate, MK801, GABA, glycine and nicotine). With the exception of tetraethylammonium (pIC50 3.07) and 4-aminopyridine (pIC50 3.68), all non-anaesthetic agents failed to displace [3H]amethocaine. Collectively our data suggest that it is unlikely that there is a single target site for all local anaesthetic agents.

The expression of vascular endothelial growth factor and transforming growth factor-beta associates with angiogenesis in epithelial ovarian cancer.

The object of this study was to clarify the association between microvessel density and clinicopathologic factors, as well as the association of angiogenic factors (vascular endothelial growth factor [VEGF] and transforming growth factor-beta [TGF-beta]) with tumor angiogenesis and patient survival in epithelial ovarian cancer. The expression of VEGF and TGF-beta was evaluated by immunohistochemical staining in 60 patients with epithelial ovarian cancer. The microvessel density, assessed by immunostaining for factor VIII-related antigen in the most neovascularized areas, varied depending on histological types but not on International Federation of Gynecology and Obstetrics stage. Patients with stage III carcinomas and positive TGF-beta had more extensive peritoneal dissemination and a worse outcome. The microvessel density of VEGF-rich and TGF-beta positive tumors was significantly higher than that of VEGF-poor and TGF-beta negative tumors. Angiogenesis appears to be an early event in epithelial ovarian cancer and may be induced differently in tumors of different histological types. The expression of VEGF and TGF-beta associates with the promotion of angiogenesis, and the expression of TGF-beta is a prognostic indicator in epithelial ovarian cancers.

Dantrolene sodium reverses the increase in cAMP response element and TPA responsive element DNA-binding activity in the rabbit brain following haloperidol administration and heat stress.

Using electrophoretic mobility-shift assay (EMSA), we examined DNA-binding activity of cAMP response element (CRE), onto its responsive element CRE, as well as TPA responsive element (TRE) in the medial hypothalamus and striatum of the experimental rabbits administered with haloperidol under heat stress exposure and studied the effects of dantrolene sodium to the transcriptional factor. In EMSA with nuclear extracts from the rabbit brain, the DNA-binding activities of CRE and TRE in medial hypothalamus and striatum increased following haloperidol and heat stress. These increases were maintained by coadministration with atropine. The treatment with dantrolene sodium markedly reversed such increases. The alterations of activities of these transcriptional factors may reflect the therapeutic effect of dantrolene sodium.

Malignant melanoma of the uterine cervix: a case report.

We present case of a 57-year-old Japanese woman a 9 cm-size primary malignant melanoma of the uterine cervix. MRI demonstrated a low-signal T1-weighted spin echo image and hyperperfusion indicated by Gd-DTPA enhancement. The sequence analysis of the MTS1/CDK4I gene revealed no deletion or mutation. The results of cytological, pathological, and electron microscopy tests are also presented.

Purification, characterization, and nucleotide sequence of an intracellular maltotriose-producing alpha-amylase from Streptococcus bovis 148.

An intracellular alpha-amylase from Streptococcus bovis 148 was purified and characterized. The enzyme was induced by maltose and soluble starch and produced about 80% maltotriose from soluble starch. Maltopentaose was hydrolyzed to maltotriose and maltose and maltohexaose was hydrolyzed mainly to maltotriose by the enzyme. Maltotetraose, maltotriose, and maltose were not hydrolyzed. This intracellular enzyme was considered to be a maltotriose-producing enzyme. The enzymatic characteristics and hydrolysis product from soluble starch were different from those of the extracellular raw-starch-hydrolyzing alpha-amylase of strain 148. The deduced amino acid sequence of the intracellular alpha-amylase was similar to the sequences of the mature forms of extracellular liquefying alpha-amylases from Bacillus strains, although the intracellular alpha-amylase did not contain a signal peptide. No homology between the intracellular and extracellular alpha-amylases of S. bovis 148 was observed.

Three of the seven bphC genes of Rhodococcus erythropolis TA421, isolated from a termite ecosystem, are located on an indigenous plasmid associated with biphenyl degradation.

Rhodococcus erythropolis TA421, a polychlorinated biphenyl and biphenyl degrader isolated from a termite ecosystem, has seven bphC genes expressing 2,3-dihydroxybiphenyl dioxygenase activity. R. erythropolis TA421 harbored a large and probably linear plasmid on which three (bphC2, bphC3, and bphC4) of the seven bphC genes were located. A non-biphenyl-degrading mutant, designated strain TA422, was obtained spontaneously from R. erythropolis TA421. TA422 lacked the plasmid, suggesting that the three bphC genes were involved in the degradation of biphenyl. Southern blot analyses showed that R. erythropolis TA421 and Rhodococcus globerulus P6 have a similar set of bphC genes and that the genes for biphenyl catabolism are located on plasmids of different sizes. These results indicated that the genes encoding the biphenyl catabolic pathway in Rhodococcus strains are borne on plasmids.

Depressed pituitary-adrenal response to surgical stress in chronic schizophrenic patients.

We evaluated whether pituitary-adrenal response to surgical stress is modified in chronic schizophrenic patients. Twenty-two schizophrenic patients on chronic antipsychotic therapy of phenothiazine derivatives over 10 years underwent orthopedic surgery of the extremities under general anesthesia by isoflurane and nitrous oxide. In chronic schizophrenic patients, responses of plasma epinephrine, norepinephrine, ACTH and cortisol levels during surgical stress were significantly lower than those of control patients. The attenuated response to surgical stress in these patients may be associated with their autonomic dysfunction and decreased pituitary-adrenal activity due to chronic administration of antipsychotic agents.

Isolation of extradiol dioxygenase genes that are phylogenetically distant from other meta-cleavage dioxygenase genes.

meta-Cleavage dioxygenase genes were isolated from the benzoate-assimilating bacteria Pseudomonas stutzeri A401 and Pseudomonas mendocina KC37. The gene products had the enzymatic activity of 2,3-dihydroxy-biphenyl-1,2-dioxygenase. Phylogenetic analysis based on the alignment of amino acid sequences suggests that these enzymes are distantly related to other meta-cleavage enzymes and may be members of a new family of extradiol dioxygenases.
