Facilitation of agonist motoneurons upon initiation of rapid and slow voluntary movements in man.

The time course of facilitation of the agonist motoneurons upon initiation of voluntary ankle dorsiflexion was investigated in eight healthy subjects. The H-reflex and visually guided tracking methods were used for testing the excitability of the motoneuron pool and for controlling the initiation of movement as well as speed and force. Since the onset of voluntary EMG activity (EMG reaction time: EMGvRT) was delayed and/or obscured by test H-reflexes which were evoked very close to the behavioral responses, the subject was instructed to make response movements bilaterally, and EMGvRT was measured on the side without stimulation. In every subject, the EMGvRT was invariably longer in the ramp movement than in the step movement. The onset of H-reflex facilitation prior to EMG onset, which was regarded as indicating the arrival time of the descending motor command to the motoneuron pool, always started earlier in the ramp movement than in the step movement. The difference in facilitation onset between the two tasks was smaller than that in EMGvRT. Since the amplitude of the H-reflex at the onset of the voluntary EMG was equivalent in both movements, the development of H-reflex towards the behavioral EMG onset was more gradual in the ramp movement than in the step movement. The present results demonstrate that the longer reaction time in the slow ramp task depends on 2 factors: delay in the arrival of descending facilitatory impulses to the agonist motoneuron pool, and its slow recruitment thereafter.

[Diffusely increased uptake in the skull in normal bone scans].

Diffusely increased skull uptake (a hot skull) is often seen in patients with bone metastases and metabolic diseases. This finding is also, however, noticed in normal bone scans of aged women. To determine whether the hot skull could be considered a normal variant in elderly women and is associated to menopause, we studied 282 normal bone scans (166 women and 116 men without metabolic and hormonal disease; age range 11 to 84 yr). We divided the patients into eight age groups--ages 10-19, 20-29, 30-39, 40-49, 50-59, 60-69, 70-79, and 80-89 yrs. Measurements of skull uptake were obtained from anterior total body views using contrast-to-noise ratio (CNR). CNR for the skull was calculated using the following equation: (counts skull - counts femur)/square root of counts skull+counts femur, where the denominator signifies a measure of the noise and the numerator represents the contrast between the skull and the midfemur. Counts skull and femur are the number of average counts per one pixel recorded over the skull and midfemur, respectively. The sex dependent difference in skull uptake began to develop in the age group 30-39 yrs (p less than 0.05). The skull showed greater activity in women than in men for age groups from 30-39 to 80-89 yrs. In the age groups 50-59 and 60-69, the difference was particularly large (p less than 0.001).(ABSTRACT TRUNCATED AT 250 WORDS)

[A case of motor neuron disease with dementia--cerebral blood flow and cerebral oxygen metabolism].

A 51-year-old man developed muscle weakness of the bilateral upper extremities, and mental changes beginning with personality change. There was no history of mental illness in his family. A neurological examination 1 year after the onset revealed muscle atrophy and fasciculation of his bilateral upper extremities Neuropsychological examination revealed concrete speech, paraphasia, and lack of judgment. Disorientation, amnesia, dyscalculia, and spatial agnosia, however, were not recognized. These neuropsychological findings were compatible with dementia of frontal lobe type. EMG and muscle biopsy revealed neurogenic muscular atrophy. There was no abnormal findings in the brain X-CT and the brain MRI. PET study using C15O2 and 15O2 revealed reduction of cerebral blood flow (CBF) and cerebral metabolic rate of oxygen (CMRO2) in the bilateral medial frontal cortex, the left temporal cortex and the bilateral thalamus. From these these findings the patient was diagnosed as having motor neuron disease with dementia. Muscle atrophy and dementia worsened gradually. A second PET study 2 years and 6 months after the onset revealed severe reduction of CBF and CMRO2 in the bilateral temporal cortex and the thalamus. These PET findings suggested that dysfunction of the temporal cortex and the thalamus related to dementia in this case.

Molecular cloning of cDNA for antimycin A-inducible mRNA and its role in cyanide-resistant respiration in Hansenula anomala.

A cDNA for mRNA induced by antimycin A in Hansenula anomala was cloned. The mRNA for the cDNA was expressed in the yeast under the conditions expressing the cyanide-resistant respiration activity. The nucleotide sequence revealed a long open reading frame of 342 codons encoding a protein with a molecular weight of 40,282 in the cDNA. An antibody recognizing the protein encoded by the open reading frame was produced. Immunoblotting of H. anomala proteins with this antibody showed that a 36 kDa protein localized in mitochondria was a mature form of the protein encoded by the cDNA. It is suggested that the cloned cDNA encodes a protein involved in the cyanide-resistant respiratory pathway.

Replacement of P1 Leu18 by Glu18 in the reactive site of turkey ovomucoid third domain converts it into a strong inhibitor of Glu-specific Streptomyces griseus proteinase (GluSGP).

Turkey ovomucoid third domain with P1 Leu18 at its reactive site is an excellent inhibitor of chymotrypsin and elastase and of many other serine proteinases with related specificities. Semisynthetic replacement of P1 Leu18 by Lys18 causes the expected change into a trypsin inhibitor. Strikingly, semisynthetic replacement P1 Leu18 to Glu18 changes turkey ovomucoid third domain into a powerful inhibitor of Glu-specific Streptomyces griseus proteinase, GluSGP. Of the 131 natural avian ovomucoid third domains we have sequenced none have P1 Glu18, but several avian ovomucoid first domains have P1 Glu24. They are weak to moderate inhibitors of GluSGP.

Antagonist inhibition during rest and precontraction.

The excitability of antagonist soleus motoneurons was tested during fast voluntary contractions of tibialis anterior (TA). Contractions of TA started either from zero or higher levels of tonic contraction of ankle extensors. Results showed that H reflex depression under 3 different conditions: 25% (R25) or 50% (R50) of the maximal isometric dorsiflexion from the resting state and the sequential isometric response (SW) appeared about 20-40 msec prior to the EMG onset of TA. In particular, H reflex depression was clear in the SW response, and the stronger the prior contraction of extensors the greater the H reflex depression. There was a significant difference in the integrated EMG between R25 and SW but none between R50 and SW. The different amounts of inhibition found for dorsiflexion from the resting state and sequential movement is most probably explained by presynaptic inhibition. If presynaptic inhibition were increased throughout the sequential movement, in comparison with the resting state, change in the H reflex gain would occur.

[2'-5' oligoadenylate synthetase activity in patients with acute viral hepatitis and chronic type B hepatitis, and its clinical significance].

The levels of 2'-5' oligoadenylate synthetase activity (2-5AS) were measured in the lymphocytes and sera of 32 patients with acute viral hepatitis and 20 patients with HBeAg positive chronic type B hepatitis, and the relationship between the effectiveness of interferon (IFN) therapy and 2-5AS reactivity was then studied. The lymphocyte 2-5AS (L.2-5AS) and the serum 2-5AS (S.2-5AS) levels in patients with any type of acute viral hepatitis at the acute stage were significantly higher than in the healthy subjects. The 2-5AS level in cases of acute type A viral hepatitis was significantly higher than in type B and type non A non B hepatitis. However, no significant difference in the 2-5AS levels between type B hepatitis and type non A non B hepatitis was observed. After recovery, the L.2-5AS and S.2-5AS levels decreased in patients with any type of acute viral hepatitis. No significant difference between the L.2-5AS level in healthy subjects and patients with chronic type B hepatitis was observed. The S.2-5AS level was significantly higher in patients with chronic type B hepatitis than in healthy subjects. During IFN administration, the L.2-5AS and S.2-5AS levels increased, with the maximum level and maximum rate of increase obtained on the 3rd day with respect to the L.2-5AS, and after one week with respect to the S.2-5AS. Of the 20 patients with chronic type B hepatitis with positive HBe antigen, 11 showed normalization of DNA-P, 4 disappearance of HBe antigen and 2 seroconversion of HBe antigen. In cases with a higher maximum rate of increase of the L.2-5AS, the effect of IFN therapy was observed to be greater.

Essential role of ferrous iron in cyanide-resistant respiration in Hansenula anomala.

Antimycin A-dependent induction of cyanide-resistant respiration in Hansenula anomala was completely blocked by o-phenanthroline, alpha,alpha'-dipyridyl, or 8-hydroxyquinoline. Pulse-labeling of the cells with [35S]methionine in the presence of both antimycin A and o-phenanthroline indicated that the 36-kDa protein previously reported to be involved in cyanide-resistant respiration [(1989) J. Biochem. 105, 864-866] was formed in mitochondria even under these conditions. The addition of Fe2+, but not Fe3+, ions to these cells in the presence of cycloheximide resulted in the rapid expression of cyanide-resistant respiration activity. These results suggest that in the presence of both antimycin A and o-phenanthroline an inactive form of the 36-kDa protein was formed and Fe2+ ions converted it to the active form. It is also likely that Fe2+ ions are involved in the reaction mechanism of cyanide-resistant respiration.

[Alleviation of cisplatin toxicity by high-dose bismuth subnitrate and pharmacokinetics of bismuth subnitrate and cisplatin].

Twelve patients with several malignant neoplasms, including lung and gastrointestinal carcinoma were treated with high-dose Cisplatin and high-dose Bismuth Subnitrate. Therapeutic efficacy and protective effect of high-dose Bismuth Subnitrate on toxicity of Cisplatin were evaluated in twenty-five courses of treatment. The Pharmacokinetics of Bismuth Subnitrate and Cisplatin were also studied in several courses. Bismuth Subnitrate was administered orally at a dose of 150 mg/kg/day for 10 days and a dose of 80-150 (Mean 108) mg/m2 of Cisplatin was administered intravenously on the day six of Bismuth Subnitrate administration. Toxicities of high-dose Cisplatin were minimal under administration of high-dose Bismuth Subnitrate and therapeutic efficacy was observed in several patients. The pharmacokinetics of Bismuth Subnitrate in plasma and urine demonstrated that 10 days administration of high-dose Bismuth Subnitrate was appropriate to maintain adequate concentration of Bismuth for preinduction of Metallothionein in organs. The pharmacokinetics of Cisplatin in plasma and urine demonstrated that deposition of total and ultrafilterable (free) platinum in blood were well described by a biphasic manner with a very rapid first phase and a very prolonged second phase, and that urine excretion of platinum was similar to the conventional manner. This study demonstrated that concurrent administration of high-dose Bismuth Subnitrate can permit the administration of high-dose Cisplatin with minimal toxicity and appropriate antitumor effect.

A 36-kDa mitochondrial protein is responsible for cyanide-resistant respiration in Hansenula anomala.

Antimycin A-dependent induction of cyanide-resistant respiration in Hansenula anomala was reversibly blocked by carbonylcyanide-m-chlorophenylhydrazone (CCCP). When the cells were pulse-labeled with [35S]methionine in the presence of both antimycin A and CCCP, the radioactivity was incorporated into a 39 kDa mitochondrial protein. Upon removal of CCCP, this protein was processed into a 36 kDa form. The increase in the 36 kDa protein completely paralleled that in cyanide-resistant respiration activity, suggesting that the 39 kDa protein is the precursor of the 36 kDa protein, which is responsible for cyanide-resistant respiration.

The differences in human spinal motoneuron excitability during the foreperiod of a motor task.

Changes in excitability of the spinal motoneuron pool during the foreperiod, which was fixed at 0.8 s, in simple and choice reaction time experiments using ankle dorsiflexion and plantar flexion were studied in fourteen healthy normal subjects by combining the visually guided tracking and H-reflex testing methods. Almost all cases showed a significant facilitation in the soleus H-reflex within the time interval between 100 and 300 ms after a warning signal (Phase I), irrespective of movement direction and task modality. The pretibial H-reflex was also facilitated. On the other hand, variable effects were noted in the later half of the foreperiod, particularly within the 200 ms prior to the response signal (Phase II). Using a simple reaction task with dorsiflexion, six cases showed no changes in the soleus H-reflex, while four others showed statistically significant inhibitory changes and the remaining four showed facilitation. The inhibition and facilitation were often accompanied with very weak and unintended EMG activities in the pretibial and So1 muscles respectively. A similar finding was obtained in the simple plantar flexion task and the choice reaction task with dorsiflexion or plantar flexion. We suggest that the facilitation at Phase I represents a perceptual orienting response to a warning signal and the effects seen in Phase II represent the difference in the waiting attitude of each subject anticipating initiation of the coming task, or the preparatory "set" which primed the spinal motor structure in a biased position.

Characterization of cyanide-resistant respiration and appearance of a 36 kDa protein in mitochondria isolated from antimycin A-treated Hansenula anomala.

Mitochondria exhibiting cyanide-resistant respiration were isolated from Hansenula anomala which had been incubated in the presence of antimycin A to induce cyanide-resistant respiration. The cyanide-resistant respiration in isolated mitochondria was not inhibited by antimycin A or myxothiazol, suggesting that the branching of the pathway from the normal cyanide-sensitive pathway takes place at the coenzyme Q level. Analysis of mitochondrial proteins by sodium dodecyl sulfate gel electrophoresis indicated that a 36 kDa protein was induced by antimycin A treatment of the yeast. It is suggested that this protein is a component of the cyanide-resistant respiratory pathway.

Selection experiment for increased egg production under 23 H and 24 H light-dark cycles in the domestic fowl.

A selection experiment was conducted for increased rate of lay under 23-h (23 HS line) and 24-h (24 HS line) light-dark cycles over 5 generations. In generation 5, rate of lay was higher in the 23 HS line than in the 24 HS line, and the proportion of hens with mean intra-sequence intervals of less than 24 h in the 23 HS line increased to 88% in the generation 5 from 15% in the base generation. The realised heritability for rate of lay was 0.25 plus/minus 0.04 in the 23 HS line and 0.15 plus/minus 0.05 in the 24 HS line. Egg weight and shell weight decreased in both lines, and the decrease in the proportion of shell was larger in the 23 HS line than in the 24 line. It is suggested that a regimen of 23 h light and dark may be an effective environment for selection to improve the laying performance of a population approaching a plateau for egg production.

Possible role of a 36 kDa protein induced by respiratory inhibitors in cyanide-resistant respiration in Hansenula anomala.

The antimycin A or cyanide-dependent appearance of a 36 kDa protein in the particulate fraction was observed in L-[35S]methionine pulse-labeling experiments on cells of Hansenula anomala, in which cyanide-resistant respiration was induced. The combined addition of cycloheximide or anaerobiosis, which block the induction of cyanide-resistant respiration, repressed the synthesis of this protein. These results suggest the involvement of the particulate 36 kDa protein in cyanide-resistant respiration.

Serum amphotericin B concentration in a very premature infant with disseminated candidiasis.

A 1,040-g premature baby was diagnosed to have disseminated candidiasis and treated with amphotericin B (AMB) and 5-fluorocytosine. During the treatment, an unexpectedly large dose of AMB was infused unintentionally. AMB level was as high as 1.73 micrograms/mL soon after 5 mg/kg infusion instead of 0.5 mg/kg. However, it dropped rapidly to 0.83 micrograms/mL after 24 hours. AMB was detected in patient's serum at a higher level than minimal inhibitory concentration as long as one month after treatment was stopped. The patient showed liver dysfunction but no nephrotoxicity. The further studies are needed to establish safe and effective treatment regimen in premature infants with disseminated candidiasis.

[Delayed lung scintigraphy with N-isopropyl-I-123-p-iodoamphetamine in lung cancer and inflammatory disease].

Lung studies with N-Isopropyl-I-123-p-Iodoamphetamine (IMP) were performed on patients with lung cancer or inflammatory disease. In the present study, we evaluated the usefulness of the delayed scintigraphy. The subjects consisted of 27 patients with lung cancer (34 lesions), 3 with radiation pneumonitis, 2 with interstitial pneumonitis, 2 with old tuberculous lesion (tuberculomas), 1 with diffuse panbronchiolitis, 1 with pneumonia and 1 with lung abscess. The delayed scintigraphy was performed 24 hr after intravenous injection of 3 mCi IMP, in sitting position. In 10 patients, SPECT images were obtained following the delayed scintigraphy. Delayed scintigraphic appearances of lung cancer were classified into 5 types, high IMP uptake in the area congruent with the lesion of atelectasis and/or obstructive pneumonia (Type I), high IMP uptake in the area surrounded the tumor (Type II), a defect in the area consistent with the tumor and no high IMP uptake in the area surrounded the tumor (Type III), high IMP uptake in the area almost congruent with the tumor (Type IV) and no significant change (Type V). Excluding 10 lesions with Type IV or V, no IMP uptake was seen in the areas congruent with the tumors. Type II was the most frequently observed pattern. Normal scintigrams (Type V) were observed in 8 lesions, whose sizes were fairly small. There was no definite trend caused by difference in histological types of cancers. In 8 patients with viable inflammatory disease of the lung, the delayed scintigrams showed high IMP uptake in the areas congruent with the abnormalities on chest roentgenograms.(ABSTRACT TRUNCATED AT 250 WORDS)

The timing and the amount of agonist facilitation and antagonist inhibition of varying ankle dorsiflexion force in man.

Experiments were conducted to investigate whether or not reciprocal descending commands occur simultaneously in the brain as well as the phasic changes in the agonist facilitation and the antagonist inhibition involved in the initiation of varying forces of ankle dorsiflexion. The H-reflex and reaction time method were combined. Results showed that: (1) the agonist and the antagonist motoneurones via Ia inhibitory interneurones simultaneously receive a subliminal facilitation and inhibition from the brain before the EMG onset; and (2) the onset of the agonist facilitation and the antagonist inhibition were not modified by different contraction forces; however, the extent of facilitation and inhibition just after the EMG onset were closely related to the amount of contraction forces. These findings suggest that the descending motor command activates parallel agonist alpha-motoneurones and Ia inhibitory interneurones. However, direct action of the descending command prior to the EMG onset may not be strong enough to drive the Ia interneurone for suppressing the antagonist motoneurones, and the agonist facilitation and the antagonist inhibition just after the EMG onset increase as the voluntary effort is strengthened.
