Nutrient starvation induces cross protection against heat, osmotic, or H2O2 challenge in Vibrio parahaemolyticus.

Glucose-starved cells of Vibrio parahaemolyticus were compared with non-starved counterparts with respects to heat, osmotic, and oxidative challenges. The starved cells demonstrated greater thermal and oxidative resistance than did the non-starved cells. The starved cells also showed greater resistance against low osmotic challenge than did the non-starved cells although both cells showed a comparable resistance against high osmotic challenge.

Molecular and immunological characterization of a 64-kDa protein of Actinobacillus actinomycetemcomitans.

The 64-kDa protein to which about half the sera from patients with localized juvenile periodontitis and rapidly progressive periodontitis reacted strongly was purified from Actinobacillus actinomycetemcomitans Y4. Determination of the N-terminal sequence of the protein revealed that it was a GroEL-like protein. The DNA fragment containing the groEL gene of A. actinomycetemcomitans was amplified by polymerase chain reaction, and the groESL operon was cloned by using colony hybridization with the amplified fragment from A. actinomycetemcomitans chromosomal DNA. Sequence analysis revealed that structures of the operon and its products were typical in gram-negative bacteria. Rabbit polyclonal antibodies to the 64-kDa protein cross-reacted with approximately 65-kDa proteins of Haemophilus aphrophilus, Haemophilus influenzae, Haemophilus paraphrophilus, Escherichia coli and Eikenella corrodens but not with any cellular proteins of Porphyromonas gingivalis, Prevotella intermedia and Fusobacterium nucleatum. It is possible that antibodies reactive to the 64-kDa protein in periodontitis patients are induced by the cross-reactivity with the hsp60 proteins of other bacteria.

Role of the capsular polysaccharide-like serotype-specific antigen in resistance of Actinobacillus actinomycetemcomitans to phagocytosis by human polymorphonuclear leukocytes.

Serotype b-specific polysaccharide antigen (SPA) of Actinobacillus actinomycetemcomitans Y4 consists of D-fucose and L-rhamnose. To clarify the role of SPA in phagocytosis of the organism by human polymorphonuclear leukocytes (PMNs), monoclonal antibodies (MAbs) against SPA and SPA-defective mutants, which were constructed by inserting the transposon Tn916 into strain Y4, were used in a chemiluminescence (CL) assay and a phagocytic killing assay. The CL responses of human PMNs to strain Y4 were very low, and the organism was not killed by PMNs. In contrast, SPA-defective mutants induced strong CL responses. The addition of immunoglobulin G MAbs against Y4 SPA enhanced significantly both the CL responses to strain Y4 and the killing of the organism in the presence of complement. The CL responses to SPA-defective mutants were little affected by the addition of these MAbs. We conclude that SPA of A. actinomycetemcomitans plays an important role in the resistance to host defenses by PMNs.

Macrophage colony-stimulating factor and interleukin-1 alpha maintain the survival of osteoclast-like cells.

When mouse bone marrow cells were co-cultured with a stromal cell line. ST2, numerous osteoclast-like multinucleated cells (MNCs) were formed. To enrich the MNCs which were tartrate-resistant acid phosphatase (TRAP)-positive, we treated the cultures with dispase. Enriched TRAP-positive MNCs adhering to the bottom of the dish were cultured for a further 24h with medium containing 15% fetal calf serum. More than 80% of TRAP-positive MNCs were detached from the dish during the culture. However, when ST2 cells were included during the culture period, survival of TRAP-positive MNCs were significantly increased. Moreover, among various factors examined for the survival of TRAP-positive MNCs, only M-CSF and IL-1 alpha were effective in prolonging viability of TRAP-positive MNCs.

Novel antiasthmatic agents with dual activities of thromboxane A2 synthetase inhibition and bronchodilation. V. Thienopyridazinone derivatives.

Synthesis and pharmacological evaluation of novel thienopyridazinones and related compounds are described. A thiophene ring was found to be able to replace the benzene ring of a phthalazinone without loss of biological activities. This observation supports our hypothesis that the benzene ring plays an important role in both thromboxane A2 (TXA2) synthetase-inhibitory and bronchodilatory activities. Further, it was shown that the carbonyl moiety of a phthalazinone is not necessary for these activities.

Novel antiasthmatic agents with dual activities of thromboxane A2 synthetase inhibition and bronchodilation. VI. Indazole derivatives.

Synthesis and pharmacological evaluation of novel indazole derivatives are described. These compounds were found to exhibit both thromboxane A2 (TXA2) synthetase-inhibitory and bronchodilatory activities. This observation supports the idea that the partial structure of the 3-pyridyl and phenyl groups with a methylene insertion is an important component for well-balanced activities.

Immunochemical crossreactivity between globulins from buckwheat and indigo seeds.

Immunochemical relationships between salt-soluble proteins (albumins plus globulins) from buckwheat and indigo seeds were shown by immunoblot and immunodiffusion analyses using rabbit antisera raised against buckwheat globulins. These antigenic crossreactivities were roughly consistent with their polypeptide components judged by two-dimensional electrophoresis and SDS-PAGE analyses.

Experimental myocardial infarction with cartilaginous and osseous metaplasia in SHR and WKY rats.

Myocardial infarction was induced by ligation of the coronary artery in fifteen 32-week-old male spontaneously hypertensive rats (SHR) and fifteen 32-week-old male normotensive Wistar-Kyoto rats (WKY). Age-matched male SHR (n = 6) and WKY (n = 7) without the operation were used as non-infarcted control rats. Four weeks after the operation, cartilaginous metaplasia and osseous metaplasia were observed histopathologically in all SHR and 13 of the 15 WKY with myocardial infarction, but not in the control rats. The mean rates of infarction to the heart and the mean areas of metaplasia per rat showing metaplasia were greater in SHR than in WKY. The metaplastic areas were well correlated with the infarct percentage in SHR. It is therefore considered that extensive myocardial infarction is associated with the pathogenesis of cartilaginous and osseous metaplasia.

[Morphological study on the gastric mucosa in diabetes mellitus rats induced by streptozotocin].

To clarify the pathogenesis of vulnerability to acute gastric mucosal lesion (AGML) in diabetes mellitus (DM), we investigated histopathologically the gastric mucosa of the fundic gland in streptozotocin-induced DM rats. The length of the mucosa and the thickness of the surface epithelial cell (SEC) layer were measured, and the number of proliferating cell nuclear antigen (PCNA) positive cells was counted. As results, compared with control rats, the ratio of the length of SEC layer to that of mucosa, and the number of PCNA positive cells were significantly decreased in DM rats, whereas the length of the mucosa tended to extend in DM rats. Consequently, it is considered that attenuation of mucosal barrier in the SEC layer can be closely associated with susceptibility to occurrence of AGML in DM.

[Age-related memory impairment and hippocampal damage in ddY male mice].

The relationship between behavioral change and hippocampal lesion was studied in ddY male mice, at 4.5, 20, 40 and 60 weeks of age. In passive avoidance response, the aging mice (40 and 60 weeks of age) showed shorter latency than young mice (4.5 weeks of age). The degenerated pyramidal cells were more numerous after 40 weeks of age in the CA3 than CA1. The mean incidence of the degenerated pyramidal cells in the CA3 was 20.8% at 60 weeks of age. In 71.9% of mice, impairment of passive avoidance response was associated well with hippocampal lesions. These results indicate that the hippocampus plays some role in the memory in mice. The ddY mice may be possible to use as models for reversible hippocampal lesion.

A novel adult T cell leukemia-derived cell line (SALT-3) susceptible to human immunodeficiency virus type 1 infection.

A novel human T cell line (SALT-3) was established from the pleural effusion of a patient with adult T cell leukemia (ATL) of lymphoma type. SALT-3 showed atypical T cell markers such as CD1-CD2-CD3-CD4+CD5+CD7+CD8-CD19-CD20-CD25+HLA-DR+. T cell receptor alpha/beta and gamma/delta were undetectable. Human T cell lymphotropic virus type 1 (HTLV-I) particles were seen on SALT-3 cells by electron microscopic analysis. HTLV-I gag p19, proviral DNA and mRNA of HTLV-I genes were also detected in the cells. Chromosome analysis showed abnormal karyotypes as 47, XY, partial trisomy of No.3 chromosome, and trisomy of No. 7 chromosome. Furthermore, SALT-3 were susceptible to the infection of human immunodeficiency virus type 1 (HIV-1) and the cells were rapidly killed after HIV-1 infection. This newly established HTLV-I-infected human T cell line would be a useful tool to study biological activities of atypical type of ATL cells and to examine the cytotoxic effects of HIV-1 and it's modulators.

Semiquantitative analysis of periodontopathogens by gene amplification.

Actinobacillus actinomycetemcomitans and Porphyromonas gingivalis, which are known as major causative organisms of periodontitis, were semiquantitatively identified by two-step polymerase chain reaction (PCR). The sets of specific primers for A. actinomycetemcomitans and P. gingivalis were prepared on the basis of the nucleotide sequences of the lktA and the fimA genes, respectively. The set of universal primers for eubacteria was designed from the nucleotide sequence of a highly conserved region in the eubacterial 16S rRNA sequence. The number of bacteria detectable by one-step PCR assay was no fewer than 10(3) cells. Less than 10(3) bacterial cells were detectable by two-step PCR assay. Subgingival plaque samples from 37 sites of 16 patients were obtained with paperpoints and analyzed by two-step PCR assay. More than 2 x 10(6) bacterial cells were found in the subgingival plaque samples from all diseased sites. In contrast, the number of total bacteria in those from more than half of healthy sites estimated by PCR assay was less than 2 x 10(6) cells, suggesting that subgingival plaque in diseased sites consists of a relatively larger number of bacteria compared with the population in healthy sites. While A. actinomycetemcomitans was detected in both healthy and diseased sites, P. gingivalis was observed only in diseased sites. Both periodontopathic bacteria occupied a minor part (less than 0.1%) of the total subgingival plaque bacteria.
