Morphological, chemical, and biological characterization of Japanese encephalitis virus virion and its hemagglutinin.

Three morphologically distinct structures, inner core, envelope, and surface projections, were observed in purified Japanese encephalitis virus virions by electron microscopy. The average diameter of each structure was 29.8 +/- 2.5, 44.8 +/- 3.2, and 53.1 +/- 4.5 nm, respectively. Double staining with uranyl acetate and phosphotungstic acid preserved these structures well. Treatment of virions with proteolytic enzymes resulted in the loss of hemagglutinating activity, surface projections, and the major polypeptide band in polyacrylamide gel electrophoresis, which corresponds to glycoprotein, one of the three virion polypeptides. Surface projections were purified by cesium chloride density gradient centrifugation after treatment of virions with Nonidet P-40. The purified materials had a density of 1.256 g/cm(3) and were composed of only glycoprotein, as revealed by polyacrylamide gel electrophoresis. Purified surface projections carried hemagglutinating activity, as well as neutralizing antibody-blocking activity, and induced neutralizing antibody in mice.

Efficacy of an antibiotic coated indwelling catheter: a preliminary report.

This study was conducted to examine if an aminoglycoside (dibekacin sulfate, DKB) incorporated into a coating layer on outer and inner surface of indwelling catheters would be released into urine both in vitro and in clinical cases on a sustained basis and if the released DKB would have any efficacy on delaying lower urinary tract infections. Released amounts of DKB from the catheter silicone rubber (SR) catheter were periodically measured both in vitro and in clinical applications. During the clinical applications, the catheters were indwelled in 14 patients. Organism counts in the patients' urine were determined and organisms were isolated from bacteriuria (defined as greater than or equal to 10(4) CFU/mL) and MICs to DKB were measured. Observations on several combined symptoms frequently associated with indwelling catheterization were performed. Results showed that sustained release of DKB continued for more than 25 days and 13 days in vitro and in clinical cases, respectively. Clinical studies suggested that 8 days of sterile urine after catheterization might be expected in patients without systemic administration of antibiotics and more than 2 weeks if combined with it. No particular problems in its use and associated symptoms were recognized.

Gene diversity of chimpanzee ABO blood group genes elucidated from intron 6 sequences.

The human and nonhuman primate ABO blood group gene shows relatively large numbers of nucleotide differences around the exon 7 region. In this study we determined intron 6 sequences for 9 alleles of common chimpanzee and for 3 alleles of bonobo to estimate nucleotide diversities among them. Sequence length polymorphisms are observed in this region as a repeat appears one to five times. From a phylogenetic network of intron 6 sequences of ABO blood group genes for humans, common chimpanzee, and bonobo, parallel substitutions and/or some kinds of convergent events are predicted in the chimpanzee lineage. We also estimated nucleotide diversities for common chimpanzee and bonobo ABO blood group genes; these values were 0.219% and 0.208%, respectively.
