Inability of a strain of Seoul virus to transmit itself vertically in rats.

SR-11 strain of Seoul virus was inoculated into new-born female rats. These rats at the age of 13 weeks were mated with uninfected male rats. The virus was transmitted to the male rats, but the offsprings were not infected from their mather rat owing to the maternal antibody. No intrauterine transmission of the virus was observed. We assumed that SR-11 type strain of Seoul virus could not be disseminated among rats in nature.

Minute carcinoma of the pancreas measuring 1 cm or less in diameter--collective review of Japanese case reports.

BACKGROUND/AIMS: According to Tsuchiya's collective review on small pancreatic cancer measuring 2 cm or less in diameter (5), more than half of them had obstructive jaundice and the 5-year survival rate was as low as 30%. Thus, a more aggressive diagnostic approach is needed to detect a smaller and more curable cancer of the pancreas. METHODOLOGY: Thus, we collected 36 reported cases of "minute" pancreatic cancer measuring 1 cm or less in diameter, from Japanese medical literature, to analyze the relationships between the diagnostic processes and long-term results. RESULTS: Excluding 3 patients with obstructive jaundice, the other 33 patients did not show any specific initial symptoms. However, 28 (78%) out of 36 patients showed an elevation in serum pancreatic enzyme levels and/or glucose intolerance. Among the 35 patients who had received ultrasonography (US) and/or computed tomography (CT), 20 (57%) patients showed duct dilation alone, whereas only 9 patients (26%) showed tumor mass. Among 35 patients who received an endoscopic retrograde pancreatography (ERP), all patients showed positive findings such as obstruction/stenosis, filling defect or duct dilation. All 36 patients underwent pancreatectomy and the 5-year survival rate was 57%. However, the 5-year survival rate was 34% in the 13 patients with jaundice and/or tumor mass depicted in US/CT, while it was 69% for the 22 patients without these two findings (p < 0.05). CONCLUSIONS: These data lead us to conclude that an elevation of serum pancreatic enzyme levels, glucose intolerance, and duct dilation alone depicted by US/CT should not be overlooked. ERP should be more widely applied to such patients, instead of persisting in delineating the tumor mass by US/CT or follow-up by tumor marker.

Combination therapy of percutaneous mitoxantrone injection, percutaneous ethanol injection, and transcatheter arterial embolization for intrahepatic hepatocellular carcinoma and adrenal metastasis.

We treated a 63-year-old man who had recurrent large hepatocellular carcinomas (> 5 cm in diameter) and left adrenal metastasis with the combination approach of percutaneous intratumoral chemotherapy with mitoxantrone, percutaneous ethanol injection, and transcatheter arterial embolization. He received repeated transcatheter arterial embolization and percutaneous ethanol injection combination therapy for intrahepatic hepatocellular carcinomas, which controlled his disease for 6 months from the first treatment. After that, left adrenal metastasis was detected by biopsy specimen. Therefore, we repeated more transcatheter arterial embolization and percutaneous ethanol injection to the liver and left adrenal gland, but this combination therapy could not control the hepatocellular carcinomas in these organs. With the patient's consent, he was treated with the combination approach of percutaneous intratumoral chemotherapy with mitoxantrone, percutaneous ethanol injection, and transcatheter arterial embolization for hepatocellular carcinomas of the liver and left adrenal gland. After this combination therapy, we followed-up the viable lesions by color Doppler ultrasonography and computed tomography examination. However, we could not detect these viable lesions of hepatocellular carcinomas in his body until one month before he died. When the degree of hepatic failure worsened due to the natural course of cirrhosis, this combination therapy was stopped 7 months before he died. He died of pulmonary tumor emboli from metastasis of inferior vena cava 24 months after the combination therapy started. However, on autopsy there was almost no remaining hepatocellular carcinoma found in the main lesions of liver and left adrenal gland. We suggest that a combination approach of percutaneous intratumoral chemotherapy with mitoxantrone, percutaneous ethanol injection, and transcatheter arterial embolization may be indicated in elderly cases of intrahepatic large hepatocellular carcinoma and adrenal metastasis, which are not under control only by transcatheter arterial embolization and percutaneous ethanol injection.

Obstruction due to persimmon bezoars: computed tomography detection.

BACKGROUND/AIMS: Small-bowel obstructions caused by persimmon bezoars are uncommon, and the diagnosis is difficult before operation. The aim of this study was to present computed tomography findings of persimmon bezoars in the small bowel. METHODOLOGY: We reviewed 3 cases diagnosed with small-bowel obstruction caused by persimmon bezoars between April 1997 and March 1999 at Chikamori Hospital. RESULTS: The cause of the small-bowel obstruction was bezoars in the 3 patients. Abdominal computed tomography performed before operation revealed a well-defined, intraluminal inhomogeneous mottled mass containing gas bubbles. CONCLUSIONS: Computed tomography is useful for diagnosing obstructed small-bowel persimmon bezoars.

Evaluation of a selective prostaglandin E receptor EP1 antagonist for potential properties in colon carcinogenesis.

BACKGROUND: Cyclooxygenases (COXs) and prostanoids play pivotal roles in colon carcinogenesis. This study was designed to determine the chemopreventive effects of ONO-8711, a selective prostaglandin E receptor EP1 antagonist, on the development of azoxymethane (AOM)-induced colonic aberrant crypt foci (ACF) in male F344 rats and to compare its potential with that of nimesulide, a well-documented selective COX-2 inhibitor. MATERIALS AND METHODS: Five-week-old male F344 rats received s.c. injections of AOM (15 mg/kg body weight) or the saline vehicle once weekly for two weeks and were fed the control diet (AIN-76A) or the experimental diets containing 400 or 800 ppm of ONO-8711 or 400 ppm nimesulide for 5 weeks. RESULTS: Administration of ONO-8711 at 800 ppm significantly reduced the total number of ACF/colon and 5-bromodeoxyuridine (BrdUrd) labeling index as compared to the control diet group (by 31% and 66%, respectively). As expected, dietary administration of nimesulide also suppressed the development of ACF and BrdUrd labeling index in the colon, by about 39% and 54%, respectively. CONCLUSION: Our finding that ONO-8711 significantly suppresses colonic ACF formation and cell proliferation strengthens the hypothesis that the selective prostaglandin E receptor EP1 antagonists possesses chemopreventive activity against colon cancer development.

Effect of lignins on HIV-induced cytopathogenicity and myeloperoxidase activity in human myelogenous leukemic cell lines.

We have previously reported the potent stimulation effect of lignin on the iodination of myeloperoxidase (MPO)-positive cells. We investigated here the anti-HIV (human immunodeficiency virus) activity of lignins in the MPO-positive (HL-60) and -negative (U-937) human myelogenous leukemic cell lines. Natural lignified material and dehydrogenation polymers, but not their precursors, effectively inhibited the cytopathic effect of HIV infection in both these cells as well as in MT-4 and MOLT-4 cells. HIV infection caused significant reduction of MPO activity in HL-60 cells, regardless of the presence or absence of lignins. These data suggest that MPO might not be involved in the anti-HIV activity induction by lignins.

Ultrasonographic evaluation of pancreatic cancers: size, contour, echo texture, and ductal configuration.

To improve the detection rate of small pancreatic tumors, it is important to know the characteristic differences in echographic patterns that distinguish small tumors from larger ones. We have studied the echographic patterns of histologically confirmed pancreatic cancers, with special reference to tumor size. The tumor size in 243 cases of adenocarcinoma determined by measurement of the pathological specimen correlated with that determined by ultrasonography. The ultrasonographic pancreatic configurations, tumor echo patterns, and pancreatic duct size and configurations were reviewed in accordance with the size of the tumor. Many pancreatic cancers showed localized swelling of the pancreas, however, more than half of all T1 cases had normal pancreatic contour, suggesting that we cannot rely on abnormal findings in configuration for the early diagnosis of pancreatic cancer. The very low echo pattern of tumor was seen only in T1 tumors. In cases in which the tumors were not detected by ultrasonography, the detection of a dilated pancreatic duct and common bile duct were very important clues to diagnosing a small tumor in the pancreas.

A hybrid artificial vascular graft based upon an organ reconstruction model. Significance and design criteria of an artificial basement membrane.

The design of an artificial basement membrane is one of the central concerns in a hybrid artificial vascular graft. The design criteria defined here include 1) structural matrix, 2) enhanced adhesion and growth of endothelial cells, and 3) ensured antithrombogenicity. The designed artificial basement membrane is a complex gel of type I collagen with dermatan sulfate that drastically alters the cellular responses for both endothelial cells and platelets. The incorporation of a very small amount of dermatan sulfate was sufficient to enhance adhesion and growth of endothelial cells and to suppress platelet adhesion and activation.

Induction of endometrial adenocarcinomas in persistent estrous Donryu rats by a single intra-uterine administration of N-ethyl-N'-nitro-N-nitrosoguanidine.

Induction of endometrial adenocarcinomas in persistent estrous Donryu rats by a single intra-uterine administration of N-ethyl-N'-nitro-N-nitrosoguanidine (ENNG) was examined. Female Donryu rats were divided into four groups at 7 weeks of age. Rats in Croup 1 were controls under a 12-h light/12-h dark regimen while Groups 2-4 were persistent estrous animals maintained under 24-h light conditions. At 10 weeks of age, rats in Group 1 and 2 were given a single intra-uterine administration of polyethylene glycol(PEG), and those in Groups 3 and 4 received ENNG(20 mg/kg body weight), dissolved in PEG, in the same manner. Group 4 rats also received an s.c. 17 beta-estradiol(E2) implant one week thereafter. At the termination of the experiment (week 42 after the ENNG-treatment), all surviving rats were killed and the development of uterine proliferative lesions assessed. All groups demonstrated hyperplasias, the incidence and severity being higher in ENNG-treated animals. The incidence of uterine adenocarcinomas in Group 2(2/20, 10%) was slightly higher than that in Group 1(0/16, 0%), although this difference was not significant. In Groups 3(7/19, 37%) and 4(6/20, 30%) statistically significant induction of adenocarcinomas was noted, two lesions in the latter case being very large with prominent invasion of tumor cells into the perimetrium. At 10 and 25 weeks after ENNG-treatment, the E2-progesterone(E2:P) ratios in Groups 2, 3 and 4 were significantly higher than in Group 1.(ABSTRACT TRUNCATED AT 250 WORDS)

Effect of age on the induction of endometrial lesions by a single intra-uterine administration of N-ethyl-N'-nitro-N-nitrosoguanidine in F344 rats.

A total of 54 10-week-old or 49 10-month-old female F344 rats were given a single intrauterine administration of 20 mg/kg N-ethyl-N'-nitro-N-nitrosoguanidine (ENNG) dissolved in polyethylene glycol (PEG) via the vagina using a non-laparotomy approach. Control animals (32 10-week-old or 29 10-month-old rats) were given PEG alone in the same manner. The animals were killed at 15 and 18 months of age, respectively. One animal in the young treated group presented an endometrial adenocarcinoma (incidence: 2.1%), although the incidence and degree of endometrial hyperplasias were significantly increased in both groups of treated animals, as compared to corresponding controls. Histologically, many of these lesions were polypous glandular hyperplasias (adenomatous polyps) with stromal proliferation. The incidence and grade of proliferative lesions in the young treated group were slightly higher than those in the old treated group. ENNG administration did not appear to affect the hormonal situation in treated animals. These results indicate that young animals may be more sensitive to ENNG than aged ones for induction of endometrial lesions, although ENNG-treatment alone is not sufficient for early and high induction of carcinomas even in young rats of this strain.

A case of hepatocellular carcinoma effectively treated with epirubicin aqueous vesicles in monodispersed iodized poppy-seed oil microdroplets.

A 64-year-old woman with a solitary mass (18 mm in diameter) of hepatocellular carcinoma underwent hepatic arterial infusion therapy with water-in-oil-in-water emulsion (W/O/W) prepared by the membrane emulsification technique using a controlled pore glass membrane. In the W/O/W, numerous microdroplets of iodized poppy-seed oil (IPSO), which contained many vesicles of aqueous solution of epirubicin, were suspended in the external aqueous phase. The mean diameter of the IPSO microdroplets was 30 microns. Seventeen milliliters of the W/O/W containing 40 mg of epirubicin and 5 ml of IPSO was infused into the proper hepatic artery. No severe side effects were encountered. Computed tomography taken 3 days after the treatment revealed a clearly defined deposition of IPSO within the tumor. Serum alpha-fetoprotein levels, 5.35x10(3) ng/ml, before treatment, decreased to 131 ng/ml on 38 days after treatment. The tumor was extirpated and histopathological examination revealed complete necrosis of the tumor.
