The modes of genotoxicity of a novel macromolecular antitumor antibiotic (SN-07) were examined using both prokaryotic and eukaryotic cells in vitro. The antibiotic induced a frameshift-type reverse mutation in Ames Salmonella typhimurium TA98 at 1.6-400 ng/plate with and without S9 mix. SN-07 also induced chromosomal aberrations and a forward mutation (6-TGr) in Chinese hamster V79 cells after 1 h treatment at 12.5-100 ng/ml without metabolic activation. The alkaline elution technique revealed that SN-07 induced interstrand DNA cross-linking dose-dependently after treatment with 2.5-10 micrograms/ml for 1 h followed by elution at pH 12.1, but it did not induce the dose-dependent cross-linking after the same treatment followed by elution at pH 12.6. It was also found that SN-07 induced single-strand DNA breaks (pH 12.1) and alkali-labile (pH 12.6) sites after treatment with 0.1-10 micrograms/ml for 1 h followed by 24-h post-incubation.
Oral administration of Xiao-Chai-Hu-Tang, the extract of a mixture of seven herbs, attenuated the hepatic fibrosis developed in mice after repeated doses of carbon tetrachloride. Its pre-administration reduced the derangement of liver function tests seen after a single dose of carbon tetrachloride as well as that seen after d-galactosamine intoxication. Xiao-Chai-Hu-Tang may be effective in the treatment of chronic liver injury through hepatocytoprotection.
The sera from two patients with chronic neutropenia in infancy were examined for the presence of antineutrophil antibodies and their specificity against neutrophil antigen by using granulocyte indirect immunofluorescence test and microleukocyte agglutination test. In the microleukocyte agglutination test, the patients' sera reacted with neutrophils from their parents and normal unrelated donors having the neutrophil antigen NA1, but not with neutrophils from NA1- donors. After the absorption of patients' sera with NA1+ neutrophils, the antibody activity was completely abolished, resulting in the confirmation of the anti-NA1 antibody. In contrast, the granulocyte indirect immunofluorescence test showed positive reactions against both NA1+ and NA1- neutrophils, and the specificity for anti-NA1 was found in the results of the sera absorbed with NA1+ neutrophils. This suggested that the absorption experiment might be necessary to determine the specificity of the antibody for neutrophil antigen. Thus, we confirmed two cases with autoimmune neutropenia caused by anti-NA1 antibody. A combination of agglutination and immunofluorescence techniques would be recommended for investigation of neutrophil antibodies against the neutrophil-specific antigen.
Serum and urinary levels of ubiquinol-10 (UQH2-10) as well as total ubiquinone-10 (total UQ-10, sum of UQH2-10 and oxidized UQ-10) were determined in healthy Japanese subjects using high performance liquid chromatography with an electrochemical detector. The mean serum and urinary levels of UQH2-10 were 0.75 +/- 0.21 microgram/ml (86% of total UQ-10, n = 77) and 0.045 +/- 0.016 microgram/mg creatinine (59% of total UQ-10, n = 30), respectively. After daily oral administration of 60 mg of UQ-10 for 7 days, the human serum level of UQH2-10 increased twice as compared to that before the treatment. However, the ratio of the UQH2-10 to the total UQ-10 remained unchanged during such administration. The serum level of UQH2-10 correlated significantly with those of vitamin E (p less than 0.001), phospholipids (p less than 0.01), total cholesterol (p less than 0.05), and total lipids (p less than 0.05). There was no correlationship between the serum levels of UQH2-10 and of triglycerides and lipid peroxides.

A convenient and reliable liquid chromatographic (LC) method with electrochemical detection (ED) was developed for the determination of reduced (ubiquinol) and total ubiquinones in biological materials. After extraction of samples with n-hexane, ubiquinol was separated on a reversed-phase column and assayed directly by ED. In order to determine the total amount of a ubiquinone in biological samples, the unbiquinone was converted into the corresponding reduced form by treatment with sodium borohydride. No significant interfering peak (plastoquinol-9, ubichromenol-9, etc.) was observed in the elution areas of ubiquinol-7 to -11. This LC-ED method was about 70 times more sensitive than the previous LC-UV method and was able to detect 150 pg of ubiquinol-10. The method was applied satisfactorily to the determination of the contents of ubiquinol homologues in biological materials. The content of ubiquinols is a major component of the total ubiquinones in human plasma and urine and rat plasma and liver, but a minor component in rat heart and kidney.
We developed an enzyme immunoassay for determining human cuprozinc superoxide dismutase (h-SOD) using two kinds of monoclonal antibodies prepared by immunizing h-SOD to BALB/c mice. This method was sensitive and specific enough to determine exogenous h-SOD injected into rats. When intravenously injected into rats, much of the immunoreactive h-SOD accumulated in the kidney and was rapidly excreted in the urine. We observed both a modified and an unmodified form of exogenous h-SOD in rat urine.
Abnormal beating patterns were induced in spontaneously contracting cultured embryonic mouse myocardial cells either by elevating or by lowering extracellular calcium. At low calcium (0.4 mM), the number of beating cells and beating rate decreased while the number of arrhythmic cells increased. By contrast, at high calcium (20 mM), the number of beating cells decreased while beating rate and the number of arrhythmic cells increased. Addition of taurine (20 mM) to the medium attenuated the response to varying calcium; the taurine effect appeared to be specific since neither taurine analog tested (beta-alanine nor glycine) provided much protection against these abnormalities. The protective effect of taurine also appeared to differ from that of verapamil, which was effective only in decreasing beating rate in the high calcium condition. Uptake of 14C-taurine by the cells was higher at both low and high extracellular calcium when compared to the normal calcium (2 mM) concentration. The results raise the possibility that the protective effect of taurine on beating abnormalities caused by low or high calcium is related to taurine uptake.
From July 1985 to March 1987, the occurrence of motile Aeromonas sp. in stool, food and environmental specimens was investigated to assess their pathogenic significance and to determine sources and routes of infection. A total of 9366 stool specimens were examined; Aeromonas was isolated from 11.1% of diarrhoeal stools and 2.2% of normal stools (P less than 0.001). Aeromonas counts in food specimens, which included minced beef, pork and chicken, seafood and various vegetables and their products, were unexpectedly high suggesting that infection might be food-borne rather than water-borne. About 70% of the isolates from meat products were A. hydrophila and A. sobria, while A. caviae was the most common in sea-fish, vegetables and their products. Most A. hydrophila and A. sobria strains produced haemolysin, but haemagglutinin was found more frequently in A. sobria.

Aeromonas has recently been recognized as an important enteropathogen. In faecal specimens the organism is readily identified and differentiated from coliforms by the positive oxidase reaction. However, few media have proved useful for foods and environmental specimens. Bile salts brilliant green starch agar (BBGS) was studied for this purpose by testing artificially contaminated foods as well as natural potential sources. The results indicate the suitability of the medium for the isolation of Aeromonas from foods and from environmental sources.
