Radioimmunoassay of hCG as an early diagnosis of cerebral metastases in choriocarcinoma patients.

To determine the early cerebral involvement of choriocarcinoma, the following studies were employed. By comparing hCG titers in measurements of serum and cerebrospinal fluid (CSF) hCG-beta through radioimmunoassay (RIA), serum/CSF ratios of titers were calculated in patients with normal pregnancies and in those with trophoblastic neoplasia. In normal pregnancies, the mean ratio of serum/CSF hCG was more than 45.5 +/- 6.0 (mean +/- SEM):1. In neoplasia without metastases to the brain, the ratio was more than 41.0:1. However, 2 patients with cerebral choriocarcinoma showed low ratios of 23.0:1 and 20.9:1, respectively. This suggests that determination of the beta-hCG concentration ratio of serum to CSF using RIA might give a more reliable evaluation for the early detection of choriocarcinoma metastasized to the brain.

[Influence of oocytes upon proliferation of cultured granulosa cells (author's transl)].

Granulosa cells (G-C) collected from mature follicles of human and bovine ovaries were cultured with an oocyte from mature follicles of human, bovine and porcine ovaries to investigate the influence of the oocyte upon the proliferation of G-C as well as the differentiation of G-C. For this purpose, G-C were compared in mitotic index (MI) between those cultured with oocyte and without oocyte control at the fourth day of culture. 1) G-C cultured with an autogenous oocyte--A bovine oocyte cultured with bovine G-C increased in MI 2.8 times compared with the control. Human G-C cultured with a human oocyte also appeared to raise the MI 2.7 times compared with the control. 2) G-C culture with heterogeneous oocyte--Moreover, a porcine oocyte cultured with bovine G-C acted to increase the MI to 2.7 times that of the control. An addition of a porcine oocyte to human cultured G-C also almost doubled the MI as compared with that of the control. 3) Within 2 days after culture of G-C with an auto- or hetero-genetic oocyte from which the cumulus oophorous had been removed, the oocyte was surrounded by a dense mass of small type of G-C, which resembled follicular G-C morphologically and indicated a high value of MI. The cells on the periphery from the oocyte, however, were clearly enlarged differing from those close to the oocyte and began to resemble lutein cells as the culture days went on. 4) Instead of the oocyte, sterilized muscle, cartilage and bone tissues were cultured with the G-C as controls. However, no binding of G-C with these tissues were found as was verified between G-C and the oocyte. These results suggest that the oocyte may characteristically bind with G-C to accelerate the multiplication and inhibit the luteinization of G-C.

Early termination of heterogeneous nuclear RNA transcripts in mammalian cells: accentuation by 5,6-dichloro 1-beta-D-ribofuranosylbenzimidazole.

Labeling of RNA in isolated HeLa cell nuclei in vitro reveals an abundance of short RNA chains made by RNA polymerase II. These short chains were initiated prior to isolation of the nuclei. The short abundant chains are increased in amount in nuclei isolated from cells treated with 5,6-dichloro-1-beta-D-ribofuranosylbenzimidazole (DRB). Kinetic evidence indicates that the bulk of the putative heterogeneous nuclear RNA (hnRNA) precursor molecules that are terminated early in vivo are terminated approximately 100-300 nucleotides from sites of initiation. DRB increases the frequency of early termination, but there is a fraction of hnRNA precursor molecules whose elongation is not affected by DRB. Heparin is useful in studies of hnRNA transcription in isolated nuclei because it enhances chain elongation.

Purification and partial characterization of three forms of alpha-glucosidase from the fruit fly Drosophila melanogaster.

Three forms of alpha-glucosidase, I, II, and III, have been purified from the whole body extract of adult flies of Drosophila melanogaster in yields of 2.1, 5.3, and 6.7%, respectively. The purification procedures involved ammonium sulfate fractionation, Con A-Sepharose 4B affinity chromatography, DEAE-Sepharose CL-6B ion exchange chromatography, Sephacryl S-200 gel filtration, and preparative gel electrophoresis. Each purified enzyme showed a single band on polyacrylamide gel on both protein and enzyme activity staining. The molecular weights of alpha-glucosidases I, II, and III were estimated to be 200,000, 56,000, and 76,000, respectively, by gel filtration. SDS gels indicated that alpha-glucosidases II and III were each composed of a single polypeptide chain, whereas alpha-glucosidase I was composed of two identical subunits. Both alpha-glucosidases II and III hydrolyzed sucrose and p-nitrophenyl-alpha-D-glucoside (PNPG), but alpha-glucosidase I hydrolyzed PNPG to a much lesser extent than sucrose. For sucrose the pH optima of alpha-glucosidases I, II, and III were pH 6.0, 5.0, and 6.0 and the Km values were 13.1, 8.9, and 10 mM, respectively. For PNPG the pH optima of alpha-glucosidases II and III were pH 5.5 and 6.5 and the Km values were 0.77 and 0.21 mM, respectively.

Phagocytic activity in the ischemic retinal pigment epithelial cells. An electronmicroscopic histochemical study of acid phosphatase.

The role of lysosomal enzymes in ischemic retinal pigment epithelial cells (RPE) of albino rabbits was examined with the modified Gomori technique for acid phosphatase. Ischemia was produced by cutting the lateral posterior ciliary artery and short posterior ciliary arteries (PCA). Five days after PCA-cut RPE in the ischemic region were disorganized by increased enzymatic digestion. In RPE of the border between the ischemic and normal region a lot of fragmented outer segments were phagocytosed 24 h to 7 days after PCA-cut. At this time phagosomes appeared much more frequently than in the normal retina, showing several variations in their shape, localization, and histochemical reaction. However, after 10 and 14 days, RPE in the border had a tendency to reduce the phagocytic activity. A strong acid phosphatase activity was encountered in the phagosomes of macrophages, which was considered to be derived from RPE, and seemed to play the major role in scavenging destructed retinal elements.

A case of pituitary apoplexy with spontaneous recovery.

A patient, 38-year-old man, with hemorrhage into a prolactin-secreting pituitary adenoma, or pituitary apoplexy, is reported. On his admission, clinical examinations revealed typical stigmata indicating that he suffered from an acute attack of pituitary apoplexy probably induced by acute meningitis. He survived the acute attack and recovered spontaneously without an urgent operation. Although there was no suspicious sign and symptom of hypopituitarism, the first study performed immediately after the attack suggested strongly that hypopituitarism might acutely developed during the hemorrhage into the tumor. Moreover, the follow-up studies indicated that TSH, LH and ADH recovered spontaneously from the initial damage following the resorption of hemorrhage for the next 3 months.
