We describe a simple method for determining magnesium in serum by using hexokinase (EC 2.7.1.1) and glucose-6-phosphate dehydrogenase (EC 1.1.1.49). The method is based on determination of the reaction rate of hexokinase activated by Mg2+, which participates in the hexokinase reaction as the substrate in the form of a Mg X ATP2- complex. The reaction rate is determined from the change in absorbance at 340 nm as NADPH is produced by the action of glucose-6-phosphate dehydrogenase. This simple and rapid spectrophotometric method does not require expensive instrumentation, but results correlate satisfactorily with those obtained by atomic absorption spectroscopy. Thus, the present method gives a "true" value for magnesium in serum, a value appreciably lower than that obtained by an earlier colorimetric method, the Xylidyl Blue II method (Biochem Med 7: 208-217, 1973), which lacks specificity.

The reactivities of anionic nitroalkanes with 2-nitropropane dioxygenase of Hansenula mrakii, glucose oxidase of Aspergillus niger, and mammalian D-amino acid oxidase have been compared kinetically. 2-Nitropropane dioxygenase is 1200 and 4800 times more active with anionic 2-nitropropane than D-amino acid oxidase and glucose oxidase, respectively. The apparent Km values for anionic 2-nitropropane are as follows: 2-nitropropane dioxygenase, 1.61 mM; glucose oxidase, 16.7 mM; and D-amino acid oxidase, 11.1 mM. Anionic 2-nitropropane undergoes an oxygenase reaction with 2-nitropropane dioxygenase and glucose oxidase, and an oxidase reaction with D-amino acid oxidase. In contrast, anionic nitroethane is oxidized through an oxygenase reaction by 2-nitropropane dioxygenase, and through an oxidase reaction by glucose oxidase. All nitroalkane oxidations by these three flavoenzymes are inhibited by Cu and Zn-superoxide dismutase of bovine blood, Mn-superoxide dismutases of bacilli, Fe-superoxide dismutase of Serratia marcescens, and other O2-. scavengers such as cytochrome c and NADH, but are not affected by hydroxyl radical scavengers such as mannitol. None of the O2-. scavengers tested affected the inherent substrate oxidation by glucose oxidase and D-amino acid oxidase. Furthermore, the generation of O2-. in the oxidation of anionic 2-nitropropane by 2-nitropropane dioxygenase was revealed by ESR spectroscopy. The ESR spectrum of anionic 2-nitropropane plus 2-nitropropane dioxygenase shows signals at g1 = 2.007 and g11 = 2.051, which are characteristic of O2-.. The O2-. generated is a catalytically essential intermediate in the oxidation of anionic nitroalkanes by the enzymes.
Serum parathyroid hormone (PTH) and renal functions were measured in thirty subjects with cadmium-induced renal damage and in the same number of control subjects. The negative correlations between PTH and creatinine clearance (Ccr) and between PTH and percentage renal phosphorus reabsorption (% TRP) were significant in the cadmium (Cd)-exposed group. Serum PTH concentrations were clearly high in two cases of the cadmium-exposed group. Their renal functions decreased severely compared with those of the other Cd-exposed people. The present study indicates that chronic cadmium intoxication could cause increased secretions of PTH, which may be related to cadmium-induced bone damage.
Alpha1-Microglobulin (alpha 1-m) was determined in the urine of both the cadmium-exposed and nonexposed subjects and was compared with those of other urinary parameters such as beta 2-microglobulin (beta 2-m), glucose, total protein or amino acids. Large amounts of alpha 1-m were detected in the urine of cadmium-exposed subjects. There were significant correlations between alpha 1-m and other urinary parameters. The sensitivity and specificity of both alpha 1-m and beta 2-m as indices of tubular dysfunction caused by cadmium was similar. It was concluded that alpha 1-m in urine seemed to be a useful indicator of renal tubular dysfunction caused by cadmium.
Iron (Fe) concentrations in liver and kidney of cadmium (Cd)-exposed people (11 Itai-Itai patients, 13 persons requiring observation and 4 Cd-exposed persons) were compared with those of nonexposed people. No significant difference in Fe levels in liver was observed between the groups. This fact indicates that anaemia in environmentally Cd-exposed people was not due to iron deficiency in contrast to findings in animal experiments.
The authors describe a case of intestinal obstruction due to a mesodiverticular band, in which both vitelline artery and vein remnants were affirmed to be present histologically. Reviewing case reports in literature, it was revealed that the vein found in the band was the right vitelline vein remnant and that this was an important fact which proved the hypothesis that the superior mesenteric vein derives from the right vitelline vein.

A method for the automated analysis of creatinine in serum using immobilized enzymes in column form which does not require a blank correction and can be run in a single buffer of pH 7.5 is described. The method was based on the determination of ammonia formed by the action of creatinine deiminase. Endogenous ammonia in serum was removed by an immobilized glutamate dehydrogenase column prior to the action of creatinine deiminase also immobilized and used in column form. The present method was found to give perfect linearity of the data up to 0.10 g creatinine/liter with satisfactory precision, reproducibility, and accurate reaction recoveries. The immobilized enzyme reactor unit showed good operational stability for a 2-month period, during which time it was repeatedly used for analyses over 2000 times. The results correlated satisfactorily with those obtained by other well-established methods.
