Protective effects of glutathione against lipid peroxidation in chronically iron-loaded mice.

To elucidate the protective effects of glutathione against iron-induced peroxidative injury, changes in the hepatic glutathione metabolism were studied in chronically iron-loaded mice. When the diets of the mice were supplemented with carbonyl iron, iron deposition occurred primarily in the parenchymal cells of the liver. In addition, expiratory ethane production was elevated, suggesting an enhancement in lipid peroxidation. In iron-loaded mice, the total hepatic glutathione contents were higher (6.21 +/- 0.53 mumol/g wet wt.) than in control mice (4.61 +/- 0.31 mumol/g wet wt.), primarily due to an increase in the reduced glutathione contents. The value of oxidized glutathione was also higher (98.5 +/- 8.1 nmol/g wet wt.) than in the controls (60.8 +/- 9.5 nmol/g wet wt.), and the ratio of oxidized glutathione to total glutathione increased. The excretion rate of glutathione from the hepatocytes in iron-loaded mice also increased. These observations suggest that chronic iron-loading of mice stimulates lipid peroxidation and oxidation of glutathione and that peroxidized molecules may be catabolized using reduced glutathione.

Evidence that aspartic proteinase is involved in the proteolytic processing event of procathepsin L in lysosomes.

Our recent studies have shown that cathepsin L is first synthesized as an enzymatically inactive proform in endoplasmic reticulum and is successively converted into an active form during intracellular transport and we postulated that aspartic proteinases would be responsible for the intracellular propeptide-processing step of procathepsin L accompanied by the activation of enzyme (Y. Nishimura, T. Kawabata, and K. Kato (1988) Arch. Biochem. Biophys. 261, 64-71). To better understand this proposed mechanism, we investigated the effect of pepstatin, a potent inhibitor of aspartic proteinases, on the intracellular processing kinetics of cathepsin L analyzed by pulse-chase experiments in vivo with [35S]methionine in the primary cultures of rat hepatocytes. In the pepstatin-treated cells, the proteolytic conversion of cellular procathepsin L of 39 kDa to the mature enzyme was significantly inhibited and considerable amounts of proenzyme were found in the cell after 5-h chase periods. Further, the subcellular fractionation experiments demonstrated that the intracellular processing of procathepsin L in the high density lysosomal fraction was significantly inhibited and that considerable amounts of the procathepsin L form were still observed in the light density microsomal fraction after 2 h of chase. These results suggest that pepstatin treatment caused a significant inhibitory effect on the intracellular processing and also on the intracellular movement of procathepsin L from the endoplasmic reticulum to the lysosomes. These findings provide the first evidence showing that aspartic proteinase may play an important role in the intracellular proteolytic processing and activation of lysosomal cathepsin L in vivo. Therefore, we suggest that cathepsin D, a major lysosomal aspartic proteinase, is more likely to be involved in this proposed model in the lysosomes.

Prerequisites in distance running performance of female runners.

We investigated which attribute or what combination of attributes would best account for distance running performance of female runners. The subjects were 30 well-trained female distance runners, aged 19 to 23 years. Anthropometric and body composition characteristics, pulmonary function characteristics, blood properties, and cardiorespiratory function characteristics were measured at rest or during submaximal and maximal exercise. Analyses of the data showed that the relationship of oxygen uptake corresponding to lactate threshold (VO2T, ml.kg-1.min-1) with each distance running performance was substantially higher as compared with the relationship of other independent variables including maximal oxygen uptake (VO2max). Furthermore, multiple regression analysis indicated that running performances in 3,000m, 5,000m, and 10,000m are best accounted for by a combination of VO2/LT (X1), fat-free weight (X2), and/or mean corpuscular volume (X3). A multiple regression equation for predicting the 5,000m (Y, s) running performance was formulated as Y = -14.75X1-3. 03X2-5.79X3 + 2282.1. We suggest that VO2max would not stand alone as a decisive factor of distance running success in female runners, and that the distance running performance could be better accounted for by a combination of several attributes relating to lactate threshold, body composition, and/or hematological status. The linear regression of the predicted running performance on the actually measured running performance can be accepted in the range of 986-1197s.

Iron release from haemosiderin and production of iron-catalysed hydroxyl radicals in vitro.

Isolated haemosiderin contained iron and nitrogen in a weight ratio of 6.75, with phosphorus and no detectable haem. Considerably more iron was released from haemosiderin under acidic conditions than under neutral conditions in the presence of ascorbate, nitrilotriacetate or dithionite. Unlike the situation with ascorbate, chelators such as citrate, ADP or succinate induced the release of only some iron, with almost no pH-dependence. Dehydroascorbate (the oxidized form of ascorbate with no reducing capacity) behaved like citrate, ADP, succinate or desferal, rather than like ascorbate itself, in releasing iron. GSH had less effect on the release of iron than these chelators, but in the presence of a small amount of chelator the release of iron increased, especially under acidic conditions. Thus reduction, chelation and pH were all found to be important factors involved in the release of iron from haemosiderin. Investigation by e.p.r. of hydroxyl-radical production by the released iron showed high radical productivity at an acidic pH. However, at a physiological pH, almost no radical formation was detected, except in the presence of nitrilotriacetate. These findings suggested that, under physiological conditions, haemosiderin was not an effective iron donor and was almost not involved in radical production. Under acidic conditions, however, such as in inflammation, hypoxia and in a lysosomal milieu, it could possibly be an iron donor and is thought to be implicated in radical production and tissue damage in iron-overloaded conditions.

Identification of latent procathepsins B and L in microsomal lumen: characterization of enzymatic activation and proteolytic processing in vitro.

Procathepsins B and L in the hepatic endoplasmic lumen were identified as having a molecular weight of 39,000 by immunoblot analysis. The proenzymes were then purified to remove the mature enzymes by concanavalin A-Sepharose chromatography. The concanavalin A-adsorbed fractions containing the proenzymes showed no appreciable activities of cathepsins B and L. When those fractions were incubated at pH 3.0, the enzymatic activities markedly increased: the activities of cathepsins B and L after 36 h incubation were 60 and 210 times those of the controls, respectively. Immunoblot analysis showed that after 36 h incubation the proenzymes disappeared and the mature enzymes increased. Thus the proenzymes were processed to the mature enzymes under acidic conditions of pH 3.0. The marked increases of enzymatic activities and the conversion of the proenzymes to the mature forms were completely blocked with pepstatin, which is a potent inhibitor of aspartic proteases. The results strongly suggested that a processing protease for procathepsins B and L might be cathepsin D, a major lysosomal aspartic protease. Indeed, lysosomal cathepsin D could convert microsomal procathepsin B to the mature enzyme in vitro. Therefore, procathepsins B and L seem first to be synthesized as enzymatically inactive forms in endoplasmic reticulum and successively may be converted into active forms by cathepsin D in lysosomal compartments.

Endocrine profiles and embryo quality in superovulated Japanese Black cattle.

The relationships between plasma concentrations of progesterone (P4), estradiol-17beta (E2) and lutenizing hormone (LH) and embryo yield and quality were examined in 40 superovulated Japanese Black cattle. The results indicated that the ovarian function, especially the function of corpus luteum on the first treatment day, is an important factor for reliable superovulation in cattle. The levels of plasma E2 and LH at estrus were related to embryo yield and quality.

Plasma progesterone profiles and embryo quality in superovulated Japanese Black cattle.

The relationship between plasma progesterone (P(4)) levels before and after superovulation treatment and embryo yield and quality was examined in Japanese Black cattle. It is concluded that a plasma P(4) level at the start of follicle stimulating hormone (FSH) injections is related to embryo quality. The P(4) level of over 3.0 ng/ml appears to be useful in preselection of donors when we want to get more than 8 normal (transferable) embryos on the average.

Effect of ascorbic acid on the metabolism of N-nitrosopiperidine.

Guinea-pigs were fed diets containing 48, 200 or 2000 mg/kg ascorbic acid (AsA) for 20-30 days and given a single oral dose of 50 mg N-nitrosopiperidine (NPIP)/kg body weight. The levels of glutaric acid, 3-hydroxy-NPIP and 4-hydroxy-NPIP, but not of 1,5-pentanediol, in urine increased with the dose of AsA in the diet.

Lipid peroxidation and cytotoxicity of Ehrlich ascites tumor cells by ferric nitrilotriacetate.

Ferric nitrilotriacetate, which causes in vivo organ injury, induced lipid peroxidation and cell death in Ehrlich ascites tumor cells in vitro. The process was inhibited by butylated hydroxyanisole and enhanced by vitamin C and linolenic acid, indicating a close relationship between cytotoxicity and the lipid peroxidizing ability of Fe3+ NTA. The cytotoxicity was suppressed by glucose and a temperature below 20 degrees C. Lipid peroxidation of Fe3+ NTA-treated cells was greater at 0 degree C than at 37 degrees C, contrary to results with Fe3+ NTA-treated plasma membranes of Ehrlich ascites tumor cell. These results suggested that metabolism and membrane fluidity are important factors in the expression of the Fe3+ NTA-induced cytotoxicity. H2O2 showed a lower cytotoxicity than did Fe3+ NTA but a greater lipid peroxidizing ability. H2O2 appeared to damage the cells less, and was quenched rapidly by cellular metabolism unlike Fe3+ NTA. In transferrin-free medium, Ehrlich ascites tumor cell readily incorporated Fe3+ NTA, and iron uptake was greater than NTA-uptake in Fe3+ NTA-treated cells, suggesting that Ehrlich ascites tumor cell incorporated iron from Fe3+NTA and metabolized it into an inert form such as ferritin.

Synthesis and biological properties of two dimeric forms of human alpha-atrial natriuretic peptide.

Two dimeric forms of human alpha-atrial natriuretic peptide (alpha-ANP) were synthesized by solution methods and compared with monomeric alpha-ANP in terms of some biological and immunochemical properties. The parallel form (beta'-ANP) and the antiparallel form (beta-ANP) were equipotent in smooth muscle relaxant activity in isolated rat aorta and their ED50 values were estimated to be 1.7 X 10(-8) M and 1.6 X 10(-8) M, respectively. Diuretic and natriuretic responses induced by beta-ANP and beta'-ANP anesthetized rats were equally less potent but exhibited a significantly longer duration than those induced by alpha-ANP. beta-ANP and beta'-ANP possessed immunoreactivities of 60-100% and 50-90% (alpha-ANP, 100% on a molar basis), respectively, with three different antisera raised against alpha-ANP-related peptides.

Generation of active oxygen species by iron nitrilotriacetate (Fe-NTA).

Ferric nitrilotriacetate (Fe3+-NTA) solution showed maximum absorbance at pH 7.5. The iron was in ferric high-spin state and coordinated octahedrally with a relatively symmetric structure and also probably pentagonally. A spin trapping technique employing 5,5-dimethyl-1-pyrroline-N-oxide (DMPO) yielded a DMPO spin adduct of unknown radical with three doublets (DMPO-Z) and a simple nitroxide radical (Y-NO.) in serum from rats injected intraperitoneally with Fe3+-NTA. When the Fe3+-NTA solution was diluted 500-fold with 50 mM NTA solution, DMPO-Z, Y-NO. and an additional signal, DMPO-OH were observed. The DMPO-Z signal was suppressed by a decrease in oxygen tension, alpha-tocopherol and 3-tert-butyl-4-hydroxy-anisole (BHA). The DMPO-OH signal was suppressed in the presence of ethanol and catalase. Fe2+-NTA solution hardly produced DMPO spin adducts. The Fe3+-NTA solution produced a strong DMPO-OH signal in the presence of H2O2. Rose Bengal solution, a singlet oxygen generating system, produced the same DMPO adducts. Fe3+-NTA reacted with oxygen in solution. The oxygen was activated and might be similar to singlet molecular oxygen. In the presence of H2O2, the Fe3+-NTA solution generated a hydroxyl radical. Fe3+-NTA itself generated free radicals, but Fe2+-NTA did not.

[Bacterial and clinical studies on imipenem/cilastatin sodium in urological infection].

Antibacterial activities of imipenem (MK-0787), cefazolin (CEZ), cefmenoxime (CMX) and cefoperazone (CPZ) were measured by using an agar dilution method against 243 strains of bacteria isolated from the patients with urinary tract infections. The MIC80 of MK-0787 against S. epidermidis, E. faecalis, E. coli, K. pneumoniae, E. cloacae, S. marcescens, P. mirabilis, P. vulgaris and P. aeruginosa was less than or equal to 0.10, 3.13, less than or equal to 0.10, 0.20, 0.78, 1.56, 3.13, 3.13 and 6.25 micrograms/ml, respectively. MK-0787 was more active than CEZ, CMX and CPZ against all bacterial species except Proteus spp. Imipenem/cilastatin sodium (MK-0787/MK-0791) 500 mg/500 mg two times a day for 5 days was given to 31 cases of chronic complicated urinary tract infections and a case of acute prostatitis. Overall clinical effectiveness rate judged by the criteria proposed by the Japanese UTI committee in 22 cases of chronic complicated urinary tract infection treated with MK-0787/MK-0791 was evaluated to be 86% including excellent in 6 cases, moderate in 13 cases and poor in 3 cases. As side effects, 2 cases encountered eruption or anorexia. Abnormal laboratory findings were observed in 6 cases.

Aldosterone antagonists. 1. Synthesis and biological activities of 11 beta,18-epoxypregnane derivatives.

Several steroid derivatives having the 11 beta,18-epoxypregnane skeleton, 7, 8, 19, 20, 21, and 31, were synthesized to evaluate their antialdosterone activity. Among them, 3-(9 alpha-fluoro-17 beta-hydroxy-3-oxoandrost-4-en-17 alpha-yl)propionic acid gamma-lactone (31) possessed fairly strong binding affinity for the cytoplasmic mineralocorticoid receptor of rat kidney and exhibited good aldosterone antagonist activity in an in vivo assay. However, its agonistic nature cannot be ignored. The properties of 31 as an aldosterone antagonist were enhanced by its very low to negligible binding affinity for the androgen, progestin, estrogen, and glucocorticoid receptors.

Effect of ascorbic acid and some reducing agents on N-nitrosopiperidine metabolism by liver microsomes.

The effect of ascorbic acid on the oxidation of N-nitrosopiperidine by a microsomal preparation obtained from guinea-pig liver has been examined. The metabolites from the oxidation of N-nitrosopiperidine were found to be 5-hydroxypentanal (about 1.93%), N-nitroso-3-hydroxypiperidine (about 0.02%), and N-nitroso-4-hydroxypiperidine (about 0.15%). With microsomes separated from the liver of guinea-pigs pretreated with phenobarbital and 3-methylcholranthrene, the yields of N-nitroso-3-hydroxypiperidine and N-nitroso-4-hydroxypiperidine were found to decrease upon increasing the concentration of ascorbic acid in the reaction mixture. In the presence of small amounts of ascorbic acid, the yields of these compounds with untreated liver microsomes apparently increased, while the presence of larger amounts of ascorbic acid caused a decrease in the yields of these metabolites. In contrast with the yields of N-nitroso-3-hydroxypiperidine and N-nitroso-4-hydroxypiperidine, that of 5-hydroxypentanal decreased with increasing amounts of ascorbic acid.
