Human kidney glomeruli, with special reference to those in the aged person: scanning electron microscopic study of microvascular corrosion casts.

Blood vascular beds of fetal, adult and aged human kidneys were reproduced with methyl methacrylate and observed with a scanning electron microscope. The kidney glomeruli, including those from the fetal kidneys, had anastomosing capillaries. The glomeruli in the kidneys of an aged person contained many more capillaries which were much more tortuous than those of the adult and fetal kidneys. Furthermore, it was observed that the glomeruli in the kidneys of the aged person usually received tortuous afferent vessels and frequently emitted multiple efferent arterioles. The glomeruli in the juxtamedullary layer of the kidneys of the aged person were rather small in size and contained degenerative capillary networks. This observation suggests that the medulla of the kidneys of the aged is poorly supplied with blood.

A modified method of fine-granular cationic iron colloid preparation: its use in light and electron microscopic detection of anionic sites in the rat kidney glomerulus and certain other tissues.

Ferric chloride, when boiled with hydrazine hydrate and cacodylic acid, is converted into a fine cationic iron (ferric hydroxide) colloid which consists of 0.5-1.5 nm electron-dense granules, and gives a distinct Prussian blue reaction. This colloid allows light and electron microscopic detection of ionized anionic sites in tissues at a wide pH range of 0.8-7.6. It is smaller in size and more stable, and assures longer and greater staining of tissues, especially at low pH levels, than the iron colloid prepared with sodium or ammonium cacodylate by Seno and his associates (1983, 1984, 1985). Some light and electron micrographs of the rat kidney, spleen and other organs stained with our colloid are presented as examples. These micrographs confirm that the glomerular podocyte end-foot surface facing the Bowman's capsular space is strongly negative-charged. They also show that almost all lymphoid cells around the arteries in the splenic white pulp and thymic cortex contain strongly negative-charged nuclei and that the distal convoluted and collecting urinary tubules are more negative-charged than the proximal convoluted tubules.

Mechanism of hematuria. I. Electron microscopic demonstration of the passage of a red blood cell through a glomerular capillary wall in rat masugi nephritis.

Masugi nephritis was induced in Sprague-Dawley rats by an intravenous injection of rabbit anti-rat kidney serum. In the autologous phase of the disease, three of 18 rats manifested continuous hematuria. Ultrastructural examination of renal glomeruli by transmission and scanning microscopy revealed gaps in the basement membranes, and the transcapillary passage of red blood cells through the discontinuous regions in the hematuric rats. Control animals revealed no gaps in the glomerular basement membranes regardless of the method used in their preparation for electron microscopy. These data support the hypothesis that hematuria is a result of the passage of red blood cells through gaps in the glomerular basement membrane in Masugi nephritis.

T4-labeling technique and its applications with particular reference to blood group antigens in bladder tumors.

A variety of visual markers have been developed to detect specific cell surface components with scanning electron microscopy. Among them bacteriophage T4 (T4) has the most distinctive shape consisting of a hexagonal head and tail. This characteristic shape of T4 makes it possible to survey the cell surface at relatively low magnification and to analyze the site specificity at high magnification. Although precise localization of surface molecules cannot be mapped due to the relatively large size of T4, T4-labeling is most suitable for the analysis of wide surface areas and simultaneous observation of many cells as well as for the analysis of the tissue materials. In this paper, T4-labeling method and technical problems in actual practice including specimen preparation as well as the results obtained are reviewed with particular reference to blood group antigens in bladder tumors.

[Clinical effect of UFT on bladder cancer].

UFT (a mixture of futraful and uracil) was administered orally to 15 cases of recurrent and superficial bladder cancer principally for more than 4 weeks at the doses of 300 or 600 mg per day. The result was evaluated by Koyama-Saito's response criteria. Of 14 evaluable cases, complete remission was noted in 5. Two cases of advanced bladder cancer were treated with UFT combined with irradiation therapy, and one case was found that cancer cells degenerated severely in the primary lesion pathologically. Another case was recognized that lung metastasis disappeared completely. As the side effects, anorexia occurred in 2 cases with 600 mg per day and 1 case with 300 mg, but it was not so severe to terminate UFT therapy. From above-mentioned results, UFT seems to be a useful drug for the treatment of bladder cancers.

Leukocytes in the glomerular capillary lumen in Masugi nephritis. A scanning electron microscopic study of freeze-cracked specimens.

Masugi nephritis was induced in rats by a single intravenous injection of rabbit anti-rat kidney serum, and studied with a scanning electron microscope. A characteristic finding was the presence of white cells, probably polymorphonuclear leukocytes, with many microspikes which penetrated through degenerated glomerular endothelial cells to be in direct contact with the glomerular basement membrane. This finding confirms the pathogenic role of leukocytes in glomerulonephritis induced by anti-glomerular basement membrane antibody.

Primary testicular plasmacytoma: a case report.

A 73-year-old man with a left testicular tumor is presented. Orchiectomy was performed and the tumor was diagnosed as a plasmacytoma. There has been no bone lesion or immunoglobulin abnormality during a follow-up period more than one year. For several reasons, the tumor was regarded as a primary plasmacytoma of the testicle. The literature concerning testicular plasmacytoma is reviewed briefly.

The effects of intravesical instillation of Thio-Tepa on the recurrence rate of bladder tumors.

Seventy-eight patients were treated with intravesical instillation of Thio-Tepa in an attempt to prevent postoperative recurrences of bladder tumors. Fifty-six patients who were given no preventive treatment against recurrences were taken as the control group. The patients in this series presented at the Okayama University Hospital between 1961 and 1976 and only the first recurrence after the primary operation was taken into consideration. There was no significant difference in the recurrence rates of the control and instillation groups.

[Studies on the pudendal nerve. Part 4. A macroscopical observation of the branches of the pudendal plexus in dogs and cats (author's transl)].

The pudendal plexus and its branches of 20 dogs and 20 cats have been observed macroscopicaly. The main findings are summarized as follows: 1) The pudendal plexus in dogs is formed most frequently by the ventral rami of the first, second and third sacral nerves. 2) The bigeminal nerve of dogs and cats is formed respectively by the union of the ventral rami mentioned above. 3) The perineal nerve in dogs arises with one root from the bigeminal nerve and that of cats two roots, of which the main root arises from the points where the bigeminal nerve joins the asciatic nerve and the accessory one from the bigeminal nerve. 4) In dogs and cats the dorsal nerve of the penis or clitoris arises from the main trunk of the perineal nerve (Fig. 5, type IB) at an average rate of 57.5%. 5) In the dogs and cats, the inferior rectal nerve consists of two divisions; a division had the branches which are derived the perineal nerve to supply the skin around the anus and the other branches which arises from the muscular branch of the dorsal nerve of the penis or clitoris to supply the Sphincter ani externus. 6) The muscular branch of dogs and cats which is analogous to that of the perineal nerve in man, arises from the dorsal nerve of the penis or clitoris. 7) In dogs and cats the muscular branch to the Obturator internus is derived from the dorsal nerve of penis or clitoris. 8) The pelvic splanchnic nerve of dogs originates from the ventral rami of the first, second and third sacral nerves and that of cats from the ventral rami of the first and second sacral nerves. 9) In the posterior femoral cutaneous nerve in dogs, as well as in rats and rabbits, the lateral deviation of the origin from the sciatic to the perineal nerve is observed.
