Relationship of primer specificity of fatty acid de novo synthetase to fatty acid composition in 10 species of bacteria and yeasts.

Fatty acid compositions of lipids from six bacteria and four yeasts were determined. Fatty acid de novo synthetases were investigated with respect to chain length specificity towards acyl-CoA primers of various chain lengths. Four species of bacteria (Bacillus subtilis, Corynebacterium cyclohexanicum, Micrococcus luteus, and Pseudomonas maltophilia) possess branched-chain fatty acids of the iso and anteiso series as the major acids. De novo synthetases from these organisms exhibited specificity towards the chain length of the primer in the order butyl-CoA > priopionyl-CoA > acetyl-CoA. The remainder, two bacteria and all four yeasts, have the straight-chain type of fatty acids only and fall into two groups: (1) Eschericia coli B, Pseudomonas fluorescens, and Saccharomyces cerevisiae, which utilize the primers in the order acetyl-CoA > priopionyl-CoA > butyryl-CoA; and (2) Candida sake, Candida tropicalis, and Rhodotorula glutinis, which show the order propionyl-CoA > acetyl-CoA > butyryl-CoA. L-alpha-Keto-beta-methylvalerate, a precursor of the branched-chain primers, can be used as a source of primer for fatty acid synthesis by the organisms with branched-chain acids but not by those with the straight-chain type. The results indicate that organisms having straight-chain fatty acids lack the branched-chain equivalents for two reasons: first, their enzymes are not active toward primers with more than three carbons, and second, they lack a system of supply suitable branched-chain primers. It appears that activities of de novo synthetases from the organisms having straight-chain fatty acids generally have much higher activities than those from the organisms possessing branched-chain fatty acids

Effect of autoxidized methyl linoleate on glutathione peroxidase.

To determine the effect of several oxidation stages of autoxidized methyl linoleate (AOML) on glutathione peroxidase (GSH-Px), several experiments were carried out. In the first experiment, GSH-Px was prepared from gastrointestinal tract and liver which were excised from mice, and the effects of AOML rich in hydroperoxides (HP) and that rich in the decomposition products of HP on the activity of GSH-Px were compared. The results indicate that GSH-Px indiscriminately metabolized both types of AOML without strict recognition of the specificity of the substrate. In the second experiment, it was noted that the inhibitory rate of GSH-Px by in vitro incubation with various AOML closely correlated with POV of the sample esters, but did not correspond with the toxicity revealed by intraperitoneal injection or intragastric feeding. In the next experiment, mice were given AOML with a low vitamin E diet for 45 days, and an increase of GSH-Px activity in gastrointestinal tract in proportion to the POV of the administered oil occurred, but the level in liver remained unchanged. On the contrary, the increase of GSH-Px activity by intraperitoneal injection of AOML in mice was marked in liver, but not in gastrointestinal tract. From these results, most of the orally administered AOML seemed to be reduced in the mucosa of the gastrointestinal tract. However, the marked increase of fluorescence in the lipid fraction of heart and kidney by oral administration of AOML suggested that the damage caused by it was not limited to gastrointestinal tract but was spread through the whole body.

Quantitative analysis of cholesterol in foods by gas-liquid chromatography.

In order to develop a simple and exact analytical method for cholesterol determination in foods by gas-liquid chromatography, several experiments were carried out in collaboration with several universities and institutes. For the extraction of lipids from foods, it was decided that chloroform-methanol (2:1) extraction was the most suitable procedure. Since pretreatment procedures using methods such as thin-layer chromatography and florisil column chromatography to purify the unsaponifiable matters reduced the recovery of cholesterol, and good results were obtained without applying the pretreatment, these procedures were concluded to be unnecessary. Gas chromatograms obtained with free sterol showed results similar to those of the trimethylsilyl ether derivative and acetate. 5-alpha-Cholestane is used as a good internal standard.

Desmoplastic fibroma of the mandible: report of case.

Desmoplastic fibroma of the bone is a rare tumor; only 48 cases of the disease have ever been reported. The tumor has most often been observed in the metaphysis of long bones. Nine cases of the lesion have been reported in the mandible. The lesion occurred in the left mandibular body of one of our patients. We treated the entity by surgically removing the tumor and concurrently reconstructing the jaw with a bone graft taken from the iliac crest. There has been no recurrence a year and eight months postoperatively.

[Comparison of the effects of dobutamine with dopamine and isoproterenol on inotropism and chronotropism in the mammalian heart (author's transl)].

The effects of dobutamine on inotropism and chronotropism of the heart were studied in vivo and in vitro and were compared with those of dopamine and isoproterenol. These compounds increased epicardial contractile force and the heart rate of the open-chest, bilaterally vagotomized dog under pentobarbital anesthesia. The dose-ratio for the contraction was [dobutamine: dopamine: isoproterenol = 1:0.8:40] and for the heart rate, [= 1:1:300]. Both drugs augmented the twitch contraction of the isolated dog ventricular papillary muscle with the dose-ratio of [dobutamine: dopamine: isoproterenol = 1:0.7:11]. This mechanical response was associated with an elevation of the plateu voltage and an increase in repolarization of the action potential, but with no alteration of the maximum rate of rise of the action potential, the resting potential and the input membrane resistance. The discharge frequency of the rabbit S-A node pacemaker potential was accelerated chiefly due to an increase in the slope of the diastolic slow depolarization. With concentration of these catecholamines for the equivalent positive inotropic potency on the papillary muscle, this effect of isoproterenol was more potent than the effects of dobutamine and dopamine. These positive inotropic and chronotropic actions of the catecholamines were abolished by a beta-receptor antagonist. Those actions of dopamine were markedly reduced by reserpine pretreatment. In addition, dobutamine had little vascular effect. These results indicatte that dobutamine has a positive inotropic effect and a less positive chronotropic effect and that such is due to the direct action on the ventricular myocardium and the S-A node through beta 1-adrenergic receptors.

Indication and appreciation of operative procedures for mandibular ankylosis.

Mandibular ankylosis may be of various etiology. A number of operative procdures have been presented and criticized, but the most appropriate procedure for ankylosis is not universally accepted. During the past decade, 17 cases of mandibular ankylosis have been treated surgically in the Nagoya University Hospital. The etiology includes both temporomandibular ankylosis and extraarticular origin. Three different surgical procedures were used: (1) High operation, in which skin incision is made in the preauricular region. This method is applied for moderate arthrogenic cases. (2) Low operation, in which skin incision is made in the submandibular region. This is applied for severe arthrogic cases and extraarticular ossification cases. (3) Tubed pedicle skin grafting is applied for buccal contracture cases caused by noma etc. A long-term follow-up to 10 years has disclosed many interesting findings and made it possible to evaluate each operative procedure.

Problems involved in grading the severity of hemophilia by test results.

Some problems involved in grading the severity of hemophilia were discussed. No definite correlation existed between the clinical severity of hemophilia and the results of coagulation tests (activity of deficient factors, whole blood clotting time, thromboplastin screening test). There was also no correlation between the presence or absence of oral bleeding or hemarthrosis and the test results. It was concluded that the clinical severity of the disease can be graded more reasonably on the basis of clinical hemorrhagic symptoms than according to the activity of deficient factors.
